목적 : 종양의 성장은 세포의 증식과 소실의 순수한 결과이며, 대부분의 종양들에서 아포프토시스는 계속되는 세포 소실의 가장 중요한 부분을 차지하고 있다. 본 연구에서는 비호지킨림프종 환자들을 REAL 분류에 따라 재분류한 다음 면역조직화학 염색을 이용하여 아포프토시스 지수, Ki-67 세포증식지수, Bcl-2 단백 발현, P53 단백 발현을 관찰하여 종양의 성장에 영향을 주는 여러 인자들의 관련 양상을 알아보고자 하였다. 대상 및 방법 : 비호지킨림프종 환자 67명을 대상으로 하였다. Working Formulation을 이용하여 분류하였을 때 저등급이 3명, 중등급이 64명이었다. 세포 표현형은 전체 67명의 환자 중 47명$(70\%)$이 B세포 표현형이었고, 18명$(27\%)$이 T세포 표현형이었으며, 2명에서는 분류할 수 없었다. 환자의 파라핀 포매 조직을 이용하여 면역조직화학 염색을 실시하여 아포프토시스 지수와 Ki-67 세포증식지수, Bcl-2 단백발현, P53 단백발현을 관찰하였다. 결과 : Bcl-2 단백의 발현은 $40\%$ (26/65)에서 양성 반응을 보였다. P53 단백의 발현은 $31\%$ (20/65)에서 보였다. 아포프토시스 지수는 $0\%$와 $15\%$사이의 범위에 있었으며 평균은 2.16이고 중앙값은 1.2이었다. 아포프토시스 지수는 세포 표현형이나 P53 단백발현 여부에 따라 의미 있는 차이를 보이지 않았으나, Bcl-2 단백발현 여부에 따라서는 통계학적으로 의미 있는 차이를 보였다(p=0.005). Bcl-2 단백발현이 양성이면 아포프토시스 지수가 낮았다. Ki-67 세포증식지수는 $1\%$와 $91\%$ 사이의 범위에 있었으며 평균은 $55.4\%$이었다. Ki-67 세포증식지수는 세포표현형이나 B치-2 단백발현 여부에 따라 의미 있는 차이를 보이지 않았으나, P53 단백발현 여부에 따라서는 통계학적으로 의미있는 차이를 보였다(p=0.000). 전체 환자군에서는 아포프토시스 지수와 Ki-67 세포증식지수 사이에 관련이 없었으나, Bcl-2 단백발현 양성인 환자에서는 아포프토시스 지수가 증가하면 Ki-67 세포증식지수가 증가하는 경향이 있었다(p=0.012). 결론 : Bcl-2 단백발현이 양성이면 아포프토시스 지수가 낮았고, P53 단백발현이 양성이면 Ki-67 세포증식지수는 높았다. 또한 Bcl-2 양성인 환자에서는 아포프토시스 지수와 Ki-67 세포증식지수사이의 양성 연관성을 보였는데 이는 아포프토시스가 종양의 성장에 있어서 세포의 증식과 별도로 분리하여 생각할 수 없는 것임을 시사해준다고 본다.
Objectives: To study the clinical features of the primary nasal/nasopharyngeal non-Hodgkin's lymphomas and to evaluate the implication of immunophenotyping as a prognostic factor. Patients and Methods: From January 1990 to December 1997,41 patients(median age, 41 years) of primary nasal/nasopharyngeal non-Hodgkin's lymphoma were studied. The clinical records and paraffin-embedded tissue blocks were reviewed retrospectively. The histologic features, immunophenotypic findings(pan-T, pan-B, CD3, CD56) and Epstein-Barr virus in situ hybridizatios were examined. The prognostic factors for clinical outcome were evaluated in these patients. According to Ann-Arbor system, there were 30 patiets(73%) with stage IE, 4(10%) with stage IIE, 3(7%) with stage IIIE, 4(10%) with stage IVE lymphoma. Among the patients with stage IE/IIE, 4 patients received local radiation alone, 4 received chemotherapy alone, 25 received combination chemotherapy and radiotherapy and 1 refused treatment. The patients with stage IIIE/IVE were given combination chemotherapy and radiotherapy. Results: Immunophenotyping were performed in 40 patients and staining results were as follows: 3(7%) patients with B cell, 17(42%) with T cell, 18(44%) with NK/T cell(CD56 positive), and two patients with unclassifiable result. Epstein-Barr(EB) virus in situ hybridization were performed in 28 patients and 23(82%) patients had positive EBV-encoded RNAs(EBERs). 21(55%) patients achieved a complete remission. There was no difference in complete remission between radiation alone and combination therapy. With median follow-up of 30 months, 5-years disease free survival of complete responders was 60% and 5-years overall survival rate was 36%. Multivariate analysis showed that better overall survival was related with absence of B symptoms, ECOG performance${\leq}1$ and non-NK cells. Conclusion: Most of all cases were positive for EBER. Since NK/T phenotype carried the worst prognosis, analysis for CD56 expression should be done. Further prospective studies were warranted to evaluate the role of chemotherapy in stage IE/IIE.
본 연구는 베타세포에서 라이코펜의 항사멸 효과와 그 기전에 대해 조사하기 위해 실시하였다. 라이코펜에 의한 베타세포독성을 조사하기 위해 다양한 농도 (0.1, 1, 10 nM)로 처리하였을 경우, 저농도에서 세포독성이 나타나지 않음을 관찰하였다. 선택한 농도를 사이토카인 혼합물과 함께 처리하였을 경우, 세포 생존율이 증가하는 것을 관찰하였고, 세포사멸 유도 단백질인 Bax의 발현양은 감소하고, 세포사멸억제 단백질인 Bcl-2 발현양은 증가하는 것을 관찰하였다. 또한 사이토카인 혼합물에서 증가하였던 세포내 산화스트레스가 라이코펜과 함께 처리하였을 경우 감소되는 것을 관찰하였고 이러한 효과는 항산화 유전자인 GCLC, NQO1, HO-1의 발현양이 증가함으로서 일어난 현상임을 알 수 있었다. 라이코펜은 미토콘드리아의 생성 및 기능과 관련된 유전자의 발현을 증가시키고 사이토카인 혼합물에 의해 감소되었던 세포내 ATP 생성량을 증가시켰다. 이러한 결과는 라이코펜의 항산화효과와 미토콘드리아 기능 개선 효과가 사이토카인에 의한 베타세포 사멸을 억제하는 기전 중의 하나로 작용할 수 있음을 의미한다. 향후 라이코펜이 베타세포를 타겟으로 하는 제 2형 당뇨 치료의 기능성 소재로 개발될 가능성이 있음을 시사하는 바이다.
The authors reviewed 167 malignant effusions from 110 patients, of which the primary site was established on the basis of either biopsy or surgical resection of the primary neoplasm. Main factors analysed were the distribution of primary organs and the cytohistoiogic correlation of body cavity effusions. The 167 fluid specimens from 110 patients consisted of 90 cases(53.9%) of pleural, 68(40.7%) of peritoneal, and 9(5.4%) of pericardial origins. Histologically they consisted of 82 cases(74.5%) of adenocarcinoma, 8(7.3%) of malignant lymphoma, 6(5.5%) of squamous ceil carcinoma, and 3(2.7%) of small cell carcinoma. The most common site among the primary lesions was the stomach in 25 cases(22.7%) followed by the lung in 21(19.1%), ovary on 17(15.5%), and breast in 7(6.4%). As for the distribution of primary tumors in adenocarcinoma, the most common site was lung un 16 cases (48.5%) in pleural fluid and stomach in 22(48.9%) in peritoneal fluid. In pericardial effusions, all 5 cases were from the lung. As a whole, the cytologic findings of malignant effusion were fairly representative of histologic characteristics of primary lesions. Thus, when the primary lesion Is unknown, careful evaluation of effusion cytology is presumed to be a helpful tooi for tracing the primary tumor.
Tuberculous lymphadenitis is not uncommon in Korea. Therefore, an inexpensive, safe and rapid method is needed to diagnose the tuberculous lymphadenitis. Flne needle aspiration cytology Is a good method for this purpose, but has several limitations in the diagnosis of tuberculous lymphadenitis, especially when the presence of acid-fast bacilli is not proved. To evaluation the usefulness of the polymerase chain reaction with enzyme immunoassay technique in the detection of Mycobacterium tuberculosis (M. tuberculosis) In the cervical Iymph node asplrates, the authors performed fine needle aspiration cytology and M. tuberculosis PCR with enzyme immunoassay for mycobacterial DNA sequences from 15 cases of the fine needle aspirates. Cytomorphologically, the cases were categorized into three types: predominantly necrotic materials; typical epithelioid cell granulomas with or without slant cells and caseous necrosis; and non-tuberculous lesions, such as reactive lymphadenitis, abscess, metastatic carcinoma and malignant lymphoma. M. tuberculosis DNA was found in 8 of 15 cases by PCR with enzyme immunoassay. Negative findings on PCR were achieved in 7 cases, which revealed non-tuberculous tymphadenopathy. In conclusion, we suggest that M. tuberculosis PCR with enzyme immunoassay using the fine needle aspirates is a very useful tool for the diagnosis of tuberculous lymphadenitis.
${\delta}$-Catenin, a member of the p120-catenin subfamily of armadillo proteins, reportedly increases during the late stage of prostate cancer. Our previous study demonstrates that ${\delta}$-catenin increases the stability of EGFR in prostate cancer cell lines. However, the molecular mechanism behind ${\delta}$-catenin-mediated enhanced stability of EGFR was not explored. In this study, we hypothesized that ${\delta}$-catenin enhances the protein stability of EGFR by inhibiting its lysosomal degradation that is mediated by c-casitas b-lineage lymphoma (c-Cbl), a RING domain E3 ligase. c-Cbl monoubiquitinates EGFR and thus facilitates its internalization, followed by lysosomal degradation. We observed that ${\delta}$-catenin plays a key role in EGFR stability and downstream signaling. ${\delta}$-Catenin competes with c-Cbl for EGFR binding, which results in a reduction of binding between c-Cbl and EGFR and thus decreases the ubiquitination of EGFR. This in turn increases the expression of membrane bound EGFR and enhances EGFR/Erk1/2 signaling. Our findings add a new perspective on the role of ${\delta}$-catenin in enhancing EGFR/Erk1/2 signaling-mediated prostate cancer.
Nodular lymphoid hyperplasia is a very rare benign disease and usually shows consolidation on chest X-ray with symptoms of pneumonia due to the proliferation of lymphoid cells in the lung parenchyma. It is common for patients to be diagnosed with pneumonia and treated with antibiotics, but patients often enter a cycle of repeated improvement, followed by aggravation of symptoms. At this point, surgical diagnostic tools are considered in order to differentiate between malignant disease and interstitial lung disease. Here, we report 2 cases of patients with nodular lymphoid hyperplasia and review the associated references.
Objectives : Artemisia princeps is used as moxa in moxibustion and traditional herbal medicine. And its extracts or compounds is known to have an efficacy of antioxidant, anti-diabete, anti-cancer, anti-inflammation and neuroprotection. This study was performed to investigate the cytoprotective effect of Artemisia princeps extract (APE) against arachidonic acid (AA)+iron-induced oxidative stress on HepG2 cell. Methods : The effects of APE on cell viability has been assessed using MTT assay. And flow cytometric analysis was performed to estimate APE's effects on mitochondrial function. To investigate its underlying mechanism, related protein was analysed by using immunoblot analysis. Results : Treatment of APE increased relative cell viability, prevented a decline of B-cell lymphoma-extra large (Bcl-xL) and cleavage of poly(ADP-ribose) polymerase (PARP) and procaspase-3, and also protected mitochondrial membrane permeability (MMP) against oxidative stress induced by AA+iron. In addition, APE treatment increased phosphorylation of AMP-activated protein kinase (AMPK) exerts a cytoprotective effect. Conclusions : This results demonstrate that APE has an ability to activation of AMPK which protects cells from AA+iron-induced oxidative stress and restores MMP.
전신방사선조사를 시행할 때 우선적으로 해결되어야 할 과제는 총선량의 결정과 머리, 목, 폐, 복부, 골반과 다리등 모든 부위에 동일한 선량을 조사하는 것이다. 본 연구에서는 머리, 목, 폐등 밀도와 두께에 따른 선량보정을 A1으로 만든 조직보상체를 이용하는 미네소타대학 방법(방법 1) 과 소아들이 장시간 동안 편안한 자세로 움직이지 않고 치료받을수 있도록 본원에서 개발한 고정치료틀에 조직등가물질로 머리, 목, 폐등을 보정한 방법(방법 2)의 선량분포를 비교검토하였다. 방법 1에서는 95.6%에서 100%의 선량분포를 보였으며, 방법 2 에서는 95.4%에서 100%의 선량분포를 보였다. 또한 방법 2 에서 중심 부위와 표면 부위의 선량분포는 머리 부위에서 103.4%, 목 부위에서 101.5%, 배꼽 부위에서 102.3%를 나타내었다.
Sophoricoside was isolated as the inhibitor of IL-5 bioactivity from Sophora japonica (Leguminosae). It has been reported to have an anti-inflammatory effect on rat paw edema model. To develop as an anti-allergic drug, genotoxicity of sophoricoside was investigated in bacterial and mammalian cell system such as Ames bacterial test, chromosomal aberration assay, Comet assay and MOLY assay. In Ames test, sophoricoside of 5000 ∼ 313 $\mu\textrm{g}$/plate concentrations was not shown significant mutagenic effect in Salmonella typhimurium TA98, TA100, TA1535 and TA1537 strains. The cytotoxicity (IC$\_$50/ and IC$\_$20/) of sophoricoside was determined above the concentration of 5000 $\mu\textrm{g}$/ml in Chinese hamster lung (CHL) fibroblast cell and L5178Y mouse lymphoma cell line. At concentrations of 5000, 2500 and 1250 $\mu\textrm{g}$/ml, this compound was not induced chromosomal aberration in CHL fibroblast cell in the absence and presence of S-9 metabolic activation system. Also in comet assay, DNA damage was not observed in L5178Y cell line. Also in MOLY assay, sophoricoside of 5000 ∼ 313 $\mu\textrm{g}$/ml concentrations was not shown significant mutagenic effect in absence of S-9 metabolic activation system. However, the higher concentration of 5000 and 2500 $\mu\textrm{g}$/ml of sophoricoside induced the increased mutation frequency (MF) in the presence of S-9 metabolic activation system. From these results, no genotoxic effects of sophoricoside observed in bacterial systems whereas, genotoxic effects observed in mammalian cell systems in the presence of metabolic activation system. These results suggested that the metabolite(s) of sophoricoside can cause some genotoxic effects in mammalian cells.
