Identification and Functional Analysis of LsMNPV Anti-apoptosis Genes

  • Three anti-apoptosis genes, Ls-iap2, iap3 and p49 were found in Leucania separata multiple nuclear polyhedrovirus. Amino acid sequence homology of Ls-IAP2 and Ls-IAP3 with Op-IAP2 and Op-IAP3 from Orgyia pseddotsugata MNPV were 20% and 42%, while that of Ls-P49 is 28% with Sl-P49 from Spodoptera littorolis MNPV. Ls-IAP2 contains one baculoviral IAP repeat (BIR) domain followed by a RING domain, while Ls-IAP3 contains two BIRs and a RING. Ls-P49 contains a reactive site loop, predicted cleavage site (KKLD$^{74}{\downarrow}$G) that is different from Sl-P49 (TVID$^{94}{\downarrow}$G). Expressed Ls-iap3 or Ls-p49 under presence of actinomycin D in SF9 cells, DNA ladder assayrevealed that Ls- IAP3 or Ls-P49 could block the apoptosis of SF9 cells induced by actinomycin D. Replication of p35 deficient-mutant Autographa californica MNPV in SF9 cells was also rescued when Ls-iap3 or Ls-p49 was expressed transiently. No anti-apoptotic activity was observed for Ls-IAP2. The results showed that both of Ls-IAP3 and Ls-P49 were functional apoptotic suppressors in SF9 cells.

Helicobacter pylori 감염 진단 시 $^{14}C$-요소호기검사의 계수측정 방법에 따른 진단성능 비교 (Comparison of the Diagnostic Performance of $^{14}C$-urea Breath Test According to Counting Method for the Diagnosis of Helicobacter pylori Infection)

  • 목적: $^{14}C$-요소호기검사(UBT)는 Helicobacter pylori (H. pylori) 감염을 진단하는데 비침습적이고 유용한 방법으로 알려져 있다. 그러나 기존의 $^{14}C$-UBT는 액체 섬광계수기(LS)를 이용하여 측정하므로 검사과정이 복잡하고 시간이 많이 걸리는 단점이 있다. 저자들은 동일한 환자에서 최근에 개발되어 간편해진 $Geiger-M\ddot{u}ller$계수기(GM)를 이용한 측정과 기존의 LS를 이용한 측정을 시행하여 진단성능을 비교하여 GM을 이용한 방법이 기존의 LS를 이용한 방법을 대체할 수 있는지를 알아보고자 하였다. 대상 및 방법: 상부위장관 증상이 있어 일주일 이내에 위십이지장 내시경, LS와 GM을 이용한 $^{14}C$-UBT가 모두 시행된 49명(M:F=30:19, 나이 $41{\pm}14$ yrs)을 대상으로 하였다. 모든 환자에서 최소 6시간 금식을 한 후 37 KBq의 $^{14}C$-urea 캡슐을 복용한 후 LS 와 GM 측정법에 따른 호기 샘플을 얻어 각각 측정하였다. 위십이장 내시경시 시행된 조직 검사 결과를 금과옥조로 H. pylori 감염에 대한 두 계수측정 방법의 진단성능을 비교하였고, 두 측정법의 일치 정도와 측정값 사이의 상관관계를 알아보았다. 결과: 49명중 조직검사를 시행받은 사람은 18명이었으며 이중 H. pylori 감염은 13명(72.2%)에서 관찰되었고 LS와 GM 측정법은 모두 동일한 결과를 보여 민감도 100%, 특이도 100%, 정확도 100%를 보였다. 49명의 대상 환자 전체에서 시행된 두 계수측정법의 일치율은 98%(48/49)이었으며 불일치률은 2% (1/49)였다. LS와 GM에 의한 측정값은 통계학적으로 유의한 상관관계 (r=0.859, p<0.001)를 보였다. 결론: GM을 이용한 $^{14}C$-UBT는 LS 측정법과 동일한 진단성능을 보여, 보다 간편하고 빠른 검사법으로 LS측정법을 대체하여 임상적으로 유용하게 이용될 수 있을 것으로 생각된다.

담즙산염과의 고체분산체로부터 로바스타틴의 용출 및 십이지장 점막 투과 특성 (Dissolution and Duodenal Permeation Characteristics of Lovastatin from Bile Salt Solid Dispersions)

  • Although lovastatin (LS) is widely used in the treatment of hypercholesterolemia, its bioavailability is known to be around 5%. This study was aimed to increase the solubility and dissolution-permeation rates of LS using solid dispersions (SDs) with bile salts. The solubilities of LS in water, aqueous bile salt solutions and non-aqueous vehicles were determined, and effects of bile salts on the cellulose or duodenal permeation of LS from SDs were evaluated using a horizontal permeation system. SDs were prepared at various ratios of LS to carriers, such as sodium deoxycholate (SDC), sodium glycocholate (SGC) and/or 2-hydroxypropyl-$\beta$-cyclodextrin (HPCD). The addition of bile salts (25 mM) in water increased markedly the solubility of LS by the micellar solubilization. Some non-aqueous vehicles were effective in solubilizing LS. From differential scanning calorimetric studies, it was found that the crystallinity of LS in SDs disappeared, indicating a formation of amorphous state. The SDs showed markedly enhanced dissolution compared with those of their physical mixtures (PMs) and drug alone. In the dissolution-permeation studies using a cellulose membrane, the donor and receptor solutions were maintained as a sink condition using pH 7.0 phosphate buffer containing 0.05% sodium lauryl sulfate (SLS). The flux of LS alone was nearly same as that of LS-SDC-HPCD (1:3:6) PM. However, the flux of LS-SDC-HPCD (1:3:6) SD slightly increased compared with drug alone and PM, suggesting that entrapment of LS in micelles does not significantly hinder the permeation across cellulose membrane. In the dissolution-duodenal permeation studies using a LS-HPCD-SDC (1:3:6) SD, the addition of various bile salts in donor solutions (25 mM) enhanced the permeation of LS markedly, and the fluxes were found to be $0.69{\pm}0.41$, $0.87{\pm}0.51$, $0.84{\pm}0.46$, $0.47{\pm}0.17$ and $0.68{\pm}0.32{\mu}g/cm^2/hr$ for sodium cholate (SC), SDC, SGC, sodium taurodeoxycholate (STDC) and sodium taurocholate (STC), respectively. The stepwise increase of donor SGC concentration increased the flux dose-dependently. From the relationship of donor SGC concentration and flux, the concentration of SGC initiating the permeation across the duodenal mucosa was calculated to be 11.1 mM, which is nearly same as the critical micelle concentration (CMC, 11.6 mM) of SGC. However, with no addition of bile salts and below CMC, the permeation was very limited and irratic, indicating that LS itself is very poor permeable. Higher protions of bile salt in SD such as LS-SDC or LS-SGC (1 : 49 and 1 : 69) showed highly promoted fluxes. In conclusion, SD systems with bile salts, which may form their micelles in intestinal fluids, might be a promising means for providing enhanced dissolution and intestinal permeation of practically insoluble and non-absorbable LS.

  • To evaluate the algicidal impact of a newly developed algicide thiazolidinedione derivative, TD49, on dinophyceae Akashiwo sanguinea in aquatic ecosystems, tentative culture experiments for the target species were conducted in small (SS), middle (MS), and large scale (LS) culture vessels. When TD49 was introduced at the final concentration of $2{\mu}M$ in SS and MS, as well as $1{\mu}M$ in LS, the abundance of A. sanguinea decreased significantly in all the treatments. On the other hand, total phytoplankton abundance, except A. sanguinea in the TD49 treatments, gradually increased with culture time, which implies that a cell destruction of A. sanguinea by TD49 is a major cause of the population growth by other phytoplankton species. Also, A. sanguinea was easily destroyed, which was likely to be a source of extracellular substances. In particular, a pH decrease was significant in the treatments than in the control, which indicates that the water in the treatments has been acidified, due to an increase in the heterotrophic metabolisms of bacteria and degradation of A. sanguinea cells. Our results indicate that the TD49 substance is the potential agents for the control of A. sanguinea in the enclosed and eutrophic water bodies.

에틸렌이민 유도체를 갖는 아조계 분산염료의 염색성에 관한 연구(II) - 나일론 6.6 섬유에의 응용 - (A Study on Dyeing Property of Azo Disperse Dyes Containing Ethyleneimine Derivatives Part (II) -Application to Nylon 6.6 Fabrics-)

  • 이 연구의 목적은 에틸렌이민 유도체를 갖는 아조계 분산 염료의 나일론 6.6 섬유에의 응용과 최적화된 나일론 염색 조건의 확립이다. 두 쌍의 염료 A-1/A-2와 D-1/D-2의 최대흡수파장(&{\lambda}_{max}&)을 비교할 경우, 아세톤 최대흡수파장(λ$_{max}$)의 차이가 없거나 적어졌다. 세가지 아지리디닐 모노 아조 염료와 이들 염료의 가수분해 된 형태 그리고 디메닐아닐린계 아조 염료의 나일론 6.6 섬유에 대한 염색 및 견뢰도 성질이 조사되었다. 아지리디닐 염료의 나일론 섬유상의 최대 염착은 pH 5에서 얻어졌으며, 염색된 나일론 섬유에서의 용매추출의 경우 최소 염료 추출은 pH 8.0-10.0 에서 얻어졌다. 아지리디닐 염료 염색된 나일론 섬유상의 세탁 견뢰도와 광 견뢰도는 염색 시 pH의 증가에 따라 증가하였으나, pH 8에서의 견뢰도 성질이 pH 10에서와 비교하여 보다 개선되었다. 세가지 아지리디닐 염료가 이들 염료의 가수분해 된 형태 그리고 디메틸아닐린계 아조 염료와 비교해서 나일론 6.6 섬유상에서 보다 개선된 견뢰도를 나타내었고 이는 염료와 섬유간의 공유 결합으로 기인한 것으로 여겨진다.

베일러 작업 시 트랙터 소요동력 분석 (Analysis of Power Requirement of Agricultural Tractor during Baler Operation)

  • Purpose of this study was to analyze power requirement of an agricultural tractor for baler operation. First, a power measurement system was developed and installed in a 75 kW agricultural tractor. Strain-gages with a telemetry system were used to measure torques of transmission and PTO input shafts. An engine tachometer was used to measure rotational speed of transmission and PTO input shafts. The measurement system also included pressure sensors to measure pressure of hydraulic pumps, an I/O interface to acquire the sensor signals, and an embedded system to determine power requirements. Second, field experiments were conducted at two PTO speed levels, and proportion of utilization ratio of rated engine power and power consumption of major parts (transmission input shaft, PTO input shaft, main hydraulic pump, and auxiliary hydraulic pump) were analyzed. Results of usage proportion of engine power for PTO speed level 1 and 2 were 4.1 and 2.2%, 31.5 and 16.3%, 49.6 and 59.7%, 14.4 and 20.8%, and 0.4 and 1.0%, respectively, for ratio of measured engine power to rated engine power of less than 25%, 25 ~ 50%, 50 ~ 75%, 75 ~ 100%, and greater than 100%. The results showed that the usage proportion increased in the range with the ratio of power requirement to rated engine power of over than 50% when the PTO gear was shifted from P1 to P2. Averaged engine power requirement for baling operation, tying and discharging operation, and total operation were 43.3, 37.3, and 42.0 kW and 49.0, 37.0, and 47.4 kW, respectively, for PTO speed level 1 and 2. Paired t-test showed significant difference in power consumption of engine, transmission input shaft, and PTO input shaft for different PTO speed levels. Therefore, the power consumption of engine for baler operation increased when the PTO gear was shifted from P1 to P2. It was indicated that the power requirement of tractor was affected by the PTO rotational speed for baler operation.

다슬기로 제조한 칼슘 락테이트가 반죽의 발효와 빵의 품질 및 저장성에 미치는 영향 (Effect of Calcium Lactate Prepared from Black Snail on Dough Fermentation, Quality and Shelf-life of Bread)

  • 다슬기분말(PBS)과 그 회분(ABS)으로 제조한 칼슘락테이트(CaL-A및 CaL-P를 0.5%되게 첨가한 반죽의 발효와 빵의 품질 및 저장성에 미치는 영향을 조사하였다. 칼슘락테이트를 첨가하여 1차발효시킨 후 반죽의 pH는 5.98~5.86으로 대조군의 5.50보다 높았다. 반죽의 부피와 빵의 loaf volume index는 CaL-A 첨가군이 대조군 보다 현저하게 낮았으나 pH를 5.5로 조정한 후의 반죽부피와 빵의 loaf volume은 대조군과 비슷하였다. 처리군의 칼슘함량은 39.36~49.70 mg/100 g으로 대조군(13.43 mg/100 g)보다 현저하게 높았다. 처리군 빵의 색상은 대조군과 유의적인 차이가 없었다. 빵의 경도와 점착성은 CaL-A 첨가군이 대조군에 비해 높고, 탄력성과 응집성은 낮았으나 pH를 5.5로 조정한 경우는 유의적인 차이가 없었다. 처리군 빵의 현미경 관찰결과 대조군에 비하여 기공이 작고 air cell이 크며 표면이 거칠었다. 맛, 색상, 냄새 및 종합적인 기호도를 평가한 결과 대조군>CaL-A>CaL-P 순으로 첨가군의 관능적 품질이 낮았다. 그러나 pH를 5.5로 조정하여 제조한 빵에서는 종합적인 기호도가 크게 향상되었다. 칼슘락테이트를 첨가한 빵은 곰팡이 발생이 지연되었으며 곰팡이의 발생정도로 평가한 2$0^{\circ}C$에성의 저장수명은 대조군이 1, CaL-A 첨가군 3일, CaL-P 첨가군은 5일이었다.

암분 매립 농경지 토양의 표면 침투 및 삼투 특성 (Infiltration and Percolation Characteristics of Water in Agricultural Land Filled with Rock-Dust)

  • 이 연구는 Rock-dust로 채워진 농업 용지(Technosol)의 환경에서 물이 보이는 침투성과 삼투성의 특정을 파악하고자 수행되었다. 이 실험은 A와 B 두 장소에서 이루어졌으며 피실험지의 토양층은 각각 4가지 층으로 분류되었다. 모든 토양층의 토성은 미사질 양토로 분석되었으며 장소 A의 가장 낮은 층에서만은 양질사토였다. 두 토양에서, 각 토양층에 대한 용적밀도는 $1.49g{\cdot}cm^{-3}$을 대체로 상회하였으며, 이는 통상적인 medium textured mineral soil의 $1.25g{\cdot}cm^{-3}$을 크게 웃도는 값이다. 경운 직후 측정된 장소 A의 표토는 이보다 낮았다. 두 토양 표면에서의 $P_2O_5$의 집적은 각각 1962 (A), 1613 (B)$mg{\cdot}kg^{-1}$이었다. 이러한 집적은 3.2~6.5번에 걸쳐서 $300{\sim}500mg{\cdot}kg^{-1}$만큼 작물 생장에 알맞은 규모로 이루어졌다. 두 토양 표변에서의 Infiltration rate는 3.54 (A), 2.85 (B)$cm{\cdot}hr^{-1}$이었으나 각 토양층에 대한 percolation rate는 0.3 이하 (A), 0.003 이하 (B)$cm{\cdot}hr^{-1}$였다. 이러한 결과들은 표토가 동의 작용을 받은 반면 심토는 암분의 농업용지 형성 과정에서 생겨난 구조적인 문제인 바위 부스러기의 응집과 농업기계에 대한 다짐현상으로 인한 것임을 의미한다고 여겨진다.

간호사의 가정간호를 위한 교육요구 분석 (A study on educational need of nurses for home care)

  • This study was conducted from July to December 1990, in order to diagnose nurses' educational need for home care. The study subjects consisted of 145 nursing educators, and the 3 groups of nurses, namely 250 senior nursing students of diploma and collegiate program, 235 health center nurses, 521 university' hospital nurses in Seoul. Four types of questionaires were formulated by Delphi method. Two questionaires for the nursing educators were designed to measure their expectations of nurses' knowledge and of their skill for home care, and another two questionaires for the nurses to measure their actual home care knowledge and skill. The results of the study were as follows : 1) The mean scores of educators' expectation for home care knowledge were 17.68 for the care of dependence on medical equipment, 17.44 for the care of mobility impairment patient, 16.56 for the care of cardiopulmonary impairment patient, 16.40 for the care of nutrition and elimination impairment patient, '1.20 for the care of psychiatric disorder patient and 9.03 for the care of cancer and terminally ill patient,. 2) The mean scores of nurses' home care knowledge tested by 20 items were 14.36 for the care of mobility impairment patient, 13.28 for the c8;re of dependence on medical equipment, 13.78 for the care of cardiopulmonary impairment patient, 12.92 for the care of nutrition and elimination impairment patient, and those of tested by 10 items were 7.08 for the care of psychologic disorder patient, 7.80 for the care of cancer and terminally ill patient. The sum of means marked 69.23. As for the nurses' home care knowledge categorized by tasks in terms of the group, significant difference were shown in the care of mobility impairment(P=0.00), cancer and terminally ill(P=0.03), nutrition and elimination impairment(P=0.00) and psychologic disorder patient(P=0.00). No significant difference were shown in the care of dependence on medical equipment and cardiopulmonary impairment patient. 3) Regard to educational need of nurses' home care knowledge categorized by task according to the group it was found that all sampled nurses had educational need in the care of mobility impairment, dependence on medical equipment, cardiopulmonary impairment, cancer and terminally ill patient. It was found that health center nurses had educational need in the care of psychologic disorder. No educational need were found in the health center nurses whose career less than 2 years, in the care of mobility impairment, cardiopulmonary impairment and psychologic disorder patient, and in those of career with 2-5 year in the care of psychologic disorder patient. No educational need were found in the hospital nurses whose career more than 15 years, in the care of cardiopulmonary impairment patient and in those of career with 11-15 year, in the care of cancer and terminally ill patient. 4) The mean scores of educators' expectation for home care skill measured by Likert 5 points scale were 4. 21 for assessing, 4.49 for planning, 4.29 for basic care, 4.42 for curative care, 4.40 for rehabilitative care, 4.36 for emergency care, 4.53 for medication, 4.31 for nutritional care, 4.32 for other means for care, and 4.38 for evaluation. 5) Regard to nurses' home care skill measured by Likert 5 points scale of self evaluation, there was a significant difference between the nurses' home care skill and group(P=0.00l). The higher scores reported by students were vital sign checking and basic care while the scores of below medium were curative care and emergency care. The higher scores reported by health center nurses were vital sign checking, other means for care and care of specimen while the scores below medium were curative, emergency and nutritional care. The higher scores reported by hospital nurses were vital sign checking, care of specimen and basic care, while the score below medium was emergency care. 6) Regard to educational need of nurses' home care skill by nursing process activity according to the group it was found that health center nurses had educational need in all nursing skills including vital sign checking, care of specimen, health assessment, socioeconomic assessment, nursing diagnosis, care plan, basic care, curative care, rehabiitative care, psychological care, emergency care, medication, nutritional care, other means for care and evaluation. And students had educational need in all nursing skills except vital sign checking, and hospital nurses had educational need in all nursing skills except vital sign checking, care of specimen and basic care. 7) In short, the result of this study suggests that the curriculum should be organized in accordence with nurses' educational background and their career for the education of nurses for home care. It should be considered to develop the short term educational program focused on curative and rehabilitative care for health center nurse or community health nurse practitioner and which was focused on family care for hospital nurse. Concerning about this field practice for home care nurse, they are required not only community practice but also . clinical practice including emergency, curative and rehabilitative care.
