식이 다가 불포화 지방산의 n-6/n-3 비율에 의한 흰쥐의 지질수준 및 Prostaglandin 생성에 대한 연령별 비교 (The Age-Related Effect of n-6/n-3 Ratio of Dietary Fats on Lipid Levels and Prostaglandin Production in Rats)

  • The effects of various n-6/n-3 ratios(about 2, 4, 6, 8) of dietary fatty acids on various lipid levels and prostaglandin production were studied at the constant P/S ratio (1.5-1.6) in young (5 weeks old) and adult(8 months old) Sprague-Dawley rats using palm oil, safflower oil and sardine oil. The concentration of serum cholesterol tended to increase with the increasing n-6/n-3 ratio. The tendency of HDL-cholesterol levels was similar to serum cholesterol levels. These were not apparent differences between young and adults rats. Serum triglyceride levels increased according to increasing n-6/n-3 ratio in young rats. These were generally high in the adult rats compared with the young rats. Though liver cholesterol level tended to increase according to the increasing n-6/n-3 ratio in the young rats. The liver triglyceride level did not change according to the n-6/n-3 ratio. However, these levels were apparently higher in the adult than in the young rats. The fatty acid compositions of phosphatidylcholine(PC) were similar in serum and liver. The arachidonate/linoleate ratios in tissue PC were influenced by the n-6/n-3 ratio. They tended to be lower in the adult rats compared with the young rats. It was suggested that the activity of $\Delta$6-desturase was decreased by aging. Production of platelet thromboxane A2(TXA2)and aortic prostacyclin(PGI2) was not apparently influenced with n-6/n-3 ratio. Whereas the ratio of TXA2/PGI2 was the lowest value at 3.8 of n-6/n-3 ratio, expecially in the young rats. Thus this ratio seemed to be a desirable level to protect atherosclerosis. These results indicate that the lipid level and prostaglandin production were influenced not only by n-6/n-3 ratio(under constant P/S ratio) but by aging, particulary triglycerde level and arachidonic/linoleic acid ratio.

Effects of Dietary Lipid Sources on Growth and Body Composition of Snail (Semisulcospira gottschei)

  • This study was conducted to investigate the effects of dietary lipid sources on survival, growth anc body composition of snail (Semisu1cospira gottschei). Three replicate groups of snail (average weighing 152mg) were fed the diets containing different lipid sources such as lauric acid (LA), squid liver oil (SO), linseed oil (LO), corn oil (CO), SO+ LO, SO+CO, LO+CO and SO+LO+CO for 8 weeks. Survival was not affected by dietary lipid sources (p>0.05). Weight gain of snail fed the SO, SO+LO and SO+LO+CO diets was significantly higher than that of snail fed the LA and LO diets (P<0.05), and the lowest weight gain was observed in snail fed the LA diet (P<0.05). No significant difference was found in crude lipid content of edible portion in snail fed the different diets (P>0.005). Contents of 12:0, 18:2n­6, 18:3n-3 and 20:5n-3 from snail fed the LA, CO, LO and SO diets were higher than those from snail fed the other diets, respectively (p<0.05). The highest 22:6n-3 content was observed in snail fed the SO+ LO but was not significantly different from that of snail fed the SO, SO+CO and SO+LO+CO diets (p>0.05). The n-6 highly unsaturated fatty acids such as 20:4n-6 and 22:4n-6 contents of snail were not affected by dietary lipid sources (p>0.05). These results suggested that squid liver oil and mixture of squid liver oil and linseed and/or corn oil are good dietary lipid sources for the normal growth of snail. However lauric acid may not be a good lipid source for snail diet.

들기름, 옥수수기름의 섭취와 2-Acetylaminofluorene 투여가 지질과산화물 및 PG $E_2$ TX $B_2$ 생성에 미치는 영향 (Effect of Intake of Perilla oil or Corn oil and 2-Acetylaminofluorene Treatment on Lipid Peroxidation PG $E_2$ and TX $B_2$ Productions in Rats)

  • This study was conducted to compare the effects of perilla oil or corn oil on lipid peroxidation and eicosanoid productions which are associated with the promotion of carcinogenesis. in liver or blood in rats. Male Sprague-Dawley8 weaning rats were fed on semisynthetic diets containing 15%(w/w) beef fat(BF). corn oil(CO) or perilla oil(PO) Three weeks after the half of rats in each diet group were injected with a single dose of 50mg 2-acetylaminofluorene (AAF)/Kg BW hepatocarcinogen intraperitoneally 3 times at 2-day interval and all of the rats were sacrificed after 8 weeks from the first injection. The rats fed on different dietary fats without 2-AAF treatment had not different MDA produc-tion and conjugated diene content in liver microsome. CO+AAf group had significantly higher conjugated diene content than BF+AAF and PO+AAF groups. and lower glucose-6-phospha-tase activity than BF+AAF group But PO+AAF had similar conjugated diene content to BF+AAF group and significantly lower MDA production than BF+AAF and CO+AAF groups. The hepatic mocrosomal lipid peroxidation was slightly greater in CO group than in PO group though perilla oil(P/S=9.67) has much more polyunsaturated fatty acids than corn oil(P/S=2.92) PG E2 level in liver and TX B2 level in plasma were significantly higher in CO group than in BF and PO groups. TX B2 level was lowered in CO and BF groups by 2-AAF treatment. These results reach to the contclousion than the type of dietary fatty acid as well as the P/S ratio has effect on hepatic microsomal lipid peroxidation and eicosanoid production and perilla oil or linolenic acid(n3) might be less effective on lipid peroxidation or PG E2 and TX B2 mediated tumor promotion than corn oil or linoleic acid(n6).

죽엽황금복합추출물 BS21의 고지방식이 유도 비만 동물모델에서의 지방생성 및 고요산혈증 개선 효과 (BS21, a combination of Phyllostachys pubescens and Scutellaria baicalensis extracts, reduces adiposity and hyperuricemia in high-fat diet-induced obese mice)

  • Objectives : Phyllostachys pubescens and Scutellaria baicalensis are considered to be effective in promoting blood circulation in traditional medicine. In this study, we examined whether a mixture of P. pubescens leaves and S. baicalensis root (BS21) had any anti-obesity, anti-hyperlipidemia, or anti-hyperuricemia effects and the possible mechanisms of action. Methods : We examined the effects of BS21 in high-fat diet (HFD)-induced obese mice. Mice were fed HFD with BS21 (75, 150, or 300 mg/kg) or Garcinia cambogia extracts (245 mg/kg) as a positive control for 8 weeks. At the end of 8 weeks, body weight, liver and adipose weight, adipocyte size, plasma lipid profiles, adipokine and uric acid levels, and adipose tissue expression levels in obesity and uric acid production-related genes were examined. Results : BS21 decreased body weight gain, white adipose tissue, liver weight, adipocyte size, and liver triglyceride accumulation. It also reduced levels of plasma glucose, triglycerides, non-esterified fatty acids, total cholesterol, low-density lipoprotein cholesterol, alanine transaminase, leptin, and uric acid. In contrast, BS21 increased adiponectin levels. Furthermore, BS21 decreased the expression levels of adipogenesis-related genes, such as peroxisome proliferator-activated receptor γ, sterol regulatory element binding protein-1c, and fatty acid synthase, as well as xanthine oxidoreductase, which is involved in uric acid production. Conclusions : These results suggest that BS21 may exert anti-obesity, anti-hyperlipidemia, and anti-hyperuricemia effects in HFD-induced obese mice by regulating the expression of xanthine oxidoreductase and adipogenesis-related genes.

Voluntary exercise training improves body weight of leptin-deficient ob/ob mice by altering hepatic stearoyl-CoA desaturase 1 and deleted in breast cancer 1 protein levels

  • [Purpose] Deleted in breast cancer 1 (DBC1) ablation causes obesity, and stearoyl-CoA desaturase 1 (SCD1) induces the biosynthesis of monounsaturated fatty acids. This study examined whether voluntary wheel running (VWR) alters SCD-1 and DBC1 protein levels in the liver of leptin-deficient ob/ob mice. [Methods] Twenty-five Ob/Ob mice were divided into two groups (ob/ob-Sed and ob/ob-Ex). The expression of DBC1 and SCD1 in the mouse liver was determined using western blotting. [Results] After 10 weeks, VWR significantly reduced body weight without affecting the fatty acid synthase and CD36 protein levels. The average daily running distance was 4.0±1.0 km/day. This improvement was associated with changes in the hepatic SCD1 and DBC1 levels. Hepatic SCD-1 protein levels increased significantly, and DBC1 protein levels decreased in ob/ob-Sed animals. On the other hand, VWR inhibited the obesity-induced increase in SCD1 expression and impaired the obesity-induced decrease in DBC1 expression in the liver of leptin-deficient ob/ob mice. [Conclusion] This is the first study showing that VWR has strong effects on hepatic SCD1 and DBC1 in ob/ob mice, and provides key insights into the effects of exercise on obesity.

한국산 잣기름이 정상토끼의 혈중 지방질 및 지단백질의 대사에 미치는 영향 (Effect on Blood Lipids and Lipoproteins of A Supplement of Korean Pinenut Oil, rich in 5-Olefinic Acids, in Normocholesterolemic New Zealand White Rabbits)

  • The present study was carried out in normocholesterolemic New Zealand white(NZW) rabbit, to evaluate the effect of dietary supplementation with Korean pinenut oil, on plasma lipids, plasma lipoproteins, liver lipids and platelet aggregation. NZW rabbits were fed for 80 days on a commercial chow diet supplemented with 5% of energy as fats(soybean oil or pinenut oil) or 10% of energy as fats(soybean oil or pinenut oil). A control group was fed a commercial stock diet. There were no significant effects of pinenut oil on plasma free cholesterol, HDL-cholesterol, triglycerides, glucose and lecithin-cholesterol acyltransferase, as compared with those obtained from rabbits fed the soybean oil diet. After 80 days, the concentration of plasma free fatty acid in only the pinenut oil group was significantly decreased by about 50% relative to the control diet. At the end of the dietary treatment, liver triglycerides and phospholipids were significantly decreased in the pinenut oil group, compared to the how diet, whereas the soybean oil-consuming rabbits had only significantly decreased phospholipid levels. Cholesterol contents of liver were unaffected by type of dietary fat. At the end of 80 days, a diet containing pinenut oil resulted in a decrease in apolipoprotein B and the apo B/apo AI ratio as compared with the stock diet or soybean oil diet. Platelet aggregation induced by collagen or arachidonic acid was depressed significantily in pinenut oil diet.

산화안정성이 높은 어유 캡슐 제품 개발

  • 강원도 동해안에서 다량 어획되어 가공되는 오징어 및 명태 내장으로부터 추출되는 내장유 용도 확대 및 고부가가치화를 위한 미세 캡슐화를 제조를 위한 조건을 시험하였다. 미세 캡슐 소재로 wall material은 gum arabic과 유화제로 gelatin이 적절하고, gum arabic과 gelatin을 소재로하여 배합비를 달리하였을 때, 전체적인 경향을 유화제가 wall material 보다 많은 것이 양호한 미세 캡슐이 된다는 것을 확인하였다. 상기한 소재로 제조된 미세 캡슐의 캡슐화율은 25-45.5% 정도의 범위로 전체적으로 미세캡슐화율은 낮았다. 향후 CMC, maltodextrin 그리고 키토산 등을 소재로 미세 캡슐화 조건을 좀 더 검색할 필요가 있을 것으로 판단되었다.

Involvement of Endoplasmic Reticulum Stress in Palmitate-induced Apoptosis in HepG2 Cells

  • The results of recent studies indicate that high levels of free fatty acids(FFAs) and adipokines may be the main causes of non-alcoholic liver disease; however, the molecular mechanism that links FFAs to lipotoxicity remains unclear. In the present study, we treated HepG2 cells with FFA(either palmitate or oleate) to investigate the mechanisms involved in lipotoxicity in the liver cells. We also treated cells with palmitate in the presence of a chemical chaperone, 4-phenylbutyric acid(PBA), to confirm the involvement of ER stress in lipotoxicity. Palmitate significantly induced cytotoxicity in dose- and time-dependent manners. Apoptosis was also significantly induced by palmitate as measured by caspase-3 activity and DAPI staining. Palmitate led to increased expressions of the spliced form of X-box-protein(Xbp)-1 mRNA and C/EBP homologous transcription factor(CHOP) protein, suggesting activation of the unfolded-protein response. PBA co-incubation significantly attenuated apoptosis induced by palmitate. The above data demonstrate that high levels of palmitate induce apoptosis via the mediation of ER stress in the liver cells and that chemical chaperones act to modulate ER stress and accompanying apoptosis.

배추김치 용매획분이 고 콜레스테롤 식이를 섭취한 토끼의 항산화 효소계 및 인지질 지방산 조성에 미치는 영향 (Effects of Solvent Fractions of Korean Cabbage Kimchi on Antioxidative Enzyme Activities and Fatty Acid Composition of Phospholipid of Rabbit Fed 1% Cholesterol Diet)

  • The antioxidant effects of dichloromethane, ethylacetate or water fraction of kimchi added to the 1% cholesterol diet were studied. Six New Zealand white rabbits in each group were fed either control diet (basal diet containing 1% cholesterol) or experimental diet containing dichloromethane (CH$_2$Cl$_2$), ethylacetate (EtOAc) or water ($H_2O$) fraction of kimchi in the control diet for 16 weeks. The amount of each solvent fraction of kimchi added to the experimental diet was equivalent to 5% of freeze-dried kimchi. Levels of hepatic lipid oxidation expressed as TBARS or peroxide value for the experimental groups were lower than that of control (p<0.05). Liver homogenated of the experimental group containing dichloromethane fraction of kimchi inhibited LDL oxidation in the presence of Cu++ by 46% (p<0.05). The activities of catalase, Glutathione peroxidase (GSH-Px), Cu, Zn-superoxide (Cu, Zn-SOD) and Mn-superoxide (Mn-SOD) of experimental groups were lower than those of control group. Low enzyme activities observed from the kimchi solvent fraction groups might be due to the level of lipid oxidation progressed less in these groups. The most significant antioxidant effects were observed from dichloromethane fraction of kimchi among the experimental groups. The major fatty acids of hepatic phospholipid of rabbit were C18:2 and C18:0. But the major fatty acid profile was changed into C16:0, C18:0, C18:1, and C18:2 when rabbit was fed 1% cholesterol diet for 16 weeks, and this profile was almost the same as in rabbit fed diet containing kimchi solvent fraction. The ratio for unsaturated fatty acid to saturated fatty acid decreased by cholesterol induced diet and it was not corrected by kimchi solvent fractions.

과량의 비타민 E 첨가 및 다불포화지방 식이가 Age가 다른 Chick의 혈청과 조직 비타민 E 농도에 미치는 영향 (Effects of Dietary Polyunsaturated Fatty Acids and Vitamin E Supplementation on Vitamin E Levels of Serum, Liver and Brain in Chicks at Different Ages)

  • 혈청내 비타민 E 농도는 부화후 1주된 군과 4주된 군 모두에서 비타민 E를 첨가해 준 II(CE)군과 IV(PE)군이 그렇지 않은 I(C)군과 III(P)군보다 각각 더 높았고, 식이내 비타민 E의 혈중 비타민 E 농도에 대한 반영은 부화후 1주된 군이 부화후 4주된 군보다 더 컸다. 2. 부후화 1주된 군의 경우 실험 3주에 비타민 E를 더 첨가해준 군(II군과IV)에서 PUFA에 의한 혈청 비타민 E 감소효과가 나타났다. 3. 간조직내 비타민 E 농도는 혈청에서와 마찬가지로 부화후 1주된 군과 4주된 군 모두에서 II(CE)군과 IV(PE)군 I(C)군과 III(P)군보다 높았고 (p<0.05), 실험 3주에 부화후 1주된군과 4주된 군에서 비타민 E를 더 참가해 준 경우에 과량의 PUFA에 의한 간조직내 비타민E 농도 감소를 보였다. 4. 실험 3주에 부화후 4주된 군의 뇌조직내 비타민 E 농도가 실험 초기에 비해 증가하는 경향을 보인것 이외에는 식이에 따라 뇌조직내 비타민 E 농도에 큰 변화가 없었고, 모든 실험 군에서 뇌조직의 비타민 E 농도는 혈청과 간조직 내의 비나민 E 농도에 비해 전반적으로 변화가 적으므로 혈청이나 간조직보다는 식이내의 비타민 E의 영향을 적게 받는 것으로 나타났다.
