Antioxidant vitamins and Lipid Peroxidation in Patients with Type 1 Diabetes Mellitus

  • It has been postulated that oxidative stress may be increased and antioxidant defenses reduced in diabetes patients. Twenty-four patients with type 1 diabetes melitus (DM) (12.8$\pm$1.7 years) and 24 nondiabetics (12.5$\pm$2.1 years) were included in this study. Serum total cholesterol and LDL -cholesterol levels were significantly higher in diabetic than in nondiabetic control subjects, but serum levels of triglyceride , retinol , tocopherol, and $\beta$-carotene were significantly lower. Both $\beta$carotene and tocopherol levels inversely correlated with HbAlc, suggesting perhaps that low serum antioxidant level enhance theglycosylation of hemoglobin. Subjects with type 1 DM had lipid peroxide levels similar to those of nondiabetics control subjects, suggesting that per-oxdation of circulating lipid is not increased in uncomplicated diabeteics. The correlation between antioxidants and serum lipids were as follows ; retinol and LDL (r--0.36, p=0.019) ; retinol and total cholesterol(r=-0.35, p=0.020), tocopherol and LDL(r=-0.47, p=0.002) ; tocopherol and cholesterol (r=-0.49, p=0.001) ; $\beta$-carotene and LDL (r=-0.51, p=0.001). Overall , the results of this study were that serum lipid peroxide in patients with type 1 DM was similar to those of control subjects and antioxidants such as retinol, tocopherol and $\beta$-carotene were lower than those of nondiabetic cotnrol subjects, and negatively correlated with serum total cholesterol and LDL-cholesterol.

제조가 Bromobenzene에 의(依)한 흰쥐의 간손상(肝損傷)에 미치는 영향(影響) (Effects of Holotrichia on damages of liver tissue induced by bromobenzene in rats)

  • Holotrichia was tested for the effects on damages of liver tissue induced by bromobenzene. Holotrichia was treated firstly into samples, and then bromobenzene intoxicated animal models were set with them. In vitro, the level of lipid peroxide in tissue of liver proportinally decreased with the level of concentration of extract prepared from Holotrichia It was much more decreased, when lipid peroxidation was induced with ferrous iron ($Fe&{+2}$). In vivo, after the extract was administered to the animal model for twenty days, the level of lipid peroxide in liver decreased compared to that of bromobenzene-treated group. The enzyme activities of epoxide hydrolase and glutathione S-transferase in liver highly increased in Holotrichia pre-medicating group compare with the group treated with only bromobenzene. And we can get the same results in the enzyme activities of superoxide dismutase, catalase and glutathione peroxidase. The level of glutathione followed by Holotrichia pre-medicationg administration, increased as highly as normal group in compare with the group treated with only bromobenzene. Also, the enzyme activities of AL T, AST and $\{gammer}-GTP$ in liver considerably decreased. In conclusion, Holotrichia recovers the damage of liver due to bromobenzene intoxication by the increased activities of lipid peroxidation and bromobenzene scavenging enzymes.

식이 아연 결핍과 나이가 흰쥐 혈액과 간의 지질과산화물 수준과 아연 함량에 미치는 영향 (Effect of Dietary Zinc Deficiency and Age on Lipid Peroxides and Zinc Levels in Rat Blood and Liver)

  • The effect of dietary zinc deficiency and age on lipid peroxide level was investigaed in rats. Zinc level in serum and liver were also measured. Fifty Sprague-Dawly male rats aging 8 months(older rats) and 2 months(younger rats) were used as experimental animal. Zinc deficient diet(1.1ppm) and normal zinc diet(36.5ppm) were used as experimental diets. Rats in each age group were divided into zinc deficient(ZnDF), zinc pair-fed(ZnPF) and zinc ad-libitum(ZnAL) to remove the variances of food intake. After 4 weeks of experimetal period, rats were sacrificed. Thiobarbituric acid reactive substance(TBARS) levels in plasma and liver, lipofuscin and conjugated diene levels in liver were measured as lipid peroxide index. Food intakes of all groups were not different because zinc deficiency did not reduce food intake in ZnDF group. Younger rats gained weight continuously, while older rats lost weight in the begining of experiment and regained afterwards. In older rats, serum zinc level was decreaed while plasma TBARS. level was increased in ZnDF group. In younger rats, plasma TBARS concentration was increased in dietary zinc deficient rats although serum zinc concentration was not reduced. Liver zinc concentration was significantly higher in older rats comparing to younger rats. However, there was no difference among the three dietary groups. Liver TBARS level was not different by age or dietary zinc level. However it was tended to be higher in older rats. However there was no difference by the dietary zinc level. In both age groups, ZnDF group significantly increased plasma TBARS levels, which suggested dietary zinc deficiency could increase lipid peroxidation in part. Significantly higher levels of lipofuscin and conjugated diene in older rats suggested lipid peroxidation was accelerated by aging.

Butylated Hydroxytoluene첨가 식이 및 2-Acetylaminofluorene 투여가 식이지방을 달리한 쥐간의 Microsomal Mixed Function Oxidase계에 미치는 영향 (Effect of Butylated Hydroxytoluene and 2-Acetylaminofluorene Administration and Microsomal Mixed Function Oxidase System in Young Rats fed different Fats)

  • Sprague-dawley 숫쥐를 식이지방을 달리하여(I:Soybean oil p/s 4.0, II:beef tallow p/s 0.08) 이유후 8주동안 사육하였다. 이때 I,II군은 각각 기초식이군, 기초식이군에 0.3% butylated hydroxytoluene(BHT)를 첨가신킨 군, 생후 5~7주 사이에 4번의 2-Acetylaminofluorene(2-AAF)를 투여한 군 2-AAF를 투여하고 BHT도 먹인 군으로 나누었다. BHT는 이유 후부터 식이에 섞어 먹였으며 2-AAF를 투여하지 않은 군읜 2-AAF 주사에 의해 얻는 stress와 같은 효과를 주기위해 placebo로 polyethylene glycol 300을 투여하였다. Mixed function oxidase(MFO)계의 효소인 cytochrome p-450, cytochrome b$_5$및 cytochrome p-450 reductase와 과산화지질 등을 측정하였다. 성장기에 2-AAF의 투여는 성장지연을 초래하였으며 지질과산화물은 지방의종류, 2-AAF,BHT 등에 의해 큰 차는 없었다. Cytochrome p-450은 2-AAF에 의해 I-BHT-AAF와 II-AAF군에서 증가되었고 BHT에 의해서는 차이가 나지 않았다. 불포화지방을 먹인경우 cytochrome p-450과 cytochrome p-450 reductase가 2-AAF에 의해 증가되기보다는 오히려 감소하거나 I,II군에 비해 별 차이가 없었는데 2-AAF의 농도와 식이의 불포화도가 높아 세포막이 손상되었기 때문이라 사료된다. Cytochrome $b_5$는 각군 사이에 별 차가 없었다. Cytochrome p-450과 과산화지질(r=0.2475, p<0.05), cytochrome p-450 reductase와 cytochrome $b_5$ (r=0.2475, P<0.05)가 각각 양의 상관관계를 나타내었다. 따라서 2-AAF를 대사시키는 MFO계는 식이지방의 종류 및 BHT의 존재에 따라 영향을 받고, 특히 불포화지방식이인 경우 2-AAF를 대사시킬 수 있는 cytochrome p-450 유도 및 합성능력이 매우 저조함을 알 수 있으며 2-AAF는 어린 쥐의 성장을 지연시켰다.

저온저장기간에 따른 고등어(Scomber japonicus)의 이화학적 지표 변화 (Changes in Physicochemical Indicators in Mackerel Scomber japonicus Muscle according to Refrigerated Storage Duration)

  • This study examined changes in physicochemical indicators [K-value, lipid peroxide, and thiobarbituric acid reactive substances (TBARS)] in mackerel muscle during refrigerated storage at $4^{\circ}C$. Analyses were conducted every 3 days for 15 days. First, we visually observed changes in the mackerel. After 3 days, there were rapid changes in color and a rotten smell developed. The K-value of mackerel muscle was 4.21, 19.00, 42.13, 51.71, 71.19, and 80.34% on days 0, 3, 6, 9, 12, and 15, respectively; lipid peroxide level also increased, to 5.34, 5.44, 6.59, 7.29, 8.32, and 8.71 nmol/mL, respectively. TBARS increased from day 0 to 6, but did not change significantly after day 6 (0.98, 1.74, 2.73, 2.09, 1.86, and 1.93 mg Eq. malondialdehyde (MDA)/kg, respectively). The changes in K-value and lipid peroxide level demonstrated a loss of value as fresh food after day 6. The freshness of the stored mackerel had decreased by day 6 and mackerel stored for 9 days was less marketable.

Lipid Peroxidation Induced by the Reaction of Cytochrome c with Hydrogen Peroxide

  • Lipid peroxidation induced by the reaction of cytochrome c with $H_2O_2$ was investigated. When linoleic acid micelles or phosphatidyl choline liposomes were incubated with cytochrome c and $H_2O_2$, lipid peroxidation was increased in cytochrome c and $H_2O_2$ concentrations-dependent manner. Radical scavengers, azide, formate and ethanol prevented lipid peroxidation induced by the cytochrome c/$H_2O_2$ system. Iron specific chelator, desferoxamine also prevented the cytochrome c/$H_2O_2$ system-mediated lipid peroxidation. These results suggest that lipid peroxidation may be induced by the cytochrome c/$H_2O_2$ system via the generation of free radicals. Carnosine, homocarnosine and anserine are present in the muscle and brain of many animals and human. Previous studies show that these compounds have an antioxidant function. In the present study, carnosine, homocarnosine and anserine significantly prevented the cytochrome c/$H_2O_2$ system-mediated lipid peroxidation. Carnosine and related compounds also inhibited the free radical-generating activity of cytochrome c. The results suggest that carnosine, homocarnosine and anserine may prevent lipid peroxidation induced by the cytochrome c/$H_2O_2$ system through a free radical scavenging.

Induction of Lipin1 by ROS-Dependent SREBP-2 Activation

  • Lipin1 was identified as a phosphatidate phosphatase enzyme, and it plays a key role in lipid metabolism. Since free radicals contribute to metabolic diseases in the liver, this study investigated the effects of free radicals on the regulation of Lipin1 expression in Huh7 and AML12 cells. Hydrogen peroxide induced mRNA and protein expression of Lipin1 in Huh7 cells, which was assayed by quantitative RT-PCR and immunoblotting, respectively. Induction of Lipin1 by hydrogen peroxide was confirmed in AML12 cells. Hydrogen peroxide treatment significantly increased expression of sterol regulatory element-binding protein (SREBP)-2, but not SREBP-1. Moreover, nuclear translocation of SREBP-2 was detected after hydrogen peroxide treatment. Hydrogen peroxide-induced Lipin1 or SREBP-2 expression was significantly reduced by N-acetyl-$\small{L}$-cysteine treatment, indicating that reactive oxygen species (ROS) were implicated in Lipin1 expression. Next, we investigated whether the hypoxic environments that cause endogenous ROS production in mitochondria in metabolic diseases affect the expression of Lipin1. Exposure to hypoxia also increased Lipin1 expression. In contrast, pretreatment with antioxidants attenuated hypoxia-induced Lipin1 expression. Collectively, our results show that ROS activate SREBP-2, which induces Lipin1 expression.

  • The effective components of Korean ginseng showing the lipid-peroxide depressing activity were isolated. From the ether-soluble acidic fraction of fresh ginseng three phenolic acids were obtained. Salicylic acid and vanilic acid exhibited the potent antioxidant activity, where p-hydroxycinnamic acid did not.

Glutathione suppresses lipid oxidation of Clanis bilineata larvae meat during frozen storage

  • The lipids of Clanis bilineata larvae meat (CBLM) are susceptible to oxidation, and thus the commercial and consumption values of CBLM decrease during frozen storage. In the present study, peroxide values (PV), thiobarbituric acid-reactive substances (TBARS), free fatty acid (FFA) content, fatty acid composition, and likeness (palatability) score of CBLM were determined to investigate the effect of glutathione on the oxidation of CBLM lipids. Glutathione decreased the PV, TBARS, and FFA content, maintained fatty acid composition, and increased the likeness (palatability) score of the CBLM, indicating that glutathione can be used as a cryoprotectant to extend the shelf life of CBLM.

칼로리 제한에 노화과정의 조절 (Modulation of the Aging Process by Food Restriction)

  • Aging is the progressive accumulation of changes with time associated with responsible for the ever-increasing susceptibility to disease and death which accompanies advancing age. Lipid peroxides easily produced in the membrane system by the chain reaction of free radicals which occurred from various environmental factors. The amount of lipid peroxides produced in biological system increased with aging process, and lipid peroxidation damages involved in aging process and pathological disorders. Although lipid peroxides have such deleterious effects on the organisms, there are numerous substances and mechanisms which prevent the reaction of peroxide formation and protect the subject from its toxicity. This review provides an overview of how does lipid peroxidation of unsaturated lipids take place by free radical, and what is the intervention of lipid peroxides in pathogenesis of some human diseases, and also how does food restriction influences the aging process and various pathological disorders. The major focus of this paper is to review the evidence indicating that food restriction retards the aging process, and possible mechanisms of its actions. Therefore, it discussed the effects of age and food restriction on life-span, membrane yield, lipid peroxidation, fatty acid composition and peroxidizability, cholesterol and triglyceride levels, prostaglasndin and thromboxane synthesis, which may be concerned with blood flow, membrane fluidity, homeostasis and glomerular filtration rate in living body.
