Promoting Effect of Hydrogen Peroxide on 1-Methyl-4-phenylpyridinium-induced Mitochondrial Dysfunction and Cell Death in PC12 Cells

  • The promoting effect of hydrogen peroxide ($H_2O_2$) against the cytotoxicity of 1-methyl-4-phenylpyridinium ($MPP^+$) in differentiated PC12 cells was assessed by measuring the effect on the mitochondrial membrane permeability. Treatment of PC12 cells with $MPP^+$ resulted in the nuclear damage, decrease in the mitochondrial transmembrane potential, cytosolic accumulation of cytochrome c, activation of caspase-3, increase in the formation of reactive oxygen species (ROS) and depletion of GSH. Addition of $H_2O_2$ enhanced the $MPP^+-induced$ nuclear damage and cell death. Catalase, Carboxy-PTIO, Mn-TBAP, N-acetylcysteine, cyclosporin A and trifluoperazine inhibited the cytotoxic effect of $MPP^+$ in the presence of $H_2O_2$. Addition of $H_2O_2$ promoted the change in the mitochondrial membrane permeability, ROS formation and decrease in GSH contents due to $MPP^+$ in PC12 cells. The results show that the $H_2O_2$ treatment promotes the cytotoxicity of $MPP^+$ against PC12 cells. $H_2O_2$ may enhance the $MPP^+$-induced viability loss in PC12 cells by promoting the mitochondrial membrane permeability change, release of cytochrome c and subsequent activation of caspase-3, which is associated with the increased formation of ROS and depletion of GSH. The findings suggest that $H_2O_2$ as a promoting agent for the formation of mitochondrial permeability transition may enhance the neuronal cell injury caused by neurotoxins.

Dietary Ascorbate Supplementation Reduces Oxidative Tissue Damage and Expression of iNOS in the Kidney of Streptozotocin Induced Diabetic Rats

  • Reactive oxygen species (ROS) have been suggested to be contributory factors in complications of diabetes mellitus. In the present study, we investigated the generation of superoxide, the lipid peroxide level measured as thiobarbituric acid reactive substances, the vasorelaxation of isolated thoracic aorta and the iNOS expression in kidney of streptozotocin induced diabetic rats. Sprague Dawley rats were divided into four groups: control, ascorbate (400 mg/kg rat weight daily in drinking water), diabetic (single dose of 50 mg of STZ/kg i.p.) and diabetic simultaneously fed with ascorbate for 12 wk. Rats in groups were studied at tri-weekly intervals (0 to 12 wk). Diabetic rats were evaluated periodically with changes of plasma glucose levels and body weight. The ascorbate supplimentation attenuated the development of hyperglycemia and weight loss induced by STZ injection in rats. In the present experimental condition, the ascorbate supplimentation had no significant effect on plasma glucose levels and changes in body weight of normal rate. The superoxide generation, formation of thiobarbituric acid reactive substance and iNOS expression in kidney were significantly increased in STZ-treated rats that were decreased by ascorbate supplimentation. The ascorbate supplimentation had no effect on vasorelaxation of isolated thoracic aorta. These results indicate that ascorbate supplimentation may exert an inhibitory effect on STZ-induced oxidative tissue damage through protection of pancreatic islet cells by scavanging reactive oxygen species. The ascorbate supplimentation may possibly attenuate the renal complication of diabetes mellitus.

Evaluation of Hydrophobic Performance and Durability of Concrete Coated with Cellulose Nanofiber Mixed Antifouling Coating Agent (셀룰로오스 나노섬유 혼합 방오코팅제가 도포된 콘크리트의 소수성능과 내구성능 평가)

  • Marine and hydraulic structures are subject to durability damage not only due to the penetration of sea water but also due to the attachment of marine organisms. Therefore, in this study, we tried to develop an antifouling coating agent with self-cleaning function for marine concrete. It was confirmed that the antifouling coating agent mixed with AKD, cellulose nanofibers and BADGE had sufficient antifouling performance at a well hydrophobicity of around 140° in contact angle and an inclination angle of 15°. In the abrasion resistance test of the surface, only a maximum loss of 0.015 g occurred. In the durability test, as a result of the chloride ion permeation test, almost no chloride ion permeation occurred in the variable where the coating agent was applied, and carbonation and freeze-thaw damage also rarely occurred, so it was analyzed that it was effective in securing durability of concrete.

The Effects of Mannyeon-hwan on Acetic Acid-induced Ulcerative Colitis in Rats (흰쥐에서 acetic acid로 유발된 궤양성 대장염에 대한 만년환의 효과)

  • Objectives: This study was designed to verify the effects of Mannyeon-hwan (MNH) on acetic acid-induced ulcerative colitis in rats. Methods: Ulcerative colitis was induced in male Sprague-Dawley rats weighing approximately 250 g by injecting acetic acid through the anus. Rats were classified into four groups: normal group, control group (acetic-acid, AA), low concentration group (AA+MNH(L)), and high concentration group (AA+MNH(H)). Body weight, visual evaluation of the colonic mucosa, anatomical histological changes, and changes in the expression of proteins in colon tissue were compared and analyzed. Results: Compared to the control group, the MNH groups showed significant recovery from weight loss and mucosal damage. The expression of inflammatory proteins such as TNF-α, IL-6, IL-1β, NF-κB p65, MPO, and COX-2 were significantly decreased in the MNH groups compared to those in the AA group. In the MNH groups, significant changes in proteins involved in apoptosis such as caspase-3, BAX, and Bcl-2 were observed compared to those in the AA group. Additionally, changes in the expression of TNF-α, IL-6, IL-1β, NF-κB p65, BAX, and Bcl-2 were greater in the MNH(H) group than those in the MNH(L) group. Conclusion: Mannyeon-hwan was found to be effective in suppressing inflammation of the colonic mucosa in ulcerative colitis, inhibiting the expression of inflammation-related proteins, and suppressing damage to the colonic mucosa by regulating the expression of apoptosis-related proteins.

Analysis for Properties of Particle or Short Fiber Reinforced Composites based on Micromechanics under Pure Shear (전단응력하의 분산형 복합재료에 미시역학적인 특성평가)

  • Cracking of the reinforcements is a significant damage mode in particle or short-fiber reinforced composites because the broken reinforcements lose load carrying capacity. This paper deals with elastic stress distributions and load carrying capacity of intact and cracked ellipsoidal inhomogeneities. Three dimensional finite element analysis has been carried out on intact and broken ellipsoidal inhomogeneities in all infinite body under pure shear. For the intact inhomogeneity, the stress distribution is uniform in the inhomogeneity and non-uniform in the surrounding matrix. On the other hand, for the broken inhomogeneity, the stress in the region near crack surface is considerably released and the stress distribution becomes more complex. The average stress in the inhomogeneity represents its load carrying capacity, and the difference of average stresses between the intact and broken inhomogeneities indicates the loss of load carrying capacity due to cracking damage. The broken inhomogeneity with higher aspect ratio maintains higher load carrying capacity.

Antimicrobial efficacy of endophytic Penicillium purpurogenum ED76 against clinical pathogens and its possible mode of action

  • This study was aimed to evaluate the antimicrobial activity of Penicillium purpurogenum ED76 on several clinically important microorganisms. The endophytic fungus P. purpurogenum ED76 was previously isolated from Swietenia macrophylla leaf. The antimicrobial efficacy of P. purpurogenum ED76 dichloromethane extract was determined via disc diffusion and broth microdilution assay. A kill curve study was conducted and the morphology of extract treated bacterial cells were viewed under scanning electron microscope. The dichloromethane extract showed significant inhibitory activity on 4 test bacteria and 2 test yeasts. The minimal inhibitory concentration of the extract ranged from 125 to $1,000{\mu}g/ml$, which indicates the different susceptibility levels of the test microorganisms to the fungal extract. The kill curve study has revealed a concentration-dependent inhibition for all test microorganisms. With the increase of the extract concentration, the microbial growth was significantly reduced. The scanning electron micrograph of dichloromethane extract-treated Staphylococcus aureus cells showed the total damage of the cells. The cell wall invagination of the bacterial cells also indicates the loss of cellular materials and metabolic activity. The gas chromatography mass spectrometry analysis of the extract also showed that the major compound was stigmasterol, which constitutes 45.30% of the total area. The dichloromethane extract of P. purpurogenum ED76 exhibited significant inhibitory activity on several clinically important bacteria and yeasts. The study proposed a possible mode of action that the extract cause significant damage to the morphology of S. aureus cells.

Effects of Bacterial Leaf Blight Occurrence on Rice Yield and Grain Quality in Different Rice Growth Stage (벼 생육시기별 흰잎마름병 발병이 쌀 수량감소 및 미질에 미치는 영향)

  • We investigated the first symptom emerging date of rice bacterial leaf blight disease during four years from 2002 to 2005. The disease occurrence date was earlier 20$\sim$30 days in 2005 than that of 2002. The damage in different rice growth stage by the bacterial disease on rice yield and grain quality in southern part of Korea was examined. The disease decreased rice yield following by increased infection rates. Slight loss in rice yield and brown head rice rate were observed at below 10% level of infected leaf area, while over 25% infected leaf area caused the significant decrease in rice yield and brown head rice rate. More than 50% of the infected leaf area rate caused 29% yield reduction in case of infection at panicle formation stage and 18% of brown head rice rate. Cooked rice quality was also affected in the diseased rice by damage in palatability score and viscosity.

Corrosive Injury Due to Edible Vinegar (식이 식초 음독 후 발생한 부식성 손상)

  • Vinegar is a very popular ingredient used in many cuisines. It is also known for its beneficial health, beauty and possible weight-loss properties. The authors report on a patient who presented to the emergency department with unstable vital signs complaining of generalized abdominal pain after ingestion of 450 ml of apple cider vinegar. We documented a case of corrosive gastrointestinal injury with persistent metabolic acidosis occurring after ingesting apple cider vinegar with an acetic acid concentration of 12~14%. Toxic damage to the liver and kidney were also observed, peaking on post-ingestion day 3. The patient received supportive care and hemoperfusion for three days without much clinical improvement and died in the seventh day of intensive care due to disseminated intravascular coagulation and multi organ failure. Edible vinegar, when taken in large amounts, is capable of inducing corrosive injuries of the GI tract as well as severe systemic toxicities, such as metabolic acidosis. Safety precautions regarding vinegar deserve more public attention and clinicians also should be astute enough to recognize the potential damage accompanying vinegar ingestion.

Two-Part Tax for Polluting Oligopolists with Endogenous Entry (내생적 시장진입 구조에서 오염배출 과점기업에 대한 이부 환경세)

  • This paper constructs the two-part tax-a combined form of output tax and entrance fee-for polluting oligopolists under endogenous entry. In the presence of external damage that varies exogenously with aggregate output, we show that the two-part tax produces the ex post Pigouvian rule and thus achieves the first-best optimum. We also examine a detailed analysis of the impact of the two-part tax on social welfare and government revenues. Finally, when estimation errors exist in the process of regulation, we identify the incentive conflicts between interest groups and analyze the effects of estimation errors on determining optimal tax. In particular, we show that if the regulator takes care of both welfare loss and revenue gain under the proposed two-part tax, not only over-estimation on the slope of external damage but also under-estimation on the slope of market demand should be taken into the policy consideration.

Fatigue Damage Evaluation of Cr-Mo Steel with In-Situ Ultrasonic Surface Wave Assessment (초음파 시험에 의한 배관용 Cr-Mo강의 피로손상의 비파괴평가)

  • Although the ultrasonic method has been developed and used widely in the fields, it has been used only for measuring the defect size and thickness loss. In this study, the relationship between surface wave attenuation through micro-crack growth and variation of velocity under repeated cyclic loading has been investigated. The specimens are adopted from 2.25Cr-1Mo steel, which is used for power plant and pipeline system, and have dimensions of $200{\times}40{\times}4mm$. The results of ultrasonic test with a 5MHz transducer show that surface wave velocity gradually decreases from the point of 60% of fatigue life and the crack length of 2mm with the increasing fatigue cycles. From the results of this study, it is found that the technique using the ultrasonic velocity change is one of very useful methods to evaluate the fatigue life nondestructively.
