Enhancement of wear resistance of components used in fast reactors is necessary for long service life of the components. Plasma nitriding is a promising surface modification technology to impart high hardness and improved wear resistance of various steel components. This study discusses the characterization of chrome nitrided SS316L casing ring used in secondary sodium pump of fast breeder reactor and its stability under long term sodium exposure. Microstructural and hardness analysis showed that stress relieved component could be chrome nitrided successfully to a thickness of about 100 ㎛. Assessment of in-sodium performance of the chrome nitrided casing ring subjected to long term exposure up to 5000h at 550℃, showed retention of chrome nitrided layer with a case depth almost similar to that before sodium exposure. A slight decrease in the hardness was observed due to prolonged high temperature sodium exposure. Tribological studies indicate very low coefficient of friction indicating the retention of good wear resistance of the coating even after long term sodium exposure.
We have previously shown that paraquat (PQ)-induced oxidative stress causes dramatic damage in various human cell lines. Naringenin (NG) is an active flavanone, which has been reported to have beneficial bioactivities, including antioxidative, anti-inflammatory, and antitumorigenic activities, with a relatively low toxicity to normal cells. In this study, we intended to assess the cytoprotective effect of NG against PQ-induced toxicity in the human bronchial epithelial BEAS-2B cell line. Co-treatment with NG in PQ-treated BEAS-2B cells can reduce PQ-induced cellular toxicity. NG can also decrease the generation of intracellular ROS caused by PQ treatment. We also observed that treatment with NG in PQ-exposed BEAS-2B cells can significantly induce the expression of antioxidant-related genes, including GPX2, GPX3, GPX5, and GPX7. NG co-treatment can also activate the NRF2 transcription factor and promote its nuclear translocation. In addition, NG co-treatment can induce the expression of NRF2-downstream target genes such as that of heme oxygenase-1 (HO-1) and NAD(P)H:quinone oxidoreductase 1 (NQO1). A small interfering RNA study revealed that the knockdown of NRF2 can abrogate NG-mediated protection of the cells from PQ-induced cellular toxicity. We propose that NG effectively alleviates PQ-induced cytotoxicity in human bronchial epithelial BEAS-2B cells through the NRF2-regulated antioxidant defense pathway, and NG might be a good therapeutic candidate molecule in oxidative stress-related diseases.
Objectives: Accumulation of cellular reactive oxygen species (ROS) leads to oxidative stress. Increased production of ROS, such as superoxide anion, or a deficiency in their clearance by antioxidant defences, mediates cellular pathology. Grewia Spp fruits are a source of bioactive compounds and have notable antioxidant activity. Although the antioxidant capacity of Grewia Spp has been studied, there is very limited evidence that links the antioxidant activities of Grewia bicolor and Grewia flava to the inhibition of free radical formation associated with damage in biological systems. Methods: This study evaluated the protective effects of Grewia bicolor and Grewia flava extracts against free radical-induced oxidative stress and the resulting cytotoxicity effect using HeLa cells. Antioxidant properties determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and total phenolic content (TPC) assays showed significantly higher (p < 0.05) antioxidant activity in Grewia flava (ethanol extract) than Grewia flava (water extract) and Grewia bicolor (ethanol and water extracts). Results: Using 3-(4,5-dimethylthiazol-2-yl)-2,5diphenyltetrazolium bromide or MTT assay, cytotoxicity results showed that extracts of Grewia bicolor and Grewia flava were less toxic to HeLa cells at tested concentrations compared to the untreated control. This confirmed the low toxicity of these edible fruits at the tested concentrations in HeLa cells. Furthermore, hydrogen peroxide (H2O2)-induced cell loss was effectively reduced by pre-incubating HeLa cells with Grewia bicolor and Grewia flava extracts, with Grewia flava (ethanol extract) revealing better protection. Conclusion: The effect was speculated to be associated with the higher antioxidant activity of Grewia flava (ethanol extract). Additional studies will warrant confirmation of the mechanism of action of such effects.
In order to investigate the effects of copper alloy on abalone physiology, we studied survival rate, respiration, excretion rate, and heavy metal accumulation in each organ of adults and spats. The survival rate of spats and adults showed 27-60% and 63-83% respectively, higher survival rate in adults. In particular, 100% of copper panel led to lowest survival rate and there was no sharp distinction according to copper alloy composition. The respiration rate and excretion rate of ammonia nitrogen was $1.81mgO_2/g$ D.W./h and 0.43 mg $NH_4-N/g$ D.W./h respectively at 100% of copper panel. In other words, there was a high significant difference at the level, but no significant difference at other test levels (P < 0.05). The atomic ratio (0: N) hit the lowest at the 100% of copper panel showing 3.79 and no significant differences were seen among other test groups with 6.57-7.18 of a very low range. This means that the species might have undergone nutritional stress. In case of copper accumulation, the 100% copper panel group showed the highest level in hepatopancreas and muscle showing 6.91 mg/kg and 1.60 mg/kg respectively but the rest of groups showed similar levels. Zinc accumulation raised at Cu-Zn alloy panel had high significance showing 18.50 mg/kg and 1.10 mg/kg in hepatopancreas and muscle respectively (P < 0.05). To sum up, a cage net made of 100% pure copper is expected to have a negative effect on abalone in light of survival rate, heavy metal accumulation, and atomic ratio (0: N). Moreover, given that the substratum used for the high adhesive species and nutritious stress that is represented through the atomic ratio need to be considered, the copper alloy net is thought not to be suitable for abalone aquaculture.
Objectives : The level of 4-hydroxyproline (4-Hyp) in human urine was measured using high performance liquid chromatography (HPLC) with a fluorescence detector. This method is useful for medical examinations and investigating the radicals induced by physical, chemical, mental stresses. This method is superior to many published several methods in terms of its low cost and ability to analyze many samples. Methods : The urine from workers in a tire manufacturing company (22 male pre- and post-shift workers) and 18 office-workers as controls were analyzed. Data concerning age, the cumulative drinking amount and the cumulative smoking amount was collected with a questionnaire. The optimum applied amount of dansyl-Cl, the optimum reaction temperature and time, the recoveries and the optimum pH of the eluent and buffer were determined.4-Hyp from human urine was derivatized with dansyl-Cl (dimethylamino-naphthalene-1-sulfonyl chloride) after removing the a-amino acid by a treatment with phthalic dicarboxaldehyde (OPA) and cleaned with Bond Elut C18 column. The 4-Hyp derivatives were separated on a reversed phase column by gradient elution with a phosphate buffer (5 mmol, pH 8.0) and acetonitrile, and detected by fluorescence measurements at 340 nm (excitation) and 538 nm (emission). Results : The detection limit for the urinary free 4-Hyp was $0.364{\mu}mol/l$. The recovery rate of 4-Hyp was 99.7%, and the effective pH of the phosphate buffer and borate buffer were 3.0 and 8.0, respectively. From statistical analysis, age, drinking and smoking did not affect the urinary free 4-Hyp in both the controls and workers. The range of urinary 4-Hyp in the controls, pre-shift, and post-shift workers were 0.33-16.44, N.D-49.06, and $0.32-56.27{\mu}mol/l$. From the pared-sample t-test, the urinary 4-Hyp levels in post-shift workers ($11.82{\pm}6.73\;nmmol/mg\;Cre$) were 2-fold higher than in pre-shift workers ($5.36{\pm}5.53\;nmol;/mg\;Cre$) and controls ($4.91{\pm}4.89\;nmol;/mg\;Cre$). Conclusions : This method was developed with high sensitivity, accuracy, and precision. The present method was effectively applied to analyze the urinary free 4-Hyp in both controls and workers.
Appropriate types and concentrations of starch-based fat replacers which are low-pH stable, shear resistant and heat stable were selected to develop reduced-fat mayonnaise without affecting taste, texture and emulsion stability. Four kinds of commercial fat replacers [Neutral DR (DR), Stellar 100X (SX), Sta Slim 150 (SS) and N-Lite L (NL)] were thermally activated after appropriately hydrated and applied to mayonnaise formulation. For each fat replacer, eight recipes having various ratios of soybean oil/fat replacer(95/5 to 60/40) were formulated and compared with standard formulation(100% soybean oil) using rheological and sensory evaluations. All tested starch pastes showed pseudoplastic behavior, time-dependent thixotropic gel characteristics. The increasing order of pseudoplasticity and yield stress was RM, NL, SS and ST. The hardness and adhesiveness of each fat replacer-added mayonnaise was increased in the same order as above. Formulated mayonnaise which contained higher ratio of fat replacer showed lower values in hardness and adhesiveness. ST at up to 25% oil replacement exhibited the highest oil replacing ability among four replacers. ST-added mayonnaise also exhibited the highest sensory score and emulsion stability among all products including standard formula. The high sensory score in spreadability of RM-added mayonnaise well reflected the consistent index of RM paste. Use of fat replacers reduced oily odor and greasy aftertaste of mayonnaise.
This experiment was conducted to investigate the effects of stocking density on performance and blood characteristics of broiler chicken in summer. One day-old 648 male commercial broiler chicks(Ross strain) were divided into 3 groups with 4 replicates(12 pens), 41 to 69 birds per pen, to compare the stocking density difference on growth performance and blood characteristics. Birds in T1, T2 and T3 were reared in different size 0.050, 0.066, 0.083 $m^2$/bird floor, respectively. The birds were fed the same experimental diet ad libitum for 5 weeks. Chickens were weekly weighed and one bird was selected from each pen to measure blood characteristics at the end of experiment. Body weight and feed intake increased significantly in low density treatment(T3) compare to that of high density treatment(P<0.05). Feed conversion was significantly improved in customary treatment(T2) relative to that of other treatments(P<0.05). No significant difference was observed in moisture contents of bedding among treatments. In blood characteristics, low density treatment(T3) decreased the total protein, cholesterol and albumin, but were no significant difference. From the result of this study, it could be concluded that the higher stocking density might give more stress to the birds from physical stimuli and thus were harmful environment causing lower productions compared to the lower ones
The aim of this study was to identify the effects of dithiopyr (DTP), a herbicide, on behavior in zebrafish. The toxicity of DTP has rarely been investigated in fish. In the present study, zebrafish were exposed to different concentrations of DTP in the range of 10-20 μM for 48 h in a test container, in order to measure the value of median lethal concentrations (LC50). Behavioral experiments were performed, including the novel tank test (NTT) and the open field test (OFT), to assess stress responses or locomotion. After exposure to the DTP solution at a sublethal concentration of 2.5–10 μM for 6 min, the behavior of the zebrafish was observed for 6 min. In the acute toxicity test, the LC50 value of DTP showed as 14.49 μM in the zebrafish. The NTT showed that the duration of immobility and the velocity were significantly increased by exposure at a concentration of 5 μM of DTP, compared with a control group (p<0.05). However, compared with the control group, DTP significantly decreased the distance moved and the frequency at the top of the tank, and significantly increased the turn angle and duration at the bottom, in a concentration-dependent manner (p<0.05). In addition, in the OFT, exposure to DTP significantly decreased the distance moved and velocity compared with the control group (p<0.05). Exposure to DTP also significantly increased the duration of immobility, the turn angle, and the meandering movement, in a concentration-dependent manner (p<0.05). Further, exposure to DTP at a low concentration elevated whole-body cortisol levels in the zebrafish. The results of this study thus suggest that DTP induces a toxic response and negative effects on behavior and the endocrine system in zebrafish.

Objectives: Based on data from Chinese and Indian traditional herbal medicines, gum resin of Ferula assa-foetida (sometimes referred to asafetida or asafoetida) has several therapeutic applications. The authors of various studies have claimed that asafetida has cytotoxic, antiulcer, anti-neoplasm, anti-cancer, and anti-oxidative effects. In present study, the anti-aging effect of asafetida on senescent human dermal fibroblasts was evaluated. Methods: Senescence was induced in in vitro cultured human dermal fibroblasts (HDFs) through exposure to $H_2O_2$, and the incidence of senescence was recognized by using cytochemical staining for the activity of ${\beta}$-galactosidase. Then, treatment with oleo gum resin of asafetida was started to evaluate its rejuvenating effect. The survival rate of fibroblasts was evaluated by using methyl tetrazolium bromide (MTT) assays. Real-time quantitative reverse transcription-polymerase chain reaction (qRT-PCR) and western blot assays were performed to evaluate the expressions of apoptotic and anti-apoptotic markers. Results: Our experiments show that asafetida in concentrations ranging from $5{\times}10^{-8}$ to $10^{-7}g/mL$ has revitalizing effects on senescent fibroblasts and significantly reduces the ${\beta}$-galactosidase activity in these cells (P < 0.05). Likewise, treatment at these concentrations increases the proliferation rate of normal fibroblasts (P < 0.05). However, at concentrations higher than $5{\times}10^{-7}g/mL$, asafetida is toxic for cells and induces cell death. Conclusion: The results of this study indicate that asafetida at low concentrations has a rejuvenating effect on senescent fibroblasts whereas at higher concentrations, it has the opposite effect of facilitating cellular apoptosis and death.
The effect of N forms ($NO_3{^-}-N$, $NH_4{^+}-N$) and concentrations (4, 8, $16meq/{\ell}$) which were changed at head formation stage on the tip-burn incidence of chinese cabbage was studied under the three levels (0, 8, 16 meq/l) of Ca. All of the plants grown on $NH_4{^+}-N$ showed the symptoms of tip-burn and low yields regardless of Ca levels. Roots of plants grown on $NH_4{^+}-N$ were severely damaged. The pH of the leachate from $NH_4{^+}-N$ pot was decreased to below pH 5. Plants which had been grown on $NH_4{^+}-N$ before the head formation stage, but changed to $NO_3{^-}-N$ were recovered from abnormal growth. But, the reverse showed the tip-burn symptoms. $NH_4{^+}-N$ treatments increased the T-N contents, but lowered K and Ca contents of inner leafblades. Icreased applications of Ca did not affect the T-N and K contents of the inner leafblades. $NH_4{^+}-N$ suppressed Ca translocation into the inner leaves from outer leaves after the head formation stage, but $NO_3{^-}-N$ accelarated it. Ca contents were much lower in leaves showing tip-burn symptoms than in healthy leaves. Internal rot which is a tip-burn symptom occuring after head formation were noted in plants applied with high concentration of $NO_3{^-}-N$ both before and after the head formation stage. Ca contents correlated well with the rates of Ca application, but there was no correlation between ca conents and internal rot incidence. Chinese cabbage tip-burn is not caused by Ca deficiency, as is commonly believed, but rather by the water stress (95% water contents in $NO_3{^-}$-fed plants, 91% in $NH_4{^+}$-fed plants) resulting from root damage caused by ammonium toxicity. Internal rot is also caused by excess applications of nitrogen, and is unaffected by calcium levels.
