해당화 뿌리의 추출물 모두 0.5g/L 이하의 농도로 투여 시는 정상세포 생존율을 80%이상으로 유지시켜 시료자체에 의한 독성은 나타나지 않으며, 각 추출물의 암세포에 대한 생육억제활성은 0.1 mg/ml의 낮은 농도에서도 비교적 $40{\sim}60%$의 높은 억제율을 나타내었다. 추출물들은 0.5mg/ml 이상의 농도에서 MCF7에 대하여 66%이상, A549에 대하여서는 70%이상의 생육억제활성을 나타내었다. Hep3B에 대하여서는 에탄을 추출물은 78%이상의 높은 암세포 생육억제활성을 나타내었으며, AGS에 대하여 물 추출물이 약 68%정도의 암세포 생육억제활성을 나타내었다 각 추출물의 selectivity에 있어서 각 추출물이 0.1mg/ml에서 1.0mg/ml의 농도에서 4이상의 수치를 나타내어 해당화 뿌리 추출물들이 암세포를 선택적으로 사멸하는 기작을 지닌 것을 확인하였다. 또한 암세포와 정상세포의 시간에 따른 산화 정도를 Microphysiometer를 이용하여 측정한 결과 시료 투여 후 점차적으로 산화도가 빠르게 증가하는 것을 확인하였다. 각 시료들의 면역세포생육 증진 기능은 0.5 mg/ml 이상의 농도에서 B세포의 생장을 $1.2{\sim}1.3$배 이상 증가시켰으며, T세포의 경우 $1.2{\sim}1.5$배 이상의 생장율의 증가를 나타내었다 에탄올 추출물이 물 추출물들에 비하여 높은 면역세포의 생육증진활성을 나타냄을 확인하였고, cytokine의 생성측정정도를 측정한 결과, IL-6는 에탄을 추출물이 배양 6일째 0.5mg/ml의 농도에서 최고 61.3 pg/ml의 분비량을 나타내었으며, $TNF-{\alpha}$도 에탄올 추출물이 배양 6일째 최고 61.9 pg/ml의 분비량을 나타내었다. acridine orange와 ethidium bromide로 형광염색하여 인간 면역 T cell에 대한세포 사멸형태를 측정한 결과 배양 4일째부터 15% 이상의 세포들이 자가 사멸 형태로 사멸함을 확인할 수 있었다.
목적 : 피부나 폐, 간, 신장 등의 섬유화가 방사선 조사 후 주요한 부작용 중의 하나이나 아직까지 이 과정에서 $TGF-\beta$가 주요한 역할을 하는 것 이외에는 밝혀진 것이 거의 없는 상태이다. 또 matrix metalloproteinase (MMP) 및 tissue inhibitor of metalloproteinase (TIMP) 유전자 활성 및 발현이 조직의 재구성과 손상, 암세포의 침윤과 전이에 관여하고 있고, 방사선이 종양의 치료에 널리 이용되고 있음에도 방사선에 의한 MMP와 TIMP 유전자의 발현에 대한 연구는 거의 없는 상태이다. 이에 저자들은 방사선 조사가 TIMP-1, TIMP-2의 발현에 어떤 영향을 끼치는지 알아보고자 본 연구를 시작하였다. 대상 및 방법 : C57BL/6 생쥐를 Varian CL4/100을 사용하여 각각 0, 2, 10 Gy의 조사량으로 전신조사를 하였다. 방사선 조사 후 24, 48 시간에 생쥐를 희생하여 폐, 간, 신장을 얻어 paraffin 포매 조직 절편을 만들어 Avidin-Biotin Complex 방법으로 면역 염색을 한 후 광학 현미경으로 관찰하여 염색의 강도와 면적에 따라 점수화 하였다. 결과 : TIMP-1은 대조군에 비해 방사선 조사 후 폐조직에서는 변화가 없는 것으로 나타났다. 간조직에서는 간세포와 Kupffer 세포에서 발현이 증가하였고, 신장조직에서는 주로 세뇨관 세포에서 발현이 증가하였고 방사선 조사 후 경과 시간에 따른 변화는 거의 없었다. 방사선량에 따른 발현의 정도는 신장을 제외하고는 변화가 거의 없었다. TIMP-2는 폐조직에서는 2 Gy 조사 후 발현이 증가되었고, 간조직에서도 2 Gy 24 시간에는 활성이 증가했으나 10 Gy에서는 대조군 수준이었다. 신장조직에서는 10 Gy에서는 발현의 정도의 변화는 없었고 2 Gy에서는 발현이 감소하였다. 결론 : 각 장기에서 TIMP-1과 TIMP-2의 발현의 정도는 차이가 있었으며, 각 장기의 특정한 세포에서만 발현이 되었다. TIMP-1의 경우 간과 신장에서는 방사선에 의해 발현이 증가되었으나 폐에서는 발현이 증가되지 않았다. TIMP-2의 경우 폐에서는 방사선에 의해 발현이 증가하였으나 간과 폐에서는 방사선에 의한 발현의 변화는 불규칙적이었다. 방사선 조사 후 경과 시간과 방사선량에 따른 발현의 변화도 일정하지 않았다.
Background: The aim of this study was to evaluate how CYP2C19 affects icotinib and metabolite' exposure, and to determine whether the exposure and EGFR genotype influences survival time, tumor metastasis and adverse drug reactions. Materials and Methods: 274 NSCLC patients who accepted 125mg icotinib/t.i.d. were chosen from a phase III study. Blood samples were obtained in $672^{nd}$ ($4^{th}$ week) and $1,680^{th}$ hours ($10^{th}$ week), and plasma was used to quantify the concentration of icotinib and blood cells were sampled to check the genotypes. Clinical data were also collected at the same time, including EGFR genotypes. Plasma concentrations were assessed by HPLC-MS/MS and genotype by sequencing. All data were analyzed through SPSS 17.0 and SAS 9.2. Results: CYP 2C19 genotypes affected bio-transformation from icotinib to M24 and M26, especially in poor-metabolisers. Higher icotinib concentrations (>1000 ng/mL) not only increased patient PFS and OS but also reduced tumor metastasis. Patients with mutant EGFR experienced a higher median PFS and OS (234 and 627 days), especially those with the 19del genotype demonstrating higher PR ratio. Patients who suffered grade II skin toxicity had a higher icotinib exposure than those with grade I skin toxicity or no adverse effects. Liver toxic reactions might occur in patients with greater M20 and M23 plasma concentrations. Conclusions: CYP2C19 polymorphisms significantly affect icotinib, M24 and M26 exposure. Patients with mutant EGFR genotype and higher icotinib concentration might have increased PFS and OS and lower tumor metastasis. Liver ADR events and serious skin effects might be respectively induced by greater M20, M23 and icotinib concentrations.
본 연구는 톳에서 푸코이단의 추출시 초음파 공정을 이용하여 처리함으로써 유래 푸코이단의 저분자화를 통한 면역활성 및 항암활성의 증진 효과를 알아보고자 톳 유래 푸코이단 및 저분자 푸코이단(분자량 14,000 이하)을 이용하여 수행하고, 푸코이단 표준물질과 비교하였다. 시료 첨가를 통한 면역 B세포와 T세포의 생육도에서 톳 유래 저분자 푸코이단이 1.0 mg/mL의 농도에서 무첨가 대조구와 비교해 각각 367%와 387%의 생육도를 나타내며 가장 높은 활성을 나타내었다. 면역세포의 cytokine 분비량 측정에서도 생육도 측정에서와 유사한 결과를 나타내며 저분자 푸코이단이 가장 많은 cytokine 분비량을 나타내었으며, 이는 NK 세포의 생육에도 영향을 끼치는 것으로 사료되는 바, NK 세포 생육 증진에서도 가장 높은 수치를 나타냈다. 인간 정상세포를 이용한 세포독성 측정에서는 시료 간 큰 차이를 보이지는 않았으나 1.0 mg/mL에서 푸코이단 표준물질이 21.45%로 가장 높은 수치를 나타내었고, 저분자된 톳 유래 푸코이단이 19%로 가장 낮은 수치를 나타내었다. 인간폐암세포인 A549와 유방암세포인 MCF-7에 대한 생육억제 활성 측정에서도 저분자 푸코이단이 1.0 mg/mL의 농도에서 각각 69.8%와 83.3%를 나타내었으며, 푸코이단 표준물질도 55% 이상의 활성을 나타내었다. 이상의 결과를 통해 푸코이단 시료는 저분자화를 통해 면역 및 항암활성의 증진이 가능함을 확인하였다.
본 연구에서는 인간 골육종암세포주인 Saos-2 세포를 이용하여 Eucheuma cottonii 추출물(Extract of Eucheuma cottonii, EE)의 항암 활성 및 분자적 작용기전을 분석하였다. 먼저 EE가 세포증식에 미치는 영향을 WST-1® assay를 통해 확인한 결과 EE는 인간 위암세포 AGS, 인간 간암세포 SK-Hep 1, 인간 뇌교모세포종 U87MG, 인간 정상 신장세포 HEK-293의 생존율에는 영향을 미치지 않고 Saos-2 세포주의 생존율만을 농도의존적으로 감소시킴을 확인 하였다. 또한 처리 농도가 증가함에 따라 Saos-2 세포의 외형적 변화가 나타남을 도립현미경을 통해 확인할 수 있었다. 그리고 DAPI staining을 통해 apoptosis classical hall marker라고 할 수 있는 DNA fragmentation이 EE 처리 농도 의존적으로 나타남을 관찰할 수 있었다. 이를 토대로 EE가 Saos-2 세포에서 세포 내의 어떠한 기작을 통해 apoptosis를 유도하는지 Western blot analysis를 통해 확인한 결과, Fas-Associated Death Domain(FADD)에 의한 caspase cascade signal pathway 발현이 증가하였고, apoptosis의 key protein 이라고 할 수 있는 cleaved caspase-3와 하위 인자인 cleaved PARP가 증가 함을 확인할 수 있었다. 이와 관련된 분자적 기전 분석을 위한 immunofluorescence staining과 Flow cytometry analysis를 추가로 수행한 결과, EE 처리시 caspase cascade signal pathway의 시발점인 FAS와 cleaved caspase-3의 발현증가가 실제 세포 내에서 일어남을 관찰 할 수 있었으며 apoptosis 유발군인 sub G1기가 증가 함을 알 수 있었다. 이상의 결과를 통해 EE는 인간 골육종암세포에서 FADD에 의한 caspase cascade signal pathway 발현증가가 유도되어 apoptosis를 유발시킨다는 것을 증명하였으며 새로운 골육종암 치료제로서의 개발 가능성과 기전연구를 위한 중요한 기초자료가 될 수 있음을 시사한다.
Background : Peripheral nerves more rapidly recover than central nerves. However, it has been known that the degree of reaction of axons of peripheral nerves is affected by distinctive characteristics of axons and environmental factors near the axons. Taxol is a widely used medicine as for ovarian, breast, lung and gastric cancer. However it causes patients difficulties under treatment due to its toxic and side effects, which include persistent pain. Objectives : This study reviewed how SJT extract in vitro and in vivo affects nerve tissues of a sciatic nerve damaged by Taxol. It also studied how SJT extract in vivo affects axons of the sciatic nerve after the sciatic nerve was damaged by pressing. Methods : After vehicle, Taxol, and Taxol plus SJT were treated respectively for tissue of the sciatic nerve in vitro and then tissues were observed using Neurofilament 200, Hoechst, ${\beta}$-tubulin, $S100{\beta}$, caspase-3 and anti-cdc2. SJT was also oral medicated by injecting Taxol into the sciatic nerve of in vivo rats. Tissues of the sciatic nerve and axons of DRG sensory nerves were then observed using Neurofilament 200, Hoechst, ${\beta}$-tubulin, $S100{\beta}$, caspase-3 and p-Erk1/2. After inflicting pressing damage to the sciatic nerve of in vivo rats, tissues of the sciatic nerve and DRG sensory nerve were observed using Neurofilament 200, Hoechst, $S100{\beta}$, caspase-3, anti-cdc2, phospho-vimentin, ${\beta}1$-integrin, Dil reverse tracking and p-Erk1/2. Results : The group of in vitro Taxol plus SJT treatment had meaningful effects after sciatic nerve tissue was damaged by Taxol. The group of in vivo SJT treatment had effects of regenerating Schwann cells and axons which were damaged by Taxol treatment. The group of in vivo SJT had effects of regenerating axons in damaged areas after the sciatic nerve was damaged by pressing, and also had variations of distribution in Schwann cells at DRG sensory nerves and axons. Conclusions : This study confirmed that SJT treatment is effective for growth of axons in the sciatic nerve tissues and improvement of Schwann cells after axons of the sciatic nerve tissues was damaged. After tissues of sciatic nerve was damaged by pressing in vivo, SJT treatment had effects on promoting regeneration of axon in the damaged area and reactional capabilities in axons of DRG sensory nerves.
Background: Ginsenoside Rh2 is well known for many pharmacological activities, such as anticancer, antidiabetes, antiinflammatory, and antiobesity properties. Glycosyltransferases (GTs) are ubiquitous enzymes present in nature and are widely used for the synthesis of oligosaccharides, polysaccharides, glycoconjugates, and novel derivatives. We aimed to synthesize new ginsenosides from Rh2 using the recombinant GT enzyme and investigate its cytotoxicity with diverse cell lines. Methods: We have used a GT gene with 1,224-bp gene sequence cloned from Lactobacillus rhamnosus (LRGT) and then expressed in Escherichia coli BL21 (DE3). The recombinant GT protein was purified and demonstrated to transform Rh2 into two novel ginsenosides, and they were characterized by nuclear magnetic resonance (NMR) techniques and evaluated by 3-(4, 5-dimethylthiazol-2-yl)-2-5-diphenyltetrazolium bromide assay. Results: Two novel ginsenosides with an additional glucopyranosyl (6→1) and two additional glucopyranosyl (6→1) linked with the C-3 position of the substrate Rh2 were synthesized, respectively. Cell viability assay in the lung cancer (A549) cell line showed that glucosyl ginsenoside Rh2 inhibited cell viability more potently than ginsenoside Rg3 and Rh2 at a concentration of 10 μM. Furthermore, glucosyl ginsenoside Rh2 did not exhibit any cytotoxic effect in murine macrophage cells (RAW264.7), mouse embryo fibroblasts cells (3T3-L1), and skin cells (B16BL6) at a concentration of 10 μM compared with ginsenoside Rh2 and Rg3. Conclusion: This is the first report on the synthesis of two novel ginsenosides, namely, glucosyl ginsenoside Rh2 and diglucosyl ginsenoside Rh2 from Rh2 by using recombinant GT isolated from L. rhamnosus. Moreover, diglucosyl ginsenoside Rh2 might be a new candidate for treatment of inflammation, obesity, and skin whiting, and especially for anticancer.

Background: This study was carried out to investigate the effect of the steaming process on chemical constituents, free radical scavenging activity, and antiproliferative effect of Vietnamese ginseng. Methods: Samples of powdered Vietnamese ginseng were steamed at $120^{\circ}C$ for various times and thei extracts were subjected to chemical and biological studies. Results: Upon steaming, contents of polar ginsenosides, such as Rb1, Rc, Rd, Re, and Rg1, were rapidly decreased, whereas less polar ginsenosides such as Rg3, Rg5, Rk1, Rk3, and Rh4 were increased as reported previously. However, ocotillol type saponins, which have no glycosyl moiety at the C-20 position, were relatively stable on steaming. The radical scavenging activity was increased continuously up to 20 h of steaming. Similarly, the antiproliferative activity against A549 lung cancer cells was also increased. Conclusion: It seems that the antiproliferative activity is closely related to the contents of ginsenoside Rg3, Rg5, and Rk1.
Sunlight, and in particular its UV component, is the major environmental trigger that underlies the major signs of human skin and skin cancer in general. Therefore, this study was carried out to investigate the UV protection effects of R. coreanus. R. coreanus was extracted by ultra high pressure extraction process at 500 MPa and $30^{\circ}C$ for 5 and 15 minutes. The cytotoxicity of the extracts extracted by ultra high pressure process on human dermal fibroblast cell CCD-986sk, human kidney normal cell HEK293, and human lung normal cell HEL299 was measured as 17.5%, 16.5% and 14.0%, respectively in adding $1.0\;mg/m{\ell}$ of the samples, which was much lower than that from conventional water extraction method at $100^{\circ}C$ as 23.2%, 22.5%, 21.2%. The secretion of $NO^-$ from macrophage showed $15.9\;{\mu}M$ on the R. coreanus extract from this process, which was higher than others. Prostaglandin $E_2$ ($PGE_2$) production from UV-induced human skin cells was also greatly decreased down to $510\;pg/m{\ell}$, compared to the control. From the results, we considered that the extracts from R. coreanus could be potent natural materials for skin anti-inflammation agent, and could be used as a potential anti-aging for the photo-damaged skin.
Carbon nanotubes (CNTs) are one of the nanomaterials that were discovered by Iijima in 1991 for the first time. CNTs have long cylindrical and axi-symmetric structures. CNTs are made by rolling graphene sheets. Because of their large length-to-diameter ratio, they are called nanotubes. CNTs are categorized as single-walled carbon nanotubes (SWCNTs) or multi-walled carbon nanotubes (MWCNTs) based on the shell structures. CNTs are broadly used in various fields, such as scanning probe microscopy, ultra fine nano balance and medicine, due to their extraordinary thermal conductivity, electrical and mechanical properties. Because long, straight CNTs have the same shape as asbestos, which cause cancer in cells lining the lung, there have been many studies on the effects of MWCNTs on human health that have been conducted. Stable atomization of CNTs is very important for the estimation of inhalation toxicity. In the present study, electro-static assisted axial atomizer (EAAA), which is the instrument that uses MWCNTs and aerosolizes them by transforming the single fiber shape using ultrasonic dispersion and electric field, was invented. EAAA consists of a ultrasonic bath for dispersion of MWCNTs and a particle generator for atomizing single fibers. The performance evaluation was conducted in order to assess the possibilities of 6-hour straight atomization with stability, which is the suggested exposure time in a day for the estimation of inhalation toxicity.

③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
