Effects of Gamisaenggan-tang on High Fat Diet-induced Nonalcoholic Fatty Liver Disease (가미생간탕(加味生肝湯)이 고지방식이로 유발된 지방간에 미치는 영향)

  • Objectives : The aim of this study was to investigate the effects of Gamisaenggan-tang on high fat diet induced nonalcoholic fatty liver disease. Methods: Rats were randomly divided into four groups. The Normal group was fed a solid diet containing 10% fat. The Gamisaenggan-tang (GS) group was fed a solid diet containing 10% fat and Gamisaenggan-tang (90mg/100g body weight). The Control group was fed a solid diet containing 60% fat. The HFD-Gamisaenggan-tang (HFD-GS) group was fed a solid diet containing 60% fat and Gamisaenggan-tang (90mg/100g body weight). Six weeks later, rats body weight, liver weight, serum ALT, GGT, ALP levels were measured. Histological findings (Oil red O staining), hepatic triglyceride, TNF-${\alpha}$, and TGF-${\beta}$ levels in the liver tissue were studied. Results: Average body weight of the HFD-GS group was significantly less than that of the Control group. There were no significant liver weight differences among each group. The GGT levels of the HFD-GS group were significantly less than those of the Control group. However, there were no significant differences in the ALT or ALP levels among the groups. TNF-${\alpha}$ protein production assessed by western blot analysis was reduced by Gamisaenggan-tang. Greater fat accumulation was observed in the liver tissue of the Control group than in the HFD-GS group, which means the Gamisaenggan-tang has an inhibitory effect on the accumulation of fat in the liver. Conclusion : The results suggest that Gamisaenggan-tang can be potential candidate for the treatment of nonalcoholic fatty liver disease in clinics.

  • Liver is an oragan which has various functions. For this functions in liver disease change variously, many methods of examination of them are invented. Each liver disease impairs these functions diversly rather than uniformly, thus it is possible to differentiate each disease by using various methods of testing it. Liver has high reproductive capacity and despite partial impairment of its function localized disease may not show any abnormality in liver function test due to compensation of remaining intact tissue of the liver. Thus the result of liver function test should be interpreted under adequate clinical investigation with other test, not depending on the liver function test only. This thesis deals with the composition, distribution in living body and clinical significance of liberating enzyme, GOT, GPT, and LDH which are included in serum enzyme that show abnormality in liver disease.

Effects of Dietary Zinc and Ethanol on the Mineral Contents of Serum and the Changes of the Liver Tissue in Rat (식이성 아연과 에탄올이 흰쥐의 혈청중 무기질 함량과 간장조직에 미치는 영향)

  • This experiment was conducted to study the effects of dietary zinc and ethanol on the mineral contents of serum and changes of the liver tissue. Eighty male rats of Sprague-Dawley strain with average weight of 80$\pm$5g were divided into five groups according to zinc levels(5ppm and 100ppm), ethanol and isocaloric sucrose feedings. The rats were sacrificed after 4 and 7 weeks of feeding periods. The results are summarized as follows. 1. There was a decrease of weight gain in the ZD group compared with the C group and also decreased in the CE and ZDE groups fed the ethanol solution. The feed intake was decreased in the ZD and ZDE groups, and the feed efficiency was also greatly reduced in the ZDE group. 2. The liver weight of the ZD and ZDE groups was increased, however, the weight of testis was decreased in the same groups. And the weight of spleen was decreased in the ZD group. 3. The AST and ALT activities of serum were significantly increased in the ZDE group. 4. The content of serum zinc was influenced by the dietary zinc level, and the amount was significantly decreased in the ZD group. But the content of serum copper was remarkably increased in the ZD grou. The calcium and magnesium contents of serum were decreased in the ZD and ZDE groups. 5. The morphological changes of the liver tissue were not different among the C, CE and PF groups, but the abllooning denaturation of the liver cell together with the infiltration of the local inflammation cell was found in ZD and ZDE groups.

Liver Dysfunction and Oxidative Stress in Streptozotocin-Induced Diabetic Rats: Protective Role of Artemisia Turanica

  • Objectives: Oxidative stress plays a central role in diabetes-induced complications. In the present study, the protevtive effect of Artemisia turanica (A. turanica) was evaluated against diabetes-induced liver oxidative stress and dysfunction. Methods: Fifty male Wistar rats were randomly divided into five groups: control, diabetic, diabetic + metformin, diabetic + A. turanica extract, and diabetic + A. turanica extract + metformin. Experimental diabetes was induced by a single-dose (55 mg/kg, intraperitoneally (ip)) injection of streptozotocin (STZ). Metformin (300 mg/kg) and A. turanica extract (70 mg/kg) were orally administrated three days after STZ injection for four weeks. The levels of malondialdehyde (MDA), total thiol content and superoxide dismutase (SOD) and catalase activities were measured in the liver tissue. Serum glucose concentration, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities were also determined. Results: In the diabetic group, serum glucose concentration, serum AST and ALT activities and liver MDA level were significantly higher while tissue total thiol content as well as catalase and SOD activities were lower, compared to the control group. Serum glucose in diabetic rats treated with metformin + A. turanica extract showed a significant decrease compared with the diabetic group. In all the A. turanica extract and metformin treated groups, serum ALT, tissue MDA level, total thiol content and SOD activity significantly improved compared with the diabetic rats. However, treatment of the diabetic rats only with metformin could not significantly change the activities of catalase and AST compared with the diabetic group. Conclusion: These findings suggested that A. turanica extract had a therapeutic effect on liver dysfuncyion and oxidative stress induced by diabetes, that may be probably due to its antioxidant and antiinflammatory effects.

An Experimental Study on the Changes of Liver Tissue by the Administration of Anticlonorchial Drugs to Rabbits with Clonorchiasis (간(肝)디스토마증(症) 치료약투여후(治療藥投與後) 간장(肝臟) 변화(變化)에 관(關)한 실험적(實驗的) 연구(硏究))

  • An experimental study was done on rabbits to observe the effects of several anticlonorchial drugs on the pathology of the liver infested with Clonorchis sinensis. After two months of infestation with Clonorchis sinensis by giving $400{\sim}500$ metacercariae by mouth, hexachlorophene, chloroquine 2,2' methylenebis (3,4,6 trichlorophenoxy acetic acid) and Hetol were administered orally and follow up macro-and microscopic studies of the liver pathology were done in 2 to 3 days, one month, 2 months and 3 months after the completion of medications. The results obtained in this study are as follows: 1. In both groups which were administered hexachlorophene piperazine 20mg/kg for seven days or 8mg/kg for 18 days, the macroscopic findings of the liver after 3 months revealed only minimal changes of the color and consistency The histopathological findings were the reduction of fibrosis, pseudolobulation, proliferation and adenomatous hyperplasia of bile ducts, and regeneration of liver parenchyma. 2. In groups which were administered chloroquine phosphate 20mg/kg for 18 days or 40mg/kg for 5 days, and also in groups which were administered dithiazanine iodide 30mg/kg for 18 days or 60mg/kg for 5 days, no significant findings of recovery were observed either macroscopically or microscopically. 3. In the group which was given 20mg/kg of 2,2' methylenebis(3,4,6 trichlorophenoxy acetic acid) for 5 days, prominent healing of the damaged tissues was observed after 2 months, revealing the decrease of fibrous tissue, caliber of bile ducts and adenomatous hyperplasia of the epitherial cells of the bile ducts, and regenerationof liver parenchyma. 4. In the group which was given Hetol 200mg/kg for 5 days, swelling, congestion and eddish-brown discoloration of the liver were noted macroscopically after 3 days of completion of drug administration. Hemorrhage, congestion, necrosis and degeneration of the parenchyma were observed microscopically After 10 days, liver appeared almost normal macroscopically, but marked fat degeneration was noted microscopically. After 2 months, the liver was almost normal in gross appearence with only slight atrophy and also marked healing was observed microscopically, i. e. decrease of fibrous tissue and reduction of the previously enlarged bile duct and the regeneration of the liver parenchyma.

Histopathological Studies on the Effect of Persimmon Leaves on Cadmium Poisoning in Mice (감잎이 마우스의 카드뮴 중독에 미치는 병리조직학적 관찰)

  • This study was undertaken to find out the effect of persimmon leaves on histopathological changes of cadmium toxicity in mice. Seventy two BALB/c mice of male were divided into a control group(A) and five experimental groups (B, C, D, E, F) : group A received tap water and basal diet, group B received tap water and diet supplemented with 3% persimmon leaves alone, group C received basal diet and 300 ppm cadmium, group D, E and F received basal diet supplemented with 1, 3% and 7% persimmon leaves and 300 ppm cadmium respectively. Cadmium dissolved in tap water was used, and the persimmon leaves were mixed with feed. All mice were dissected on the 84th day. Pathological changes in liver, kidney, cortical osseous tissue of femoral shaft, bone trabecular of femur, and epiphyseal cartilage plate of femur were observed. Group B showed no significant changes as the control group. But group C showed the unclearness of specific cells in liver, the loss of architecture and necrosis of hepatocyte, degeneration and necrosis of renal convoluted tubules, desquamation and vacuolization of the greater part of the renal tubular epithelium, marked thinning of the cortical osseous tissue in femoral shaft, reduction of cancellous bone volume and decreaswe of trabecular number, and marked thinning of epiphyseal cartilage plate in femur. On the other hand, persimmon leaves-treated group showed a little convalescent changes an maintained their normal architectures in liver, kidney, cortical osseous tissue of femoral shaft, bone trabecular of femur, and epiphyseal cartilage plate of femur.

  • The following case study suggests strongly the efficacy of Danggwisusan and acupuncture therapy in the treatment of patients with severe contusion and tissue injury of the liver and kidney. The chief symptoms of this case were severe pain in the right side, purpura hemorrhagica, insomnia and third degree bums at the shoulder and face. Analysis of blood chemistry indicated high AST, high ALT, high BUN, high creatine, high creatine kinase and high LDH. Fallowing medication of the patient with Danggwisusan, the symptoms and indexes for tissue injury decreased rapidily within the first week. This results support a potential role for Danggwisusan in the treatment of severe contusion with tissue injury of the liver and kidney.

  • The role of scintigraphic imaging has moved from the detection of lesions to the tissue-specific characterization of lesions over the past 2 decades. Major advances in nuclear medicine imaging include: 1) positron imaging, 2) improved instrumentation, such as the use of multidetector (dual or triple head) gamma cameras for single photon emission computed tomography, and 3) development of numerous new radiopharmaceuticals for positron or single photon imaging (labeled glucose analogue, amino acids, fatty acids, hormones, drugs, receptor ligands, monoclonal antibodies, etc). These advances have resulted in a significantly improved efficacy of radionuclide techniques for the evaluation of various tumors, including those within the liver. The current role of nuclear medicine in the evaluation of focal hepatic tumors is reviewed in this article with an emphasis on the clinical applications of various tracer studies and imaging findings.

Cadmium contents in tissues of slaughtered deer in Korea (도축 사슴의 체조직내 카드뮴 함량에 관한 조사 연구)

  • In order to clarify the cadmium(Cd) contents of normal deer, tissue samples including kidney, liver, muscle, spleen, heart, lung, rumen, abomasum, intestine, skin and bone were collected from 18 deer which were slaughtered in Korea. All samples were burned to ashes and analyzed for Cd contents by atomic absorption spectrophotometric method. The mean Cd contents in \mu\textrm{g}$/g wet matter for 18 deer were liver 0.013, kidney 0.286, muscle 0.010, spleen 0.001, heart 0.007, lung 0.005, men 0.019, abomasum 0.016, intestine 0.018, skin 0.016 and bone 0.312, respectively. Two samples in kidney, one sample in muscle out of 18 deer showed higher Cd contents than normal limit value of 0.50\mu\textrm{g}$/g for liver and kidney, 0.10\mu\textrm{g}$/g for muscle. In addition, three deer also showed higher Cd contents in one body tissue than normal limit value which reported foreign countries.

Tissue-Specific Regulation of Angiotensinogen and Angiotensin II Receptor Gene Expression in Deoxycorticosterone Acetate-Salt Hypertensive Rats

  • Molecular regulation of the renin-angiotensin system (RAS) was investigated in deoxycorticosterone acetate (DOCA)-salt hypertension. The expression of renin, angiotensinogen and angiotensin II receptor genes in the kidney and liver was determined by Northern blot analysis in rats which were made DOCA-salt hypertensive over the period of 2 or 4 weeks. Along with the hypertension, renin mRNA was decreased in the remnant kidney. The expression of angiotensinogen gene was not significantly altered in the kidney, but was significantly decreased in the liver. The expression of angiotensin II receptor gene was increased in the kidney, while it remained unaltered in the liver. The duration of hypertension did not affect the altered gene expression. It is suggested that the components of RAS are transcriptionally regulated in DOCA-salt hypertension in a tissue-specific manner.
