단체급식에서 시금치의 조리방법에 따른 Carotenoids 및 Cis/Trans β-Carotene 함량의 변화 (Changes in the Contents of Carotenoids and Cis/Trans β-Carotenes of Fresh and Cooked Spinach in Foodservice Operations)

  • 본 연구에서는 단체급식에서 많이 이용되는 시금치의 조리방법에 따른 카로티노이드 분리 및 정량을 위해서 HPLC를 이용하여 분석하였다. 시금치의 methanol 추출물로 THF 용매 분획물에 대하여 카로티노이드 및 그 이성체 화합물을 표준품으로 해서 분리된 카로티노이드의 RT와 함량은 lutein(63%), zeaxanthin(2.1%), crytoranthin, 13-cis-$\beta$-carotene(1.8%), a-carotene, trans-$\beta$-carotene(29.6%), 9-cis-$\beta$-carotene(3.1%)의 순으로 분리되었으며, 여기서 분리된 crytoxanthin은 미량 검출되어 본 실험에서는 정량하지 않았다. Lutein은 다른 카로티노이드에 비하여 조리방법 및 가열에 의한 영향을 비교적 덜 받는 것으로 나타났고, zeaxanthin은 조리 후 감소하는 경향을 보였다. 한편, trans-cis-$\beta$-carotene의 이 성 체 인 9-cis-$\beta$-carotene과 1,3-cis-$\beta$-carotene은 조리방법과 상관없이 가열 후 증가하는 경향을 보였는데 이는 가열과정이 trans-$\beta$-carotene의 이성화를 형성시키는 것으로 설명된다. Steam 조리 및 냉동 후 microwave조리에서 trans-$\beta$-carotene은 증가하는 경향을 보였다(p<0.05). 시금치의 총 non-provitamin A의 경우, 각 처리구간에 별 차이가 없었으나, 총 provitamin A에서는 원재료와 냉동 후 조리에서 유의적 차이를 볼 수 있었다(p<0.05). 또한 provitamin A로서의 기능이 가장 큰 trans-$\beta$-carotene은 steam으로 가열한 경우 가장 높았으며 또한 카로티노이드의 총 함량도 다른 시료보다 높은 함량을 보여주었다. 그 다음은 냉동 후 microwave로 가열한 시료로 두 시료와 유의적인 차이를 보였다(p<0.05). 이상의 결과로부터 steam 가열시료, 냉동 후 가열시료, 원재료를 microwave로 가열한 시료 그리고 원재료 시금치의 순으로 카로티노이드의 총 함량이 높게 나타났다.

HPLC method for the analysis of chlorophylls and carotenoids from marine phytoplankton

  • The quantitative determination of carotenoids, chlorophylls and their degradation products from marine phytoplankton was performed by reverse-phase high performance liquid chromatography (HPLC). Separated and quantified chlorophylls and their degraded products are chlorophyll a, b, c, chlorophyllide a, phaeophytin a, b and phaeophorbide a. Concentrations of six carotenoids including fucoxanthin, 19'-butanoyloxyfucoxanthin, 19'-hexanoyloxyfucoxan-thin, prasinoxanthin, alloxanthin, zoaxanthin/lutein were also determined from natural field samples by absorbance. Results of pigment analysis of field samples collected from the southern waters of the East Sea on October 8th, 1996 were reported. Concentration of chi a ranged from 7.2 to 180.4 ng/1. Concentration of chi b and chi c ranged from 22.7 to 53.7 ng/1 and from 3.3 to 58.5 ng/1, respectively. Significant concentrations of phaeophytin a, and chlorophyllide a were also detected at different depths. Fucoxanthin, 19'-hexanoyloxyfucoxanthin and zoaxanthin/lutein were the most abundant carotenoids and 19'-butanoyloxyfucoxanthin, prasinoxanthin and alloxanthin were also detected relatively at low concentrations. These results of pigment analyses suggest predominance of diatom, prymnesiophytes and cyanobacteria and presence of crysophyte, green algae (prasinophyte and prochlorophyte), and cryptophyte in the study area on October 8th, 1996, We report prymnesiophyte for the first time as an important biomass component of marine phytoplankton in the study area. Vertical profiles for the concentration changes of the major pigments were also given.

Serum Carotenoid, Retinol and Tocopherol Concentrations and Risk of Cervical Cancer among Chinese Women

  • Background: Despite many epidemiological studies on the effects of dietary antioxidant micronutrients on risk of cervical cancer, the findings remain uncertain and little evidence is available for serum nutrient markers. The present study aimed to examine the relationship between serum carotenoid, retinol and tocopherol concentrations and risk of cervical cancer among Chinese women. Materials and Methods: We conducted a hospital-based case-control study in which 358 adults (158 incident cases and 200 controls) were recruited from Xinjiang, China. Serum levels of carotenoids (${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin, lycopene and lutein/zeaxanthin), retinol, and tocopherols (${\alpha}$-tocopherol and ${\gamma}$-tocopherol) were assessed by reverse-phase high-performance liquid chromatography. Results: We found inverse associations between serum carotenoid (${\alpha}$-carotene, ${\beta}$-carotene, and lutein/zeaxanthin) and tocopherol (${\alpha}$-tocopherol) concentrations and the risk of cervical cancer after adjusting for potential confounders, but a null association for retinol. The ORs for 1-SD increase were 0.71 (95 % CI: 0.56-0.92; p=0.003) for total carotenoids and 0.75 (95 % CI: 0.60-0.94; p=0.008) for total tocopherols. Conclusions: These results show that higher serum concentrations of some carotenoids and tocopherols are associated with a lower risk of cervical cancer among Chinese women.

Photochemical Response in 0-Year-Old and 1-Year-Old Needles of Picea glehnii during Cold Acclimation and Low Temperature

  • P. glehnii, an evergreen conifer found in northern areas, is known as a cold-resistant species. In this experiment, we measured the water content, PSⅡ efficiency, chlorophyll fluorescence, pigments of the xanthophyll-cycle and activity of enzymes of the ascorbate-glutathione cycle during cold acclimation and at subsequent low-temperature conditions to examine the importance of acclimation to cold tolerance. P. glehnii showed a decrease in PSⅡ efficiency (especially in Fv) during cold acclimation and at subsequent low temperatures. However, cold-acclimated needles showed higher PSⅡ efficiency at low temperatures than nonacclimated needles. In addition, 0-YON (first-year needles) showed an increase in $\beta$-carotene and lutein, while 1-YON (one-year-old needles) immediately developed an antioxidant mechanism in the ascorbate-gluthathione cycle as soon as they were exposed to low temperature and both 0-YON and 1-YON showed increased zeaxanthin and de-epoxidation ratios at continuous low temperature. Based on our results, we suggest that P. glehnii maintain PSⅡ efficiency at low temperature by effectively protecting the photosynthetic apparatus from photo-damage by rapid induction of an antioxidant mechanism in 1-YON and dissipation of excess energy by $\beta$-carotene and lutein in 0-YON.

재(회) 처리의 미역 색소 안정화 효과 (PIGMENT STABILIZATION OF FRESH UNDARIA PINNATIFIDA WHEN TREATED WITH ASHES)

  • In this study, changes in chlorophyll and carotenoid pigment in fresh and ash treated Undaria pinnetifida were determined by column chromatography. And pigment stabilizing effects of ashes were discussed. The results obtained are summarized as follows: 1. The main carotenoids of Undaria pinnetifida are fucoxanthin, violaxanthin, lutein and $\beta-carotene$. 2. In storage of fresh Undaria pinnetifida at $2-5^{\circ}C$ for 30 days, chlorophyll a decreased rapidly and mainly converted to pheophytin a. Among carotenoids, lutein was markedly lost while the others retained approximately $30\%$ after 30 days' storage. 3, By treating the sample with ashes, the loss of chlorophyll a was restrained whereas that of carotenoids seemed to be promoted. It is considered that pigment stabilization was attributed to both effect, the pH control effect by the alkalinity of ashes and the fixation of the pigment by heavy metals in ashes. The stabilizing effect of chlorophyll a differed from the kind of ashes used. Reed ashes showed better effect than others such as straw or pine ashes. 4. Since fucoxanthin is extremely sensitive to alkali the treatment of ashes with a high alkalinity resulted in rapid decrease of the pigment during storage. 5. In the analysis of ashes, reed ashes which had better effect on chlorophyll a retention than the other ashes showed the highest content of iron.

가열처리 및 저장조건에 따른 당근과 시금치퓨레의 Carotenoids 함량변화와 이성질화 형성에 관한 연구 (Changes in Carotenoids Contents in Pureed and Cooked Carrot and Spinach during Storage)

  • Investigations were conducted on the changes in carotenoids content, and quantification of cis-trans-${\beta}$-carotene Isomers in pureed and cooked carrot and spinach during storage. The isomerization and degradation of carotenoids were monitored by high-performance liquid chromatography on a C$\_$30/ reversed-phase column with diode-array detection. The results showed that lutein, ail-trans-${\beta}$-carotene, ${\alpha}$-carotene, 9-cis-${\beta}$-carotene and 13-cis-${\beta}$-carotene were present in carrot and spinach. Zeaxanthin and cryptoxanthin were present in raw spinach. The contents of lutein, zeaxanthin, cryptoxanthin, ${\alpha}$-carotene and all-trans-${\beta}$-carotene in pureed and cooked carrot and spinach decreased with increasing storage period. The 9-cis and 13-cis carotenoid isomers were the major types formed in cooked carrot during storage. Cooking was not found to alter the carotenoid profile of the sample, but increased the total amount of carotenoids compared with pured ones. This increase could be explained that cooking itself increased the extraction efficiency and inactivated the enzymes degradating carotenoids.

고구마 잎의 유용성분 추출조건 최적화 (Optimization of Extraction Conditions for Useful Components from Sweet Potato Leaves)

  • 본 연구는 고구마 잎의 유용성분 추출을 최적화하기 위하여 추출온도, 시료에 대한 용매 비 및 에탄올 농도에 대한 영향을 살펴보았다. 추출조건별 lutein, total chlorophyll, polyphenol 및 항산화활성에 대한 반응표면 회귀식의 결정계수 $R^2$ 값은 0.9354~0.9971 범위로 모두 0.1%의 유의수준에서 유의성이 인정되었다. 추출물의 lutein과 총 폴리페놀함량은 온도, 시료에 대한 용매 비 및 에탄올 농도의 영향을 많이 받으며, 총 클로로필 함량은 온도의 영향을 받지 않는 것으로 나타났다. 항산화활성 또한 온도의 영향보다 시료에 대한 용매 비와 에탄올 농도의 영향을 크게 받는 것으로 나타났다. 최적 추출조건과 함량은 lutein이 $46.75^{\circ}C$, 25.78 mL/g 및 81.23%에서 $381.88{\mu}g/g$, 총 폴리페놀은 $53.90^{\circ}C$, 29.10 mL/g 및 67%에서 58.44 mg/g, 총 클로로필은 $69.90^{\circ}C$, 25.98 mL/g 및 93.10%에서 769.1 mg/100 g이었다. DPPH 라디칼 소거능의 경우 $IC_{50}$ 값이 $54.63^{\circ}C$, 19.57 mL/g 및 64.56%에서 0.582 mg/mL로 나타났고, ABTS 라디칼 소거활성은 $44.51^{\circ}C$, 26.01 mL/g 및 64.42 %에서 32.54 mg AA eq/g이었다. 예측된 최적조건에서의 실험값은 예측 값과 유사한 값을 나타내었으며, 이러한 결과는 고구마 잎을 이용한 기능성식품 개발에 이용할 수 있을 것으로 판단된다.

연어과에 속하는 열목어와 산천어의 Carotenoid 색소성분의 비교 (Comparison of Carotenoid Pigments on Manchurian Trout, Brachymystax lenok and Masu Salmon, Oncorhynchus macrostomus in the Family Salmonidae)

  • 담수산 어류에 관한 비교 생화학적 연구의 일환으로서, 우리나라 특산의 냉수성 담수 어류인 천연산 열목어와 천연산 및 양식산 산천어의 표피 carotenoids를 TLC, column chromatography 및 HPLC로 분리하고 $NaBH_4$에 의한 환원반응, $I_2$에 의한 이성화반응 및 uv-visible spect-rophotometer로 동정하여 비교, 검토한 결과는 다음과 같다. 열목어 표피의 총 carotenoids 함량은 천연산이 $3.72\;mg\%$으로서 연어과의 다른 어종에 비해 함량이 높았으며 carotenoids 조성은 zeaxanthin $36.9\%$, $\beta-carotene\;14.7\%$로서 주성분을 이루고, 그 외 triol $7.8\%$, isocryptoxan-thin $7.3\%$, 4-hydroxy echinenone $5.7\%$, lutein $4.7\%$, salmoxanthin $4.5\%$ 및 astaxathin $2.2\%$의 순으로 나타났으며, canthaxanthin, tunaxanthin A, tunaxanthin B, tunaxanthin C, $\beta-cryptoxanthin$$\alpha-cryptoxanthin$이 미량성분으로 함유되어있다. 산천어 표피의 총 carotenoids 함량은 천연산 $0.82\;mg\%$인데 비하여 양식산은 $0.66\;mg\%$ 로서 천연산이 양식산에 비해 함량이 높았다. carotenoid 조성은 천연산에서 zeaxanthin $20.7\%$, isocryptoxanthin $17.0\%\;\beta-carotene$$15.8\%$로서 주성분을 이루고, 그 외 $\beta-cryptoxanthin\;6.7\%$, triol $6.2\%$, 4-hydroxy echinenone $6.1\%$, salmoxanthin $6.1\%$, canthaxanthin $5.9\%$, Butein $5.8\%$, $\alpha-cryptoxanthin\;4.9\%$ 및 astaxanthin $1.0\%$의 순으로 함유하며, 양식 산에 는 isocryptoxanthin $19.7\%$, $\beta-carotene\;18.0\%$, Beaxanthin $10.3\%$이 주성분을 이루며, 그 외 $\beta-cryptoxanthin\;8.9\%$, $\alpha-cryptoxanthin\;8.5\%$, lutein $8.0\%$, canthaxanthin $7.6\%$, triol $5.1\%$ 및 astaxanthin $2.0\%$의 순으로 함유하였다. 한편 천연산 산천어에는 zeaxanthin, salmoxanrhin 및 4-hydroxy echinenone의 함량이 높은 반면 양식산 산천어에는 $\alpha$-cryptoxanthin의 함량이 높아 서로 다른 경향을 보여 상이하였다. 양식산 산천어에는 천연산 산천어와는 달리, 연어과 특유의 carotenoid인 4-hydroxy echinenone과 salmoxanthin이 검출되지 않았고 또한 천연산 및 양식산 산천어는 천연산 열목어와는 달리 tunaxanthin A, tunaxanthin B 및 tunaxanthin C가 존재하지 않았다.
