산화동물모델 흰 쥐 간세포에 대한 자근(紫根)의 황산화 효과 연구 (Study on Antioxidant Effect of Lithospermi Radix on Liver Cells Isolated from Oxidatively Stressed Rat)

  • Objective : This study purposed to investigate the anti-oxidative effect of Lithospermi Radix (root of Lithospermum erythrorhizon S.) on liver cells isolated from oxidatvely stressed rat by AAPH. Method : We investigate effects of Lithospermi Radix(LR) and its fractions on normal liver cells' proliferation. And the amounts of SOD, GSH, catalase, NO, MDA production by liver cells isolated from the oxidatively stressed rat by AAPH also were measured after incubation with various fractions of LR extraction. Results : LR and its fracitons showed no toxicity on the normal liver cells from rat. LR and its fracitons increased the activity of SOD and reduced the amounts of NO and MDA in the liver cells from the oxidatively stressed rat. Conclusion : Lithospermi Radix could be supposed to have antioxidant effect on liver cells with no toxicity.

증례(證例)를 통해 본 한약투여(韓藥投與)가 간질환(肝疾患) 환자(患者)의 간손상(肝損傷)에 미치는 영향(影響) (The effects of herbal medicine administraion on liver injury of clinical patient)

  • Objective : The studies on liver effect in administration of western medicine have been well-established so far, but the studies on liver effect in administration or herbal medicine haven't been made. To make things worse, people who have liver disease generally believe that taking a herbal medicine is not useful to take care or their disease and even think it can cause liver disease. But this belief is not verified at all. And some doctor claim that herbal medicine is good to cure liver disease. So those various assertion makes patients to feel confuse about taking a herbal medicine. so we feel the need to study about how taking herb medicine affect to liver disease patient. Methods : First we chose the 12 patient who seems to have the liver disease on the index of biochemistry test and we administrate the herbal medicine and after several day or weeks we recheck the biochemistry test. Results : Through the this method, we find the positive effect of taking herbal medicine on the patients who have the liver disease. Conclusions : Our results give no evidence that herbal medicine is harmful for liver disease. We need to study more about this.

  • When rats are exposed to cadmium in their diet, the cadmium accumulates in the liver. This study was carried out to investigate the effect of chitin, chitosan and dithiocarbamate chitosan on the cadmium accumulation in liver of rats. For this experiment, 10 male Sprague-Dawley were used. The experimental groups were divided into four independent groups which were one control group and three experimental groups by cadmium alone treatment or chitin, chitosan and dithiocarbamate chitosan with cadmium. In order to investigate the eliminative effect of chitin, chitosan and dithiocarbamate chitosan on the cadmium accumulation in liver of rats, the cadmium concentration, the metallothionein level and the enzyme activitys were measured. The results obtained revealed the eliminative effect of cadmium in liver of rats by chitin, chitosan and dithiocarbamate chitosan. The effect of chitin on the cadmium elimination was less than that of chitosan and dithiocarbamate chitosan. Also it shows that the eliminative effect of cadmium by dithiocarbamate chitosan was the highest.

The Anti-hepatotoxic Effect of Ginseng in Rats: Meta-analysis

  • The aim of this meta-analysis was to systematically investigate the anti-hepatotoxic effect of ginseng in rats induced toxicity which damage to liver. Primary researches were gained on the ScienceDirect database, the DBpia, and the KISS, and the data about the effect factors in plasma and in enzyme were listed as many as possible. The effect factors were alanine transaminase (ALT), aspartate transaminase (AST), liver aminopyrine N-demethylase (AD), liver aniline hydroxylase (AH), liver 3,4-Methylenedioxyamphetamine (liver MDA), cytochrome P450 (P450), serum alkaline phosphatase (ALP), serum lactate dehydrogenase (LDH), cytochrome b5 (Cyto b5), glutathione reductase (GR), Liver glutathione S-transferase (GST), liver glutamyltransferase (GT), Liver (${\gamma}-GCS$), serum liver 3,4-Methylenedioxyamphetamine (serum MDA), serum sorbitol dehydrogenase (SDH), serum total protein (TP), serum ${\gamma}-glutamyltransferase$ (${\gamma}-GT$). To investigate the effect of ginseng, the mean difference (MD) between the group of rats induced by toxicity (RH) and the group of rats induced by toxicity with ginseng (RHG) were combined, and the significance of MDs were tested. The combined MDs were checked the biases caused by heterogeneity among studies and the publication biases, and adjusted by using random effect model and trim and fill method, respectively. The effect about ALT, AST, ALP, LDH, SDH, TP and ${\gamma}-GT$ in plasma factors were significant, and about AD, liver MDA, P450, Cyto b5, GR, GST, GT and ${\gamma}-GCS$ in enzyme factors were significant. The treatment with ginseng supplementation was significantly effected on plasma and enzyme factors of damaged-rats.

The combined HGs checked the biases caused by heterogeneity among studies and the publication biases. Then they were adjusted by using the random effect model and trim and fill method. Although the publication biases were assumed, among all plasma factors the HGs of ALT, AST, serum MDA, SDH, TP, and $\gamma$-GT were significant, and among all enzyme factors the HGs of liver MDA, Cyto b5, GR, GST, and GT were significant. The treatment with ginseng significantly affected the plasma and enzyme levels in rats induced with toxicity.

Effect of Toluene Application to Skin on the Enhancement of Liver Injury in $CCl_4$-Pretreated Rats

  • To evaluate an effect of toluene application to skin on the enhancement of liver damage in $CCl_4$-pretreated rats, toluene (35 mg/$cm^2$) was sequentially applied for 5 days to the skin of liver damaged rats with $CCl_4$ (6 times every other day: 0.1 ml/100 g body weight-50% $CCl_4$ in olive oil) On the basis of the functional and morphological findings in rat liver, appling toluene to the skin in liver damaged animals led to the more enhanced liver damage. In addition, by applying toluene to skin of liver damaged rats, the hepatic cytochrome P450 content was somewhat more increased, but the hepatic benzylalcohol dehydrogenase activity was significantly decreased (P<0.001), whereas benzaldehyde dehydrogenase activity was not statistically changed. In conclusion, the toluene application to skin in liver-damaged rat led to enhancement of liver injury that may be due to the accumulation of toluene metabolite in liver.

Benzo(a)pyrene에 의해 유도된 간기능 장해에 미치는 쑥의 효과 (The Effect of Mugwort Extracts on the Benzo(a)pyrene-induced Hepatotoxicity in Rats)

  • Mugwort has been used as a Korean folk medicine in treating liver diseases acting as an analgesics, sedative, diuresis, choleretics. This study was perfomed to evaluate the effect of mugwort extracts on the changes of enzyme activities, lipid accumulation of the serum and liver, when hepatotoxicity was induced by benzo(a)pyrene. The results are as follows: 1. Mugwort water extract administration prevented the increase of serum and liver AST, ALT, LDH, ${\gamma}$-GTP, liver ALP activities and bilirubin content caused by B(a)P injection. 2. The increase of serum and liver ALT, LDH, ${\gamma}$-GTP, serum AST activities and liver bilirubin contents in B(a)P treated group were decreased by mugwort methanol extract treatment. 3. Serum and liver total cholesterol, phospholipid, triglyceride level and serum HDL-cholesterol level were increased by B(a)P treatment. After combined treatment of mugwort water and methanul extracts, these lipid content were significantly decreased. 4. The hepatotropic effect of mugwort water extract and after-treatment against B(a)P induced hepatotoxicity was superior to that of methanol extract and pretreatment.

Oxidant에 의한 간독성유발에 가미행체엽탕 합 육미지황탕의 효과 (Effect of Kamihaengche-tang Plus Yukmijihwang-tang Oxidant-induced Liver Cell injury)

  • This study was carried out to determine whether Kamihaengche-tang plus Yukmijihwang-tang (KCYH) exerts the protective effect against oxidant-induced liver cell injury. Cell injurt was estimated by measuring lactate dehydrogenase (LDH) and alanine aminotransferase (ALT) release, and lipid peroxidation was estimated by measuring malondialdehyde, a product of lipid peroxidation in rabbit liver slices. $H_2O_2$increased LDH release which was significantly prevented by 1% KCYHT. The protective effect of KCYH against $H_2O_2$-induced cell injury was dose-dependent in the range of 0.05-1% concentrations. Similary, KCYH inhibited $H_2O_2$ induced lipid peroxidation in a dose-dependent manner. When liver tissuse were exposed to Hg(0.5 mM), ALT activity in the medium and lipid erpoxidation in tissues were markedly increased. These changes were prevented by 1% KCYH. KCHY restored Hg-induced inhibition of cellular GSH content. These result indicate that KCYH exerts the protective effect oxidant-induced liver cell injury, and this effect is attributed to prevented to prevention of lipid peroxidation. These dffects may be due to an increase in concentration of endogenous antioxidants.

The Effects of Anti-Inflammatory and Liver Function using Heat-Treated Cabbage

  • The cabbage extract of the research does not show cytotoxicity, and thus can be used safely. In an experiment performed on an animal model with liver injury induced by a drug (APAP), it could be seen that the cabbage extract exhibited the effects of protecting liver and improving liver function by effectively reducing AST and ALT which are liver injury markers, indicating that the cabbage extract is effective as a pharmaceutical composition for preventing or treating liver disease. In particular, the cabbage extract was effective in treating inflammation of the liver by reducing the expression of the inflammatory mediators iNOS and COX-2 and the proinflammatory cytokine IL-1β, which are involved in acute inflammatory reactions accompanying liver injury. In the research, an extract of cabbage heat-treated at a temperature of 100 to 150℃ had a better liver function-improving effect or anti-inflammatory effect than an extract of raw cabbage.
