  • Corporation, local government and government office provide useful information. Advertisement service is getting various manner. Typical example is electronic display, TV-advertisement and internet or mobile advertisement service. In this paper, I'm study about Taxi-advertisement service still. A similar case is bus or subway advertisement from display. But, Taxi have been only lapping advertisement. Because, Taxi get space restraints. Broadly speaking, Person using Gyeonggi-do to Soeul or Incheon to Soeul is 423million people. On a national basis, it's very amazing figure. However It's just so many person showing one advertisement. So people have no interest in advertisement. So I develop new advertisement system through taxi in the whole country. 254,823 car have passenger personal information and showing people needing advertisement. Also this system get geographical information. This system have high advertisement effect.

Molecular Cloning, Sequence Analysis, and in Vitro Expression of Flavanone 3β-Hydroxylase from Gypsophila paniculata (안개초(Gyposphila paniculata)로부터 Flavanone 3β-Hydroxylase 유전자의 분리 및 분석)

  • Flavanone 3$\beta$-hydroxylase (FHT) is an enzyme acting in the central part of the flavonoid biosynthesis pathway. FHT catalyses the hydroxylation of flavanone to dihydroflavonols in the anthocyanin pathway. In this paper we describe the cloning and expression of the genes encoding the flavonoid-biosynthetic enzyme FHT in Gypsophila paniculata L. A heterologous cDHA probe from Dianthus cavophyllus was used to isolate FHT-encoding cDHA clones from Gypsophila paniculata L.. Inspection of the 1471 bp long sequence revealed an open reading frame 1047 bp, including a 190 bp 5' leader region and 288 bp 3' untranslated region. Comparison of the coding region of this FHT cDHA sequence including the sequences of Arabidopsis thaliana, Citrus sinensis, Dianthus caryophyllus, Ipomoea batatas, Matthiola incana, Nierembergia sp, Petunia hybrida, Solanum tuberosum, Vitis vinifera reveals a identity higher than 69% at the nucleotide level. The function of this nucleotide sequences were verified by comparison with amino acid sequences of the amino-terminus and tryptic peptides from purified plant enzyme, by northern blotting with mRHA from wild type and mutant plants, by in vitro expression yielding and enzymatically active hydroxylase, as indicated by the small dihydrokaempferol peak. Genomic southern blot analysis showed the presence of only one gene for FHT in Gypsophila paniculata.

Anti-diabetic effect of mulberry leaf extract fermented with Lactobacillus plantarum (Lactobacillus plantarum으로 발효한 뽕잎 추출물의 항당뇨 효과)

  • The purpose of this study was to isolate novel lactic acid bacteria to ferment mulberry leaf extract (MLE) and to investigate its anti-diabetic effect. Lactobacillus plantarum SG-053 isolated from gatkimchi was selected to ferment MLE because it exhibited high α-glucosidase inhibitory activity (96.8%) and enhanced the content of 1-deoxynojirimycin (DNJ), an anti-diabetic substance, in fermented MLE up-to 2.2 times. MLE fermented with L. plantarum SG-053 (FMLE) showed growth promoting activity against L6 myotubes and increased the gene expressions of IRS-1, PI3K p85α, and GLUT-4 up-to 1.4, 2.2, and 1.4 times, respectively, and 2-deoxyglucose uptake up-to 40.7%. In rat skeletal muscle tissue, the expressions of PI3K p85α and GLUT-4 increased by 6.4 and 2.1 times, respectively. These results suggest that L. plantarum SG-053 could enhance the DNJ content of MLE by fermentation and that FMLE is effective in ameliorating insulin resistance via activation of the insulin signaling pathway.

Fast Heuristic Algorithm for Similarity of Trajectories Using Discrete Fréchet Distance Measure (이산 프레셰 거리 척도를 이용한 궤적 유사도 고속계산 휴리스틱 알고리즘)

  • A trajectory is the motion path of a moving object. The advances in IT have made it possible to collect an immeasurable amount of various type of trajectory data from a moving object using location detection devices like GPS. The trajectories of moving objects are widely used in many different fields of research, including the geographic information system (GIS) field. In the GIS field, several attempts have been made to automatically generate digital maps of roads by using the vehicle trajectory data. To achieve this goal, the method to cluster the trajectories on the same road is needed. Usually, the $Fr{\acute{e}}chet$ distance measure is used to calculate the distance between a pair of trajectories. However, the $Fr{\acute{e}}chet$ distance measure requires prolonged calculation time for a large amount of trajectories. In this paper, we presented a fast heuristic algorithm to distinguish whether the trajectories are in close distance or not using the discrete $Fr{\acute{e}}chet$ distance measure. This algorithm trades the accuracy of the resulting distance with decreased calculation time. By experiments, we showed that the algorithm could distinguish between the trajectory within 10 meters and the distant trajectory with 95% accuracy and, at worst, 65% of calculation reduction, as compared with the discrete $Fr{\acute{e}}chet$ distance.

QTL Analysis of Rice Heading-related Genes Using Cheongcheong/Nagdong Doubled Haploid Genetic Map (청청/낙동 배가반수체 유전자 지도를 이용한 쌀의 출수기 관련 양적형질유전자좌(QTL) 분석)

  • Disaster-related extreme weather is rapidly increasing due to climate change. In Korea, typhoons accompanied by rainfall usually approach in August and September, causing great damage. The purpose of this study is to find a gene that regulates the heading date of rice in order to avoid loss of harvest from climate change and typhoons. Cheongcheong/Nagdong doubled haploid (CNDH) was used as the plant material to investigate the location of heading-related genes using QTL and sequence analysis by cloning the gene. In the distribution chart, the heading dates, culm lengths, panicle lengths, numbers of panicles, and 1,000-grain weights all have normal distributions. QTL analysis found 13 contigs on chromosome 8. One QTL, named qHd8, was detected on chromosome 8. The range at qHd8 was approximately 7.7 cM, with RM72 and RM404 markers near the peak. There were 13 contigs and 1 ORF. Protein sequence analysis showed that rice was similar to Os08g0341700, AtSFH13, and AtSFH7 proteins. Os08g0341700, which is involved in signal transduction, is similar to phosphatidylinositol transfer-like protein II, and complete information is not available, but it is believed to play a role in the phosphatidylinositol-specific signaling pathway related to Sec14P.

Cultivated Orostachys japonicus Induces Apoptosis in Human Colon Cancer Cells (인체 대장암 세포주 SW480에서 재배 와송의 세포 사멸 유도 효과)

  • This study was performed to elucidate the anticancer activities and the mechanism of chloroform fractions from cultivated Orostachys japonicus (CFCOJ) in human colon cancer cells. CFCOJ markedly decreased viable cell numbers in both a dose-dependent and time-dependent manner within SW480 cells. Cell death induced by CFCOJ increased cell populations in the sub-G1 phase, as well as the formation of apoptotic bodies, nuclear condensation, and induced DNA fragmentation. CFCOJ-induced apoptosis was associated with the activation of initiator caspase-8 and -9, as well as the effector caspase-3. CFCOJ also stimulated Bid cleavage, indicating that the apoptotic action of caspase-8-mediated Bid cleavage leads to the activation of caspase-9. CFCOJ increased the expression of the proapoptotic protein, Bax, and decreased the expression of the antiapoptotic protein, Bcl-2. These results indicate that CFCOJ exert anticancer effects on human colon cancer SW480 cells through a caspase-dependent apoptotic pathway.

Ectopic expression of Bcl-2 or Bcl-xL suppresses p-fluorophenylalanine-induced apoptosis through blocking mitochondria-dependent caspase cascade in human Jurkat T cells (Jurkat T 세포에 있어서 ρ-fluorophenylalanine에 의해 유도되는 세포자살의 Bcl-2 및 Bcl-xL에 의한 저해 기전)

  • $\rho$-Fluorophenylalanine (FPA), a phenylalanine analog, is able to induce apoptotic cell death of human acute leukemia Jurkat T cells. To better understand the mechanism by which FPA induces apoptotic cell death, the effect of ectopic expression of antiapoptotic proteins, Bcl-2 and Bcl-xL, on FPA-induced apoptosis was investigated by employing lurkat T cells transfected with Bcl-2 gene (JT/Bcl-2) or Bcl-xL gene (1/Bcl-xL) and Jurkat T cells transfected with vector (JT/Neo or J/Neo). When Jurkat T cells, JT/Neo or J/Neo, were exposed to FPA at concentrations ranging from 0.63 to 5.0 mM, the cell viability determined by MTT assay declined in a dose-dependent manner. In addition, apoptotic DNA fragmentation along with several apoptotic events such as caspase-8 activation, Bid cleavage, mitochondrial cytochrome c release, caspase-9 activation, caspase-3 activation, and degradation of PARP was induced. However, the FPA-induced cytotoxic effect, activation of caspase-8, and cleavage of Bid were significantly abrogated by ectopic expression of Bcl-2 or Bcl-xL. At the same time, there was marked reduction in the level of cytochrome c release from mitorhondria, caspase-9 activation, caspase-3 activation, and degradation of PARP. These results indicate that caspase-8 activation, Bid cleavage, and mitochondrial cytochrome c release with subsequent activation of the caspase cascade are negatively regulated by Bcl-2 or Bcl-xL, and are thus required for FPA-induced apoptosis in Jurkat T cells

The Contamination of Groundwater by Acid Mine Drainage in the Vicinity of the Hanchang Coal Mine and the Efficiency of the Passive Treatment System (산성광산배수에 의한 한창탄광 부근 지하수의 오염과 자연정화처리시설 효율에 관한 연구)

  • Sampling of waters from each stage of treatment system (Successive Alkalinity Producing System; SAPS), tailings seepage, and spring near the Hanchang coal mine of Kangwon Province were carried out seasonally and analyzed to evaluate the source and possible path of groundwater contamination by acid mine drainage (AM). Sulfur isotope compositions were measured to identify the origin of groundwater contaminations and the sulfate reduction processes in the SAPS. Low pH and high metal concentration of spring water indicates possibility of the groundwater contamination by AMD. Removal efficiency of acidity of the SAPS was 18.17 g/$\textrm{m}^2$/day on an average and the metal removal efficiency was almost 100%, which was higher than those of other treatment systems. However, no appreciable decrease of sulfur content and almost similar sulfur isotope compositions of water from each stage of the treatment system may suggest incomplete or very poor sulfate reduction by sulfate reducing bacteria. Chemical and sulfur isotope compositions showed that spring water was contaminated by seepage from mine tailings. And seepage of stonewall, a part of treatment system was affected by both tailings seepage and mine adit drainage. In this study site, the treatment system was constructed for the only AMD from mine adit not for tailings seepages, which resulted in the groundwater contamination from tailing seepages. Similar situation is expected in other abandoned coal mine areas.

A Case Study on Earthquake-induced Deformation of Quay Wall and Backfill in Pohang by 2D-Effective Stress Analysis (2차원 유효응력 해석에 의한 지진시 포항 안벽구조물의 변형 사례 분석)

  • The purpose of this study is to investigate the mechanism about damages occurring at quay wall and backfill in Youngilman Port during Pohang earthquake (M5.4) on November 15, 2017. In the field investigation, the horizontal displacement of the caisson occurred between 5 cm and 15 cm, and the settlement at backfill occurred higher than 10 cm. 2D-effective Stress Analysis was performed to clarify the mechanism for the damage. The input earthquake motion used acceleration ($3.25m/s^2$) measured at bedrock of Pohang habor. Based on a numerical analysis, it was found that the effective stress decreased due to the increase of excess pore pressure in the backfill ground and the horizontal displacement of the caisson occurred by about 14 cm, and the settlement occurred by about 3 cm. In backfill, the settlements occurred between 6 cm and 9 cm. This is similar to field investigation results. Also, it was found that the backfill soil was close to the Mohr-Coulomb failure line due to the cyclic loading from the effective stress path and the stress-strain behavior. It may be related to decreasing of bearing capacity induced by the reduction of effective stress caused by the increase of the excess pore water pressure.

동물 결핵

  • 동물의 결핵은 Mycobacterium bovis의 감염에 의한 만성 소모성 질병이며 인수공통전염병이다. 동물로부터 사람으로의 결핵 전염은 생유 섭취하던 시대에 상당히 많이 보고되었다. 우유의 살균처리와 소에서 피내진단에 의한 양성우 살처분 및 보상금 지급 정책을 전개하면서 M. bovis의 사람전염은 급격히 감소하였다. 소 결핵은 우리나라에서 연간 0.15% 내외의 발생을 보이고 있으며, 발생의 주원인으로는 외부입식소, 인근발생농장, 과거발생농장의 사후관리소홀 등이다. 사람 결핵의 주원인균인 M. tuberculosis와 M. bovis는 유전체가 99.9% 유사하며, M. bovis를 M. tuberculosis의 아종으로 분류하기도 한다. 두 세균은 M. tuberculosis complex에 속하며, M. tuberculosis와 M. bovis이외에도 M. africanum, M. canettii, M. microti, M. pinnipedii 등이 있다. M. bovis는 M. tuberculosis complex중에서 가장 넓은 숙주범위를 가진다. M. bovis의 대표적인 숙주는 종이름에도 나타나 있듯이 소이다. 소결핵 전파원으로서는 M. bovis에 감염된 소가 가장 중요하다. 소 이외에도 면양, 산양, 말, 돼지, 사슴, 엘크, 영양 (antelope, kudus, elands, sitatungas, oryxes, addaxes), 개, 고양이, 흰족제비 (ferrets), 낙타, 여우, 밍크, 오소리, 쥐, 영장류, 라마, 맥 (tapirs), 코끼리, 코뿔소 (rhinoceroses), 주머니쥐, 땅다람쥐 (ground squirrels), 수달 (otters), 물개, 산토끼 (hares), 두더쥐 (moles), 너구리 (raccoons), 코요테, 사자, 호랑이, 표범, 살쾡이 (lynx) 등에 감염될 수 있으나, 대부분 종결숙주 (spillover host)로 가축의 결핵방제가 유지되고 있는 국가에서는 야생동물 결핵의 가축 전염이 문제시되고 있다. M. bovis는 주로 호흡기와 소화기를 통하여 감염되며, 결핵결절이 형성되는 부위를 관찰하면 감염경로를 추정할 수 있다. 결핵에 감염되면, 초기에는 뚜렷한 임상증상을 보이지 않으나, 아침, 추운 날씨, 또는 운동 중에 심한 기침을 하며, 호흡곤란을 일으킬 수 있다. 결핵은 감염되어도 대부분 무증상이기 때문에 피내진단, 결핵결절 병리소견, 원인균 분리 등에 의해 진단하여야 한다. 감염된 결핵균은 탐식세포에 탐식되어 특징적인 육아종성 결절 병변으로 진행된다. 현재 결핵은 피내진단과 결핵결절 병리소견 등에 의해 판정하고 있다. 최신 진단법으로는 피내진단을 대체할 수 있는 인터페론 감마 검사법과 우군의 결핵 스크리닝과 말기 결핵 검사에 우수한 항체진단법이 개발되어 있다. 그러나, 소 결핵 근절을 위해서는 일관성있는 진단법과 진단기준을 적용하는 것이 중요한 성공요인중 하나이다. 소결핵 청정국인 호주와 캐나다에서는 피내진단과 도축장 결절검사를 결핵 양성우 색출방법의 근간으로 삼고 있으며, 소결핵 근절의 최종단계에 이르러서는 특이적인 검사법을 적용하였지만, 근절목적상 민감성이 높은 피내진단법을 사용하였다. 이와 더불어, 피내진단 양성우의 부검소견과 원인균 분리를 통해 결핵을 확진하여 출처농장의 역추적 검사를 통하여 결핵 양성소를 제거하였다. 한편, 결핵의 농장간 및 지역간 전파방지를 위해 결핵 청정농장과 결핵 오염농장, 결핵 청정지역과 결핵 오염지역 구분을 통하여 결핵 오염농장과 결핵 오염지역으로부터 결핵 청정농장과 결핵 청정지역으로의 이동전 결핵 검진을 통해 개체 이동에 따른 결핵 전파를 근본적으로 차단하는 시스템을 엄격히 적용한 것이 주요한 성공 요인중 하나였다. 호주 결핵 근절정책 성공요인을 요약하면, 일관성 있는 결핵진단법 적용, 양성우 출처농장의 철저한 역추적 검사, 개체 이동전 결핵 음성증명 확인, 농가단체의 경제적 및 방역상 적극적인 지원 및 협조 결핵의 지속적인 모니터 링과 현장요구에 부응하는 방제신기술의 지속적인 연구개발 등을 들 수 있다. 최근 들어 국내 동물 결핵은 소, 특히, 한우의 결핵발생이 증가하고 있으며, 사슴 결핵발생도 증가하고 있다. 농장간 및 지역간에 결핵 감수성 가축, 특히, 소와 사슴의 거래가 아주 복잡하게 이루어지고 있는 현실을 고려할 때, 결핵전파의 주원인인 결핵감염 소나 사슴의 농장내 반입을 철저히 차단해야 할 것이다. 이때, 개체 검사는 물론이고, 출처농장에 대한 결핵 음성을 확인한 후 입식하여야 할 것이며, 입식 후에도 60일정도 격리사육하면서 피내진단등 결핵검진 후 음성인 경우에만 합사하여야 할 것이다. M. bovis는 사람을 비롯한 거의 모든 온혈동물에서 결핵을 일으킬 수 있기 때문에, 결핵 감염소로 판정된 농장 종사자는 각 시도 보건소의 협조를 받아 결핵검진을 받도록 해야 한다. 농장 가축에 접촉할 수 있는 야생동물의 접촉을 차단하여야 하며, 특히, 농장 사료의 야생동물에 의한 오염을 방지할 수 있는 사료창고관리를 철저히 해야 한다. 결핵 감염소를 다룰 때는 분비물 또는 가검물에 의해 감염될 수 있기 때문에 개인방역장비 - 방역복, 마스크, 비닐장갑, 비닐장화 - 를 착용한 상태에서 다루어야 한다. 특히, 결핵 감염소를 매몰 또는 소각하는 과정에서 결핵 감염소의 배설물 및 분비물 처리를 철저히 하여야 한다. 모든 작업을 마친 후에는 개인방역장비, 매몰 또는 소각에 사용하였던 장비 등을 청소 및 소독하고 필요시 소각 또는 매몰하여야 하며, 개인감염위험과 타인 감염위험을 방지하기 위해 노출부위를 세척하여야 한다.
