Production of Lignin Degrading Enzymes and Decolorization of Various Dye Compounds by Wood-Rot Fungi (목재 부후균의 리그닌 분해효소 활성과 염료 화합물의 탈색)

  • Wood-rot fungi produce extracellular lignin-degrading enzymes, the best known of which are lignin peroxidase, Mn-peroxidase and laccase. In this experiment, some of them produced all of three enzymes. Many other wood-rot fungi produced one or two of those enzymes with various combinations. In this experiment, we tried to clarify the relationship between the pattern of enzyme production and degradative activity of several dye compounds. From the 36 strains of 23 species of wood-rot fungi, Mn-peroxidase activity was found in 30 strains of the fungi tested, whereas the activity of lignin peroxidase and laccase was detected in 11 strains and 12 strains of species, repectively, in Kirks low nitrogen media. In relation to the activity of lignin degrading enzymes and degradation of dye compounds, the white-rot fungi with three kinds of enzymes tested showed the best dye decolorizers. The fungi with Mn-peroxidase activity only decolorized poly R-478 and remazol brilliant blue R dye in proportion to the enzyme activity, while methylene blue, bromophenol blue and congo red dye were degraded in regardless of enzyme activity. Those dyes were degraded in relation to the growth rate of mycelium. Brown-rot fungi did not degrade all the dye compounds except bromophenol blue, in spite of moderate growth rate.

Optimization of Cellulose Dyeing with Natural Indigo: Ramie dyeing by One-step Reduction/dyeing Process (천연인디고를 이용한 셀룰로오스계 직물 염색의 표준화 연구: 일단계 환원/염색에 의한 마직물 염색)

  • One-step reduction/dyeing method was applied for ramie dyeing with natural indigo powder. The effect of reduction/dyeing conditions including the pH of bath, dye temperature and time, and concentration of indigo powder and reduction agent on dye uptake and color properties were investigated. Regardless of addition of alkali, the dyed fabrics appeared in the PB Munsell color range(${\lambda}_{max}$: 660 nm) and the dye uptake was much higher with no addition of sodium hydroxide. Dyeing was carried out through the use of only sodium hydrosulfite in the bath. The maximum dye uptake was obtained at 60 for 30min. Saturated dye uptake was obtained at 2 g/L of sodium hydrosulfite concentration up to 2 g/ L of indigo powder. Whereas, at higher indigo powder concentration (4 g/L), more than 3 g/L of reducing agent concentration was required for obtaining the saturated dye uptake. Color reproducibility was reliable with color difference in the range of 0.03~0.16. Regardless of color strength, fastness to rubbing was acceptable with a 3/4~4/5. Fastness to washing, dry cleaning, and light of samples with low color strength were poor. Whereas, fastness to washing, dry cleaning, and light of samples with high color strength were very good.

A Study on the Dyeing Property of Polycarboxylic Acid Treated Cotton Fabrics with Chelidonium majus Extracts (애기똥풀 추출액에 대한 Polycarboxylic Acid 처리 면직물의 염색특성 연구)

  • The main colorant of Chelidonium majus extracts is a berberine which shows relatively good dyeability onto silk fabrics in the appropriate dyeing conditions without mordant, But cotton fabrics are difficult to dye with berberine because of the low substantivity the cationic to cellulosic fibers. we treated cotton fabrics using three types of polycarboxylic acid, DL-malic acid, citric acid, 1,2,3,4-butanetetracarboxylic acid(BTCA) to increase dyeability of Chelidonium majus extracts onto cotton fabrics. As a result the cotton fabrics treated with polycarboxylic acid could be dyed with Chelidonium majus extracts and also showed yellow vividly. The dye uptake were increased with increasing the number of carboxy groups. And so dye uptake resulted the largest in the BTCA solutions which have four carboxylic groups in the molecular unit. The optimal dyeing temperature a of Chelidonium majus extracts onto polycarboxylic acid treated cotton fabrics were $60^{\circ}C$. The dye equilibrium was reached 20 minutes after dyeing. Neutral pH of dye solutions showed in higher K/S value than acidic or alkaline conditions. But the colorfastness to washing and light according to polycarboxylic acid treat was not enhance.

  • Dye sensitized solar cell (DSSC) having high efficiency with low cost was first reported by Gr$\ddot{a}$tzel et al. Many DSSC research groups attempt to enhance energy conversion efficiency by modifying the dye, electrolyte, Pt-coated electrode, and $TiO_2$ films. However, there are still some problems against realization of high-sensitivity DSSC such as the recombination of injected electrons in conduction band and the limited adsorption of dye on $TiO_2$ surface. The surface of $TiO_2$ is very important for improving hydrophilic property and dye adsorption on its surface. In this paper, we report a very efficient method to improve the efficiency and stability of DSSC with nano-structured $TiO_2$. Atmospheric plasma system was utilized for nitrogen plasma treatment on nano-structured $TiO_2$ film. We confirmed that the efficiency of DSSC was significantly dependent on plasma power. Relative in the $TiO_2$ surface change and characteristics after plasma was investigated by various analysis methods. The structure of $TiO_2$ films was examined by X-ray diffraction (XRD). The morphology of $TiO_2$ films was observed using a field emission scanning electron microscope (FE-SEM). The surface elemental composition was determined using X-ray photoelectron spectroscopy (XPS). Each of plasma power differently affected conversion efficiency of DSSC with plasma-treated $TiO_2$ compared to untreated DSSC under AM 1.5 G spectral illumination of $100mWcm^{-2}$.

High-Fidelity Perforator Visualization for Cadaver Dissection in Surgical Training

  • In the first half of the third century B.C., Herophilus and Erasistratus performed the first systematic dissection of the human body. For subsequent centuries, these cadaveric dissections were key to the advancement of anatomical knowledge and surgical techniques. To this day, despite various instructional methods, cadaver dissection remained the best way for surgical training. To improve the quality of education and research through cadaveric dissection, our institution has developed a unique method of perforator-preserving cadaver injection, allowing us to achieve high-fidelity perforator visualization for dissection studies, at low cost and high efficacy. Ten full body cadavers were sectioned through the base of neck, bilateral shoulder, and hip joints. The key was to dissect multiple perfusing arteries and draining veins for each section, to increase "capture" of vascular territories. The vessels were carefully flushed, insufflated, and then filled with latex dye. Our injection dye comprised of liquid latex, formalin, and acrylic paint in the ratio of 1:2:1. Different endpoints were used to assess adequacy of injection, such as reconstitution of eyeball volume, skin turgor, visible dye in subcutaneous veins, and seepage of dye through stab incisions in digital pulps. Dissections demonstrated the effectiveness of the dye, outlining even the small osseous perforators of the medial femoral condyle flap and subconjunctival plexuses. Our technique emphasized atraumatic preparation, recreation of luminal space through insufflation, and finally careful injection of latex dye with adequate curing. This has allowed high-fidelity perforator visualization for dissection studies.

  • Using the new type auxiliary(K-1), dyeing experiment was done at the various dyeing conditions and the effects of auxiliary on the dyeing properties of acid milling dye for the silk fiber were dicussed. The results obtained were as follows; 1. The exhaust rate of acid milling dye was increased according to the auxiliary concentration at dyebath pH 7, But the increasing tendency of exhaust rate was decreased above auxiliary concentration 3% o. w. f.. 2. It was the same tendency of exhaust rate at dyebath pH 5, but the diffusion was more powerful at dyebath pH 7 than pH 5. 3. The decreasing ratio of exhaust rate according to dye concentration increasing was lower at auxiliary addition than auxiliary non-addition. 4. The exhaust rate curve according to increasing dyeing temperature showed the inflection point at auxiliary non-addition, but it was not observed in auxiliary addition. Consequently, it seems to be avoid that the occurrence of uneven dyeing at auxiliary addition.

Enhancement of Dye-Sensitized Solar Cell Efficiency by Spherical Voids in Nanocrystalline ZnO Electrodes

  • Light scattering enhancement is widely used to enhance the optical absorption efficiency of dye-sensitized solar cells. In this work, we systematically analyzed the effects of spherical voids distributed as light-scattering centers in photoanode films made of an assembly of zinc oxide nanoparticles. Spherical voids in electrode films were formed using a sacrificial template of polystyrene (PS) spheres. The diameter and volume concentration of these spheres was varied to optimize the efficiency of dye-sensitized solar cells. The effects of film thickness on this efficiency was also examined. Electrochemical impedance spectroscopy was performed to study electron transport in the electrodes. The highest power conversion efficiency of 4.07 % was observed with $12{\mu}m$ film thickness. This relatively low optimum thickness of the electrode film is due to the enhanced light absorption caused by the light scattering centers of voids distributed in the film.

Development of a Method to Quantify Lysine in Small Amount of Rice Grain

  • A lysine determination method for low quantity of rice was modified from the original Dye-Binding Lysine (DBL) method used in the national standard in China [GB 4801-84, 1984]. By making use of the property that lysine does not bind to the crocein orange G dye after treated with propionic anhydride, the amount of lysine in rice samples could be determined directly by calculating the difference between the absorbances of the treated and the untreated samples. Various commercial rice samples were purchased from market and evaluated. Several methods were tested by varying both the sizes of the samples and the concentrations of the dye solutions. Results showed that when using 1.284 mM of crocein orange G dye solution and 15.5 mg of sample, the results were most reproducible. The corresponding lysine content in sample were $3.36\;{\pm}\;0.09\;mg/g$ and $3.35\;{\pm}\;0.19\;mg/g$ by traditional method and modified method, respectively. Statistically, there was no significant difference between the results (p>0.05).

A Study on the Dyeability of the Aminized Cotton Fabrics (아민기를 도입한 면직물의 염색성에 관한 연구)

  • Cotton fabric was treated with acrylonitrile in t-butyl alcohol and then aminized by reduction of the resultant cyanoethyl cellulose with LiAIH4, under various temperatures and times. Aminized cotton fabric toras dyed with acid and reactive dye. Dyeability as to temperatures, pH, and color fastness were compared to the amiRe group content of treated cotton fabrics. The results of this study were as follow: 1. D.S of cyanoethyl cellulose has been increased by increasing treating temperatures and times. Maximum D.S of cyanoethyl cellulose was 1.45. By SEM and the tensile strength, the damage of the treated cotton was not observed. 2. Since aminized cotton has greater affinity on acid dye than untreated cotton, dyeability increased with the increase of amino group content, and the lower pH, $60^{\circ}C$. 3. Since amino group in aminized cotton changes surface charge of cotton, dyeability for reactive dye increased by increasing the amino group content. Optimum dyeability was obtained at pH 5-7, $40^{\circ}C$. And dyeability of the amine treated cotton increased with the NaCl conc. 4. Color fastness to washing & light of the amine treated cotton was very low except for the wash fastness of the reactive dye.

  • Up sizing of dye-sensitized solar cell(DSC) is the important technology to bring about commercialization of DSC. Several studies to obtain a stable large area DSC have been investigated in overseas laboratories, but have been hardly done in our country. In this study, up sizing technology of dye sensitized solar cells(DSCs) was investigated. We investigated low dark current materials for the current collecting grid. From the result, a new DSC module with metal grid was designed, and fabricated. For a new interconnection, both working and counter electrodes are alternately coupled on 10[cm]$\times$7[cm] substrate. We have achieved 68% of fill factor and photoelectric conversion efficiency of around 2.6% as the best results of new designed DSC structure.
