This study was conducted to find out the effects of artificial shrinkage (AS) on post-thaw development of bovine embryos. The blastocoelic cavity of blastocyst was punctured to remove its fluid contents and then incubated in the holding medium (HM) for 10 min. The punctured and non-punctured (control) blastocysts were equilibrated in vitrification solution 1 (VS1; TCM-199+20% FBS+10% EG) for 5 min and vitrification solution 2 (VS2; TCM199+20% FBS+35% EG+5% PVP+0.5 M Sucrose) for 1 min and vitrified by direct dropping into the liquid nitrogen. Vitrified blastocysts (punctured and control) were thawed and cultured in vitro (12 hr) for studying survival and hatching rates. The levels of shrinkage were measured by the volume of the blastocyst during equilibration in VS1 (at 1, 3 and 5 min of equilibration) and VS2 (at 30 and 60 sec of equilibration) that was considering the volume of non-punctured blastocyst in HM as 100%. The levels of shrinkage were higher in punctured group (62.4, 64.6, 64.3% at 1, 3 and 5 min in VS1; 50.6 and 52.7% at 30 and 60 sec in VS2) than control group (84.8, 86.6, 86.4% at 1, 3 and 5 min in VS1; 72.1 and 68.8% at 30 and 60 sec in VS2), but within each group the levels of shrinkage were similar. The survival (90.9%) and hatching (50.0%) rates of vitrified blastocysts at 12 hr post-thaw were higher in punctured group than that in control group (76.9% and 0.0% respectively). We confirmed that vitrification solutions (VS1 and VS2) have no toxic effect on the survival of blastocysts because the survival rates of blastocysts exposed to VS1 and VS2 for 24 hr were similar between punctured and control groups (94.3 vs. 96.0%; p>0.05). In conclusion, the preliminary data show that AS of blastocyst may improve survival and hatching rate after thawing.
The purpose of this study was to determine the effects of Taxol pre-treatment to in vitro matured bovine oocytes, and sucrose and trehalose added to vitrification solution on spindle morphology and embryonic development following cryopreservation. Bovine oocytes were collected from ovaries and matured in tissue culture medium 199 (TCM 199) supplemented with 10% Fetal Bovine Serum (FBS), 0.05ng/ml epidermal growth factor, 0.01 IU/ml luteinizing hormone and $1{\mu}g/ml$ estradiol for 22h in $39^{\circ}C$, 5% $CO_2$, TCM 199-HEPES containing 20% FBS was used as basic medium (BM) to prepare vitrification solution. Oocytes were pre-treated with $1\;{\mu}M$ Taxol in maturation medium for 15 min prior to vitrification. Oocytes were exposed to 1.6 M ethylene glycol (EG) and 1.3M dimethyl sulfoxide (DMSO) in BM and then were exposed to 3.2 M EG, 2.6 M DMSO and 0.5 M sucrose in BM or 3.2 M EG, 2.6 M DMSO and 0.5 M trehalose in BM. Oocytes with cumulus cells and oocytes without cumulus cells were considered as control 1 and control 2, respectively and held in TCM 199-HEPES at $39^{\circ}C$. Oocytes were frozen using modified solid surface vitrification and were stored in cryotubes in liquid nitrogen for more than 1 week. Frozen oocytes were thawed in TCM 199-HEPES containing 0.5 M, 0.25 M and 0.1 M sucrose in BM for 2 min, respectively or 0.5 M, 0.25 M and 0.1 M trehalose in BM for 2 min, respectively. Immunoflurorescence staining of oocytes was performed to assess spindle morphology and chromosome configuration of oocytes. The rates of cleavage and blastocyst were examined following in vitro fertilization. Normal spindle morphology rate of oocytes pre-treated with Taxol prior to vitrification was not higher than that of other vitrified groups. Taxol pre-treatment did not increase cleavage and blastocyst formation rates, although control groups showed significantly higher rates (p<0.05). Percentages of normal spindle and embryonic development were not significantly different among vitrified groups regardless of type of sugar. In conclusion, Taxol pre-treatment of oocytes before cryopreservation did not reduce the damage induced by vitrification and subsequently did not improve embryonic development following vitrification. Trehalose may be used as an alternative non-permeating cryoprotectant in vitrification solution.

The aim of this study was performed to evaluate the effects of ice-binding protein from the arctic yeast Leucosporidium (LeIBP) supplementation on cryopreservation of boar sperm. The collected semen was diluted ($1.5{\times}10^8/ml$) in lactose egg yolk (LEY) and cooled at $5^{\circ}C$ for 3 h. The cooled semen was then diluted ($1{\times}10^8/ml$) in LeIBP containing LEY with 9% glycerol and maintained at $5^{\circ}C$ for 30 min. The semen was divided into six experimental groups (control, 0.001, 0.005, 0.01, 0.05 and 0.1 mg/ml of LeIBP). The straws were kept on above the liquid nitrogen ($LN_2$) vapors for 20 minutes and then plunged into $LN_2$. After thawing, computer-assisted sperm analysis was used for sperm motility and flow cytometry was performed to assess the viability, acrosome integrity (FITC-PSA/PI), ROS (DCF/PI), lipid peroxidation (BODIPY C11/PI) and apoptosis (Annexin V/PI), respectively. No significant responses were observed for sperm motility. However, sperm viability was significantly increased on 0.05 and 0.1 mg/ml of LeIBP groups compared to control (P < 0.05). In addition, acrosome integrity was significantly increases LeIBP groups (P < 0.05) and both ROS and lipid peroxidation level were lower in all LeIBP groups than those of control (P < 0.05). On the other hand, a significant higher apoptosis rate was observed in 0.05 and 0.1 mg/ml of LeIBP groups compared to control (P < 0.05). It can be assumed that a supplementation of LeIBP in boar sperm freezing extender is an effective method to increase the sperm qualities after cryopreservation.
The information indicating device plays an important part in the information times. Recently, the classical CRT (Cathod Ray Tube) display is getting transferred to the LCD (Liquid Crystal Display) one which is a kind of the FPDs (Flat Panel Displays). The OLED (Organic Light Emitting Diodes) display of the FPDs has many advantages for the low power consumption, the luminescence in itself, the light weight, the thin thickness, the wide view angle, the fast response and so on as compared with the LCD one. The OLED has lately attracted considerable attention as the next generation device for the information indicators. And also it has already been applied for the outside panel of a mobile phone, and its demand will be gradually increased in the various fields. It is manufactured by the vapor deposition method in the vacuum state, and the uniformity of thin film on the substrate depends on the temperature distribution in the point-cell source. This paper describes the basic concepts that are obtained to design the point-cell source using the computational temperature analysis. The grids are generated using the module of AUTOHEXA in the ICEM CFD program and the temperature distributions are numerically obtained using the STAR-CD program. The temperature profiles are calculated for four cases, i.e., the charge rate for the source in the crucible, the ratio of diameter to height of the crucible, the ratio of interval to height of the heating bands, and the geometry modification for the basic crucible. As a result, the blowout phenomenon can be shown when the charge rate for the source increases. The temperature variation in the radial direction is decreased as the ratio of diameter to height is decreased and it is suggested that the thin film thickness can be uniformed. In case of using one heating band, the blowout can be shown as the higher temperature distribution in the center part of the source, and the clogging can appear in the top end of the crucible in the lower temperature. The phenomena of both the blowout and the clogging in the modified crucible with the nozzle-diffuser can be prevented because the temperature in the upper part of the crucible is higher than that of other parts and the temperature variation in the radial direction becomes small.
We have reported previously that intact embryogenic cells can be used instead of protoplasts for electroporation-mediated transformation of zoysiagrass and rice. In this study, conditions of the tissue electroporation were examined to optimize the procedures. Embryogenic cell suspensions were established in liquid MS medium containing 2 mg/L of 2,4-D with embryogenic calluses induced from mature embryos of Z. japonica. The suspension-cultured cell clumps were electroporated with 35S-gusA expression vector DNA, and degrees of DNA introduction into the cells were determined by histological expression rates of the gusA marker gene. DNA transfer into the cell clumps occurred in wide range of voltage (100-400 V) and capacitance (10-1980 $\mu\textrm{F}$), but more in the ranges of 200-300 V and 330-800 $\mu\textrm{F}$ DNA concentrations higher than 6 $\mu\textrm{g}$/mL were adequate for GUS expression of the electroporated cells. DNA transfers were confirmed in all three embryogenic cell lines but only in one out of eleven non-embryogenic lines. Positive GUS expressions occurred with DNAs added even 20-40 h after pulse treatments. As a promoter of gusA, Act1 and Ubi1 were effective 7 and 5 times than 35S respectively in number of GUS expression units on electroporated cell clumps. Embryogenic cell clumps survived and regenerated into plantlets after pulse treatments of wide range of conditions.
According to a series of batch-scale washing tests, SDS+$POE_5$ and $POE_5$+$POE_14$ were determined for the applicable mixed surfactants. Because SDS+$POE_5$ showed slightly negative effects on the microbes in the toxicity tests, $POE_5$+$POE_14$((1:1) 1%) was chosen for this study. In the in-situ flushing experiments, the removal rate of endosulfan was 67% for the injection rate of 1.5L/min/$\textrm{km}^2$. And when methanol and ethanol were added as cosolvent, 75% and 81% removal efficiencies were achieved, respectively. In the tests of bioremediation after the application of in-situ flushing, the removal rates of contaminated soils having 13mg/kg dry soil and 3mg/kg dry soil as initial concentrations were 86% and 81%, respectively. There were no significant degradation after 24 hours. The major rate-limiting factor for the biodegradation of endosulfan might be the mass transfer from soil phase to liquid phase after 24 hours. With the addition of surfactant, 89% removal was achieved after 120 hours. Because the surfactant improved the mass transfer rate, the biodegradation of endosulfan was enhanced. When surfactant and cosolvent were added together, the adaptation period of microorganisms to the surfactant became longer and the removal rates were 84% and 83% for methanol and ethanol, respectively.

The purpose of this study was performed to quantitatively measure the thermal conductivity of poultry slaughter waste with variation of reaction temperature for optimal design of thermal hydrolysis reactor. We continuously quantified the thermal conductivity of dehydrated sludge related to the reaction temperature. As the reaction temperature increased, the dehydrated sludge is thermally liquefied under high temperature and pressure by the thermal hydrolysis reaction. Therefore, the bond water in the sludge cells comes out as free water, which changes the dehydrated sludge from a solid phase to slurry of a liquid phase. As a result, the thermal conductivity of the its sludge was more than 2.11 times lower than that of the water at $20^{\circ}C$. However, the thermal conductivity of the sludge approached to $0.677W/m{\cdot}^{\circ}C$ of water at $200^{\circ}C$, experimentally substantiating liquefaction of the dehydrated sludge. Therefore, we confirmed that the change in physical properties due to thermal hydrolysis appears to be an important factor for heat transfer efficiency. And the thermal conductivity function related to reaction temperature was derived to give the boundary condition for the optimal design of the thermal hydrolysis reactor. The consistency of the calculated function was 99.69%.

It is well known that volatile compounds including disinfection by-products as well as emissive dissolved gas in water can be removed effectively by air stripping. The micro-bubbles of flotation unit are so tiny as microns while the diameter of fine bubbles applied to air stripping is ranged from hundreds to thousands of micrometer. Therefore, the micro-bubbles in flotation can supply very wide specific surface area to transfer volatile matters through gas-liquid boundary. In addition, long emission time also can be gained to emit the volatile compound owing to the slow rise velocity of micro-bubbles in the flotation tank. There was a significant difference of the THMs species removal efficiency between air stripping and flotation experiments in this study. Moreover, the results of comparative experiments on the removal characteristics of THMs between air stripping and flotation revealed that the mass transfer coefficient, $K_La$ showed obvious differences. To overcome the limit of low removal efficiency of dissolved volatile compounds such as THMs in flotation process, the operation range of bubble volume concentration is required to higher than the operation condition of conventional particle separation.
Soil freezing is a phenomenon arising due to temperature difference between atmosphere and ground, and physical properties of soils vary upon the phase change of soil void from liquid to solid (ice). A heat-transfer mechanism for this case can be explained by the conduction in soil layers and the convection on ground surface. Accordingly, the evaluation of proper thermal properties of soils and the convective condition of ground surface is an important task for understanding freezing phenomenon. To describe convection on ground surface, simplified coefficient methods can be applied to deal with various conditions, such as atmospheric temperature, surface vegetation conditions, and soil constituents. In this study, two methods such as n-factor and convection coefficient for the convective ground surface boundary were applied within a commercial numerical program (TEMP/W) for modeling soil freezing phenomenon. Furthermore, the numerical results were compared to laboratory testing results. In the series of the comparison results, the convection coefficient is more appropriate than n-factor method to model the convective boundary condition.

For an orbit transfer in a space exploration mission, a solid or liquid rocket booster is included at the last stage of the launch vehicle. During the orbit transfer, thrust misalignment can cause a severe orbit error. Three axis attitude control or spin stabilization can be implemented to minimize the error. Spin stabilization technique has advantages in structural simplicity and lightness. One of ways to apply the spin stabilization to the payload is to include a spin table system in the launch vehicle. In this paper, effect of the spin table system on separation dynamics of the payload is analyzed. Simple model of the spin table to mimic basic functions is designed and simulation environment is established with the model. Effect of the spin table is tested by evaluating separation dynamics of a payload with and without the spin table. Analysis on tolerance effect of separation spring constant on separation dynamics of a payload is conducted.
