Effect of Feeding Eggs on Levels of Serum Lipid, Protein, and Enzyme Activities in Rats (계란 섭취수준이 흰쥐 혈청의 지질, 단백질, 칼슘 농도 및 효소활성에 미치는 영향)

  • The effects of dietary eggs on liver and serum lipids, serum protein and mineral concentrations, and enzyme activities in male rats were studied. Sprague-Dawley rats were fed four types of diets for 4 weeks, respectively: a control diet, a control diet supplemented with 5, 10 or 15% eggs powder. In rats fed 5, 10 or 15% egg diets the body weight gain, the food intake, the weights of kidney and epididymal fat pad were similar to those in rats fed the control diet. The hepatic weight, hepatic cholesterol and triglyceride concentrations of rats fed the all egg diets were significantly higher than those of rats fed the control diet. The concentrations of total cholesterol, HDL-cholesterol, LDL+VLDL-cholesterol, triglyceride and phospholipid in the serum of rats fed the all egg diets were similar to those of rats fed the control diet. In the rats fed 5% egg diet the HDL-cholesterol/total-cholesterol ratios was significantly increased, and the atherogenic index was significantly decreased compared with those in the rats fed 10 or 15% egg diets. The serum total protein and albumin concentrations of rats fed 10 or 15% egg diets were significantly increased compared with those of rats fed the control diet. The serum calcium levels of rats fed the all egg diets were significantly increased compared with those of rats fed the control diet. No differences were noted in the concentrations of urea nitrogen, creatinine, glucose and hemoglobin, and activities of GOT, GPT, ${\gamma}-GTP$ and alkaline phosphatase in the serum among the rats with on all the experimental diets. These results showed that the all egg diets feeding increased cholesterol and triglyceride concentrations in liver, but no differences lipids and lipoprotein cholesterol concentrations in serum of rats.

  • The purpose of this paper was to study the effect of sulpyrin on the absorption, excretion, metabolism, and protein binding of ampicillin in the small intestine of the rats and rabbits. The results are as follows; The absorption of ampicillin in small intestine of rats was increased by the combination of sulpyrin and ampicillin. The blood level of ampicillin in rabbits was elevated by oral administration of sulpyrin. The bioavailability of ampicillin was increased by simultaneous administration of sulpyrin and ampicillin. The urinary excretion of ampicillin was slightly decreased by combined administration of sulpyrin. The blood level of ampicillin was decreased and the urinary excretion was increased by long term administration of sulpyrin. On the other hand, metabolising enzyme of ampicillin was influenced by long term administration of sulpyrin. Protein binding rate of ampicillin was decreased by combination of sulpyrin as compared with control.

  • The present study was carried out to investigate the effects of magnesium supplement levels and periods on lipid metabolism in male Sprague-Dawley rats given low protein and magnesium deficient diets. The effect of magnesium supplement levels and periods on lipid metabolism in rats given a low protein and magnesium deficient diet for 2 weeks were investigated. Serum total lipid and triglyceride contents were significantly lower in magnesium supplement group compared with magnesium deficient group. Serum HDL-cholesterol/total cholesterol ratio was significantly increased as magnesium supplement level was increased. Liver total lipid, triglyceride, total cholesterol and phospholipid contents were significantly lower in magnesium supplement group than those in magnesium deficient group. Serum ALP, GOT and GPT activities were significantly decreased in magnesium supplement group compared with magnesium deficient group. In summary, the effect of magnesium supplement on lipid metabolism and enzyme activities were significant and we can see that magnesium supplement level propered to be requirement level(400 mg/kg diet)in the other cases except serum lipid contents.

  • It has been known that the ginseng extracts activate the lipid metabolism in animal body. The experiments were undertaken to elucidate the effects of total, diol, and trial saponin of ginseng on the activities of acyl Co-A synthetase and hydroxyacyl Co-A dehydrogenase involved in fatty acid metabolism in normal albino rat liver. The acyl Co-A synthetase activity, in vitro, was increased by 20% with treatment of 2.5${\times}$10-3% total saponin, by 14% with 2.5${\times}$10-3% diol saponin, arid 30% with 2.5${\times}$10-4% triol saponin, respectively. And the enzyme activity was increased by 27% at 2 hours after intraperitoneal injection of total saponin. Hydroxyacyl Co-A dehydrogenase activity, in vitro, was increased by 77% with 10-4% total saponin, by 64% with 10-2% diol saponin, and by 72% with 10-3% triol saponin, respectively. Also, the enzyme activity, in vivo, was increased by 15.3% and 33% at 2 hours and 4 hours.

  • The effects of naturally occurring furanocoumarins on cytochrome P-450 have been investigated in rat liver microsomes. Incubation of microsomes with an NADPH-generating system and four furanocoumarins such as imperatorin, isoimperatorin, phellopterin and byakangelicin at $37^{\circ}$ in vitro resulted in a significant destruction of cytochrome P-450. A single treatment(50 mg/kg, i.p.) of rats with each furanocoumarin caused a rapid loss of cytochrome P-450 accompanied by the loss of heme from the microsomes but not by the loss of cytochrome $b_5$. It is suggested that cytochrome P-450 is specifically destroyed by furanocoumarins in a metabolic process involving destruction of its heme group and as a consequence, hepatic enzyme activities are depressed markedly.

  • In order to study the mechanism of the parts of omiza (Schizandra chinensis baillon) on metabolism in normal rats, the metabolites and enzyme activities both in serum and liver were determined. The rats were treated with water extract of the parts of omiza and the results showed a significant decrease of GOT, Glucose (excepted for water extract of fruits), Urea nitrogen, and increase of LDH in serum. Free fatty acid level tended to decrease in serum of rats treated with water extracts of fruits and endocarps and increase in seeds extract treated group. Serum GPT level was unchanged. The level of hapatic metabolites and enzyme activity showed a significant increase, but Pyruvate level was not significantly decreased.

  • Previous studies have shown that methanol extract and its butanol fraction of Carthamus tinctorius L. Semen have the hepatoprotective effect on the CCl4-induced hepatotoxicity. The hepatoprotective effect of subfractions has been evaluated by analyzing blood and hepatocyte biochemical analyses and biotransformation enzyme analyses. Treatment of BS-5, subfraction has significantly decreased the activities of alanine aminotransferase and aspartate aminotransferase. In addition, the levels of cholesterol and triglyceride in liver have been decreased as compared with that of CCl4 treated rats. The hepatoprotective effect of BS-5, subfraction on the CCl4-induced hepatotoxicity would be mediated of the attenuation of the level of cytochrome P450 and the enhancement of the activity of glutathion S-transferase.

  • Yeast alcohol dehydrogenase (YADH) has an acidic residue that interacts with the 2'- and 3'-hydroxyl groups of the adenosine ribose of the $NAD^+$ coenzyme. The acidic residue of Asp-223 (according to horse liver alcohol dehydrogenase amino acid sequence) is supposed to determine the coenzyme specificity for $NAD^+$ rather than $NADP^+$. We mutated Asp-223 to leucine and the mutant YADH was expressed in yeast and characterized for the coenzyme specificity. The turnover numbers of mutant enzyme for $NAD^+$ and ethanol were decreased 3.5- and 4.8-fold compared to wild-type enzyme, respectively. Contrastively, catalytic specificity for $NADP^+$ was increased 13-fold. As a result, the mutant YADH also employed $NADP^+$ as a coenzyme.

  • When rubber dissolved in toluene was used as a binding material of graphite powder, the mechanical robustness of the carbon paste was guaranteed by the fast volatility of the solvent immediately after electrode construction. This characteristic of the rubber solution met qualifications for practical use of carbon paste electrodes and enabled the design of a new enzyme electrode bound with EPDM. In order to confirm whether the electrode shows quantitative electrochemical behaviors or not, its kinetic parameters, e. g. the symmetry factor (0.2), the exchange current density ($3.66\;{\mu}A/cm^2$), the capacity of the double layer ($2.0{\times}10^{-5}\;F$), the Michaelis constant ($4.39{\times}10^{-3}\;M$), the diffusion coefficient of substrate ($2.58{\times}10^{-12}\;cm^2/sec$), the time constant (0.018 sec) and other factors were investigated.

Effect of Gam-Tea on the Metabolizing Enzyme Activity of Some Free Radical and Alcohol in Rats

  • To investigate an effect of Gam-Roa tea on the free radical or alcohol detoxicating enzyme activities, the rats got a drink at the Gam-Roa tea instead of water for 3 months, and then the animals were sacrificed and obtained the following findings. The animals receiving Gam-Roa tea showed a decreasing tendency of hepatic xanthine oxithine oxidase activity and significantly incresed content of cytochrome P-450 compared with the control. Furthermore, hepatic superoxide dismutase and glutathione S-transferase activities were also more increased in rats received Gam-Roa tea than in the control group, those receiving water. On the other hand, alcohol or aldehyde dehydrogenase activities were more increased in rats receiving Gam-Roa tea than the control. In conclusion, it is likely that the liver of rats receiving Gam-Roa tea may have the oxygen free radical or alcohol detoxication potential.
