제주도산 한우와 제주흑한우 집단의 도체형질에 대한 Lysophospholipase I (LYPLA1) 유전자형의 효과 (Genotypes of Lysophospholipase I (LYPLA1) Gene Associate with Carcass Traits of the Jeju Crossbred Cattle (Hanwoo × Jeju Black cattle) and Hanwoo Populations)

  • 본 연구는 Lysophospholipase I (LYPLA1) 유전자의 SNP 마커인 rs385360448 g.23655332G>C의 유전자형이 제주도 한우와 제주흑한우(한우${\times}$제주흑우, JCC) 집단의 도체형질에 미치는 영향을 시험하였다. 한우 거세우 집단에서 LYPLA1 rs385360448 G/-를 보유한 도체는 C/C 동형접합에 비해 유의적으로 높은 수준의 근내지방도(MARB)와 조직감지수(TI)를 나타내었다(p<0.05). 반면, LYPLA1 유전자형은 도체중, 등지방두께, 등심단면적, 육색, 지방색에서는 유의적인 차이를 보이지 않았다(p>0.05). JCC 거세우에서는 LYPLA1 G/- 도체에서 C/C 동형접합에 비해 등심단면적(EMA)의 수준이 유의적으로 더 높았다(p<0.05). 이번 연구의 결과들은 LYPLA1 유전자형이 소의 등심에서 인지질 대사를 통해 근내지방도와 조직감의 수준과 등심단면적의 수준을 변화시킬 수 있음을 보여주고 있다. 이러한 결과들은 LYPLA1 유전자형이 한우와 제주흑우-유래의 산업화축군에서 도체형질을 개선할 수 있는 유전자 마커로써의 기능을 수행할 수 있을 것으로 기대된다.

Effects of feeding high-energy diet on growth performance, blood parameters, and carcass traits in Hanwoo steers

  • Objective: Our study aimed to investigate the effects of a 2% increase in dietary total digestible nutrients (TDN) value during the growing (7 to 12 mo of age) and fattening (13 to 30 mo of age) period of Hanwoo steers. Methods: Two hundred and twenty Hanwoo steers were assigned to one of two treatments: i) a control group (basal TDN, BTDN, n = 111 steers, growing = 70.5%, early fattening = 71.0%, late fattening = 74.0%) or high TDN (HTDN, n = 109 steers, growing = 72.6%, early = 73.1%, late = 76.2%). Growth performance, carcass traits, blood parameters, and gene expression of longissimus dorsi (LD) (7, 18, and 30 mo) were quantified. Results: Steers on the BTDN diets had increased (p≤0.02) DMI throughout the feeding trial compared to HTDN, but gain did not differ appreciably. A greater proportion of cattle in HTDN received Korean quality grade 1 (82%) or greater compared to BTDN (77%), while HTDN had a greater yield grade (29%) than BTDN (20%). Redness (a*) of LD muscle was improved (p = 0.021) in steers fed HTDN. Feeding the HTDN diet did not alter blood parameters. Steers fed HTDN diet increased (p = 0.015) the proportion of stearic acid and tended to alter linoleic acid. Overall, saturated, unsaturated, monounsaturated, and polyunsaturated fatty acids of LD muscle were not impacted by the HTDN treatment. A treatment by age interaction was noted for mRNA expression of myosin heavy chain (MHC) IIA, IIX, and stearoyl CoA desaturase (SCD) (p≤0.026). No treatment effect was detected on gene expression from LD muscle biopsies at 7, 18, and 30 mo of age; however, an age effect was detected for all variables measured (p≤0.001). Conclusion: Our results indicated that feeding HTDN diet could improve overall quality grade while minimum effects were noted in gene expression, blood parameters, and growing performance. Cattle performance prediction in the feedlot is a critical decision-making tool for optimal planning of cattle fattening and these data provide both benchmark physiological parameters and growth performance measures for Hanwoo cattle feeding enterprises.

청보리 사일리지 TMR 또는 청호밀 사일리지 TME 급여 및 급여기간이 거세 한우의 증체, 사료비 및 육질특성에 미치는 효과 (Effect of Feeding Whole Crop Barley Silage- or Whole Crop Rye Silage based-TMR and Duration of TMR Feeding on Growth, Feed Cost and Meat Characteristics of Hanwoo Steers)

  • 본 연구는 주요 동계사료작물인 청보리와 청호밀을 이용한 TMR 급여가 한우 거세우의 생산성과 사료비, 그리고 도체특성과 등급판정 및 배최장근의 이화학적 특성에 미치는 효과를 조사하기 위해 한우 거세우 송아지 80두를 대상으로 육성기에서 비육후기까지 총 680일에 걸쳐 실시되었다. 시험사료는 배합사료와 볏짚을 분리 급여하는 관행사양구(Control), 청보리 사일리지 TMR을 비육 중기까지 자유채식토록 하고 비육 후기에는 관행사양구와 동일한 사료를 급여하는 처리구(BS-TMR I), 청보리 사일리지 TMR을 비육 후기까지 자유채식토록 하는 처리구(BS-TMR II), 청호밀 사일리지 TMR을 비육 중기까지 자유채식토록 하고 비육 후기에는 관행 사양구와 동일한 사료를 급여하는 처리구(RS-TMR I), 청호밀 사일리지 TMR을 비육 후기까지 자유채식토록 하는 처리구(RS-TMR II)로 구성하였다. 각 처리 당 거세된 송아지 16두 씩을 배치하였고, 처리 당 거세한우 송아지를 4두씩 4개의 pen에 수용하였으며, 완전임의배치 방법으로 각 처리구의 pen을 배치하였다. 한우 거세우의 증체량은 관행사양구에 비해 육성기 및 비육전기에서 청보리 및 청호밀 사일리지 TMR을 섭취한 처리구에서 다소 높았고 비육 중기에서 현저히 높았으나(P<0.0001) 비육 후기에서는 RS-TMR II를 제외하고는 관행사양구의 한우에서 다소 높은 것으로 나타났다. 한우 두당 kg 증체 당 사료비는 관행사양구의 한우에 비해 청보리 및 청호밀 사일리지 TMR을 섭취한 한우에서 다소 낮았다. 또한 비육 후기까지 사일리지 TMR을 섭취한 처리구(BS-TMR II 및 RS-TMR II)에 비하여 비육 중기까지 사일리지TMR을 섭취하고 비육 후기에는 관행사료를 섭취한 처리구(BS-TMRI 및 RS-TMR I)에서 두당 kg 증체 당 사료비가 다소 감소 되었다. 사일리지 TMR이 관행사료 급여에 비해 한우 거세우의 도체중, 등지방 두께 및 배최장근 면적을 다소 증가시켰으나 육량지수를 낮추었다(P<0.047). 다른 시험사료에 비해 청보리 사일리지 TMR이 근내지방도를 다소 낮추었으나 육질 1등급 이상 출현두수에는 처리 간 차이가 없었다. 한우 거세우 내 배최장근의 화학적성분, 도체특성, 대부분의 육질 특성, 육색 및 지방산 조성은 시험사료에 의한 영향을 받지 않았다. 그러나 전단력은 사일리지 TMR을 섭취한 한우에 비해 관행사양 방법으로 비육된 한우에서 낮았으나(P<0.046) 사일리지 TMR을 섭취한 처리구 간에는 차이가 없었다. 본 시험의 결과로 미루어 보아 배합사료와 볏짚 중심의 관행사양방법에 비하여 BS 또는 RS를 이용한 TMR 급여가 거세한우의 육질에 크게 영향하지 않은 반면 증체를 개선하고 사료비를 절감할수 있을 것이라 여겨진다. 또한 전체적으로는 청보리 및 청호밀 사일리지 TMR의 사료적 가치는 거의 비슷한 것으로 여겨진다.

Identification of Differentially Expressed Genes Related to Intramuscular Fat Development in the Early and Late Fattening Stages of Hanwoo Steers

  • Marbling of cattle meat is dependent on the coordinated expression of multiple genes. Cattle dramatically increase their intramuscular fat content in the longissimus dorsi muscle between 12 and 27 months of age. We used the annealing control primer (ACP)-differential display RT-PCR method to identify differentially expressed genes (DEGs) that may participate in the development of intramuscular fat between early (12 months old) and late fattening stages (27 months old). Using 20 arbitrary ACP primers, we identified and sequenced 14 DEGs. BLAST searches revealed that expression of the MDH, PI4-K, ferritin, ICER, NID-2, WDNMI, telethonin, filamin, and desmin (DES) genes increased while that of GAPD, COP VII, ACTA1, CamK II, and nebulin decreased during the late fattening stage. The results of functional categorization using the Gene Ontology database for 14 known genes indicated that MDH, GAPD, and COP VII are involved in metabolic pathways such as glycolysis and the TCA cycle, whereas telethonin, filamin, nebulin, desmin, and ACTA1 contribute to the muscle contractile apparatus, and PI4-K, CamK II, and ICER have roles in signal transduction pathways regulated by growth factor or hormones. The final three genes, NID-2, WDNMI, and ferritin, are involved in iron transport and extracellular protein inhibition. The expression patterns were confirmed for seven genes (MDH, PI4-K, ferritin, ICER, nebulin, WDNMI, and telethonin) using real-time PCR. We found that the novel transcription repressor ICER gene was highly expressed in the late fattening stage and during bovine preadipocyte differentiation. This information may be helpful in selecting candidate genes that participate in intramuscular fat development in cattle.

Genetic, management, and nutritional factors affecting intramuscular fat deposition in beef cattle - A review

  • Intramuscular fat (IMF) content in skeletal muscle including the longissimus dorsi muscle (LM), also known as marbling fat, is one of the most important factors determining beef quality in several countries including Korea, Japan, Australia, and the United States. Genetics and breed, management, and nutrition affect IMF deposition. Japanese Black cattle breed has the highest IMF content in the world, and Korean cattle (also called Hanwoo) the second highest. Here, we review results of research on genetic factors (breed and sex differences and heritability) that affect IMF deposition. Cattle management factors are also important for IMF deposition. Castration of bulls increases IMF deposition in most cattle breeds. The effects of several management factors, including weaning age, castration, slaughter weight and age, and environmental conditions on IMF deposition are also reviewed. Nutritional factors, including fat metabolism, digestion and absorption of feed, glucose/starch availability, and vitamin A, D, and C levels are important for IMF deposition. Manipulating IMF deposition through developmental programming via metabolic imprinting is a recently proposed nutritional method to change potential IMF deposition during the fetal and neonatal periods in rodents and domestic animals. Application of fetal nutritional programming to increase IMF deposition of progeny in later life is reviewed. The coordination of several factors affects IMF deposition. Thus, a combination of several strategies may be needed to manipulate IMF deposition, depending on the consumer's beef preference. In particular, stage-specific feeding programs with concentrate-based diets developed by Japan and Korea are described in this article.

Isolation, Expression Pattern, Polymorphism and Association Analysis of Porcine TIAF1 Gene

  • TIAF1 is a TGF-${\beta}$1-induced anti-apoptotic factor that plays a critical role in blocking TNF (tumor necrosis factor) cytotoxicity in mouse fibroblasts and participates in TGF-${\beta}$-mediated growth regulation. In this study, we obtained the full-length cDNA sequence of the porcine TIAF1 gene. Real-time PCR further revealed that the TIAF1 gene was expressed at the highest level in liver and kidney with prominent expressions detected in uterus, and lower levels detected in heart, spleen, lung, stomach, small intestine, skeletal muscle and fat of Large White pigs. Sequence analysis indicated that a 6 base-pair deletion mutation existed in the exon of the TIAF1 gene between Meishan and Large White pigs. This mutation induced deletion of Gln and Val amino acids. PCR-RFLP was used to detect the polymorphism in 394 pigs of a "Large White${\times}$Meishan" $F_{2}$ resource population and four purebred pig populations. The frequencies of the A allele (with a 6 bp deletion) were dominant in Chinese Meishan and Bamei pigs, and the frequencies of the B allele (no 6 bp deletion) were dominant in Large White and Landrace pigs. Association analyses revealed that the deletion mutation had highly significant associations (p<0.01) with meat marbling score of the thorax-waist longissimus dorsi (LD) muscle (MM1) and intramuscular fat percentage (IMF), and significant associations (p<0.05) with carcass length (CL). The results presented here supply evidence that the 6 bp deletion mutation in the TIAF1 gene affects porcine meat quality and provides useful information for further porcine breeding.

Variation of Meat Quality Parameters Due to Conformation and Fat Class in Limousin Bulls Slaughtered at 25 to 27 Months of Age

  • The aim of the present study was to determine the influence of age of animal, hot carcass weight, pH, conformation and fat class on basic beef quality attributes (tenderness, sarcomere length, basic chemical composition, marbling and colour) in a group of purebred animals. The object of the study was beef of Limousin bulls (25 to 27 months, hot carcass weight - $432{\pm}31$ kg, "U" conformation class, "2"-"3" fat class). Analysed cuts were Infraspinatus muscle from the blade and Longissimus dorsi muscle from the cube roll and the striploin. Tenderness was analysed with universal testing machine, colour - chromometer analysis, sarcomere length - microscopic method, basic chemical composition - near-infrared spectroscopy and marbling - computer image analysis. No differences in tenderness and sarcomere length were observed within the age groups of Limousin bulls (age of 25, 26, 27 months) (p>0.05). Moisture (p = 0.0123) and fat (p = 0.0250) content were significantly different for meat of animals slaughtered at the age of 25 and 27 months. No influence of pH value on tenderness was observed, but at the same time, influence on sarcomere length (p = 0.039) and $b^*$ component of colour (0.045) was found. For "U" conformation class, in subclasses, as well as for fat classes "2"-"3", there were no differences in tenderness, sarcomere length and colour components. Higher fat content was observed in the higher fat class, rather than in lower, however this feature was not associated with marbling.

Assessment of frozen storage duration effect on quality characteristics of various horse muscles

  • Objective: The study aimed at assessing the effects of frozen storage duration on quality characteristics, lipid oxidation and sensory quality of various horse muscles. Methods: Five representative muscles: longissimus dorsi (LD), gluteus medius (GM), semimembranosus (SM), biceps femoris (BF), and triceps brachii (TB) at 24 h post-mortem obtained from 28-mo-old Jeju female breed horses (n = 8) were used in the present investigation. The muscles were vacuumpackaged and frozen at $-20^{\circ}C$ for 120, 240, and 360 days. All the samples were analyzed for thawing and cooking losses, pH, Warner-Bratzler shear forces (WBSF), color traits, total volatile basic nitrogen (TVBN), thiobarbituric acid reactive substances (TBARS) and sensory traits. The muscle samples analyzed on day 0 of frozen storage (fresh, non-frozen) were used for comparison. Results: Results revealed that thawing and cooking losses significantly (p<0.05) increased in all the muscles after 120 days and then remained unchanged up to 360 days of frozen storage. The TBARS and TVBN contents significantly increased as increasing frozen storage time up to 360 days (p<0.05). While, significant decreases in WBSF values were observed for all the muscles with increased frozen storage time (p<0.05). Frozen storage variously affected the color traits of the muscles for instance; the redness of LD, GM, and BF muscles showed a decreasing tendency during frozen storage while it was not changed in TB and SM muscles. Furthermore, the frozen storage did not produce detrimental effects on sensory quality as it did not cause flavor and juiciness defects whereas it partially improved the tenderness of all the muscles studied. Conclusion: Based on the results obtained from our work, it is concluded that frozen storage could be applied to increase the long-term shelf life of horsemeat while still retaining its sensory quality.

Growth performance, carcass and meat quality of lambs supplemented different vegetable oils

  • Objective: The purpose of this study was to evaluate the effect of rapeseed and linseed oil supplementations on performance and meat quality of lambs. Methods: The experiment was conducted on 18 growing (100-day-old) lambs of $19.7{\pm}1.9kg$ live weight, assigned to 3 groups of 6 animals each. Control lambs were fed meadow hay and concentrate alone. Experimental animals additionally received rapeseed or linseed oils at a dose of 50 g/d. The lambs were slaughtered at an average body weight of $35.7{\pm}0.5kg$. Results: The dressing percentage was higher in lambs fed rapeseed oil. Total saturated fatty acids (SFA) and C15:0, C16:0, C17:0, C21:0, C24:0 were lower in longissimus dorsi muscle (MLD) in lambs fed linseed oil. Supplementation of diet with linseed oil decreased concentrations of total monounsaturated fatty acids and C16:1, C17:1, C18:1 cis-9 in MLD. The concentrations of n-3 polyunsaturated fatty acids (PUFA) and C18:3 n-3, C20:5 n-3 in MLD were higher in lambs fed linseed oil than in other groups. Oils supplementation to diets resulted in increased concentration of C22:6 n-3 in MLD. The inclusion of linseed oil into the diet increased the contents of total PUFA, n-3 PUFA and C18:3 n-3, C20:5 n-3, C22:6 n-3 in semitendinosus muscle in comparison to control. A tendency towards a lower n:6/n:3 ratio in MLD was observed when lambs were supplemented linseed oil. Conclusion: The supplementation of linseed oil to diets seems to reduce the concentration of SFA and increase the concentration of n-3 PUFA. The n-6/n-3 ratio is an important nutritional factor, and its value has been favorably decreased below 2, thereby achieving an important target related to human health. Due to these changes carcass fatty acid profile was improved, and so enhanced lamb meat healthy properties.

The effect of adding ethanolic saffron petal extract and vitamin E on growth performance, blood metabolites and antioxidant status in Baluchi male lambs

  • Objective: This study investigated the effects of the administration of ethanolic saffron petal extract (SPE) and vitamin E (Vit E) on growth performance, blood metabolites and anti-oxidant status in Baluchi lambs. Methods: Thirty-two Baluchi male lambs ($35.22{\pm}5.75kg$) were randomly divided into 4 groups. The 1st (control), 2nd (injectable saffron petal extract [ISPE]), and 3rd (Vit E) groups were respectively injected subcutaneously with either physiological saline (5 mL), SPE (25 mg/kg body weight [BW]) or DL-${\alpha}$-tocopheryl acetate (225 IU) once a week. An oral dose of SPE (500 mg/kg BW) was also administered to the 4th group (oral saffron petal extract [OSPE]). Feed intake and BW were measured for 42 days and blood samples were taken on days 1, 14, 28, and 42. The lambs were slaughtered, and tissue samples were taken. Results: Growth performance and many blood metabolites were not affected (p>0.05) by the treatments. Cholesterol of plasma in the ISPE and Vit E groups was similar and less (p<0.01) than both the OSPE and control groups. Although there was no significant difference between the control and other groups for plasma triglyceride, the ISPE group showed lower (p<0.05) triglyceride than the OSPE and Vit E groups. The highest (p<0.01) plasma glutathione peroxidase (GPx) was detected in the OSPE group, while the ISPE and Vit E groups showed higher (p<0.01) superoxide dismutase (SOD) of plasma than the control. Malondialdehyde of plasma in the ISPE group was lower (p<0.05) than the OSPE. No differences (p>0.05) were observed among the groups for antioxidant status of both longissimus dorsi muscle and liver. However, the activity of GPx in the kidney and heart, as well as SOD activity in the kidney, were influenced ($p{\leq}0.01$) by the treatments. Conclusion: Adding ethanolic SPE improved antioxidant status and lowered lipids oxidation in lambs. The SPE and Vit E demonstrated similar effects on antioxidant status in lambs.
