Effect of Injin Fraction on Hepatic Fibrosis induced by $TGF-{\beta}1$ (인진이 $TGF-{\beta}1$ 유도성 간섬유화에 미치는 영향)

  • Objective : The aim of this study is to investigate the effect of Injin fractions on hepatic fibrosis induced by $TGF-{\beta}1$. Method : $TGF-{\beta}1$ mRNA, protein, $TGF-{\beta}1$ receptor, Smad family and PAI-I mRNA were studied in HepG2 cell, and the proliferation, connective tissue growth factor, fibronectin and collagen type I mRNA in T3891 fibroblast by quantitative RT-PCR, ELISA and thymidine incorporation assay. Results : On $TGF-{\beta}1$ mRNA and protein synthesis in HepG2, $H_2O$, butanol and hexane fractions of Injin showed inhibitory effect in a dose-dependent way. In the study on $TGF-{\beta}1$ receptor, Smad family and PAI-1 mRNA in HepG2, $H_2O$, butanol and hexane fraction of Injin showed inhibitory effect on the expression of PAI-1 in a dose-dependent way. On the proliferation of T3891 fibroblast induced by $TGF-{\beta}1$, $H_2O$, ethylacetate and butanol fractions of Injin showed inhibitory effect. In the study on the factors affected by $TGF-{\beta}1$, $H_2O$, ethylacetate and butanol fractions of Injin showed inhibitory effect on CTGF, and $H_2O$, butanol, chloroform and hexane fractions showed inhibitory effect on the expression of collagen type I, whereas no fraction showed inhibitory effect on the expression of fibronectin Conclusion : These results show that each fraction of Injin acts as a fibrosis inhibitory factor by itself or in combination, ultimately inhibiting liver cirrhosis.

A Preliminary Analysis of the Impact of Urban Green Spaces on the Urban Heat Island Effect Using a Temperature Map

  • Temperature is one of the main issues in climate change, and the urban heat island effect in highly developed urban areas is an important issue that we need to deal with. This study analyzed the extent of the cooling effects of urban green spaces. The study used a surface temperature map of Seoul. It found that the cooling effects of green space was observed within limited distances, although it varied a little depending on the parks investigated. The cooling effect distance ranged from 240m to 360m, averaging about 300m. It also found the size of an urban green space does not make much difference in cooling the surrounding areas. Although further investigation with diverse urban areas should be conducted on this matter, the results did imply that many small green spaces in the neighborhood are more effective than a single big green space in mitigating the heat island effects of cities.

Effect of Bradykinin on Oxygen Consumption in the Distal Tubule and Cortical Collecting Tubule of Rat (흰쥐 원위세뇨관과 피질집합관의 산소소비량에 대한 Bradykinin의 영향)

  • Infusion of bradykinin (BK) into the renal arteries increases sodium excretion. However, it is not clear whether natriuresis results from the renal hemodynamic effects or from the direct effect on renal tubular sodium transport. Therefore, we examined the effects of BK on the transport-dependent oxygen consumption in the distal tubule (DT) and cortical collecting tubule (CCT) of deoxycorticosterone-treated rats. BK inhibited oxygen consumption in a dose-dependent way with a maximal reduction at $0.1\;{\mu}M$ BK. The inhibitory effect of BK was not present in the absence of sodium or in the presence of ouabain (1 mM). These data imply that the inhibitory effect of BK is restricted to the sodium transport-dependent oxygen consumption. We also investigated the relationship between the effect of BK on oxygen consumption and arachidonic acid metabolism. Mepacrine $(10\;{\mu}M)$, an inhibitor of membrane phospholipases, prevented the inhibitory effect of BK, but indomethacin (0.5 mM) didn't. These results suggest that BK decreases the sodium transport-related oxygen consumption in the rat DT and/or CCT, and that it may be mediated by products of enzymes other than cyclooxygenase.

Antioxidant Effect of Captopril and Enalapril on Reactive Oxygen Species-Induced Endothelial Dysfunction in the Rabbit Abdominal Aorta

  • Background: Reactive oxygen species (ROS) are known to be related to cardiovascular diseases. Many studies have demonstrated that angiotensin-converting enzyme inhibitors have beneficial effects against ROS. We investigated the antioxidant effect of captopril and enalapril in nitric oxide mediated vascular endothelium-dependent relaxations. Materials and Methods: Isolated rabbit abdominal aorta ring segments were exposed to ROS by electrolysis of the organ bath medium (Krebs-Henseleit solution) after pretreatment with various concentrations (range, $10^{-5}$ to $3{\times}10^{-4}$ M) of captopril and enalapril. Before and after electrolysis, the endothelial function was measured by preconstricting the vessels with norepinephrine ($10^{-6}$ M) followed by the cumulative addition of acetylcholine (range, $3{\times}10^{-8}$ to $10^{-6}$ M). The relevance of the superoxide anion and hydrogen peroxide scavenging effect of captopril and enalapril was investigated using additional pretreatments of diethyldithiocarbamate (DETCA, 0.5 mM), an inhibitor of Cu/Zn superoxide dismutase, and 3-amino-1,2,4-triazole (3AT, 50 mM), an inhibitor of catalase. Results: Both captopril and enalapril preserved vascular endothelium-dependent relaxation after exposure to ROS in a dose-dependent manner (p<0.0001). Pretreatment with DETCA attenuated the antioxidant effect of captopril and enalapril (p<0.0001), but pretreatment with 3AT did not have an effect. Conclusion: Both captopril and enalapril protect endothelium against ROS in a dose-dependent fashion in isolated rabbit abdominal aortas. This protective effect is related to superoxide anion scavenging.

Effect of Sodium deoxycholate and Sodium dodecy sulfate on Phospholipid Composition and Phospholiases of Rhizopus oryzae (Rhizopus oryzae의 인지질과 그 분해효소에 미치는 계면활성제의 영향)

  • Effect of sodium deoxycholate and sodium dodecyl sulfate on Rhizopus oryzae were investigated. Morphological change was obtained by supplement of these surfactants into culture media during the sumerged culture. In accordance with morphological changes, composition of phospholipid was changed. In case of surfactant-free culture, phosphatidylcholine, phosphatidylethanolamine, and phosphatidylserine were measured more than 95% of total phospholipid. But cardiolipin and phosphatidylinositol were conspicuously increased by treatment of both sufactants. Presence of phospolipase A, C, and D were detected from mycelium. Phospholipase A and D were activated by supplement of sodium deoxycholate and C was activated by sodium dodecyl sulfate. These results were interpreted in respect of polymorphism of phospholipid and membrane stability against solubilization effect of surfactants.

Modulation of Outward Potassium Currents by Nitric Oxide in Longitudinal Smooth Muscle Cells of Guinea-pig Ileum

  • To investigate the possible involvement of outward potassium ($K^+$) currents in nitric oxide-induced relaxation in intestinal smooth muscle, we used whole-cell patch clamp technique in freshly dispersed guinea-pig ileum longitudinal smooth muscle cells. When cells were held at -60 mV and depolarized from -40 mV to -50 mV in 10 mV increments, sustained outward $K^+$ currents were evoked. The outward $K^+$ currents were markedly increased by the addition of 10 ${\mu}M$ sodium nitroprusside (SNP). 10 ${\mu}M$ S-nitroso-N-acetylpenicillamine (SNAP) and 1 mM 8-Bromo-cyclic GMP (8-Br-cGMP) also showed a similar effect to that of SNP. 1 mM tetraethylammonium (TEA) significantly reduced depolarization-activated outward $K^+$ currents. SNP-enhanced outward $K^+$ currents were blocked by the application of TEA. High EGTA containing pipette solution (10 mM) reduced the control currents and also inhibited the SNP-enhanced outward $K^+$ currents. 5 mM 4-aminopyridine (4-AP) significantly reduced the control currents but showed no effect on SNP-enhanced outward $K^+$ currents. 0.3 ${\mu}M$ apamin and 10 ${\mu}M$ glibenclamide showed no effect on SNP-enhanced outward $K^+$ currents. 10 ${\mu}M$ 1H-[1,2,4]oxadiazolo [4,3-a]quinoxaline-1-one (ODQ), a specific inhibitor of soluble guanylate cyclase, significantly blocked SNP-enhanced $K^+$ currents. We conclude that NO donors activate the $Ca^{2+}-activated$ $K^+$ channels in guinea-pig ileal smooth muscle via activation of guanylate cyclase.

Effect of light, ultrasonication and liquid smoke on germination of proso millet (Panicum miliaceum L.) seeds

  • High quality seed of proso millet that has high germination percentage, germination speed, and uniformity demanded to increases rates of mechanization in cereal crop cultivation. In order to improve germination characteristics, proso millet seeds were treated with red light, ultrasonication and liquid smoke (LS) solution that generated from hickory wood. All treatments were performed in seed priming solution with 100mM $NH_4NO_3$ at $15^{\circ}C$ for 24hrs under aeration condition. Seeds were exposed under light intensity of 2000 lux for 15m, 30m, 60m, and 120m in priming solution. Ultrasonic treatment was performed at 60%, 80%, and 100% intensity of 21.6 KHz for 5m, 10m, and 20m in priming solution. For LS treatment seed were soaked in 0%, 0.5%, 1.0%, 5.0% and 10.0% of diluted solution with $dH_2O$ or 100mM $NH_4NO_3$ solution. The effect of seed treatment was evaluated with germination percentage (GP), mean germination time (MGT), germination index (GI), germination rate (GR), Germination uniformity (GU) and heath seed percentage (HS). Our results demonstrate that red light (15min) or ultrasonication (21.6kHz, 5min) treatment improved MGT, GI, GR, and GU comparing to untreated control. Importantly, we show that LS treatments have significant effect on the health seedling and germination characteristics. Proso millet seeds that treated with 5% LS solution for 24hrs improves HS up to 97% that similar results obtained in 100mM $NH_4NO_3$ priming for 24hrs. The combined treatment with LS solution and 100mM $NH_4NO_3$ priming were not effective in all treatments. Our results demonstrate that treating seeds with LS or 100mM $NH_4NO_3$ priming or ultrasonication improves germination characteristics. The methods described here will help advance research using this species by increasing the germination performance at which successive seed pellet process.

Effect of Overshooting on Final Masses of Type Ibc Supernova Progenitors

  • Helium mass in the envelope is one of the most important properties in progenitors of type Ib/c supernovae (SNe Ib/c), since SN Ib/c progenitors are distinguished by the presence of He I lines. However, previous progenitor models do not reproduce the required He mass limit($M_{He}$ < $0.14M_{\odot}$) suggested by a spectroscopic analysis of SN Ib/c. In this work, we investigated the effect of overshooting on the evolution of pure helium stars, focusing on the final He mass in the envelope, $M_{He,f}$. We used the MESA code to calculate single helium star models with the initial masses of $M_{init}=5{\sim}30M_{\odot}$, Z=0.02, 0.04 and overshooting parameters of $f_{ov}=0{\sim}0.4$. The final He mass $M_{He,f}$ decreases as $f_{ov}$ increases, due to larger burning core compared to weak overshooting models. Dependence of the final mass $M_{He,f}$ on overshooting is strongest for models with $M_{init}=7{\sim}10M_{\odot}$, and this effect originates from accelerated mass loss during transition between WNE and WC/O phase. However, $M_{He,f}$ exceeds $0.27M_{\odot}$ for all models, which still doesn't meet the criteria of $M_{He}$ < $0.14M_{\odot}$. This implies that mass loss during the post helium burning phase must be enhanced dramatically compared to what the standard models predict.

  • We investigated hydrogen peroxide resistance of Escherichia coli possessing acetyl xylan esterase(AxeA) of Streptomyces coelicolor A3(2). The induction of AxeA production by isopropyl-$\beta$-thiogalactoside was confirmed by SDS-polyacrylamide gel electrophoresis. The differences in growth between induced and non-induced E. coli were determined by the changes in optical density of cultures after hydrogen peroxide treatment The lethal effect of hydrogen peroxide was observed for non-induced cultures at all concentrations tested in this study (lmM, 2.5mM and 5mM). However, cultures induced for AxeA production resisted the lethal effect, except at 5mM where cells were killed irrespective of the AxeA production. The axeA induction increased survival against 1.5mM hydrogen peroxide from 59% to 74%. In addition, AxeA producing E. coli showed increased survival at $45^{\circ}C$, near maximum growth temperature. Therefore, it was concluded that AxeA conferred a cross-resistance upon the bacterium against both oxidative- and heat stress.

Studies of Pharmacological Activity on the Piperazine Derivatives of Ibuprofen (이부프로펜의 피페라진 유도체에 대한 약리활성연구)

  • To enhance the activity of ibuprofen, amides of ibuprofen, 1-piperazinyl-2-(4-isobutylphenyl)propionamide(Ibu-P.A.) and 1-(4-methylpiperazinyl)-2-(4-isobutylphenyl)propionamide (Ibu-M.P.), were synthesized and the pharmaceutical properties and the pharmacological activities of the amides were studied. The lipid:water partition coefficients and pKa values were examined in vitro, and the antiinflammatory effect, analgesic effects, acute toxicity, and intestinal absorption were studied for the amides and compared with ibuprofen in vivo. The results are summarized as belows; 1) The lipid:water partition coefficients of Ibu-M.P. were higher than those of ibuprofen. 2) The calculated pKa values of ibuprofen and Ibu-M.P. were 5.49 and 8.66, respectively. 3) The antiinflammatory effects of ibuprofen, Ibu-P.A., and Ibu-M.P. were same intensity, but the duration of the effects of Ibu-P.A. and Ibu-M.P. were longer than that of ibuprofen. 4) The analgesic effect of Ibu-M.P. was more potent than those of ibuprofen and Ibu-P.A. in the acetic acid-induced writhing test. 5) The $LD_{50}$ was 495 mg/kg for ibuprofen, 187 mg/kg for Ibu-M.P., and over 1250 mg/kg for Ibu-P.A.. 6) The absorption rate constants(k) and half-life($t_{1/2}$) were 0.74($hr^{-1}$) and 0.94(hr) for ibuprofen, and 0.72 ($hr^{-1}$) and 0.96 (hr) respectively for Ibu-M.P..
