Optimization of Extraction Conditions of Antioxidant Activity and Bioactive Compounds from Rice Bran by Response Surface Methodology (반응표면분석법을 이용한 미강으로부터 항산화 활성 및 생리활성물질의 초음파 추출조건 최적화)

  • The rice's waste byproduct is known as a rice bran and produced annually about 400,000 to 600,000 tons. Most of the rice bran are used as a livestock feed or waste disposal, and needed to be used to produce high-added substances, such as bioactive materials. In this study, extraction conditions of the ultrasound-assisted extraction (UAE) of the rice bran were optimized using a statistically-based optimization. The influence of extraction variables including the extraction time ($X_1$), extraction temperature ($X_2$) and ethanol concentration ($X_3$) were investigated using the response surface methodology in order to determine optimum extraction conditions which maximize total phenolic compounds (TPC), total flavonoid compounds (TFC) and electron donating abilities (EDA). The optimal UAE from rice bran was achieved under the extraction temperature of $94.9^{\circ}C$, extraction time of 41.6 minute and ethanol concentration of 74.0% (v/v) with maximum yields of TPC 2.78 mg GAE/g DM, TFC 1.63 mg QE/g DM and EDA 42.86%. The UAE process shows its potential to the extraction of bioactive and antioxidant compounds from rice bran in a short extraction time and low temperature. Also, it is proposed that rice bran could be considered as food additives and cosmeceutical products.

Characteristics of the spatio-temporal distributions of water quality and phytoplankton communities in the Isa Stream systems (ISS) (이사천 수계의 수질환경과 식물플랑크톤 군집의 시·공간적 분포 특성)

  • We analyzed the spatio-temporal distribution characteristics of water quality and phytoplankton communities in the Isa Stream systems (ISS) from Sangsa Lake to Suncheon Bay. Sangsa Lake showed relatively oligotrophic and mesotrophic conditions, but the freshwater and mixed brackish water zones showed more severe eutrophication than Sangsa Lake and Suncheon Bay due to the influence of industrial waste such as livestock waste. In terms of the phytoplankton community, the number of phytoplankton species was higher in freshwater and mixed brackish water zones than in Sangsa Lake and Suncheon Bay, but the cell density and Chlorophyll-a concentrations (Chl-a) were relatively high in Sanga Lake and Suncheon Bay. In particular, the mesotrophic species Fragilaria crotonensis and Asterionella formosa showed different dominance in the surface and bottom layers, and the influence of A. formosa was significant in the freshwater and mixed brackish water zones in spring and summer. However, Skeletonema costatum-ls, a eutrophic indicator species, dominated in mixed brackish water zones to seawater in autumn and winter. Thus, the severe eutrophication and rapid environmental changes in the ISS could seriously damage the coastal ecosystem in Suncheon Bay. These ecosystem changes are threatening in terms of conservation and management of the UNESCO Suncheon Biosphere Reserve and Yeoja Bay including Suncheon Bay, which recorded the first Ramsar wetland in Korea. Therefore, further research is needed to establish an in-depth management plan.

  • The purpose of this study was development and application of a multiple box model for long term prediction and control of water quality in estuary reservoir. The model was composed of one main model and two sub models for hydrology and water quality. Water quality constituents for modeling were temperature, chlorophyll-a, BOD, DO, N, and P. The model had been applied to Asan reservoir, and the reservoir had been devided into three boxes-two boxes for a upper layer and one box for a lower layer-to represent stratification. The model appeared satisfactory in representing long term trend of water quality variations by comparing measured and simulated results. According to the application of the model to study alternatives for water quality improvement to deal with increase in future pollution load, use of non-phosphorous detergent for chlorophyll-a, advanced sewage treatment for BOD and T-N, reduction of livestock waste for T-P were evaluated as more effective ones than any others, respectively.

  • Effluent from an anaerobic digestion system with an elutriated phased treatment(ADEPT, Anaeorbic Digestion Elutriated Phase Treatment) for piggery waste treatment using anaerobic ammonium oxidation(ANAMMOX) process was used as a substrate of partial nitrification reactor. In mesophilic condition($35^{\circ}C$), controlling parameters of nitrite accumulation were HRT, pH, free ammonia(FA) and hydroxylamine rather than dissolved oxygen. Bicarbonate alkalinity consumption ratio including bicarbonate stripping and buffering was 8.78 g $Alk._{comsumed}/g\;NH_4-N_{converted}$. In steady state for 1 day of HRT and $2.7{\sim}4.4mg/L$ of DO, $NO_2-N/NH_4-N$ ratio of partial nitrification effluent was about $1{\sim}3$, which was applicable to ANAMMOX reactor influent for the combined partial nitrification-ANAMMOX process.

  • 본 연구는 유기성폐기물을 처리하고 그것을 이용하여 고체연료 또는 퇴비를 생산하는 종합시스템에 관한 것이다. 유기성폐기물은 90% 전후의 높은 함수율을 나타내고 있기 때문에 처리에 중점을 두어야 할 부분이 수분 처리 방법이다. 따라서 기존의 처리방법으로는 고액분리하여 처리 후 하천에 방류하는 시스템에 의해 처리되고 있다. 하지만 본 연구에서는 기술개발의 차별화 및 선행기술의 회피 전략을 위해, 유기성폐기물의 수분을 건조시키는 방법으로 미생물이 유기성폐기물의 유기물을 분해하여 발생 되는 $80^{\circ}C$ 이상의 열에너지에 의해 수분을 제거하는 발열반응을 적용하여 수분을 제거하는 무방류시스 템이다. 뿐만 아니라 고체연료의 발열량을 높이기 위해서 무연탄, 코크스, 기름 등의 열량보조제를 첨 가하는 대신에 유기성폐기물을 첨가 한 후 적당한 발열반응 및 건조시켜 고체연료를 제조하는 방법으 로 4,000kcal/kg 이상의 높은 발열량을 얻을 수 있으며 불완전연소 등에 의한 2차 오염을 방지할 수 있는 기술이다. 따라서 친환경 미생물 발열반응에 의해 유기성폐기물을 저렴한 비용으로 액상 및 고상 을 동시에 처리 할 수 있으며, 고체연료를 제조할 수 있는 새로운 신기술이다.

In Vitro Antagonistic Effects of Bacilli Isolates against Four Soilborne Plant Pathogenic Fungi

  • Twenty isolates of Bacillus spp. obtained from livestock manure composts and cotton-waste composts were tested for in vitro antagonistic effects against soilborne plant pathogenic fungi, Fusarium oxysporum, Phytophthora capsici, Rhizoctonia solani AG-4, and Sclerotinia sclerotiorum. Seven isolates of Bacillus spp. had antagonistic effects on mycelial growth of all the isolates of F. oxysporum tested. The bacterial isolate RM43 was the most effective to inhibit the mycelial growth of the fungal isolates. Twelve isolates of Bacillus spp. had antagonistic effects on mycelial growth of all the isolates of P. capsici tested. The bacterial isolates M34 and M47 were very effective to inhibit the mycelial growth of the fungal isolates. Thirteen isolates of Bacillus spp. had antagonistic effects on mycelial growth of all the isolates of R. solani AG-4 tested. The bacterial isolates M27 and M75 were very effective to inhibit the mycelial growth of the fungal isolates. Fourteen isolates of Bacillus sp. had antagonistic effects on mycelial growth of all the isolates of S. sclerotiorum tested. The bacterial isolates M49 and M75 were very effective to inhibit the mycelial growth of the fungal isolates. The antagonistic effects of most Bacillus spp. isolates against the isolates of the four fungi differed depending on the fungal species and the isolates of each fungus. The bacterial isolates M27 and M75 were the most effective to inhibit the mycelial growth of all four fungi.

Characteristics of Malodor Pollutants and Their Dispersion Measured in Several Industrial Source Regions in Yangsan (양산시 산업단지에서 측정한 악취물질의 농도 분포 특성 및 대기확산 모델링)

  • In this study, the environmental behavior of malodor pollutants (MPs: $H_2S$, $CH_3SH$, DMS, and DMDS) was investigated around areas influenced by strong anthropogenic processes based on observations and modeling study (a CALPUFF dispersion model). The MP emission concentrations were measured from 8 industrial source regions (tire plants (S1-S3), waste water disposal plant (S4), and oil refinery (S5) in an urban center area and paper mill/incineration plant (S6) and livestock feedlots (S7-S8) in Ungsang area) in Yangsan city during a fall period in 2008 (21 October 2008). Overall, the most MPs emitted from the urban center area were found to affect the malodor pollution in their downwind areas during early morning (06:00 LST) and nighttime (18:00 and 21:00 LST), compared with those in the Ungsang area. For malodor intensity, the most MPs in the urban center area (especially S1 and S2) were found to be a significant contributor, whereas $CH_3SH$ and $H_2S$ in the Ungsnag area (especially S6) were the dominant contributor. The model study showed agreement in the spatial distributions of simulated MPs with those of the observations. The largest impact of MPs in the urban center area on the malodor pollution in its residential areas occurred at S1, S2, and S3 sites during nighttime, while that of MPs in the Ungsang area occurred at S6 and S8 sites. This may be caused mainly by the high MP emissions and in part by wind conditions (prevailing northeasterly winds with low wind speeds of 2-3 m/s).

Evaluation of Complex Odor and Odorous Compounds in a Pilot-Scale Ultra Thermophilic Aerobic Composting Process (초고온 호기성 퇴비화 공정의 복합 악취 및 악취 물질 평가)

  • This study was conducted to evaluate production of complex odor and 12 specific odorous compounds in a pilot-scale (capacity : $100m^3$) ultra thermophilic aerobic composting. There were three types input: municipal wasted sludge, livestock manure and slurry, and food waste produced in Jung-Eb city. Each raw material was mixed with seed material and operated for two periods (1st : 50 days, 2nd : 60days). During composting, the temperature hit $90{\sim}95^{\circ}C$ after every mixing in both periods. Therefore, it was concluded that increasing temperature also saves the time which required for composting and high reduction of organics and water contents. The primary odorous compounds were ammonia, methyl mercaltan, dimethyl disulfide and trimethylamine. The concentration of the primary compounds and complex odor during the operation were higher than those on final day and most compounds did not exceed the allowable exhaust standard for odor. Also, it was found that optimal mixing time and control of high temperature are the most important parameters for odor control in ultra thermophilic aerobic composting.

  • To investigate the agricultural material application possibility of the compost made by the fermenting tank. We evaluate the stability fertility of the compost In evaluation of the stability of the compost, In germination index of test, germination test of lettuce of little resistibility not germinated wholly, in the radish of resistibility, both Shinco and TM-101 is showed 61%, 55% of germination rate, length of root not showed wholly. In stability fertility of the compost, early growth of radish in a little treatment applied was superior to other treatments except for TM-101 treatment, but the latter growth of those was showed opposite result. In evaluation of the fertility of compost, Primary growth of lettuce was superior in treatment applied 1.5t/10a, the latter growth of those was superior in treatment applied 3t/10a.

In Vitro Antagonistic Characteristics of Bacilli Isolates against Trichoderma spp. and Three Species of Mushrooms

  • Twenty isolates of Bacillus species obtained from livestock manure composts and cotton-waste composts were tested for their antagonistic effects in vitro against three green mold pathogens of mushrooms (Trichoderma harzianum, T. koningii, and T. viridescens). However, there exists a possibility Bacillus species may have antagonistic effects against mushrooms themselves, and thus the same 20 isolates were tested in vitro against three species of mushrooms (Flammulina velutipes, Lentinus edodes, and Pleurotus ostreatus). Of the 20 Bacillus species isolates tested, two inhibited mycelial growth of T. harzianum, seven that of T. koningii, and eight that of T. viridescens. Importantly, the bacterial isolates M27 and RM29 strongly inhibited mycelial growth of all the Trichoderma spp. isolates tested. The isolate M27 was subsequently identified as the most effective in inhibiting mycelial growth of all the Trichoderma species. Interesting results of the effect Bacillus isolates had upon the mushroom species followed. It was found that most Bacillus isolates except 5T33 at least somewhat inhibited mycelial growth of the three mushroom species or some of the mushrooms. Furhermore, the antagonistic effects of the bacterial isolates against the three species of mushrooms varied depending on the mushroom species, suggesting a role for mushroom type in the mechanism of inhibition. The bacterial isolates M27 and RM29 were identified as having the most antagonistic activity, inhibiting mycelial growth of all the Trichoderma spp. as well as mycelial growth of the three species of mushrooms. These results suggest that the bacterial isolates and their antagonistic effects on green mold pathogens should be further studied for their practical use for biological control of green mold in the growing room of the mushrooms.
