Effects of Ginseng Alkaloidal Fraction on Lipid Metabolism (인삼(人蔘) Alkaloidal Fraction이 지질대사(脂質代謝)에 미치는 영향(影響))

  • Alkaloidal fraction, a principle of ginseng extract was administered to rats orally in the amount of 2.5 mg per day per 100 gm body weight continuously for eight weeks to assess the lipid content of the serum and liver tissue. The findings can be summarized as follows: 1. Total cholesterol contents of serum and liver tissue were found to decrease with the administration of ginseng alkaloidal fraction. 2. Phospholipid contents increased in the serum and decreased in the liver tissue with the administration of ginseng alkaloidal fraction. 3. Triglyceride contents of the serum as well as liver tissue increased with the administration of ginseng alkaloidal fraction during the first 4 weeks but decreased in the second 4 weeks to reach a level below that of the pre-experimental period.

Gastrointestinal Tissue Blood Volume Affected by Venous Pressure Change (실혈 후 및 혈압상승 후의 소화기 조직 혈액량 및 산소 섭취량 -제 1 편 정맥혈압과 소화기 조직 혈액량-)

  • Changes in gastrointestinal tissue blood volume induced by variations of venous pressure between 6 and 40 mmHg were studied in 32 rabbits. Venous pressure lowering was produced by withdrawal of appropriate volume of blood and venous pressure elevation was obtained by partial occlusion of intra-thoracic vena cava inferior. Estimation of regional tissue blood volume was performed by means of regional distribution of injected $Cr^{51}-labeled$ red blood cells. The following results were obtained. 1. At the normal control venous pressure value of 18 mmHg, spleen showed the highest value of tissue blood volume expressed on weight basis, namely, $111{\mu}l/gm$, Liver tissue blood volume was $95\;{\mu}l/gm$, small intestine 24 and stomach $21\;{\mu}l/gm$, respectively. 2. Linear relationships were observed between venous pressure change and gastrointestinal tissue blood volume. The coefficients of correlation were: in spleen r=0.723; in liver r=0.791; in stomach r=0.704, respectively. In small intestine the relationship was less clear and r=0.358. Tissue blood volume of extrabdominal tissue, such as M. gastrocnemius was not influenced by venous pressure change. 3. The highest change in tissue blood volume expressed on weight basis was observed in spleen. The liver tissue showed the next highest change. Change in total tissue blood volume, however, was greatest in liver and next greatest in small intestine. This was interpreted by the fact that total weight of these two organs was much greater than that of spleen. 4. The mechanism that the change in tissue blood volume lies in the venous system which has a great compliance was discussed.

  • To investigate the effect of 'Saengkankunbi-Tang' on the liver tissue recovery in about 70% cutted rat liver, the observation on the photographing by the Gamma-camera and the electromicroscopic section against the rabbit liver induced by $CCl_4$, these studies were conducted. The results were summarized as follows; Observed on the liver tissue recovery, function regeneration and body weight increase in about 70% cutted rat liver, the recovery of liver tissue and body weight were increased with the passage of the time and also they were showed to the high increase rate in proportion to administration amount. The glucose content in the serum was found in increasing tide as compared with the control group. The alkaline phosphate activity in the serum showed the significant difference in the 10th day and 13th day as compared with the control group. The ammonia content in the blood showed the significant difference only in the 10th day. Observed on the photographing by the Gamma-camera against the rabbit liver induced by $CCl_4$, sample groups were shown to be significant recovery as compared with the control group. Observed on the electromicroscopic section, sample groups showed the control action of the nuclear destruction against the rabbit liver induced by $CCl_4$ and also they showed the control action of the expansion of glycogen and granular endothelial reticulum in cytoplasm. According to the above finding, it is presume that 'Saengkankunbi-Tang' can be applicable to the extensive treatment of liver disease.

  • The oral admistration of deer horn extract was proved to cause a considerable amelioration to the impairment of hepatic oxidative phosphorylation and to the decrease of ATP content in liver tissue induced by cholesterol ingestion. Deer horn extract was shown to accelerate considerably incorporation of $acetate-1-C^{14}$ into cholesterol in liver tissue and it tends to restore a decrease of GOT activity of liver tissue, caused by cholesterol administration. Histological examination showed that deer horn extract had a remakable preventive effect aginst fatty infiltration of organgs such as liver, heart, spleen, and adrenal gland of cholesterol given rabbits.

Protective effect of Saenggangeonbi-tang on liver fibrosis induced by thioacetamide (Thioacetamide로 유도된 간섬유화 모델에서 생간건비탕(生肝健脾湯)의 보호 효과)

  • Objective : In modern society, liver diseases such as liver fibrosis are on the rise as inflammation and wound healing processes of the liver are repeated due to factors such as drinking, smoking, and stress. This study was conducted to evaluate the effect of Saenggangeonbi-tang (SGGBT) on thioacetamide (TAA)-induced liver fibrosis. Methods : The mice were divided into 4 groups for examination (n=6): Normal group (Nor), distilled water-treated liver fibrosis mice (Con), silymarin 50 mg/kg-treated liver fibrosis mice (Sily), SGGBT 200 mg/kg-treated liver fibrosis mice (S200). Liver fibrosis was established in the mice via TAA for 8 weeks (1 week 100 mg/kg, 2,3 weeks 200 mg/kg, 4-8 weeks 400 mg/kg, three times a week, intraperitoneal injection) and they were administered silymarin and SGGBT (every day, oral administration) with the TAA. Results : SGGBT significantly decreased the levels of aspartate aminotransferase, alanine aminotransferanse, ammonia, and myeloperoxidase in serum increased by liver fibrosis. As a result of confirming H&E and MT staining, it was confirmed that SGGBT reduced damage and inflammatory cell infiltration in liver tissue, and alleviated changes in collagen fiber deposition and histological fibrosis. Also, it was confirmed through PAS staining that it reduced glycogen deposition in liver tissue. In addition, SGGBT significantly decreased the NADPH oxidases as well as significantly modulated the expression of MMP-2 and TIMP-2. Conclusions : These results suggest that SGGBT regulates the expression of MMP/TIMP protein through inhibition of oxidative stress and alleviates liver fibrosis by reducing collagen and glycogen deposition in liver tissue.

Metabolism of Lactate Dehydrogenase in Tissues from Ldh-C Expressed Fish at Starved State (기아상태에서 Ldh-C가 발현된 어류 조직의 젖산탈수소효소의 대사)

  • Metabolism of lactate dehydrogenase (EC 1.1.1.27, LDH) was studied to identify the function of LDH-C. Tissues of LDH liver-specific Ldh-C expressed Carassius auratus and eye-specific Ldh-C expressed Lepomis macrochirus after starvation were studied. LDH activity in liver tissue from C. auratus was increased after starvation. And LDH specific activity (units/mg) and LDH/CS were increased in tissues. It means the anaerobic metabolism was taking place in C. auratus after starvation. LDH B4 isozyme was decreased in skeletal muscle and increased in heart tissue. LDH C4 isozymes those showed in eye and brain tissues were identified as liver-specific C4 isozymes and disappeared after starvation. And C hybrid in eye, A4 isozyme in brain, and both C hybrid and C4 isozyme in liver tissue were increased, respectively. In L. macrochirus, the level of variation of LDH activities was low but greatly increased especially in eye tissue and LDH A4 and AC hybrid were increased in brain tissue. The LDH activities in tissues from C. auratus and L. macrochirus remained 30.30-18.64% and 25-18.75%, respectively, as a result of the inhibition by 10 mM of pyruvate. The KmPYR values of LDH in C. auratus were increased. As a result, LDH liver-specific C4 isozyme was expressed in liver, brain and eye tissues during starvation. It seems metabolism of lactate was predominant in brain tissue. After starvation, the liver-specific LDH-C was affected more than eye-specific LDH-C.

Fatty Liver Analysis through Quantitative Measurement Study of Ultrasonography Images (초음파 검사 영상의 정량적인 측정 연구를 통한 지방간 분석)

  • This study attempted to find out the degree of agreement between ultrasound image findings along with analysis of attenuation index and scatter distribution index values within tissues through quantitative measurement analysis using liver ultrasound images. From August 2022 to October 2022, liver ultrasound was performed on 45 patients who were suspected of having fatty liver and who received a prescription for liver ultrasound. As a result of the study, as a result of analyzing the agreement between the ultrasound image findings and the tissue attenuation index, the Kappa value was 0.82 (p<0.05), showing a very high agreement between the two examination methods. In addition, as a result of the agreement analysis between the ultrasound image findings and the scatter distribution index in the tissue, the Kappa value was 0.642 (p<0.05), showing high agreement between the two examination methods. At the time of fat liver prediction, the use of liver ultrasound findings and quantitative ultrasonography techniques, such as intra-tissue attenuation index and intra-tissue scatter distribution index, may be helpful in determining the degree of progression of fatty liver patients.

Identification of MFGE8 in mesenchymal stem cell secretome as an anti-fibrotic factor in liver fibrosis

  • The beneficial paracrine roles of mesenchymal stem cells (MSCs) in tissue repair have potential in therapeutic strategies against various diseases. However, the key therapeutic factors secreted from MSCs and their exact molecular mechanisms of action remain unclear. In this study, the cell-free secretome of umbilical cord-derived MSCs showed significant anti-fibrotic activity in the mouse models of liver fibrosis. The involved action mechanism was the regulation of hepatic stellate cell activation by direct inhibition of the $TGF{\beta}$/Smad-signaling. Antagonizing the milk fat globule-EGF factor 8 (MFGE8) activity blocked the anti-fibrotic effects of the MSC secretome in vitro and in vivo. Moreover, MFGE8 was secreted by MSCs from the umbilical cord as well as other tissues, including teeth and bone marrow. Administration of recombinant MFGE8 protein alone had a significant anti-fibrotic effect in two different models of liver fibrosis. Additionally, MFGE8 downregulated $TGF{\beta}$ type I receptor expression by binding to ${\alpha}v{\beta}3$ integrin on HSCs. These findings revealed the potential role of MFGE8 in modulating $TGF{\beta}$-signaling. Thus, MFGE8 could serve as a novel therapeutic agent for liver fibrosis.

The Effect of Hasuogamibang on Hyperlipidemic SHR and Antioxidant Activity (하수오가미방이 고지혈증 SHR 및 항산화에 미치는 영향)

  • Objectives : This study was designed to evaluate the effect of Hasuogamibang (HGB) on hyperlipidemia and antioxidant activity. Methods : For this study, we divided four groups of rats (normal WKY group, normal SHR group, high cholesterol diet and saline-treated SHR group, high cholesterol diet and HGB-treated SHR group), and observed the change of body weight, weight of liver, cholesterol, triglyceride, glucose, albumin, histologic change of liver and aorta, lipid peroxidation and antioxidant activity of liver tissue, and cholesterol gene revelation control efficiency. Results : Total-cholesterol, LDL-cholesterol, triglycerides were decreased significantly by HGB. However, HDL-cholesterol increased significantly. The tissue of liver and aorta were controlled defect by HGB on histologic study. Lipid peroxidation and SOD of liver tissue was decreased significantly by HGB. Gene revelation of ACAT and HMG CoA reductase in hepatic tissue was decreased significantly by HGB. Conclusion : This study suggests that HOB is significantly effective on hyperlipidemia and antioxidant activity.

Effects of Rapeseed Oil Diet on Serum and Liver Lipid Levels in Rats (유채유 함유사료가 쥐의 혈청 및 간의 지질조성에 미치는 영향)

  • The fatty acid composition of a rapeseed oil being on the market was analyzed and the effect on gain of the body weight and lipid levels in serum and liver tissue of male rats of Sprague-Dawely strain fed the diet containing the rapeseed oil were studied. The fatty acid components of marketed rapeseed oil was oleic acid 29.4%, erucic acid 26.52%, linoleic acid 20.39% and linolenic acid 8.68%. The contents of total lipid in serum W3S Significantly higher in RSO20 group than Contr01 group(P< 0.01) . But that in the liver tissue did 001 show significant differences. The contents of triglyceride in serum was control group 84.14mg/dll, RSO15 group 100.33mg 141 and RSO20 group 122.00mg 141 and showed significant difference between each group, but that in the livertissue did not show significant differences. The contents of phospholipid in serum did not show significant differences. But that in the liver tissue showed significant difference between the control group 8.42mg /g and Rs02o group 7.34mg /g(p<0.001). The contents of total-cholesterol and free-cholesterol in serum and liver tissue of the RSO20 group showed the highest levels compared with control group, but there did not show significant differences. The contents of ester-cholesterol in serum showed significant.
