Evaluation of Nutrients Removal Efficiency in Effluents of Sewage and Wastewater using Microalgae (미세조류를 이용한 오·폐수 영양염류 제거효율 평가)

  • This study was intended to evaluate the removal efficiency of nutrients in effluents of wastewater using microalgae. Microalgae used in the culture experiment collected in stream and reservoir located in Gyeongsangbuk-do. Dominant species in prior-culture tank were Monoraphidium contortum, Scenedesmus acutus, Coelastrum microporum and Chlorella sp. Dominant species in synthetic wastewater culture under the 4000 Lux and 8000 Lux were Chlorella sp. and Scenedesmus obliquus. The removal efficiency of $NO_3-N$ under the 4000 Lux and 8000 Lux were 27.2%~88.1% and 63.0%~83.6% respectively. The removal efficiency of $PO_4-P$ under the 4000 Lux and 8000 Lux showed above 93%. Removal efficiency of nutrients of $1.0{\times}10^6cells\;mL^{-1}$ inoculation concentration was more higher than that of nutrients of $1.0{\times}10^5cells\;mL^{-1}$ and $1.0{\times}10^7cells\;mL^{-1}$ inoculation concentration. Microalgae cultured in synthetic wastewater removed 94.9% of TN and 90.0% of TP. The removal rate of TN and TP in synthetic wastewater were $1.961mg\;L^{-1}\;day^{-1}$ and $0.200mg\;L^{-1}\;day^{-1}$ respectively. Nutrient removal efficiency of microalgae according to kinds of wastewater showed the highest in the private sewage.

Basic Research on Lighting Design for Learning Effect (학습효과 증진을 위한 조명설계에 대한 기초연구)

  • This study conducted basic research on an LED lighting design to improve the learning effect from brain wave analysis. The ideal environments of mathematics, language, and creative region can be different. Inside the space where the lighting environment can be experienced directly, the test subject consisted of common elements. Other lighting was blocked completely in controlled lighting conditions. The brain waves were analyzed according to the change in color temperature and illumination. The analyzer used was fabricated by EMOTIV Company. In the variable RGB LED light, the color of the light was measured, and the brain wave of each subject was determined. LED lights have variable color temperature (3000 [K], 4500 [K]. 250 [lux], 70% -350 [lux], 100% -500 [lux]). As research results, the highest concentration in a mathematics study was in the general condition of a high color temperature, in which the optimal condition was a 6000[K] color temperature and 350[lux] illumination. The optimal condition for a language study was a 4500[K] color temperature and 500[lux] illumination, and that of the creative study was 3000[K] color temperature and 500[lux] illumination. Overall, the possibility of emotional ability and concentrated learning efficiency can be improved by the LED lighting design with the color temperature and illumination.

Bacterial Hash Function Using DNA-Based XOR Logic Reveals Unexpected Behavior of the LuxR Promoter

  • Introduction: Hash functions are computer algorithms that protect information and secure transactions. In response to the NIST's "International Call for Hash Function", we developed a biological hash function using the computing capabilities of bacteria. We designed a DNA-based XOR logic gate that allows bacterial colonies arranged in a series on an agar plate to perform hash function calculations. Results and Discussion: In order to provide each colony with adequate time to process inputs and perform XOR logic, we designed and successfully demonstrated a system for time-delayed bacterial growth. Our system is based on the diffusion of ${\ss}$-lactamase, resulting in destruction of ampicillin. Our DNA-based XOR logic gate design is based on the op-position of two promoters. Our results showed that $P_{lux}$ and $P_{OmpC}$ functioned as expected individually, but $P_{lux}$ did not behave as expected in the XOR construct. Our data showed that, contrary to literature reports, the $P_{lux}$ promoter is bidirectional. In the absence of the 3OC6 inducer, the LuxR activator can bind to the $P_{lux}$ promoter and induce backwards transcription. Conclusion and Prospects: Our system of time delayed bacterial growth allows for the successive processing of a bacterial hash function, and is expected to have utility in other synthetic biology applications. While testing our DNA-based XOR logic gate, we uncovered a novel function of $P_{lux}$. In the absence of autoinducer 3OC6, LuxR binds to $P_{lux}$ and activates backwards transcription. This result advances basic research and has important implications for the widespread use of the $P_{lux}$ promoter.

Response of Rudder Fish to the White Lights (백색광에 대한 벵에돔의 반응)

  • The purpose of this study is to find the light intensity which induced maximum gathering rate and to observe the variation of the gathering rate both in daytime and at night by suing Rudder fish, Girella punctata(Gray). An experimental tank(360L*50W*55H cm) was set up in a dark room. An illumination system was attached to the end of one side of the tank to control horizontal light intensity. Eight artificial light sources were prepared by combination of three light bulbs(10W, 60W, 100W) and eight filters. During the experiment water depth was maintained 50cm level in the tank. The tank was marked into six longitudinal sections each being 60cm long to observe the distribution of fish. The fish were acclimatized in dark condition for 50 minutes prior to the main experiment. Upon turning on the light, the number of fish in each section was counted 60 times every 30 seconds, and the gathering rate was obtained from the average number of fish in each section. The light intensity inducing maximum gathering rate was 2.98 lux(1.90-4.40 lux) at daytime and 298.56 lux(188.44-444.96 lux) at night. The variation of the gathering rate of fish in illumination time was great and irregularly fluctuated, but did not show any definite pattern. It was somewhat difference between daytime and night.

luxS and smcR Quorum-Sensing System of Vibrio vulnificus as an Important Factor for In Vivo Survival

  • Vibrio vulnificus is an opportunistic pathogen that causes a septicemia and expresses numerous virulence factors, in which luxS and smcR are genes encoding for components responsible for quorum-sensing regulation. In the present study, null mutants were constructed with lesions in each or both of these two genes from the V. vulnificus Vv$\Delta$Z strain, which is a lacZ$^{-}$ and chloramphenicol/streptomycin-resistant derivative of the wild-type ATCC29307 strain, and their phenotypes related to virulence were compared with those of the parental cells. $LD_{50}$ and histopathological findings of luxS-, smcR-, or luxS- smcR- deficient mutant were not different from those of the parent strain, a lacZ-deficient streptomycin-resistant strain in mice. However, time of death in mice was delayed, and numbers of bacteria survived in bloodstream after intraperitoneal injection in mice were decreased by mutation, especially luxS and smcR double mutant (VvSR$\Delta$ZSR). These phenomena were supported by increased serum sensitivity and delayed bacterial proliferation in both murine blood and iron-restricted medium. These results suggest that the luxS and luxR homologous genes in V. vulnificus could playa role in bacterial survival in host by enhancing proliferation and adjusting to changed environment.

Role of Alkaline Serine Protease, Asp, in Vibrio alginolyticus Virulence and Regulation of Its Expression by LuxO-LuxR Regulatory System

  • The alkaline serine protease asp, which was shown to be a virulence factor of Vibrio alginolyticus as a purified protein, was cloned from V. alginolyticus EPGS, a strain recently isolated from moribund Epinephelus coioides in an outbreak of vibriosis in a mariculture farm of Shenzhen. The asp null mutant was constructed by homologous recombination with suicide plasmid pNQ705-1. Compared with the wild-type strain, the asp null mutant exhibited a significant decrease of total extracellular protease activity, and caused a IS-fold decrease in virulence of V. alginolyticus. In our previous study, the luxO and $luxR_{val}$ genes from V. alginolyticus MVP01 were cloned and identified, and the luxO-$luxR_{val}$ regulatory couple was shown to regulate various genes expression, suggesting that it played a central role in the quorum sensing system of V. alginolyticus. In this study, the regulation of the asp gene was analyzed by using RT-PCR and quantitative real-time PCR methods; we proved that its transcription was greatly induced at the late stage of growth and was regulated by a luxO-$luxR_{val}$ regulatory system.

Intraspecific Variation in the Light Intensity Niche Component of the Diatom Skeletonema Costatum from Korean Coastal Waters (한국연안 산 규조 Skeletonema Costatum 의 조도에 대한 생태적지위 성분의 종내 변이)

  • Final biomass yields (peak optical density) and growth rates (divisions/day) of seven clones of Skeletonema costatum from Korean coastal waters were measured to understand their intraspecific variations in the light intensity niche component under 25$^{\circ}C$ condition. Daily growth rates of 6 out of 7 S. costatum clones were maximum at 6000 lux while that of YS4, a neritic clone, was maximum at 9000 lux. The final biomass yields of 4 out of the 7 s. costatum clones were maximum at the lowest light intensity of 2000 lux. Minimum final biomass yields were found at 9000 lux in all the S. costatum clones other than an estuarine clone, HDC9. The intraspecific variations of the mean growth rate and mean final biomass yield under each of the three different light intensities in terms of the coefficient of variation were not greater than 10% in any of the 7 S. costatum clones.

The Effects of Visual and Auditory Feedback on Pain Reduce (시각과 청각되먹임이 통증감소에 미치는 영향)

  • This study set out to investigate what kind of effects the consistent visual stimuli and verbal and non verbal auditory stimuli have on pain alleviation, as well as to see the influence of joint application of visual and auditory stimuli at the same time on pain alleviation, according to lightness of 50lux and 200lux, ultimately providing basic data in setting up an environment in case of treating pain. The subject were comprised of 30 male and female adults with pain in the neck and back area. The subject were treated in their pain area with Transcutaneous Electrical Nerve Stimulator(TENS) 100HZ for 20 minutes in the research set where each visual, auditory, and joint visual and auditory stimuli was given. For analysis methods, Visual Analogue Scale(VAS) and McGill Pain Questionnaire were adopted to see the changes before and after treatment, and the electrocardiogram, systolic and diastolic pressure, number of heart rate and breathing frequence and endorphin were compared and analyzed using the Wilcoxon singed-rank test. And The Kreskal-walllis test was used to compare the two subgroups from each group. Wilcoxon singed-rank test and the Kreskal-walllis test was used to compare the two subgroups from each group. The results were as follows: 1. The group of 50lux and 200lux were compared given varying degrees of visual stimuli. The group of 200lux showed more reduction in pain points, average systolic and diastolic pressure and average endorphin. 2. The group of verbal and non verbal were compared given varying degrees of auditory stimuli. The group of non-verbal showed more reduction in average systolic and diastolic pressure. 3. The group of 200lux+verbal and 200lux+non verbal were compared given varying degrees of joint visual and auditory stimuli. There was found a statistical significance(p<0.05) in endorphin between the two groups, with more endorphin reduction for 200lux+non verbal group. And there was a statistically significant reduction in VAS and McGill before and after the treatment between the two groups.

Six artificial light sources were prepared by combination of two light bulbs (5W, 150W) and seven filters. During the experiment water depth was maintained 50 cm level in the tank. The tank was marked into six longitudinal sections each being 60 cm long to observe the distribution of fish. The fish were acclimatized in dark condition for 50 minutes prior to the main experiment. Upon turning on the light, the number of fish in each section was counted 40 times every 30 seconds, and the gathering rate was obtained from the average number of fish in each section. The light intensity inducing maximum gathering rate was 1.9 lux ($1.2{\sim}2.9\;lux$) in the daytime and 0.7 lux ($0.5{\sim}1.1\;lux$) at night. The variation of the gathering rate of fish in illumination time was small and comparatively fluctuated with stability.

  • Lamp mura has become a challenge for slim LCD-TV backlight design. The aim of this paper is to apply a novel diffuser plate to improve lamp mura issue instead of increasing lamps or optical films. The results are positive and contribute to the design of slimmer TV without cost up.
