Study on the Accumulation of Ochrtoxin A in Mouse's Organs and the Establishment of ELISA Method for Ochratoxin A (Ochratoxin A가 마우스의 장기에 축적 정도와 ELISA법 확립에 관한 연구)

  • Ochratoxin A was produced from Aspergillus ochraceus ATCC 18472 which was then orally administered into the experimental mice to study the toxic levels of ochratoxin A. AELISA (enzyme-linked immunosorbent assay) method which is more rapid and safe than conventional analytical method, was developed by using ochratoxin A antibody. This method was successfully used to measure the levels of ochratoxin Ain blood, liver and kidney of mice. In order to produce a large amount of ochratoxin A to study toxicity in the mice, Aspergillus ochraceus ATCC 18412 was incubated in the rice medium and as a result 0.5 g of ochratoxin A form rice medium (kg) was produced after extraction and purification Feed consumption and gain in body weight of mice with ochratoxin A at a level of $10 \mu\textrm{g}/g$ body weight was significantly (p<0.05) reduced as compared with control during period of 3 weeks. Ochratoxin A-BSA conjugate was made by putting 13 mole of ochratoxin A on I mole of BSA. This conjugate was used to develop ELISA method. The minmum detection level of ochratoxin A by established ELISA method was 0.5 ppb. After oral adminstraton of ochratoxin A dose of $10\;\mu\textrm{g}/g$ every two day for 3 weeks, concentration of ochratoxin A was measured in the blood, liver and kidney by ELISA method. The level of ochratoxin A was $11\;\mu\textrm{g}/dl,\;0.9 \mu\textrm{g}/g\;and\;3.7\;\mu\textrm{g}/g$ in the blood, liver and kidney, respectively.

Effect of Geumsuyukgunjeon on Airway Mucus Secretion and Mucin Production (금수육군전(金水六君煎)이 이산화황으로 유발된 흰쥐의 호흡기 점액 및 뮤신생성에 미치는 영향)

  • Objectives In this study, effect of Geumsuyukgunjeon (GYJ) on the increase in airway epithelial mucosubstances of rats with acute bronchitis and EGF-induced MUC5AC mucin production from human airway epithelial cells were investigated. Materials and Methods Hypersecretion of airway mucus was induced by exposure of rats to SO2 during 3 weeks. Effect of orally-administered GYJ during 2 weeks on increase in airway epithelial mucosubstances from tracheal goblet cells of rats was assesed using histopathological analysis after staining the epithelial tissue with PAS-alcian blue. Possible cytotoxicity of GYJ was assessed by examining the potential damage of kidney and liver functions by measuring serum GOT/GPT activities and serum BUN and creatinine concentrations of rats and the body weight gain during experiment, after administering GYJ orally. Effect of GYJ on EGF-induced MUC5AC mucin production from human airway epithelial cells (A549) was investigated. Confluent A549 cells were pretreated for 30 min in the presence of GYJ and treated with EGF (25 ng/ml) for 24 hrs, to assess the effect of GYJ on EGF-induced MUC5AC mucin production using enzyme-linked immunosorbent assay (ELISA). Results (1) GYJ decreased the amount of intraepithelial mucosubstances of trachea of rats. (2) GYJ did not show kidney and liver toxicities and did not affect body weight gain of rats during experiment. (3) GYJ significantly inhibited EGF-induced MUC5AC mucin production from A549 cells. Conclusions The result from the present study suggests that GYJ might control both the mucus hypersecretion in vivo and do not show in vivo toxicity to liver and kidney functions after oral administration and the production of pulmonary mucin.

Effect of swimming exercise therapy on a fatty liver in SD-Rat dosage high fat diet (수영운동 치료가 고지방식 투여 SD-Rat의 지방간에 미치는 영향)

  • The purpose of this study is to clarify the repercussions of swimming exercise therapy that has an effect on lipid in blood and liver enzyme of the 3D-Rat dosage high fat diet. The object of this study consisted of two groups. One was the swimming exercise SD-Rat group, the other was the non-exercise SD-Rat group. Sample size was seven Rats repectively. Exercise period was ten week. Exercise group swimmed twenty minutes per a day and 5 times per a week. And then collecting blood from these two group's SD-Rats, making M Health center a request for a blood test on TC, TG, GOT, GPT, we come to a conclusion like below. The weight of the swimming exercise group has decreased 5.93% in comparison with non-exercise group. and has a significant difference(p<0.05). The liver weight of the swimming exercise group has decreased 7.83% in comparison with non-exercise group, and has not a significant difference. The TC of the swimming exercise group has decreased 39.22% in comparison with non-exercise group, and has a significant difference(p<0.05). The TG of the swimming exercise group has decreased 62.88% in comparison with non-exercise group, and has a significant difference(p<0.05). The GOT of the swimming exercise group has decreased 3.22% in comparison with non-exercise group, and has not a significant difference. The GPT of the swimming exercise group has decreased 16.14% in comparison with non-exercise group, and has a significant difference(p<0.05). In regard to above results, the regular swimming exercise therapy with dosage of high fat diet has an important role in healing and preventing a fattyliver, a hyperlipidemia, and an arteriosclerosis, intervening the lipid in blood.

  • The transgene, pFV4CAT, containing CAT reporter gene regulated by carp $\beta$-actin promoter, was expressed in independent transgenic mud loach germ lines, determined by reverse transcriptase-PCR (RT-PCR) and enzyme-linked immunosorbant assay (ELISA). Expression of the transmitted transgene was found to be tissue-specific in F1 and F2 generations. Tissue specificity of the expression was dependent on each transgenic line with reproducible patterns. Liver and spleen did express the transgene more frequently than other tissues tested, and muscle and heart revealed the higher amount of CAT than other tissues, while testes showed the lowest expression level. The highest level of CAT expression in muscle from a transgenic F1 line was corresponding to 68-fold compared to the basal levels of controls.

Effect of $\alpha$-Tocopherol and $\beta$-Carotene Supplementation on Oxidative Damage by Lipid Oxidation in Rat Liver

  • The effect of ${\alpha}$-tocopherol and ${\beta}$-carotene supplementation on reducing the oxidative damag in the liver of rats were studied. Forth-five male Sprague Dawley aged 4 weeks were randomly assigned to 9 groups of five for the 12 weeks of the study. Nine groups, sardine oil, sardine oil+Vt E, sardine oil+${\beta}$-carotene, soybean oil, soybean oil+Vt E, soybean oil+${\beta}$-carotene, lard, lard+Vt E, lard+${\beta}$-carotene group, were prepared. Sardine oil, soybean oil, or lard was used for dietary fat and 200% of ${\alpha}$ -tocopherol or 150% of ${\beta}$-carotene was supplemented to each diet. Each diet supplied 65% of total energy as carbohydrate, 15% as protein, and 20% as lipid. The MDA value and protein carbonyl contents of sardine oil group were significantly different(p<0.05) to those of other fat groups indicating that the most severe lipid oxidation occurred in the group fed diet containing highly polyunsaturated fatty acid. When ${\alpha}$-tocopherol or ${\beta}$ -carotene was supplemented to the sardine oil diet, MDA value(-35%, -15%, respectively) and protein carbonyl content(-44%, -32%, respectively) decreased significantly(p<0.05). Cu, Zn-superoxide dismutase(SOD) and catalase activities of three different sardine oil groups with or without antioxidants were lower than those of soybean oil or lard group. The reducing effect of ${\alpha}$-tocopherol on oxidative damage in sardine oil group supplemented with ${\alpha}$-tocopherol was noticeable(p<0.05). However the adverse effect of ${\beta}$-carotene was observed. SOD and catalase activities of ${\beta}$-carotene supplemented groups were that the lowest among the same fat groups, but the differences were not statistically significant. The possible cause of decreased enzyme activity seemed to be related to the vitamin A(Vt A) toxicity in the liver where retinol converted from dietary ${\beta}$-carotene in the intestinal mucosa was stored.

  • We have isolated fractions from Cordyceps militaris and Paecilomyces japonica and investigated their effects on the activity of rat liver cytosolic glucokinase, a key metabolic enzyme involved in carbohydrate metabolism. The dried powder of the C. militaris and P. japonica were successively extracted with ethanol and with 70% ethanol. The residue was exhaustively extracted with hot water. The extract was dialyzed against water, and to the non-dialyzable solution was added 2 volumes of ethanol. The precipitate was collected by centrifugation dispered in water, and lyophilized to afford fraction A. The residue after hot-water extraction was suspended in 5% sodium carbonate. The final residue was suspended in 5% NaOH. The alkaline suspension was purified in a similar manner as described above to afford fraction B. Hepatic glucokinase activities of the fraction A extracted from C. militaris and P. japonica were 371.4 and 379%, respectively. The fraction B was 314.2 and 147.4%. The activity of fraction B of C. militaris extracts was higher than that of P. japonica. Liver cytosolic glucokinase activity of rats fed normal diet supplemented with 0.1% C. militaris was 1316%. In conclusion, the present study has demonstrated that C. militaris extracts were able to prevent sudden postprandial peaks in blood glucose as a result of a marked increase in the liver cytosolic glucokinase activities.

Effects of the Antioxidative Components to Ginsenoside in the Liver of 40-week-old Mice (40주령의 생쥐 간에 미치는 ginsenoside의 항산화효과)

  • To study on antioxidant effects of saponin fractions, we investigated effects in the liver of 40-week-old mice to which were pretreated with 5 mg/kg per body weight of saponins for 5 days. The ability of saponins to protect against oxidative damage to the mouse liver was examined by determining the level malondialdehyde (MDA),hydrogen peroxide and the activity of superoxide dismutase (SOD) and catalase (CAT). The only panaxadiol (PD)among the ginseng saponin fractions significantly increased the hepatic SOD activities (p<0.01), Whereas PD, panaxatriol (PT), ginsenoside Rd (G-Rd) (p<0.01) and ginsenoside Re (G-Re) (p<0.05) significantly decreased the contents of hydrogen peroxide. It was only G-Rd that significantly increased CAT activities (p<0.05). The level of MDA was significantly decreased by G-Rd and PD. In conclusion, PD and G-Rd among the saponin fractions were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation.

The Identification of Type II DNA Topoisomerase-Associated Protein Kinase Activity from Regenerating Rat Liver (재생 쥐간에서 분리한 DNA topoisomerase II에 결합된 protein kinase 활성)

  • We have found a protein kinase activity that is tightly associated with type II DNA topoisomerase purified from regenerating rat liver. The activities of protein kinase and topoisomerase II were not separable when the enzyme was subjected to analytical chromatographies (Hydroxyapatite, phosphocellulose, and double strand DNA cellulose) and glycerol gradient sedimentation. The kinase activity from purified rat topoisomerase II was also inactivated by the topoisomerase II inhibitors such as N-ethylmaleimide or novobiocin. The evidences, however, do not rule out a possibility that the kinase activity resides in a polypeptide other than the topoisomerase II protein. The topoisomerase II-associated protein kinase required Mg++ for its activity, and this requirement was not substituted by any other mono- or divalent ions. Histone H1 act as a strong stimulator and a good substrate for the kinase activity and other histones and ${\alpha}$-casein could not substitute the effect of histone H1.

Identification of Polymorphisms in CYP2E1 Gene and Association Analysis among Chronic HBV Patients

  • Cytochrome P450 2E1 (CYP2E1) is a member of the cytochrome P450 superfamily, and it is a key enzyme responsible for the metabolic activation of many smallmolecular-weight compounds such as alcohol, which is classified as a human carcinogen. In this study, we identified 19 single nucleotide polymorphisms (SNPs) in CYP2E1 in Korean population. In these SNPs, we examined possible genetic association of CYP2E1 polymorphisms with HBV clearance and the risk of hepatocellular carcinoma (HCC). Five common polymorphic sites were selected, CYP2E1 polymorphisms at rs381-3867, rs3813870, rs2070673, rs2515641 and rs2480257, considering their allele frequencies, haplotype-tagging status and LDs for genotyping in larger-scale subjects (n=1,092). Statistical analysis demonstrated that CYP2E1 polymorphisms and haplotypes show no significant association with HBV clearance, HCC occurrence and onset age of HCC (p>0.05). Previous studies, however, have shown contradictory findings on associations of CYP2E1 polymorphisms with CYP2E1 activities and HCC risk. Comparing the contrasting results of previous researches suggest that CYP2E1 polymorphism is associated with CYP2E1 activity induced by ethanol, but is not directly associated with HCC risk. CYP2E1 variation/haploype information identified in this study will provide valuable information for future studies on CYP2E1.

Primary Hepatosplenic B-cell Lymphoma: linitial Diagnosis and Assessment of Therapeutic Response with F-18 FDG PET/CT (원발성 간비장 B세포성 림프종에서 치료전과 치료 후 F-18 FDG PET/CT 소견)

  • A 52-year-old woman with a history of general weakness, fatigue, weight loss, elevated serum levels of liver transaminase enzyme for three months underwent an F-18 FDG PET/CT scan to evaluate a cause of the hepatosplenomegaly found on abdominal ultrasonography. Initial PET/CT revealed markedly enlarged liver and spleen with intense FDG uptake. Otherwise, there were no areas of abnormal FDG uptake in whole body image. Histological evaluation by a hepatic needle biopsy demonstrated diffuse large B cell type lymphoma and final diagnosis for this patient was hepatosplenic B-cell lymphoma. She received five cycles of CHOP chemotherapy, and second PET/CT scan was followed after then. Follow-up PET-CT revealed normal sized liver with disappearance of abnormal FDG uptake. Hepatosplenic B-cell lymphoma is relatively rare and mostly presents as single or multiple nodules.1,2 Diffuse type hepatosplenic lymphoma is extremely rare and poorly recognized entity.3 The diagnosis is very difficult and complicated by the presence of misleading symptoms.4 In this rare hepatosplenic B-cell lymphoma case, F-18 FDG PET/CT scan provided a initial diagnostic clue of hepatosplenic lymphoma and an accurate chemotherapy response.
