Inhibitory Effects of Polyoxypregnane Constituents from the Roots of Cynanchum caudatum on the Aldehyde Oxidase Activity and Lipid Peroxidation (우피소근의 polyoxypregnane 화합물의 Aldehyde Oxidase 및 지질과산화 억제효과)

  • The roots of Cynanhum caudatum (Asclepiadaceae) have been used in folk medicine in Japan and China for the prevention and treatment of various geriatric diseases and also as a cardiotonic agent. Constituents of this plant have mainly been examined for glycosides: besides two steroidal alkaloids, gagaminine and gagamine which was firstly isolated by us, more than 35 polyoxypregnane glycosides and aglycones have also been identified. Gagaminine inhibits potently the hepatic aldehyde oxidase activity and lipid peroxidation in vitro. The present work deals with the comparison of antioxidative activities of gagamine, a new pregnane alkaloid, three isolated polyoxypregnanes containing a keto group at C-20 with those of gagaminine, a potent antioxidant, in order to explain the structure-activity relationships. The results of this study further prove that the cinnamoyl group of gagaminine is very important for the inhibition on the aldehyde oxidase activity while the nicotinoyl group is necessary for anti-lipid peroxidation. Besides that, the keto compounds having no ester group at C-12 were found to be more active than the others except gagaminine.

Protective Effects of Extracts of Mori Cortex Radicis on Carbon Tetrachloride-induced Hepatotoxicity (사염화탄소에 의해 유발된 간독성에 대한 상백피 추출물의 간보호효과)

  • In order to evaluate the protective effects of extracts of Mori cortex radicis on carbon tetrachloride-induced hepatotoxicity, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, lactic dehydrogenase, malondiadehyde values and glutathione S-transferase activity were measured in ICR mice. The activities of serum aminotransferase and the hepatic content of lipid peroxide after carbon tetrachloride-treatment were markedly increased than normal control but those levels were significantly decreased by the treatment of butanol fraction of Mori cortex radicis mathanol extract. Glutathione S-transferase activity was decreased by carbon tetrachloride than control, but that also inhibited by the treatment of butanol fraction of Mori cortex radicis methanol extract. These results demonstrate a possible hepatoprotective role of extract role of extract of Mori cortex radicis against ${CCl_4}-induced$ hepatoxicity in vivo.

Antioxidant Activity of Ergosterol Peroxide (5,8-Epidioxy-$5\alpha,8\alpha$-ergosta-6,22E-dien-3$\beta$-ol) in Armillariella mellea

  • Antioxidant activities of various mushroom fruiting bodies were investigated in vitro. Among the mushroom extracts examined, Armillariella mellea, Daedalea dickinsi, Fomitella fraxinea and Pleurotus cornusopiae markedly exhibited inhibition on lipid peroxidaton of rat liver microsomes. Ergosterol peroxide (5,8-epidioxy-5α,8α-ergosta-6,22E-dien-3β-ol), antioxidant from A. mellea, was isolated by solvent extraction, silica gel column chromatography and recrystallization. The structure of the compound was determined by NMR, GC/MS and X-ray crystallography. Ergosterol peroxide showed potent inhibition on lipid peroxidation and exhibited higher antioxidant activity than well-known antioxidants, α-tocopherol and thiourea.

Effects of Natrually Occurring Furanocoumarins on Lipid Peroxidation and Carbon Tetrachloride Induced Hapatotoxicity in Mice

  • Several naturally occurring furanocoumarins significantly inhibited microsomal lipid peroxidation not only mediated by endogeneous iron and NADPH but also initiated by $CCL_4$ metabolites, phellopterin, a potent inhibibitor of cytochrome p-450, exhibited an almost complete inhibition of $CCL_4$-induced hepatotoxicity as measured by sGPT activity 24 hr after $CCL_4$ intoxication, whereas other furanocoumarins such as imperation, byakangelicin and oxypeucedanin methanolate exerted no protective effect. When compared with other cytochrome P-450 inhibitors(SKF-52A, AIA) and silymarin given at the same dose level $(ED_{50})$, phellopterin still showed a significant inhibition of hepatotoxicity which was even stronger than that of AIA, known as a typical suicide inhibitor. Phellopterin was partially effective when given 30 min after $CCL_4$ treatment. Repeated administrations of phellopterin, however, resulted in a complete loss of the protection against $CCL_4$-induced hepatotoxicity.

Effect of Plant Extracts on the Activity of Alcohol Dehydrogenase and the Antioxidation in Alcohol-treated Rat Hepatocyte (알코올 투여한 흰쥐 간세포내 알코올 탈수소효소의 활성과 항산화에 미치는 식물추출물들의 영향)

  • This study was purposed to compare the activity of alcohol dehydrogenase and antioxidative effects of several plant extracts in the alcohol-treated rat liver. Sprague-Dawley rat weighing about 200 g were divided into the following 6 groups : normal, alcohol group and 4 different plant extracts administrated groups(Soybean sprout, Pine needle, Lentinus edodes, acanthopanacis cortex). Each plant extract was administrated orally by 200mg/kg b.w./day for 8 days before the alcohol treatment (5 g of 30% alcohol /kg b.w. by i.p.injection). All rats were sacrificed at 90 min after the alcohol treatment. The alcohol concentrations in serum of Soybean sprout and pine needle group were significantly lower than the Lentinus edodes and Acanthopanacis cortex group. The activity of alcohol dehydrogenase in the hepatic cytosol of Soybean sprout and Pine needle group was also significantly higher than the alcohol and the other groups However, the activity of catalase seemed not to be affected, although the extract groups showed slightly higher activities of catalase than the alcohol group. These results may indicate that the extracts of Soybean sprout and Pine needle were relatvely effective on the alcohol degradation. the activity of blutathione-peroxidase and lipid peroxidaton of all of the extract groups were significantly lower than the activity of alcohol group. These results can suggest that all of the use plant extracts more or less have an antioxidative effect on the alcohol-induced oxidation and especially, extracts of Soybean sprout and Pine needle have an stimulating effect on the alcohol absorption and degradation.
