PROTEIN SPARING EFFECT AND AMINO ACID UTILIZATION IN BROILERS FED TWO TYPES OF LYSINE

  • A growth experiment was conducted to evaluate the nutritivie values of supplemental lysine and methionine in broiler chicks. Two types of L-lysine, liquid and powder type, and DL-methionine were added to the diets at different levels of dietary protein with two growth phases, 0-3 weeks and 4-6 weeks named starter and grower, respectively. Six hundred seventy two chicks were allotted in 14 treatments; 3 controls by dietary CP level (starter-grower) with CP 23-21%, CP 21-19% and CP 20-18, 8 groups of liquid and powder lysine supplementation of 0.1, 0.2, 0.3 and 0.4%, and 3 groups of lysine and methionine supplementation. Body weight, feed intake, and excreta were measured and analyzed to determine growth performance, amino acid digestibilities, and the quantity of excreted nitrogen in feces. Chicks fed CP 23-20 with 3,200 ME kcal showed significantly better growth performance than those fed CP 21-18 for 6 weeks. The supplementation of 0.2% of either type of lysine to CP 21-19 diet improved weight gain and feed efficiecy to the extent that CP 23-21 diet was fed. Physical type of lysine did not affect chick's growth and amino acid digestibilities of the diets. The level of CP in the diet significantly affected nitrogen excretion in feces. Supplementation of lysine and methionine to CP 21-18 diet reduced fecal nitrogen by 10% compared to CP 23-21 diet. It was confirmed that 0.2% of supplemental lysine to the broiler diet spared the dietary protein by 3%, and also reduced nitrogen excretion in feces by 10%.

  • 본 연구는 $_L-lysine \; (_{-2}$, 6-Diaminohexanoic acid)의 영향을 받는 남조류와 남조류에 영향을 주는 $_L-lysine $의 농도를 파악 하고자 20종의 Microcystis.를 실험에 사용하였다. 남조류의 생장 억제 능력은 double-layered agar method와 microplate method를 이용하여 측정하였다. L-lysine의 농도를 100-${\mu}g\; ml-1~300 ${\mu}g\; ml^{-1}$이상에서 처리한 경우 Microcystis ichthyoblabe NIER-10021외 7종의 Microcystis sp.에서 투명대가 형성되었다. Microplate method서는10~500${\mu}g\; ml^{-1}$의 농도에서 생장억제 및 lysis를 나타내어TEk. 그리고 Microcystis viridis NIER-10020 외 3종은 10${\mu}g\; ml^{-1}$이하의 낮은 농도에서도 생장억제 및 분해를 나타내는 높은 활성을 보였다.

Developing Model Equation to Subdivide Lysine Requirements into Requirements for Growth and Maintenance in Pigs

  • Purified diets containing 5 graded levels of lysine were fed to young and growing pigs to determine the lysine requirement for growth and maintenance. A model was developed to subdivide the lysine requirement for the maintenance from requirement for growth. From this model, the lysine requirement for growth was 18.018 and 19.431 g/kg live weight gain and the maintenance requirement was 0.115 and 0.033 g per unit of metabolic body size at each stage of growth, respectively. In the young pigs, the lysine requirement for growth was 0.950 g/g N gain and the maintenance requirement was 0.114 g per unit of metabolic body size. The breakpoint of plasma lysine concentrations was 8.695 and 13.464 g/d, respectively. Expected requirements obtained from these formulae were in general agreement with previous estimates. Based on weight gain vs N gain equation, about 7.92% of the retained protein was comprised of lysine as compared to 7.11%, the mean lysine content of pig muscle CP.

Genotype by Dietary Lysine Interaction for Growth and Response to Sheep Red Blood Cells and Escherichia coli Inoculation in Commercial Broiler Chicks

  • An experiment was conducted to assess the interaction between genotypes and dietary lysine content in commercial broiler chicks by measuring growth, and response to sheep red blood cells (SRBC) and Escherichia coli (E.coli) inoculation. Female chicks from four genotypes (A=Anak 2000; B=Hubbard; C=Cobb and D=Synthetic broiler) were fed four levels of lysine in diet from d old till the end of experiment. The lysine content of the diet was 9.61, 10.51, 11.41 and 12.31 g/kg. Body weights at 0, 14, 28 and 42 d of age and pen-wise feed intake till 14, 28 and 42 d of age were recorded. Production of antibody against SRBC and resistance to E.coli were measured at 5 d of post inoculation (PI) at 43 d of age. Also, response to phytohemaglutinin-P (PHA-P) was measured at 12 and 24 h of PI at 48 d of age. Genotype by dietary lysine interaction was significant for body weights at 14 and 28 d of age, but not at 42 d of age. Genotype by dietary lysine interaction was not significant for feed efficiency, for antibody titers against SRBC, and for air sac lesion score, relative bodyweight change, and relative weights of bursa and spleen in response to E.coli inoculation. However, a significant interaction was observed between the levels of lysine and dosage of SRBC for antibody titers. There was significant genotype by dietary lysine interaction for cutaneous basophilic hypersensitivity (CBH) response to PHA-P at 12 and 24 h of PI. It may be concluded that to obtain optimum body weight and immunity in commercial broilers the dietary lysine requirement may be recommended specific to the genotype.

Effects of Lysine Intake during Middle to Late Gestation (Day 30 to 110) on Reproductive Performance, Colostrum Composition, Blood Metabolites and Hormones of Multiparous Sows

  • Lysine intake during gestation has a major impact on subsequent reproductive performance. The objective of this experiment was to determine the effect of lysine intake from mid-gestation until farrowing on the reproductive performance of multiparous sows. On day 30 of gestation, 200 Landrace${\times}$Large White sows were randomly assigned to one of four groups based on body weight and parity (n = 50). The gestation diets contained 0.46, 0.56, 0.65 or 0.74% lysine. Increasing dietary lysine concentration improved sow body condition at farrowing and increased litter weights (p<0.05). Dietary lysine level also had a significant effect on the dry matter (p<0.05) and protein content (p<0.05) of colostrum. Increased lysine intake increased serum insulin concentration (p<0.05) and there was a trend towards increased serum prolactin content (linear, p = 0.07). However, increased lysine tended to decrease blood urea N (quadratic, p = 0.05). These results suggest that higher lysine levels (0.65-0.75%) than those recommended by the National Research Council improved reproductive performance for multiparous gestating sows and this increase may be partially mediated through blood metabolites or metabolic hormone levels.

Generation and DNA Characterization of High-lysine Mutants by Biochemical Selection from Callus Culture of 'Hwayeongbyeo'

  • Lysine is the first essential amino acid for optimal nutrient quality in rice grain. For the narrow genetic diversities of lysine contents in rice, somaclonal variation was the source of mutation in our breeding program. Biochemical selection was conducted using 1 mM S-(2-aminoethyl) cysteine followed by two passages of 5 mM lysine plus threonine in the callus subculture medium. The lysine contents in endosperm of all progenies recovered from the biochemical selection were higher than those of their donor cultivar 'Hwayeongbyeo'. These elevated lysine levels of mutants were successfully transmitted to $M_4$ generation. The lysine contents in endosperm varied 3.85 to $4.80\%$ compare to their donor cultivar 'Hwayeongbyeo' was $3.85\%$. Three of high-lysine germplasms, Lys-l, Lys-2 and Lys-7 were selected by biochemical selection and rapid screening methods. DNA analysis showed that a new insertion of Tos 17 which mapped to rice chromosome 11 on the high-lysine mutant, Lys-2.

치어기 잉어에 있어 사료내 Lysine 부산물의 첨가효과 (Effects of Dietary Supplementation of Lysine Cell Mass (LCM) in Juvenile Israeli Carp, Cyprinus carpio)

  • 본 연구는 치어기 잉어에 있어서 어분 (fish meal, FM) 대체 단백질원으로서 lysine 부산물 (lysine cell mass, LCM)이 사료내 이용 가능성과 대체 수준을 결정하기 위해 수행하였다. 실험사료의 조단백질 함량은 $38\%$, 가용에너지는 15.2kJ/g (protein, carbohydrate and lipid: 16.7, 16.7 and 37.7kJ/g)으로 동일하게 맞추어 실험사료를 제조하였으며, 사료의 조성을 요약하면 다음과 같다: $100\%$ $FM; LCM_20$, $80\%$ $FM+20\%$ $LCM; LCM_40$, $60\%$ $FM+40\%$ $LCM; LCM_60$, $40\%$ $FM+60\%$ $LCM; LCM_100$, $100\%$ $LCM; LCM_20$l, $80\%$ $FM+20\%$ $LCM+0.07\%$ $Lysine; LCM_40$l, $60\%$ $FM+40\%$ $LCM+0.14\%$ $Lysine; LCM_60$l $40\%$ $FM+60\%$ $LCM+0.22\%$ $Lysine; LCM_100$l, $100\%$ $LCM+0.35\% Lysine. 6주 동안의 실험 결과, 증체율, 사료효율, 일간성장률, 간중량지수 및 단백질전환효율에 있어서 LCM_0와 비교하석 LCM_20는 유의적인 차이가 없는 반면에 (P>0.05), $LCM_40,\;LCM_60$$LCM_100,\;LCM_40l,\;LCM_60$l 및 $LCM_100$l는 유의적으로 낮게 나타났다 (P<0.05). $LCM_0$$LCM_20$l은 증체율, 일간성장률, 간중량지수 및 단백질 전환효율에 있어서는 유의적인 차이를 보이지 않는 반면에 (P>0.05), 사료효율에 있어서는 $LCM_0$에 비해 $LCM_20$l이 유의적으로 낮게 나타났다 (P<0.05). 헤마토그리트과 비만도는 모든 사료구에서 유의적인 차이가 없었다 (P<0.05). 따라서, 치어기 잉어에 있어서 Iysine 부산물은 어분단백질의 $100\%$ $FM; LCM_20$, $80\%$ $FM+20\%$ $LCM; LCM_40$, $60\%$ $FM+40\%$ $LCM; LCM_60$, $40\%$ $FM+60\%$ $LCM; LCM_100$, $100\%$ $LCM; LCM_20$l, $80\%$ $FM+20\%$ $LCM+0.07\%$ $Lysine; LCM_40$l, $60\%$ $FM+40\%$ $LCM+0.14\%$ $Lysine; LCM_60$l $40\%$ $FM+60\%$ $LCM+0.22\%$ $Lysine; LCM_100$l, $100\%$ $LCM+20\%까지 대체 가능하며, lysine 부산물에 결핍된 필수아미노산인 lysine을 첨가한 사료구에서는 첨가 효과가 나타나지 않았다.

Optimal Lysine:DE Ratio for Growing Pigs of Different Sexes

  • This study was conducted to evaluate changes in the lysine to digestible energy (DE) ratio on performance, apparent ileal and fecal nutrient digestibilities as well as blood urea nitrogen (BUN), and to estimate optimal lysine:DE ratios for growing pigs of different sexes. A total of 150 pigs ($(Landrace{\times}Yorkshire){\times}Duroc$, 16.78 kg average body weight, 75 barrows and 75 gilts) was randomly allotted into a $2{\times}3$ (sex by diet) factorial design. Three diets were formulated to contain a crude protein level of 19%, a DE level of 3.5 Mcal/kg with three lysine:DE ratios of 3.2 (low), 3.5 (middle) and 3.8 (high) g lysine/Mcal DE per kg diet for both barrows and gilts throughout the study. With increasing dietary lysine:DE ratio, the average daily gain (ADG) of barrows decreased but there was no significant difference among treatments (p>0.05). However, ADG was significantly higher in gilts fed the diet containing the high lysine:DE ratio (p<0.05), followed by the middle and low lysine:DE ratio dietary groups. No significant effects of lysine:DE ratios on feed intake (ADFI) and feed conversion (F/G) were observed for barrows and gilts during overall period (p>0.05), while the optimal F/G was found in barrows fed diets of low and in gilts fed high lysine:DE ratio. Blood urea nitrogen had a positive relationship with growth rate. The results showed that the optimal lysine:DE ratios were 3.2 and 3.8 g lysine/Mcal DE per kg diet for barrows and gilts of 16 to 57 kg body weight, respectively.

Corynebacterium glutamicum의 연속배양에 의한 $_{L}$-Iysine 생산 (Proguction of $_{L}$-Iysine by Continuous Culture of Corynebacterium glutamicum)

  • $_{L}$-Iysine 의 생산을 Corynebacterium glutamicum SH35을 이용하여 유가식 배양법과 연속 배양법으로 수행하였다. 유가시 배양에 의한 $_{L}$-Iysine 생산을 조사한 결과 최종 발효액 중의 lysine 농도는 129.2 g/L, lysine 수율 및 productivity는 가각 47%m 3.08g.$L^{-1}$.$h^{-1}$이였다. Single-stage 연속 배양에서 dilution rate에 따른 균의 생장 및 lysine 생산을 조사한 결과, critical diluation rate은 0.100 h$^{-1}$으로 이때 균체농동($OD_{610}$)은 67, lysine 농도는 44.0 g/L 그리고 lysine 수율 및 productivity 는 41%, 4.39 g.$L^{-1}$.$h^{-1}$이고 lysine 연속배양 시 최적의 dilution rate 이였다. 또한 lysine 생산을 위한 medium reservoir 내의 최적 탄소원 농도, 최적 질소원(ammonium sulfate) 농도 및 최적 pH는 각각 108 g/L, 25 g/L 및 6.9이었다. 유가식 배양법과 single-state 연속 배양법에 의한 lysine 생산에서, productivity 는 유가식 배양의 경우 3.08 g.$L^{-1}$.$h^{-1}$인 반면, 연속 배양에서는 4.39 g.$L^{-1}$.$h^{-1}$로 유가식 배양법에서보다 약 1.4배 정도 증가하였따. 연속 발효 공정을 통해 생산 역가의 향상은 있었으나, lysine 농도와 수율이 낮아 이에 대한 해결책으로 two-stage 연속 배양 방법을 수행하였다. 그 결과, 연속 배양 발효 수행보다 lysine 농도는 2배 정도 증가하였으며, lysine 수율은 46%로서 유가식 배양과 비슷한 정도로 얻어졌다. 이러한 실험 결과로 미루어 lysine 생산 역가 향상을 위한 방법으로 two-stage 연속배양의 산업적인 이용가능성이 있을 것으로 사료된다.

Effect of Dietary Lysine Restriction and Energy Density on Performance, Nutrient Digestibility and Meat Quality in Finishing Pigs

  • This experiment evaluated the effects of dietary lysine restriction and energy density on growth performance, nutrient digestibility and meat quality of finishing pigs. A $2{\times}2$ factorial arrangement of treatments was utilized in a randomized complete block (RCB) design, and factor 1 was lysine restriction and factor 2 was energy density. The control diet was formulated to contain 3.265 Mcal of ME/kg, 0.75% lysine in the early-finishing phase and 3.265 Mcal of ME/kg, 0.60% lysine in the late-finishing phase and other nutrients met or exceeded NRC (1998) standards. Compared to the control diet (CON), lysine levels of experimental diets were restricted to 15% (treatment EL, EEL) or 30% (treatment ELL, EELL), whereas energy level of experimental diets was increased by 0.100 or 0.200 Mcal of ME/kg. A total of 100 crossbred pigs ([Yorkshire${\times}$Landrace]${\times}$Duroc), with average initial body weight of $58.47{\pm}1.42\;kg$, were allotted to 5 dietary treatments based on sex and body weight. Each treatment had 5 replicates with 4 pigs (two barrows and two gilts) per pen. ADG, ADFI and feed efficiency were calculated in an 8-week growth trial. In the late finishing period (5-8 weeks), pigs fed ELL or EELL diets had decreased ADG and feed efficiency (p<0.01), however, when the EEL diet was provided, a similar growth performance was observed compared to those fed the CON diet during the whole experimental period (p>0.05). In a metabolic trial, 15 pigs were used to evaluate the effect of dietary lysine restriction and energy density on nutrient digestibility. The digestibility of dry matter, crude fat and crude ash was not improved by restricting dietary lysine or energy density. However, crude protein digestibility was decreased (p<0.05) as dietary lysine was restricted. When dietary lysine was restricted, fecal nitrogen was increased whereas nitrogen retention was decreased. BUN concentration was affected by dietary lysine restriction; treatments ELL and EELL had higher BUN values than other treatments (p<0.01). Carcass characteristics and meat quality were measured when average body weight of pigs reached $107.83{\pm}1.50\;kg$. Treatment ELL had higher last rib backfat depth (p<0.05) than treatment CON, but ELL and EEL did not differ significantly. The ELL and EEL treatments had higher (p<0.05) subjective marbling score than treatment CON. Treatment EEL showed higher longissimus fat content than treatment EL and CON (p<0.01). The results indicated that finishing pigs fed a diet with 15% lysine restriction and 3.465 Mcal of ME/kg energy density had no detrimental effects on growth performance and N utilization, and could achieve substantial increases in marbling and longissimus fat content of pork.
