Combination Effects of 7β-Hydroxycholesterol and Radiation in Human Lung Cancer Cells

  • The present study was performed to evaluate combination effect of 7$\beta$-hydroxycholesterol (7$\beta$-OHC) and $\gamma$-radiation in NCI-H460 human lung cancer cells. 7$\beta$-OHC in combination with $\gamma$-irradiation has an enhanced effect in decreasing clonogenic survival and increasing cellular DNA damage. Pretreatment of cells with 7$\beta$-OHC enhanced radiation-induced apoptosis. Apoptosis of the cells by combined treatment of 7$\beta$-OHC and $\gamma$-irradiation was associated with reactive oxygen species generation and loss of mitochondrial membrane potential, resulting in the activation of caspase 9 and caspase 3. The combined treatment also resulted in an increased G1 cell cycle distribution. These results indicate that 7$\beta$-OHC shows the additive effect of radiation sensitivity in human lung carcinoma cells in vitro.

Cilostazol attenuates kainic acid-induced hippocampal cell death

  • Cilostazol is a selective inhibitor of type 3 phosphodiesterase (PDE3) and has been widely used as an antiplatelet agent. Cilostazol mediates this activity through effects on the cyclic adenosine monophosphate (cAMP) signaling cascade. Recently, it has attracted attention as a neuroprotective agent. However, little is known about cilostazol's effect on excitotoxicity induced neuronal cell death. Therefore, this study evaluated the neuroprotective effect of cilostazol treatment against hippocampal neuronal damage in a mouse model of kainic acid (KA)-induced neuronal loss. Cilostazol pretreatment reduced KA-induced seizure scores and hippocampal neuron death. In addition, cilostazol pretreatment increased cAMP response element-binding protein (CREB) phosphorylation and decreased neuroinflammation. These observations suggest that cilostazol may have beneficial therapeutic effects on seizure activity and other neurological diseases associated with excitotoxicity.

Prediction and Verification of Lateral Joining Strength for Tapered-Hole Clinching using the Taguchi Method (다구찌 기법을 이용한 이종재료 경사 홀 클린칭 접합부 수평 방향 접합강도 예측 및 검증)

  • Fiber metal laminates (FMLs) are well known for improved fatigue strength, better impact resistance, superior damage tolerance and slow crack growth rate compared to traditional metallic materials. However, defects and loss of strength of a composite material can occur due to the vertical load from the punch during the joining with a dissimilar material using a conventional clinching method. In the current study, tapered-hole clinching was an alternative process used to join Al 5052 and FMLs. The tapered hole was formed in the FML before the joining. For the better understanding of static and dynamic characteristics, a clinched joining followed by a tensile-shear test was numerically simulated using the finite element analysis. The design parameters were also evaluated for the geometry of the tapered hole by the Taguchi method in order to improve and compare the lateral joining strength of the clinched joint. The influence of the neck thickness and the undercut were evaluated and the contribution of each design parameter was determined. Then, actual experiments for the joining and tensile-shear test were conducted to verify the results of the numerical simulations. In conclusion, the appropriate combination of the design parameters can improve the joining strength and the cross-sections of the tapered-hole clinched joint formed in the actual experiments were in good agreement with the results of the numerical simulations.

Performance Evaluation and Design of an Edible Fresh Corn Harvesting Machine (식용 풋옥수수 수확 시험장치 설계 및 성능평가)

  • In this study, an edible fresh corn harvest testing machine was designed and manufactured. And harvesting performance was analyzed through the field test. The testing machine is of the tractor attached type. It is connected to the tractor PTO shaft to transfer power to the each part of the harvesting machine. And it harvests fresh corn by one row through the processes of cutting, stem crushing, detaching, and collecting. The performance test was performed at PTO speed (540, 750, 1050 rpm, respectively), working speed (0.1, 0.15, 0.2 m/s, respectively), and cropping cultivation (row spacing·hill spacing 70·25 cm, 70·40 cm, 90·30 cm, respectively). The performance test was repeated three times in the 15 m section. The detachment loss ratio, uncollected crop ratio, damage ratio, and harvest ratio were analyzed. As a result of the performance test, it was analyzed that the PTO speed 540 rpm, running speed of 0.1 m/s, and row spacing·hill spacing 70·40 cm were the optimal condition.

A Study on Dynamic Analysis of Moored Ship Motions by Tsunami (쓰나미에 의한 계류 선박의 동적 동요 해석에 관한 연구)

  • Recent warnings indicate that there is a potential risk of massive earthquake. These earthquakes could produce large-scale tsunamis. Consideration of the effect of Tsunami to the moored ship is very important bemuse it brings the loss of life and vast property damage in a viewpoint of ship operations within a harbor. If a tsunami occurs, a ship in a harbor may begin drifting in case of ship entering and departing harbor, and breakage of mooring rope and drifting of moored ship are happened. And extremely serious accident, such as stranding and collision to a quay, might occur. On the other hand, since the tsunami consists of approximately component waves of several minutes, there is a possibility of resonance with the long period motion of mooring vessel. As the speed of Tsunami is much faster than tidal current in a harbor, a strong resisting force might act on the moored ships. In this paper, the numerical simulation procedure in the matter of ship motions due to the attack of large-scale tsunamis are investigated and the effects on the ship motions and mooring loads are evaluated by numerical simulation.

Strategies for Conservation and Restoration of Freshwater Fish Species in Korea (한국 담수어류의 보존 및 복원 전략)

  • The tiny fragment of freshwater body is providing home for huge biodiversity and resources for the existence of human. The competing demand for freshwater have been increased rapidly and it caused the declination of biodiversity in recent decades. Unlike the natural process of extinction in gradual progress, the current species extinction is accelerated by human activity. As a result many fish species are already extinct or alive only in captivity in the world and about fifty eight animal species are in endangered in Korea including eighteen freshwater species. Conservation of biodiversity is the process by which the prevention of loss or damage is attained, and is often associated with management of the natural environment. The practical action is classified into in-situ, or ex-situ depending on the location of the conservation effort. Recovery means the process by which the status of endangerment is improved to persist in the wild by re-introduction of species from ex-situ conservation population into nature or translocation of some population. However there are a lot of restrictions to complete it and successful results are known very rare in case. In this article the authors explore some strategies for conservation and restoration of freshwater fish species conducted in Korea for few years. The major causes are discussed in relation with the decline of freshwater fish diversity during few decades and some strategies are evaluated to advance the process of conservation. A study on the Korean bullhead, Pseudobagrus brevicorpus, is introduced as a case for ex-situ conservation and restoration in freshwater ecosystem.

Car-tire-related Crushing Injury of the Lower Leg in Children (자동차 바퀴에 의한 소아 아래다리의 압궤 손상)

  • Purpose: Crushing injuries by car tires result from a combination of friction, shearing, and compression forces and the severity of injury is influenced by the acceleration. Because car-tire injuries of the lower leg in children are common these days but they have received little attention; thus, our purpose was to look closely into this problem. Methods: A retrospective analysis was conducted of data from children under 15 years old age who visited an emergency department because of a car-tire-related crushing injury to the lower leg in pedestrian traffic accident from January 2008 to September 2012. The patient's age, sex, site of injury, degree of injury, associated injuries, type of surgery, and complications were reviewed. Results: There were 39 children, the mean age was 8.0 years, and 71.8% were boys. The dorsal part of the leg was involved most frequently. According to the severity classification, 15 children were grade I, 6 were grade II, and 18 were grade III. Among 24 patients, 13 were treated with skin graft and 3 were treated using a sural flap. Twelve patients developed complications, such as hypertrophic scarring, contractures, and deformities with significant bone loss. Conclusion: Various degrees of skin or soft tissue defects were caused in children by car tires. In this study, patients were often also had tendon or bone damage. Proper and timely initial treatments are needed to reduce the incidence of infection, the number of operative procedures, and the hospital stay.

Lysate of Probiotic Lactobacillus plantarum K8 Modulate the Mucosal Inflammatory System in Dextran Sulfate Sodium-induced Colitic Rats

  • Inflammatory bowel disease (IBD) is caused by dysregulation of colon mucosal immunity and mucosal epithelial barrier function. Recent studies have reported that lipoteichoic acid (LTA) from Lactobacillus plantarum K8 reduces excessive production of pro-inflammatory cytokine. In this study, we investigated the preventive effects of lysate of Lb. plantarum K8 in dextran sulfate sodium (DSS)-induced colitis. Male Sprague-Dawley rats were orally pretreated with lysate of Lb. plantarum K8 (low dose or high dose) or live Lb. plantarum K8 prior to the induction of colitis using 4% DSS. Disease progression was monitored by assessment of disease activity index (DAI). Histological changes of colonic tissues were evaluated by hematoxylin and eosin (HE) staining. Tumor necrosis factor-alpha (TNF-${\alpha}$), interleukin-6 (IL-6) levels were measured using enzyme-linked immunosorbent assay (ELISA). The colon mRNA expressions of TNF-${\alpha}$, IL-6, and toll like receptor-2 (TLR-2) were examined by quantitative real-time-transcription polymerase chain reaction (qPCR). Lysate of Lb. plantarum K8 suppressed colon shortening, edema, mucosal damage, and the loss of DSS-induced crypts. The groups that received lysate of Lb. plantarum K8 exhibited significantly decreased levels of the pro-inflammatory cytokines TNF-${\alpha}$ and IL-6 in the colon. Interestingly, colonic expression of toll like receptor-2 mRNA in the high-dose lysate of Lb. plantarum K8 group increased significantly. Our study demonstrates the protective effects of oral lysate of Lb. plantarum K8 administration on DSS-induced colitis via the modulation of pro-inflammatory mediators of the mucosal immune system.

  • The power of semiconductor, Korea is continuously constructing semiconductor production line for keeping a front-runner status. however, studies and data about potential risks in semiconductor factory are still short. If fire does not initially suppressed, the fire causes a great damage. To decrease fire risk factors, in addition to fire fighting safety equipment, more important thing is how to design and construct fire protection system. The current fire protection codes about semiconductor factory come under functional law, and this law is short of consideration about particularity of factory. The existing prescriptive fire codes depending on experience compose without evident engineering verifications, thus equipments which is created by the current prescriptive fire code may bring about a variety of problems. For example, the design under the current regulation can not cope with the excessive investments, low efficiencies, and the diversifying construction designs and be applied to the quick changes of new technologies. Ergo, an optimal design for fire protection is to equip fire protection arrangements with condition and environment of production field. Manufacturing factory of semiconductors is a windowless airtight space. And for cleanliness, there exists strong flow of cooperation. Therefore, there is a need for fire safety design that meets the characteristic of a clean room. Accordingly, we are to derive smoke flow according to cooperation process within a clean room and construction plan of an optimal sensor system. In this study, in order to confirm the performance of proposed smoke-exhaust equipment and suggest efficient smoke exhaust device when there is a fire of 1MW of methane in the clean room of company H, we have implemented fire simulation using fluid dynamics computation.

Study of heavy fuel oil fly ash for use in concrete blocks and asphalt concrete mixes

  • Use of heavy fuel fly ash (HFFA) (diesel and cracked fuel) for power generation in Saudi Arabia has generated and accumulated large quantities of HFFA as a byproduct. In this research, HFFA is studied with the emphasis on the utilization of this waste material in concrete blocks and asphalt concrete mixes. Two types of mixes, one with low and other with high cement content, were studied for concrete blocks. Different mixes having varying percentages of HFFA (0% to 25%), as cement/sand replacement or as an additive, were studied. The performance of concrete blocks is evaluated in terms of compressive strength, water absorption, durability and environmental concerns. The results showed that blocks cannot be cast if more than 15% HFFA is used; also there is a marginal reduction in the strength of all the mixes before and after being exposed to the sulfate solution for a period of ten months. HFFA is studied in asphalt concrete mixes in two ways, as an asphalt modifier (3&5%) and as a filler (50%) replacement, the results showed an improvement in stiffness and fatigue life of mixes. However, the stability and indirect tensile strength loss were found to be high as compared to the control mix due to moisture damage, indicating a need of using antistripping agents. On environmental concerns, it was found that most of the concerned elements are within acceptable limits also it is observed that lower concentration of barium is leached out with the higher HFFA concentrations, which indicates that HFFA may work as an adsorbent for this leaching element.
