To determine the optimum fermentation condition, the 50:50 feed mixture of both crushed corn and wet brewer’s grain were anaerobically fermented at various conditions. Three supplementation levels(0, 2%, and 5%) of molasses, five supplementation levels(0, 0.1%, 0.5%, 1.0%, and 2.0%) of yeast and five different incubation temperatures(4$^{\circ}C$, 10$^{\circ}C$, 30$^{\circ}C$, 40$^{\circ}C$ and 50$^{\circ}C$) were tested to determine the optimum fermentation conditions. During fermentation, alcohol concentration, live yeast cell number (LYCN) and pH values of the mixture were analyzed. Alcohol concentrations of the fermented feed mixtures were proportionally increased with increasing level of molasses supplementation. After 24hr fermentation, alcohol concentrations at 0.1%, 0.5%, 1.0% and 2.0% of yeast supplementation was 1.9%, 2.4%, 3.1% and 3.1%, respectively. These results indicate a proportional increase of alcohol concentration with the increasing supplementation of yeast (P<0.05) up to 1%, thereafter showing plateau at 2% supplementation. LYCN was relatively high with more than 0.5% yeast supplementation, whereas the value was significantly low(P<0.05) at 0.1% yeast supplementation. Alcohol production was significantly higher at 30$^{\circ}C$ and 40$^{\circ}C$ than other incubation temperatures (P<0.05), with the highest concentration at 30$^{\circ}C$. pH was rapidly decreased until 24 hours of incubation with steeper decrease at lower temperatures(P<0.05). Based on there results, optimum fermentation conditions for the alcoholic-fermented feed production could be achieved with 5% supplementation of molasses, 1% supplementation of yeast, and 24hr long incubation at 30$^{\circ}C$.
Probiotics are microorganisms that can benefit host health when ingested in a live state, and lactic acid bacteria are the most common type. Among fungi, Saccharomyces boulardii (SB) is the only strain known to have a probiotic function with beneficial effects on colitis; however, information on other probiotic yeast strains is limited. Therefore, this study aimed to discover yeast strains expressing intestinal anti-inflammatory activities by exhibiting probiotic properties in dextran sodium sulfate (DSS)-induced colitis mice model. Nuruk (Korean traditional fermentation starter) containing various microbial strains was used as a source for yeast strains, and S. cerevisiae 28-7 (SC28-7) strain was selected with in vitro and in vivo characteristics to enable survival in the intestines. After 14 days of pretreatment with the yeast strains, DSS was co-administered for six days to induce colitis in mice. The results revealed that the disease activity index score was lowered by SC28-7 treatment compared to the DSS group, and the colon length and weight/length ratio were recovered in a pattern similar to that of the normal group. SC28-7 administration significantly reduced the secretion of pro-inflammatory cytokines in the serum and modified the mRNA expression of inflammatory cytokines (interleukin-1β, transforming growth factor-β, and interferon-γ) and proteins involved in gut barrier functions (mucin 2, mucin 3, zonula occludens-1, and occludin) in colon tissues. These results indicate that SC28-7 attenuates DSS-induced colon damage and inflammation, supporting its future use as a probiotic yeast for treating and preventing intestinal inflammatory diseases such as inflammatory bowel disease.

We present a high-throughput continuous cell separation chip using hydrodynamic dielectrophoresis (DEP) process. The continuous cell separation chip uses three planar electrodes in a separation channel, where the positive DEP cells are moved away from the central streamline while the negative DEP cells remain in the central streamline. In the experimental study, we use the mixture of viable (live) and nonviable (dead) yeast cells in order to obtain the continuous cell separation conditions. For the conditions of the electric fields frequency of 5MHz and the medium conductivity of $5{\mu}S/cm$, the fabricated chip performs a continuous separation of the yeast cell mixture at the varying flow-rate in the range of $0.1{\sim}{\mu{\ell}/min$.; thereby, resulting in the purity ranges of $95.9{\sim}97.3\%\;and\;64.5{\sim}74.3\%$ respectively for the viable and nonviable yeast cells. present chip demonstrates the constant cell separation performance for varying mixture flow-rates.

The study was conducted to investigate the chemical composition of distillery yeast sludge and its inclusion in broiler diets to replace canola meal. Raw distillery yeast sludge was washed with water using water and sludge in the ratio 6:1, respectively. Proximate analysis of raw distillery yeast sludge and washed distillery sludge was carried out for crude protein (CP), true protein (TP), ether extract (EE), ash, acid insoluble ash and nitrogen free extract (NFE) determination. Mineral contents and amino acid profile of raw distillery yeast sludge and washed distillery sludge were also determined. After chemical evaluation, four iso-caloric and iso-nitrogenous broiler starter and finisher diets were prepared in mash form using 0 (control), 4, 8 and 12% levels of washed distillery sludge replacing canola meal. One hundred and twenty day-old broiler chicks were randomly distributed into 12 experimental units in such a way that each diet was offered to three experimental units, each comprising of 10 chicks. It was observed that washing affected the nutrients either by decreasing or increasing their concentration. It decreased the total mineral contents whereas CP, TP, EE and NFE contents increased. Washing also increased amino acid profile. Average feed intake and weight gain were higher in birds fed diet containing 8% washed distillery sludge and lower in birds fed diet containing 0% washed distillery sludge. Feed cost per kg live weight gain decreased significantly as the level of washed distillery sludge was increased in the diet. Average heart, liver and pancreas weights decreased with increased level of washed distillery sludge in the diet. The study revealed that after washing, distillery yeast sludge can be used successfully in broiler diets up to the level of 8% without any adverse effect on broiler's performance.
An experiment was carried out to study the effects of exogenous enzymes (cellulases and proteases), yeast culture and effective micro-organism (EM) culture on feed digestibility and the performance of rabbits fed rice bran rich diets over a period of ten weeks. Twenty four, 8 to 9 weeks old male and female New Zealand White rabbits were allotted to 4 dietary treatments; a basal (control) feed containing 43% rice bran, basal feed supplemented with either enzymes, yeast culture or EM. Individual feed intake, body weight gain, nutrient digestibility, carcass characteristics and feed cost were studied. Sex of the rabbits had no significant (p<0.05) influence on the parameters studied. The control group showed the lowest daily feed intake (104.8 g), body weight gain (12.8 g) and the highest feed/gain ratio (8.20 g/g). The highest daily feed intake (114.3 g), body weight gain (20.42 g) and the lowest feed/gain ratio (5.60) were observed with enzymes. Compared to the control, yeast significantly (p<0.05) improved the feed intake, body weight gain and feed/gain ratio by 4.9, 34.4 and 22.0%, respectively, while EM improved (p<0.05) them by 4.0, 32.6 and 21.6%, respectively. All the additives improved (p<0.05) the digestibility of dry matter, crude protein, crude fiber and energy by 4.9-8.7, 3.6-10.7, 5.9-8.3 and 4.3-6.4%, respectively. Higher weights of pancreas (by 38.5-56.4%) and caecum (by 13.1-26.8%, compared to the control) were recorded with all additives but liver weight was increased only by yeast (24.5%) and enzymes (26.7%). Significantly (p<0.05) higher carcass recovery percentages were observed with enzymes (60.55), yeast (60.47) and EM (56.60) as compared to the control (48.52). Enzymes, yeast and EM reduced (p<0.05) the feed cost per kg live weight by 23.8, 15.9 and 15.5%, respectively. Results revealed that enzymes, yeast culture and EM can be used to improve the feeding value of agro-industrial by-products for rabbits in Sri Lanka and thereby to reduce the feed cost. Under the present feeding system, enzyme supplement was the best.
The harpacticoid copepod, Tigriopus japonicus is one of the most important zooplankton as a live food for the production of marine fish fry. Thus, the salinity tolerance and the optimum culture environment of this copepod in terms of salinity, temperature and light were examined. The food values of 6 kinds of phytoplankters and 2 kinds of yeast were also investigated for mass culture of this copepod. The results are as follows: After 5 day culture in the experiment of salinity tolerance, the survival rates of the gravid female at $0\%\;and\;90%o\;were\;40\%\;and\;70\%$, respectively. However, at salinity ranging from $2\%o\;to\;80\%o$, high survival rates above $85\%$ were observed. It means T. japonicus is very euryhalinous species. Temperature was more important factor than salinity for the fecundity of T. japonicus. The optimum culture conditions of this species were $24^{\circ}C,\;24\%o$, and 3,000 lux with 24 L: 0D. Under these culture conditions, the average fecundity from a gravid female per spawning was 38 nauplii, and the interval time between spawnings were 2.05 days. Phaeodactylum tircornutum seemed to be the most suitable phytoplankton as a live food for T. japonicus, and the large chlorophyta, Tetraselmis suecica showed the lowest food value among 6 phytoplankters and 2 yeasts. The food value of w-yeast was better than that of baker's yeast, and it is similar to that of phytoplankton such as Amphora sp., Chlorella ellipsoidea and Nannochloris oculata. So, the w-yeast seems to be appropriate food source for mass culture of T. japonicus.
This study evaluated the effect of Candida norvegensis (C. norvegensis) viable yeast culture on in vitro ruminal fermentation of oat straw. Ruminal fluid was mixed with buffer solution (1:2) and anaerobically incubated with or without yeast at $39^{\circ}C$ for 0, 4, 8, 16, and 24 h. A fully randomized design was used. There was a decrease in lactic acid (quadratic, p = 0.01), pH, (quadratic, p = 0.02), and yeasts counts (linear, p<0.01) across fermentation times. However, in vitro dry matter disappearance (IVDMD) and ammonia-N increased across fermentation times (quadratic; p<0.01 and p<0.02, respectively). Addition of yeast cells caused a decrease in pH values compared over all fermentation times (p<0.01), and lactic acid decreased at 12 h (p = 0.05). Meanwhile, yeast counts increased (p = 0.01) at 12 h. C. norvegensis increased ammonia-N at 4, 8, 12, and 24 h (p<0.01), and IVDMD of oat straw increased at 8, 12, and 24 h (p<0.01) of fermentation. Yeast cells increased acetate (p<0.01), propionate (p<0.03), and butyrate (p<0.03) at 8 h, while valeriate and isovaleriate increased at 8, 12, and 24 h (p<0.01). The yeast did not affect cellulolytic bacteria (p = 0.05), but cellulolytic fungi increased at 4 and 8 h (p<0.01), whereas production of methane decreased (p<0.01) at 8 h. It is concluded that addition of C. norvegensis to in vitro oat straw fermentation increased ruminal fermentation parameters as well as microbial growth with reduction of methane production. Additionally, yeast inoculum also improved IVDMD.
Methods of separating yeast cells from oil-water-cell emulsion and subsequent purification of the recovered yeast have been studied. In addition, the results of preliminary feeding experiments in which a yeast grown on gas oil was incorporated into chick rations are reported. According to the present study, it appears that the recovery of the yeasts would be easier at pH 9, since the emulsion is relatively more unstable. A class of surface active agent at a concentration of 0.3% was found to facilitate the separation of the yeast from the emulsion. The use of electrolytes such as NaCl and KCl were found to be most effective in breaking the emulsion. Solvent treatment using iso-propyl alcohol and its azeotropic mixture with hexane at $58^{\circ}C$ are particularly suitable for purification of the yeast. In the feeding experiment it was found that 5 percent of the fishmeal in the control ration could be replaced by the yeast with no adverse effect on performance. However, when 8 percent of the fish meal in the control ration was replaced by the yeast, some effect on live-weight gain of the chicks was observed.
This study was designed to evaluate the effects of dietary prebiotic (mannan oligosaccharide = MOS) and probiotic (Saccharomyces cerevisiae = SC) in finishing turkey diets on carcass, cut yield, meat composition and colour. A total of 72 ten-weeksold Big6 male turkey poults were used in the trial. There were eight replicate floor pens per floor with three birds in each. The experiment lasted up to 20 wks of age. The trial was set up as a completely randomized design with 3 dietary treatments. The treatments were: i) negative control (C, no additive); ii) MOS 1 g per kg of diet and iii) SC 1 g per kg of diet (strain SC47, $300{\times}10^{10}$ CFU/kg). Body weight (BW) and feed intake were determined for each of the two week intervals. Twenty-four birds were slaughtered and eviscerated to determine carcass, carcass parts and internal organ weights at 20 wks of age. Meat colour and pH levels were measured 24 h after slaughter. The dietary treatments did not affect BW and average daily gain during the trial (p>0.05). The average daily feed intake and feed conversion ratio of turkey toms fed with MOS were higher than those of control and SC groups during the overall period (p<0.05). The dietary treatments did not affect carcass yield, breast meat, thigh, wing, liver, heart, empty gizzard, intestine, and abdominal fat pad proportions and meat pH, composition and pigmentation (p>0.05). These results suggest that the addition of MOS and SC is not likely to produce any performance or carcass characteristics in finishing turkeys at 10 to 20 wks of age.
The HIS5 gene of Saccharomyces cerevisiae host was encoded histidinol phosphate aminotransferase(E.C.: 2.6. 1.9). The HIS5 gene of Saccharomyces cerevisiae was cloned on plasmid pSH 530. This gene mighted be transcripted from a promoter of yeast gene both in E. coli and yeast hosts. We have determined the nucleotide sequence of the yeast HIS5 gene and its 5' and 3' flanking sequences. There are no large differences between the relative levels of HIS5 mRNA molecules with different 5' termini in represent and derepressed cell. In the DNA sequence upstream from the 5' termini of HIS5 mRNA we have found live closely related copies of a 9 base pair sequence. The sequence is also repeated in the 5' noncoding regions of HIS1, HIS3, HIS4, HIS5 and TRP5. Closely related sequence are not found flanking repeat sequence plays a role in the regulation of amino acid biosynthetic genes subject to the general amino acid control.
