카드뮴이 랫드의 Heat Shock Protein 발현에 미치는 영향과 독성학적 변화에 관한 연구 (Effects of Cadmium on Heat Shock Protein Induction and on Clinical Indices in Rats)

  • Exposure indices are important tools which enable scientists to reliably predict and detect exposures to xenobiotics and resultant cell injury. Since the de novo synthesis of stress proteins can be detected early after exposure to some agents, analysis of toxicant-induced changes in gene expression, i.e. alterations in patterns of protein synthesis, may be useful to develop as biomarkers of exposure and toxicity. The acute and chronic effects of cadmium(Cd, $CdCl_2$ 20 mg/kg) on Wistar male rats were evaluated concerning cadmium contents, tissues enzyme activity, HSP expression. The results of the study were as follows: 1. Less cadmium was absorbed through the digestive tracts, but the ratio of contents in renal to hepatic cadmium was higher at 8 weeks after treatment. 2. ALT(alanine aminotransferase), AST(aspartate aminotransferase), glucose, BUN(blood urea nitrogen), creatinine, the key indices of the clinical changes in hepatic and renal function were significantly changed by the cadmium treatment after 1 week in liver, after 4 weeks in kidney. 3. Enhanced synthesis of 70 KDa relative molecular mass proteins were detected in 2 hours after cadmium exposure, with maximum activity occurring at 8~48 hours. Induction of $HSP_{70}$ was evident at proximal tubules and glomeruli in kidney. Testicular cells produced enough HSP to be detected normally. From the above results, it could be concluded that $HSP_{70}$ induction by the cadmium treatment was a rapid reaction to indicate the exposure of xenobiotics.

돼지 간중의 Monooxygenase 가 Diazinon 의 분해에 미치는 영향 (The degradation o Diazinon by hepatic monooxygenase of Pig)

  • 돼지간을 두 fraction($105,000{\time}g$ pellet, superatant)으로 나누어 Diazinon의 분해에 관여하는 두 효소(Monooxygenase와 esterase)의 활성 및 저해제(EPN, Beam, PBO)첨가에 따른 Diazinon의 분해억제 능력을 비교 검토하였다. 1. 돼지간의 microsomal fraction과 soluble fraction에서 M. O.와 β-esterase 활성은 20분과 60분에 최고활성을 나타내었다. 2. Microsomal fraction에서의 M. O.활성은 NADPH generating system의 첨가로 1.6배의 활성증가와 지속성이 확인되었다. 3. Microsomal fraction에서의 Diazinon분해량은 30분에 최고량(0.060nM/ mg/ min)을 보였으나, soluble fraction에서는 60분에 최고량(0.018nM/ mg/ min)을 보여 1/3수준이었다. 4. M. O.활성에 미치는 Diazinon에 대한 EPN, Beam, PBO의 첨가효과는 Beam(14%)< EPN(15%)

Intestinal Parasites in an Ottoman Period Latrine from Acre (Israel) Dating to the Early 1800s CE

  • The aim of this study is to determine the species of parasites that affected the inhabitants of the city of Acre on the coast of the eastern Mediterranean during the Ottoman Period. This is the first archaeological study of parasites in the Ottoman Empire. We analysed sediment from a latrine dating to the early 1800s for the presence of helminth eggs and protozoan parasites which caused dysentery. The samples were examined using light microscopy and enzyme-linked immunosorbent assay (ELISA) kits. We found evidence for roundworm (Ascaris lumbricoides), whipworm (Trichuris trichiura), fish tapeworm (Dibothriocephalus sp.), Taenia tapeworm (Taenia sp.), lancet liver fluke (Dicrocoelium dendriticum), and the protozoa Giardia duodenalis and Entamoeba histolytica. The parasite taxa recovered demonstrate the breadth of species present in this coastal city. We consider the effect of Ottoman Period diet, culture, trade and sanitation upon risk of parasitism in this community living 200 years ago.

Conjugated Linoleic Acid (CLA) Supplementation for 8 Weeks Reduces Body Weight in Healthy Overweight/Obese Korean Subjects

  • In the present study, a randomized, double-blind, placebo-controlled trial to determine the effect of conjugated linoleic acid (CLA) supplementation (50:50 ratio of cis-9, trans-11 and trans-10, cis-12 isomers) for 8 weeks on body composition and biochemical parameters in healthy overweight/obese (body mass index, BMI${\geq}23\;kg/m^2$) Korean subjects was performed, Thirty participants (3 males and 27 females) were randomized to receive placebo (2.4 g olive oil/day) or 2.4g/day CLA (mixture containing 36.9% of cis-9, trans-11 and 37.9% of trans-10, cis-12). Eight weeks of CLA supplementation significantly decreased body weight by -0.75kg, BMI by $-0.27\;kg/m^2$, and hip circumference by -1.11 cm. The reduction of body weight was ascribed to the reduction of body fat mass (-0.59 kg) and lean body mass (-0.18 kg), although these changes were not significant. No significant differences in serum lipid profiles, liver function enzyme activities, and protein concentration were observed in either the CLA or placebo groups. These results indicate that short tenn supplementation (8 weeks) with CLA (2.4 g/day) may decrease body weight in Korean overweight/obese subjects.

흰쥐에 있어서 구기자 알콜 추출물이 Oxygen Free Radical 및 Alcohol 대사효소 활성에 미치는 영향 (Effect of the Ethanol Extract of Lycium chinense on the Oxygen Free Radical and Alcohol Metabolizing Enzyme Activities in Rats)

  • To investigate an effect of the ethanol extract of Lycium chinense(EELC) on the activities of enzymes scavenging oxygen free radicals or detoxicating alcohol. The ground Lycium chinense was extracted with 30% edible ethanol and then diluted with 6% ethanol to contain 2% EELC(w/v). Three different groups of male Sprague-Dawley rats had taken a drink EELC, ethanol(ETH) or water(control), respectively for 2 months. At the end of experimental period, the animals were sacrificed and obtained the following findings. The EELC-treated animals showed the highest activity of hepatic glucose-6-phosphatase among three groups. The activities of xanthine oxidase and cytochrome p-450 from EELC treatment group were lower than those from ETH-treated group. However, the activity of superoxide dismutase was higher in the EELC-treated group than the ETH-treated(p<0.005). Furthermore, hepatic alcohol or aldehyde dehydrogenase activity, and glutathione content and glutathione peroxidase were significantly higher in EELC-treated animals than in ETH-treated those. The activity of glutathione S-transferase in liver was appeared the orderly higher value in EELC, ETH and control-treated group. As the result, EELC may affect the reduction of oxygen free radical production and help the detoxication of ethanol.

경부 지방종증의 치험 (Treatment of Benign Symmetric Lipomatosis)

  • Benign symmetric lipomatosis was initially described in 1846 by Sir Benjamin Brodie. In 1888, Otto Madelung presented 33 cases of benign symmetric lipomatosis and described the classic“horse collar”cervical distribution of the lipomatous tissue. Launois and Bensaude described benign symmetric lipomatosis as a distint syndrome characterized by a diffuse, symmetric, fatty accumulation in the cervical region. This disease is rare condition affecting mostly middle aged alcoholic men and associated with many systemic diseases such as diabetes mellitus, hyperuricemia, renal tubular acidosis, liver enzyme abnormality etc. The condition does not spontaneously involute and surgical excision is the only proven method of treatment, and recurrence is frequent. We experienced six patients of benign symmetric lipomatosis who underwent surgical excision via collar incision which afford wide exposure of the entire cervical area. We report them with the review of literature.

Acute Pancreatitis after Additional Trauma in Chronic Traumatic Pancreatic Diaphragmatic Hernia

  • Traumatic diaphragmatic injuries (TDIs) are a rare complication in thoraco-abdominal trauma. The diagnosis is difficult and if left untreated, TDI can cause traumatic diaphragmatic hernia (TDH). Through an injured diaphragm, the liver, spleen, stomach, small intestine, and large intestine can be herniated to the thoracic cavity, but pancreatic herniation and pancreatitis are quite rare in TDH. This paper reports a case of pancreatitis developed by additional trauma in a patient with asymptomatic chronic TDH. A 58-year-old male visited the emergency department with a left abdominal injury after a fall 6 hours earlier. The vital signs were stable, but the amylase and lipase levels were elevated to 558 U/L and 1,664 U/L, respectively. Abdominal computed tomography (CT) revealed a left diaphragmatic hernia and an incarceration of the stomach, pancreatic ductal dilatation, and peripancreatic fatty infiltration. Additional history taking showed that he had suffered a fall approximately 20 years ago and had an accidentally diaphragmatic hernia through a chest CT 6 months earlier. A comparison with the previous CT revealed the pancreatitis to be caused by secondary pancreatic ductal obstruction due to the incarcerated stomach. For pancreatitis, gastrointestinal decompression was performed, and after 3 days, the pancreatic enzyme was normalized; hence, a thoracotomy was performed. A small ruptured diaphragm was found and reposition of the organs was performed. This paper reports the experience of successfully treating pancreatitis and pancreatic hernia developed after trauma without complications through a thoracotomy following gastrointestinal decompression.

Protective effects of Hizikia fusiforme and Chlorella sp. extracts against lead acetate-induced hepatotoxicity in rats

  • In the present study, the protective effects of Hizikia fusiforme and Chlorella sp. extracts on lead acetate-induced hepatotoxicity were investigated. Hepatic damage was induced in rats by intraperitoneal (i.p.) injection of lead acetate and the protective effects of H. fusiforme (HZK) and Chlorella sp. (CHL) extracts on lead acetate-induced hepatic damage in rat liver were examined. The results revealed significantly increased glutamic oxaloacetate and glutamic pyruvic transaminase levels in the group treated with lead acetate only (Pb group); oral administration of HZK and CHL extracts tended to decrease the enzyme levels similar to those observed in the control group. Regarding antioxidant enzymes, superoxide dismutase activity was increased in the Pb group and decreased in a concentration-dependent manner in the HZK- and CHL-treated groups. Glutathione levels were increased in a concentration-dependent manner in the HZK- and CHL-treated groups. There was no significant difference in catalase activity. Western blot analysis showed inflammation-related protein expression in mitogen-activated protein kinase and Nrf2 pathways was affected in the HZK- and CHL-treated groups. Therefore, HZK and CHL extracts exerted antioxidant and anti-inflammatory effects against lead acetate-induced hepatotoxicity. Development of functional health foods containing HZK and CHL extracts, which have hepatoprotective effects against inhaled lead acetate, should be considered.

A Study on the Effects of Turmeric Intake after Weight Training on Blood Alcohol Concentration

  • The purpose of this study was to investigate the effects of turmeric intake and weight training on blood alcohol concentration, liver enzyme levels of and their effects on health promotion in adult males. There was no significant difference of taking turmeric powder combined with weight training exercise on blood alcohol levels in adult men who consumed alcohol on a regular basis. There was also no change on the change of body composition. The results were collected from elite athletes that had spent more than 10 years in their respective sports so it is difficult to observe any significant results from 8 weeks, short-term exercises of 1RM 70~80%, 3 times per week. In the next study, it is necessary to divide the subjects into more diverse groups and subject them by varying amounts of turmeric intake, exercise, etc., in order to fully study and understand the effects on blood alcohol level, change, and health promotion. Consequently, this study demonstrated there were no significant differences in the effects of continuous drinking habits of adult men's turmeric powder intake and weight training exercise on changes in GOT, GPT, γ-GTP, and blood alcohol concentrations. Moreover, health improvements themselves didn't affect changes in body composition.

Comparison of the trometamol-balanced solution with two other crystalloid solutions for fluid resuscitation of a rat hemorrhagic model

  • Currently, the optimal resuscitation fluid remains debatable. Therefore, in the present study, we designed a trometamol-balanced solution (TBS) for use as a resuscitation fluid for hemorrhagic shock. Hemorrhagic shock was induced in 18 male Wistar-Kyoto rats, which were assigned to normal saline (NS), Ringer's solution (RS), and TBS groups. During the hemorrhagic state, their hemodynamic parameters were recorded using an Abbott i-STAT analyzer with the CG4+ cartridge (for pH, pressure of carbon dioxide, pressure of oxygen, total carbon dioxide, bicarbonate, base excess, oxygen saturation, and lactate), the CG6+ cartridge (for sodium, potassium, chloride, blood glucose, blood urea nitrogen, hematocrit, and hemoglobin), and enzyme-linked immunosorbent assay kits (calcium, magnesium, creatinine, aspartate aminotransferase, alanine aminotransferase, bilirubin, and albumin). Similar trends were found for the parameters of biochemistries, electrolytes, and blood gas, and they revealed no significant changes after blood withdrawal-induced hemorrhagic shock. However, the TBS group showed more effective ability to correct metabolic acidosis than the NS and RS groups. TBS was a feasible and safe resuscitation solution in this study and may be an alternative to NS and RS for resuscitation in hemorrhagic shock patients without liver damage.
