Antiviral activity of 20(R)-ginsenoside Rh2 against murine gammaherpesvirus

  • Background: Ginsenosides are the major components of Panax ginseng Meyer, an herbal medicine used for the treatment of various diseases. Different ginsenosides contribute to the biological properties of ginseng, such as antimicrobial, anticancer, and immunomodulatory properties. In this study, we investigated the antiviral effects of 15 ginsenosides and compound K on gammaherpesvirus. Methods: The antiviral activity of ginsenosides was examined using the plaque-forming assay and by analyzing the expression of the lytic gene. Results: 20(R)-Ginsenoside Rh2 inhibited the replication and proliferation of murine gammaherpesvirus 68 (MHV-68), and its half-maximal inhibitory concentration ($IC_{50} $) against MHV-68 was estimated to be $2.77{\mu}M$. In addition, 20(R)-ginsenoside Rh2 inhibited 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced lytic replication of human gammaherpesvirus in the Kaposi's sarcoma-associated herpesvirus (KSHV)-positive cell line BC3. Conclusion: Our results indicate that 20(R)-ginsenoside Rh2 can inhibit the replication of mouse and human gammaherpesviruses, and thus, has the potential to treat gammaherpesvirus infection.

Structure-Antifungal Activity Relationships of Cecropin A-Magainin 2 and Cecropin A-Melittin Hybrid Peptides on Pathogenic Fungal Cells

  • In order to investigate a relationship of the structure-antifungal and hemolytic activities between cecropin A(1-8)-magainin 2(1-12) and cecropin A(1-8)-melittin(1-12) hybrid peptides, several analogues with amino acid substitution at positions 10 (Ile) and 16 (Ser) were designed and synthesized. The increase of the hydrophobicity by substituting with Leu, Phe, and Trp at position 16 in cecropin A(1-8)-magainin 2(1-12) did not have a significant effect on antifungal activity but caused a remarkable increase in hemolytic activity. These results indicate that the hydrophobic property at position 16 of cecropin A(1-8)-magainin 2(1-12) is more correlated to hemolytic activity than to antifungal activity. Replacement with Pro at position 10 of cecropin A(1-8)- magainin 2(1-12) and cecropin A(1-8)-melittin (1-12) caused a remarkable decrease in a-helical contents in the 50% TFE solution and induced a reduction in lytic activity against Aspergillus flavus, and Aspergillus fumigatus. These results demonstrate that flexibility at the central hinge region is essential for lytic activity against fungal cells and $\alpha$-helicity of the peptides.

  • Arthrobacter luteus로부터 분리(分離)한 zymolyase 조효소(粗酵素) 표품(標品)의 효모(酵母) 세포벽(細胞壁)에 대한 용해(溶解) 활성(活性)을 측정(測定)함에 있어서, 보다 적합(適合)한 조건(條件)을 찾기 위하여, 맥아즙(麥芽汁) 배지(培地)에서, 진탕 배양(培養)한 S. sake의 용해(溶解)에 관여(關與)하는 제(諸) 인자(因子)의 영향(影響)을 검사(檢射)하였다. 1. 본(本) 용해(溶解) 효소(酵素)에 대한 S. sake 생세포(生細胞)의 용해(溶解) 감수성(感受性)은 대수(對數) 증식기(增殖期)의 세포(細胞)는 높았고, 지체기 및 정상기(定常期)의 세포(細胞)는 낮았다. 그 중에서 특(特)히 18시간(時間) 배양(培養)된 세포(細胞)는 가장 높은 용해(溶解) 감수성(感受性)을 보였다. 2. 본(本) 용해(溶解) 효소(酵素)는 S. sake의 생세포(生細胞)에 대해서 아주 낮은 용해(溶解) 활성(活性)을 나타냈으나, 효모(酵母) 세포(細胞)를 아유산(亞硫酸) 소-다(0.05 M)로 전처리(前處理)하므로서 그 용해(溶解) 활성(活性)은 4배(倍) 이상(以上)으로 증가(增加)되었다. 3. 시중(市中)의 빵 효모(酵母)에 대해 본(本 ) 용해(溶解) 효소(酵素)의 활성(活性)은 지급히 미약(微弱)하였고 아유산(亞硫酸) 소다의 효과(效果)도 약하였다. 4. 동결(凍結) 건조(乾燥)시킨 빵 효모(酵母)의 세포(細胞)는 생세포(生細胞)의 경우 보다 높은 용해(溶解) 감수성(感受性)을 보였다. 그러나 동결(凍結) 건조(乾燥) 세포(細胞)의 경우, S. sake나 빵 효모(酵母)의 그 어느 세포(細胞)도 아유산(亞硫酸) 소-다에 의한 용해(溶解) 감수성(感受性)의 영향(影響)은 인정(認定)되지 않았다. 5. 효모(酵母) 세포벽(細胞壁) 용해(溶解)에 있어서, 그 반응(反應) 속도(速度)와 조효소(粗酵素)의 농도(濃度)와의 관계(關係)는 효소(酵素) 반응(反應) 속도론(速度論)에 준(準)하는 경향(傾向)을 보이는 것 같았으나, 반응(反應) 속도(速度)와 효모(酵母) 세포(細胞)의 농도(濃度)와의 관계(關係)는 효소(酵素) 반응(反應) 속도론(速度論)과는 다른 패턴을 보여주었다. 6. 0.05 M phoshate buffer (pH 7.5)에 용해(溶解)시킨 zymolyase 조세포(粗細胞) 표품(標品)을 $7^{\circ}C$에서 10일간(日間) 보존(保存) 시킨 결과(結果), 용해(溶解) 활성(活性)의 잔존(殘存)율은 약(約) 80 %였다.

Prediction of Lytic Segments from Bacillus thuringiensis var israelensis 130 kDa and 72 kDa Proteins

  • The amino acid sequences of 130 kDa and 72 kDa proteins responsible for the larvicidal activity of Bacillus thuringiensis var israelensis (Bti) were analyzed by hydrophobic moment plots. A search for highly amphiphilic $\alpha$-helices was made in these proteins using the helical hydrophobic moment as a criterion of amphiphilicity The protein segments of the largest hydrophobic moments were analyzed. In the present communication we report the surface seeking helices in 130 kDa and 72 kDa proteins of Bacillus thuringiensis var israelensis. It is assumed that the surface seeking segments may participate in one of the membrane-related functions of Bacillus thuringiensis.

유산간균 Bacteriophage의 증식억제물질 (Effect of Phosphates on Lytic Activity of Bacteriophages Infected in Lactobacilus Cells)

  • 본 실험의 결과를 요약해 보면 인산염의 종류와 농도에 따라서 유산균의 증식과 phage 감염후의 유산균 증식에 미치는 영향을 검토하였다. 특히, phage 증식억제를 나타내는 물질은 sodium metaphosphate, sodium pyrophosphate, sodium phosphate (dibasic), potassium phosphate(dibasic)으로써 각각 0.2∼0.4%, 0.1∼0.2%, 0.2∼0.3%, 0.2∼0.35%에서 phage 감염 후 유산균의 증식을 가능하게 하였다. 그리고 phage 감염 후에도 증식한 유산균 배양액을 새로운 배지에 1,2차 계대 하였을 때, 역시 인산염의 종류와 농도에 따라 배양액에 존재하는 phage에 의해 유산균의 증식에 큰 영향을 미쳤다. 특히 sodium pyrophosphate의 경우, 0.17%이상에서는 1차 계대 후에도 phage의 증식이 억제되어 유산균이 증식하기 때문에 starter 배양액용 P.R.M제조에 이용이 가능한 것으로 보여 진다.

Biocontrol of Pectobacterium carotovorum subsp. carotovorum Using Bacteriophage PP1

  • Pectobacterium carotovorum subsp. carotovorum (formerly Erwinia carotovora subsp. carotovora) is a plant pathogen that causes soft rot and stem rot diseases in several crops, including Chinese cabbage, potato, and tomato. To control this bacterium, we isolated a bacteriophage, PP1, with lytic activity against P. carotovorum subsp. carotovorum. Transmission electron microscopy revealed that the PP1 phage belongs to the Podoviridae family of the order Caudovirales, which exhibit icosahedral heads and short non-contractile tails. PP1 phage showed high specificity for P. carotovorum subsp. carotovorum, and several bacteria belonging to different species and phyla were resistant to PP1. This phage showed rapid and strong lytic activity against its host bacteria in liquid medium and was stable over a broad range of pH values. Disease caused by P. carotovorum subsp. carotovorum was significantly reduced by PP1 treatment. Overall, PP1 bacteriophage effectively controls P. carotovorum subsp. carotovorum.

Characterization of Endolysin LysECP26 Derived from rV5-Like Phage vB_EcoM-ECP26 for Inactivation of Escherichia coli O157:H7

  • With an increase in the consumption of non-heated fresh food, foodborne shiga toxin-producing Escherichia coli (STEC) has emerged as one of the most problematic pathogens worldwide. Endolysin, a bacteriophage-derived lysis protein, is able to lyse the target bacteria without any special resistance, and thus has been garnering interest as a powerful antimicrobial agent. In this study, rV5-like phage endolysin targeting E. coli O157:H7, named as LysECP26, was identified and purified. This endolysin had a lysozyme-like catalytic domain, but differed markedly from the sequence of lambda phage endolysin. LysECP26 exhibited strong activity with a broad lytic spectrum against various gram-negative strains (29/29) and was relatively stable at a broad temperature range (4℃-55℃). The optimum temperature and pH ranges of LysECP26 were identified at 37℃-42℃ and pH 7-8, respectively. NaCl supplementation did not affect the lytic activity. Although LysECP26 was limited in that it could not pass the outer membrane, E. coli O157: H7 could be effectively controlled by adding ethylenediaminetetraacetic acid (EDTA) and citric acid (1.44 and 1.14 log CFU/ml) within 30 min. Therefore, LysECP26 may serve as an effective biocontrol agent for gram-negative pathogens, including E. coli O157:H7.
