In computer simulation models the output from the computer code is often deterministic, i.e., running the code twice with the same values for the input variables would give the same output. It is discussed why the response surface method with polynomial approximation for the true response function is a good approximation to the computer experiments model. A sequential strategy to find the proper reduced quadratic polynomial model is illustrated with a case study in the military war game computer simulation model.
Defense M&S(Modeling & Simulation) requires weapon effectiveness index which indicates Ph(Probability of hit) and Pk(Probability of kill) values on various impact and environmental conditions. The index is usually produced by JMEM(Joint Munition Effectiveness Manual) development process, which calculates Pk based on the impact condition and circular error probable. This approach requires experts to manually adjust the index to consider the environmental factors such as terrain, atmosphere, and obstacles. To reduce expert's involvement, this paper proposes a meta-model based method to produce weapon effectiveness index. The method considers the effects of environmental factors during calculating a munition's trajectory by utilizing high-resolution weapon system models. Based on the result of Monte-Carlo simulation, logistic regression model and Gaussian Process Regression(GPR) model is respectively developed to predict Ph and Pk values of unobserved conditions. The suggested method will help M&S users to produce weapon effectiveness index more efficiently.

Modern systems have become more and more complex due to the ever-increasing user requirements and rapid advance of technology. As such, the frequency of accidents due to system design errors or failure has been increasing. When the damage incurred by accidents to human beings or property is serious, the underlying systems are referred to as safety-critical systems. The development of such systems requires special efforts to ensure the safety of the human beings operating them. To cope with such a requirement, in this paper an approach is employed in which we consider safety starting from the conceptual design phase of the systems. Specifically, a systems design method that can detect functional failure is proposed by utilizing meta-models and M&S methods. To accomplish this, the safety design data from international safety standards are first extracted and also a meta-model is generated using SysML (systems modeling language). Then, a SysML-based system design method is proposed based on the use of the developed meta-model. We also discuss how the safety requirements can be created and verified using a simulation method. Finally, through a case study in automotive design, it is demonstrated that the detection of a functional failure and the verification of a safety requirement can be accomplished using the SysML-based M&S method. This study indicates that the use of meta-models can be useful for collecting and managing safety data and that the meta-model based M&S method can make it possible to satisfy the system requirements by reducing the design errors.
Background: To evaluate the role of the X-ray repair cross complementing group 3 (XRCC3) T241M polymorphism in head and neck cancer susceptibility. Materials and Methods: We performed a meta-analysis of all available studies, which included 3,191 cases and 5,090 controls. Results: Overall, a significant risk effect of the T241M polymorphism was not found under homologous contrast (MM vs TT: OR=1.293, 95% CI=0.926-1.805; TM vs TT: OR=1.148 95% CI=0.930-1.418) and recessive models (MM vs TT+TM): OR=1.170, 95% CI=0.905-1.512, but a significantly increased risk was observed under a dominant model (MM+TM vs TT): OR=1.243, 95% CI=1.001-1.544. In stratified analyses, there were no significant associations for Asians or Caucasians. Conclusion: Our meta-analysis suggested the XRCC3 241M allele (MM+TM) might act as a head and neck cancer risk factor among all subjects, and the effect of T241M polymorphism on head and neck susceptibility should be studied with a larger, stratified population.
Growing evidence suggests an elevated risk for colorectal neoplasia among individuals with low levels of vitamin D, the biological actions of which are mediated by the vitamin D receptor (VDR). To investigate the association among vitamin D status, VDR polymorphisms (FokI, and BsmI), and colorectal adenoma, we conducted a meta-analysis of nine studies of circulating levels of 25-hydroxyvitamin D (25(OH)D) and five studies of FokI or BsmI polymorphisms in relation to colorectal adenomas. Study-specific relative risks (RRs) and 95% confidence intervals (CIs) were pooled using a random-effects model. A total of 3398 coloreetal adenomas for 25(OH)D and 1754 colorectal adenomas for VDR were included in the meta-analysis. We identified a significant inverse association between colorectal adenoma (combined RR, 0.93; 95% CI, 0.87-0.98 per 10 ng/mL increase in 25(OH)D levels). When we examined FokI and BsmI polymorphisms in the meta-analysis, we found no association for either FokI (combined RR, 1.00; 95% CI, 0.95-1.06) or BsmI (combined RR, 0.99; 95% CI, 0.93-1.05) in the additive model. These data suggest an inverse association between circulating 25(OH)D levels and colorectal adenoma risk.
Background: Alpha-methylacyl-CoA racemase(AMACR) is thought to play key roles in diagnosis and prognosis of prostate cancer. However, studies of associations between AMACR gene polymorphisms and prostate cancer risk reported inconsistent results. Therefore, we conducted the present meta-analysis to clarify the link between AMACR gene polymorphisms and prostate cancer risk. Materials and Methods: A literature search was performed in PubMed, Embase, China National Knowledge Infrastructure (CNKI), Wanfang and Weipu databases. Odds ratios (ORs) and 95% confidence intervals (95%CIs) were calculated to assess the strength of any association between AMACR polymorphisms and prostate cancer risk. Subgroup analyses by ethnicity, source of controls, quality control and sample size were also conducted. Results: Five studies covering 3,313 cases and 3,676 controls on five polymorphisms (D175G, M9V, S201L, K277E and Q239H) were included in this meta-analysis. Significant associations were detected between prostate cancer and D175G (dominant model: OR=0.89, 95%CI=0.80-0.99, P=0.04) and M9V (dominant model: OR=0.87, 95%CI=0.78-0.97, P=0.01) polymorphisms as well as that in subgroup analyses. We also observed significant decreased prostate cancer risk in the dominant model (OR=0.90, 95%CI=0.81-0.99, P=0.04) for the S201L polymorphism. However, K277E and Q239H polymorphisms did not appear to be related to prostate cancer risk. Conclusions: The current meta-analysis indicated that D175G and M9V polymorphisms of the AMACR gene are related to prostate cancer. The S201L polymorphism might also be linked with prostate cancer risk to some extent. However, no association was observed between K277E or Q239H polymorphisms and susceptibility to prostate cancer.

Purpose To systematically compare the performance of liver imaging reporting and data system treatment response (LR-TR) with the modified Response Evaluation Criteria in Solid Tumors (mRECIST) for diagnosing viable hepatocellular carcinoma (HCC) treated with locoregional therapy (LRT). Materials and Methods Original studies of intra-individual comparisons between the diagnostic performance of LR-TR and mRECIST using dynamic contrast-enhanced CT or MRI were searched in MEDLINE and EMBASE, up to August 25, 2021. The reference standard for tumor viability was surgical pathology. The meta-analytic pooled sensitivity and specificity of the viable category using each criterion were calculated using a bivariate random-effects model and compared using bivariate meta-regression. Results For five eligible studies (430 patients with 631 treated observations), the pooled per-lesion sensitivities and specificities were 58% (95% confidence interval [CI], 45%-70%) and 93% (95% CI, 88%-96%) for the LR-TR viable category and 56% (95% CI, 42%-69%) and 86% (95% CI, 72%-94%) for the mRECIST viable category, respectively. The LR-TR viable category provided significantly higher pooled specificity (p < 0.01) than the mRECIST but comparable pooled sensitivity (p = 0.53). Conclusion The LR-TR algorithm demonstrated better specificity than mRECIST, without a significant difference in sensitivity for the diagnosis of pathologically viable HCC after LRT.
This study is performed to reexamine the association between ambient air pollution and daily mortality in seven major cities of Korea using a method of meta-analysis with the data filed for the period 1998-2001. These cities account for half of the Korean population (about 23 million). The observed concentrations of carbon monoxide (CO, mean=1.08 ppm), ozone ($O_3$, mean=33.97 ppb), particulate matter less than 10 ${\mu}m$ ($PM_{10},\;mean=57.11\;{\mu}g/m^3$), nitrogen dioxide ($NO_2$, mean=25.09 ppb), and sulfur dioxide ($SO_2$, mean=9.14 ppb) during the study period were at levels below Korea's current ambient air quality standards. Generalized additive models were applied to allow for the highly flexible fitting of seasonal and long-term time trends in air pollution as well as nonlinear associations with weather variables, such as air temperature and relative humidity. Also, we calculated a weighted mean as a meta-analysis summary of the estimates and its standard error. In city-specific analyses, an increase of $41.17{\mu}g/m^3(IQR)\;of\;PM_{10}$ corresponded to $1{\sim}12%$ more deaths, given constant weather conditions. Like most of air pollution epidemiologic studies, this meta-analysis cannot avoid fleeing from measurement misclassification since no personal measurement was taken. However, we can expect that a measurement bias be reduced in district-specific estimate since a monitoring station is better representative of air quality of the matched district. Significant heterogeneity was found for the effect of all pollutants. The estimated relative risks from meta-like analysis increased compared to those relative risks from pooled analysis. The similar results to those from the previous studies indicated existence of health effect of air pollution at current levels in many industrialized countries, including Korea.
O-ethyl S-methyl ethylphosphonothioate [$LD_{50}$ (rat, oral) 4.6mg/kg ; $K_i$(bovine erythrocyte acetylcholinesterase) 303 $M^{-1\;min-1}$] was selected as a model compound to study the mode of action of O, S-dialkyl alkylphosphonothioates which have been hypothesized to be toxic via a bioactivation process. Two chemical oxidants, meta-chloroperoxybenzoic acid and monoperoxyphthalic acid, and rat liver microsomal oxidases were used to mimic the action of mixed function oxidases on the model compound. The formation of S-oxide, a very unstable active intermediate, was proposed based on the identification of metabolic products.Furthermore, a trapping experiment with ethanol showed that the unstable intermediate S-oxide had the ability to phosphorylate acetylcholinesterase which is an important enzyme in nerve systems. The S-oxide intermediates are presumed to be responsible for the toxicity of O,S-dialkyl alkylphosphonothioates.
