Effect of Sawdust Culture on Oak Mushroom, Lentinula edodes (Berk.) Pegler by Inoculation of the Liquid Spawn (액체종균(液體種菌) 접종(接種)에 의한 표고톱밥재배(栽培) 효과(效果))

  • For cultivation on sawdust-bed of oak-mushroom until present time, inoculation of spawn on sawdust bed has been performed by sawdust spawn. But, liquid spawn may have advantages for rapid mass production of spawn, and now, sawdust-cultivation by liquid spawn inoculation should be applied instead of sawdust spawn. Therefore, investigations were performed to evaluate the effect of sawdust-cultivation by liquid spawn inoculation. The results were as follows: 1. When 11 kinds of liquid media were applied, the oak-mushroom culture medium was the most excellent in growth. Most suitable temperature at PDA was $25^{\circ}C$, and $22.5\sim27.5^{\circ}C$ in range were optimal for liquid culture. In liquid culture, amount of mycelial growth increases rapidly up to 40 days of cultivation. Incubation at fermentor brought yield of 106mg dry mycelia per 40ml media after 17 days. 2. In 1l-spawn bottle, growth of mycelium by inoculation of 20ml-liquid spawns were faster than 6g-sawdust spawn in spread of mycelia. On 2kg-bag culture, inoculations of 10ml-, 20ml- and 30ml-liquid spawns were all slower than 20g-sawdust spawn in mycelial spread. So, amount increasement in ampunt of liquid spawn should be discussed. Yields of mushrooms until third sproutings of 2kg-bag culture were 580g in 30ml-liquid spawn inoculation, but 510g, 486g and 470g from 20g-sawdust spawn, 20ml-liquid spawn and 10ml-liquid spawn, respectively. Thus, 30ml-liquid spawn inoculation was highest in yield.

Cultural Characteristics by Sawdust and Liquid Spawn for the Cultivation of Neolentinus lepideus (잣버섯(Neolentinus lepideus) 재배를 위한 액체 및 톱밥종균의 배양특성)

  • This study was carried out to elucidate suitable spawn culture for an artificial cultivation of Neolentinus lepideus. The optimum culture conditions of the liquid spawn were deffated soy flour for main material, 12 days for culture period and 0.9 vvm for aeration volume, respectively. Sawdust spawn was appropriate for douglas fir sawdust and deffated corn flour (95 : 5, v/v) for mycelia growth and fruiting body formation. In case of liquid spawn, cultivation period was two shorter than sawdust spawn and mushroom yield was 111.9 g per 850ml cultivation bottle. In conclusion, the suitable spawn for fruiting body production was found to be liquid spawn than sawdust spawn with considering cultivation period and mushroom productivity.

Culture Condition and Inoculum Volume of Liquid Spawn on the Bottled Cultivation of Agrocybe cylindracea (버들송이(Agrocybe cylindracea)의 액체종균 배양조건과 접종량)

  • In order to shorten cultivation periods of Agrocybe cylindracea, liquid spawn was used in place of sawdust spawn. Optimum culture condition of liquid spawn was PSB (potato extract sugar broth) medium, $25^{\circ}C$ temperature and pH 7.0. And, optimum inoculum volume was $10{\sim}15ml$. Two grams per liter of dry weight of the fungal mycelia were obtained with 10l of culture bottle with liquid spawn after 10 days. In 850 ml-bottle of sawdust media, cultivation period using liquid spawn (30 days) was five days shorter compared with the cultivation period in sawdust spawn (35 days). Also, complete growth of sawdust media was increased to 92% (liquid spawn) from 75% (sawdust spawn) and yield of fruiting bodies was increased to about 10%.

  • Mushroom cultivation has gained increased attention in recent years. Currently, only four types of spawn, including sawdust spawn, grain spawn, liquid spawn, and stick spawn, are commonly available for mushroom cultivation. This limited spawn diversity has led to difficulty in selecting suitable inoculum materials in some cultivation. In this study, three small blocks of lignocellulosic agro-wastes and one block of a synthetic matrix were prepared as support for growing Pleurotus ostreatus in liquid medium. Mycelium-adsorbed blocks were then evaluated for their potential as block spawn for fructification. Our results indicated that the edible fungus was adsorbed and abundantly grew internally and externally on loofah sponge and synthetic polyurethane foam (PUF) supports and also has the ability to attach and grow on the surface of sugarcane bagasse and corncob supports. The mycelia of P. ostreatus adhered on corncob exhibited the highest metabolic activity, while those on the PUF showed the least activity. Mycelial extension rates of block spawns made of agro-waste materials were comparable to that of sawdust spawn, but the block spawn of PUF showed a significantly lower rate. No significant differences in cropping time and yield were observed among cultivations between experimental block spawns and sawdust spawns. Moreover, the corncob block spawn maintained its fruiting potential during an examined period of 6-month storage. The developed block spawn could be practically applied in mushroom cultivation.

Studies on the production of liquid spawn of Pleurotus ostreatus (Pleurotus ostreatus의 액체 종균 생산에 관한 연구)

  • For the improvement of productivity of Pleurotus ostreatus, the production of liquid spawn was studied. The highest liquid spawn production was obtained after shaking culture for 4 days in the culture medium containing 5%(W/V) wheat flour, 0.2%(W/V) yeast extract, 0.1%(W/V)$KNO_3$ 0.05% (W/V) $MgSO_4{\cdot}7H_2O$, 0.05%(W/V) $KH_2PO_4$. The optimum pH and temperature was 7.0 ana $30^{\circ}C$. The period required to complete the mycelial growth after spawning were 28, 22, 10 and 9 days, respectively, when the 2%(V/V) of solid spawn and 2%(V/V), 5% (V/V) and 10%(V/V) of liquid spawn were inoculated. The days required from spawning to fruiting bodies were 38, 34, 28 and 27 days.

The effect of different culture conditions of liquid spawn on the quality characteristics of shiitake mushroom (Lentinula edodes) (표고의 액체종균 배양 조건 및 품질 특성)

  • To improve the productivity of shiitake mushroom (Lentinula edodes), seven different types of media for liquid spawn (denoted as "A" to "G") were prepared with 0.3% soybean meal and varying sugar and glucose concentrations. During 14 days of incubation, the pH of the liquid culture gradually acidified with increasing incubation period. Additionally, there was a significant, but not prominent, difference in the degree of acidification depending on the sugar to glucose ratio. Liquid spawn culture "G," which had the highest sugar content was the most acidic on the last day of incubation. Mycelium dry weight increased significantly with increasing incubation period, and there was no significant difference in mycelium dry weight irrespective of the sugar to glucose ratio even after 14 days of culture. The inoculation of liquid spawn in sawdust medium with an inoculation volume ${\geq}45mL$ and incubation period of 15 to 18 days were the optimal culture conditions. Productivity of fruit bodies in sawdust medium and mushrooms treated with liquid spawn was significantly higher compared to solid spawn treatment. The mushrooms treated with liquid spawn had better chewiness, and the free amino acid content, which is associated with savory taste, was higher in these mushrooms compared to those treated with solid spawn.

  • The effects of liquid spawn to sawdust substrate on the growth of Flammulina velutipes were conducted for two years. The duration of optimal incubation for preculture and main culture of liquid spawn was 4 days and 6 days, respectively. When using liquid spawn, the application time for the first pinhead formation was similar with sawdust spawn, and incubation time of sawdust substrate was variable with liquid spawn and inoculum quantity. But, the overall yield of mushroom fruitbody was increased by using liquid spawn, excepting sesame hull extract medium.

  • Lentinula edodes liquid spawn growth under explosive aeration (supplying air with tiny bubbles) and soybean meal addition to liquid culture medium were investigated in terms of mycelial growth and residual free sugar content. The two treatments were effective for homogeneous culturing of mycelial spawn and for separating colonies during multiplication after an exponential growth period without limiting sustaining nitrogen nutrients. The mycelial growth and carbon dioxide concentration were greatest on the 13th day since the inoculation. At 12th day, however, free sugars were almost depleted in the upper part of the liquid medium. Total nitrogen content within precipitated mycelia was the highest at the 13th day. Chitin and sucrose contents in the mycelia were the highest at the 18th day, but ergosterol content became highest at 22 days. These results suggest that Lentinula edodes liquid spawn is ready in 18 days after inoculation.

  • In this study, to produce Flammulina velutipes mushroom liquid spawn efficiently and effectively the effects of explosive aeration (supplying air with tiny bubbles) of the liquid culture medium on carbon dioxide concentration and residual sugar content in the medium on carbon dioxide concentration and residual sugar contentin the medium were measured. Carbon dioxide concentrations were measured at the outlet of the incubator. On the third day the explosive aeration greatly increased mycelial growth of the liquid spawn, and carbon dioxide concentration also greatly increased but decreased after 5 days. Free sugar contents in the liquid culture consistantly decreased up to 7 days and thereafter was not detected. The weight of the mycelia were maintained similar levels after 3 days. Total nitrogen content in the liquid medium constantly decreased during the 11days of explosive aeration. The content of free sugars in 7 days of culture was the lowest level, thus the inoculum incubated for 6~7 days was thought to be the most effective. Carbon dioxide concentration measurement at the outlet of the container during the liquid spawn incubation required low cost but was efficient to estimate the degree of mycelial growth to be used as a simple indicator.

Culture conditions of liquid spawn and the growth characteristics of Pleurotus ostreatus (느타리버섯의 액체종균 배양 조건과 생육 특성)

  • To improve the productivity of Pleurotus ostreatus, different conditions of liquid spawn culture were tested. The optimum culture conditions were potato dextrose broth, incubation temperature of $22^{\circ}C$, and pH 6. Fifteen different media ('A' to 'O') containing 0.3% soybean meal (SM) were prepared by varying sugar and glucose contents. The cultures were propagated in SM media for 14 days, at $22^{\circ}C$, and pH 6. Their absorbances were higher after 14 days of incubation in the media containing both sugar and glucose. In particular, the absorbance of media containing 5 to 20% of glucose alone ('C', 'D', 'E', and 'F') tended to increase in the incubation period. Dry cell weight was lower in media containing less than 20% sugar or glucose alone than in media containing 30% sugar ('A') or 30% glucose ('B') alone. In sawdust media, in 900 mL-bottle, the optimum inoculation volume of liquid spawn was 15 to 20 mL. The texture of the mushroom cultivated with the liquid spawn was superior to that cultivated in the solid spawn.
