Effects of the Antioxidative Components to Ginsenoside in the Liver of 40-week-old Mice (40주령의 생쥐 간에 미치는 ginsenoside의 항산화효과)

  • To study on antioxidant effects of saponin fractions, we investigated effects in the liver of 40-week-old mice to which were pretreated with 5 mg/kg per body weight of saponins for 5 days. The ability of saponins to protect against oxidative damage to the mouse liver was examined by determining the level malondialdehyde (MDA),hydrogen peroxide and the activity of superoxide dismutase (SOD) and catalase (CAT). The only panaxadiol (PD)among the ginseng saponin fractions significantly increased the hepatic SOD activities (p<0.01), Whereas PD, panaxatriol (PT), ginsenoside Rd (G-Rd) (p<0.01) and ginsenoside Re (G-Re) (p<0.05) significantly decreased the contents of hydrogen peroxide. It was only G-Rd that significantly increased CAT activities (p<0.05). The level of MDA was significantly decreased by G-Rd and PD. In conclusion, PD and G-Rd among the saponin fractions were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation.

Optimal Process of Eel Hot-Water Extract for Proper Edible Volume (장어고음의 적정 음용량을 위한 제조공정의 설정)

  • Eel is Anguilla japonica and Anguillidae family as a freshwater fish, and Japanese name is Unagi. The content of vitamin A of eel muscle are about 4,000IU/100g and they have many effective components, so it is known as healthy food. Therefore, many people have eel that cooked roast and hot-water extract in Korea. WHO encouraged that daily edible content of vitamin A is 2,000IU. Now, Japan government proposes the weight of roasted eel in lunch basket as 50g. But, Korea government does not proposes in every related aspects. Therefore, we researched content of vitamin A in hot-water extract from eel and optimal processing condition of hot-water extract from eel, in order to determine a proper edible volume. The content of vitamin A in eel hot-water extract sold at a market was about 200 IU. This study resulted from the fact that many eel hot-water extracts lacked the vitamin A. The reason was that the lipids were removed for taste on processing. But, The content of vitamin A in eel hot-water extract which didn't removed lipid on processing was about 800IU. Therefore, in drinking for 3packs per day, eel hot-water extract might have adjusted 100mL per pack which is almost consistent with the recommended dietary allowance of vitamin A in WHO.

Influence of Cooking on Carotenoid Contents in Provitamin A-Biofortified Rice (가열조리에 따른 프로비타민 A 강화 쌀의 카로티노이드 함량)

  • This study was conducted to investigate the effects of different cooking methods on the carotenoid content in genetically modified(GM) provitamin A-biofortified rice. The proximate components (moisture, protein, lipid, and ash) of GM rice were similar to those of conventional non-GM rice. Provitamin A-biofortified rice was cooked in various ways, including boiling in water and steaming/roasting. Carotenoid composition was determined by HPLC. Total carotenoid contents of provitamin A-biofortified brown and milled rice were $122.79{\mu}g/100g$ and $125.44{\mu}g/100g$, respectively. Compared with raw unprocessed samples, boiling caused approximately 20% carotenoid loss. In contrast, seaming/roasting decreased carotenoid content by 78%.

Studies on the growth and enlargemet of tuber in tropical yams (Dioscorea alata L.) (열대산 도입마(Dioscorea alata L.)의 생육과 괴경비대)

  • This study was carried out to determine agronomic and genetic characteristics of the production process in tropical yams(Dioscorea atata L.). To make clear the possibility of cultivation of tropical yams in the Suwon area, the growth of the aboveground parts and the enlargement of tuber of Dioscorea alata, were investigated. The aboveground parts of the plants whose sprouted mother tuber were planted in late April were grown slowly until the 70-days after planting and grown rapidly after. from about 150-days after plantation, total length of vines slowly decreased due to the burning of the branch vines of lower modes. With regard to the branching progressed to 4th for the solo yam and to 6th for purple yam. The secondary branches tended to grow better. When the formation of daughter tubers was observed at the 50-days after planting, their enlargement was very slow. The rapid growth of tubers began at the 140-days and continued to the 190-days. General components such as protein, fiber, and lipid were higher in D. opposita rather than in D. alata. Hardness of D. alata was 2696.2 while that of D. opposita was 4946.9. Lightness of D. alata was 73.99, being higher than that of D. opposita.

Determination of Monoglycoceramides in Biological Samples using Enzymatic Deacylation and Reverse-phase HPLC (역상HPLC컬럼을 이용한 생체 내 단당세라마이드 분석)

  • Glycosphingolipids are structural components of mammalian cell membranes and are involved in essential cellular physiology such as cell-cell interaction, recognition, transmembrane signaling, proliferation and cell death. In this study, the simple quantitative method of monoglycoceramides-containing glucosylceramide and galactosylceramide was developed. The glycosylceramides extracted from culture cells and rat plasma were resolved by TLC, deacylated by SCDase and analyzed by HPLC-fluorescence detector at an excitation wavelength of 340 nm and an emission wavelength of 455 nm. Limit of detection was approximately 0.1 pmol and limit of quantification was about 1 pmol for both monoglycoceramide standards. The recoveries of standard glucosylceramides from intra- and inter-day assays were 113.8 and 88.8% and those of galactosylceramides were 110.7 and 123.9%, respectively. The monoglycoceramide contents of SW-620 cells and rat plasma were $141.5{\pm}5$ pmol/$1{\times}10^6$ cells and $3.9{\pm}0.3{\mu}M$, respectively. The present analytical method provides a reproducible quantification and total content of monoglycoceramide which may be as a potential biomarker for lipid imbalance-related human diseases.

Lipids and Fatty Acid Composition of Free and Bound Lipids in Barley Grain (보리의 유리(遊離) 및 결합지질(結合脂質)의 조성에 관한 연구)

  • Lipids and fatty acid composition of free and bound lipids isolated from four barley varieties were studied. The average content of purified free and bound lipids were 1.57% and 0.48%, respectively. The average contents of neutral lipids, glycolipids and phospholipids in the free and bound lipids contain 81.1% and 16.4%, 11.4%, and 6.8%, 6.7% and 75.5%, respectively. Among the neutral lipids in both free and bound lipids, triglycerides were the predominant with free fatty acids, monoglycerides, diglycerides, free sterol esters being present as minor components. Linoleic, oleic, palmitic and linolenic acid were the principal fatty acids in both free and bound lipids. And fatty acid composition of lipid classes in free and bound lipids were determined.

Composition of Lipids Associated with Dense Coat-enriched Fractions of Bovine Milk Fat Globule Membrane (우유 지방구막의 고밀도 표피에 결합된 지질의 조성)

  • To analyze the components of dense coat fractions associated with fat globule membrane, The membrane was treated with various concentrations of Triton X-100, a non-ionic detergent, and the composition of lipids associated to the detergent insoluble material was analyzed. The amount of protein, phospholipid, cholesterol and ganglioside in milk fat globule membrane was reduced consistently with increasing concentrations of Triton X-100. Butyrophilin (band 12), xanthine oxidase (band 3) and band 16 as constituents of insoluble coat materials was revealed after electrophorisis on SDS-polyacrylamide gels. Phosphatidyl ethanolamine, phosphatidyl choline, phosphatidyl serine, phosphatidyl inositol and sphingomyelin were identified as the major phospholipids of the coat materials without selective concentration relative to the original membranes. Percentages of total phospholipid were not changed by any of the treatments. Fatty acids of total lipid were myristate, palmitate, stearate (major saturated acids), oleate and linoleate (major unsaturated acids). Cholesterol contents on a protein basis were slightly reduced with increasing concentrations of Triton X-100. Cholesterol adhered to protein more tightly than other constituents The contents of gangliosides was proportionally refuted with increasing concentration of Triton X-100.

A Clinical Study on Blood Lipids as a Risk Factor of Cerebral Infarction (뇌경색 위험인자로서의 혈중 지질에 대한 임상적 연구)

  • Objective The purpose of this case-control study was done to examine the relationship among the acute brain infarction, silent brain infarction and blood lipids. Methods We compared the components of blood lipids among acute brain infarction patients group (n=99), silent brain infarction patients group(n=101) and controls group (n=153). The analysis of the data was done by Frequency Analysis, one-way ANOVA, Post Hoc Test(Duncan), Pearson's Correlation. Results The results were as follows. High density lipoprotein cholesterol(HDL-Chol) was significantly lower in patients of acute brain infarctions group. The blood levels of total cholesterol(T-Chol) was significantly higher in patients of silent brain infarctions. A study on the relationship among the significant variables ; In T-Chol and HDL-Chol, DM(No=0, Yes=1) and Age, they had positive correlation each other. Between Sex(Female=0, Male=1) and HDL-Chol, Age and HDL-Chol, DM and HDL-Chol, they had negative correlation each other. Conclusions These results suggest that low HDL-Chol may be risk factor of acute brain infarction.

Isolation, Characterization and Numerical Taxonomy of Novel Oxalate-oxidizing Bacteria

  • The present work is aimed at providing additional new pure cultures of oxalate utilizing bacteria and its preliminary characterization for further work in the field of oxalate-metabolism and taxonomic studies. The taxonomy of 14 mesophilic, aerobic oxalotrophic bacteria isolated by an enrichment culture technique from soils rhizosphers, and the juice of the petiole/stem tissue of plants was investigated. Isolates were characterized with 95 morphological, biochemical and physiological tests. Cellular lipid components and carotenoids of isolates were also studied as an aid to taxonomic characterization. All isolates were Gram-negative, oxidase and catalase positive and no growth factors were required. In addition to oxalates, some of the strains grow on methanol and/or formate. The taxonomic similarities among isolates, reference strains or previously reported oxalotrophic bacteria were analysed by using the Simple Matching (S/ sub SM/) and Jaccard (S$\_$J/) Coefficients. Clustering was performed by using the unweighted pair group method with arithmetic averages (UPGMA) algorithm. The oxalotrophic strains formed five major and two single-member clusters at the 70-86% similarity level. Based on the numerical taxonomy, isolates were separated into three phenotypic groups. Pink-pigmented strains belonged to Methylobacterium extorquens, yellow-pigmented strains were most similar to Pseudomonas sp. YOx and Xanthobacter autorophicus, and heterogeneous non-pigmented strains were closely related to genera Azospirillum, Ancylobacter, Burkholderia and Pseudomonas. New strains belonged to the genera Pseudomonas, Azospirillum and Ancylobacter that differ taxonomically from other known oxalate oxidizers were obtained. Numerical analysis indicated that some strains of the yellow-pigmented and nonpigmented clusters might represent new species.

Anticoagulant and Antiplatelet Activities of Artemisia princeps Pampanini and Its Bioactive Components

  • Artemisia princeps Pampanini (AP) has been used as a traditional medicine in Korea, China and Japan and reported to exhibit various beneficial biological effects including anti-inflammatory, antioxidant, anti-atherogenic and lipid lowering activities; however, its antiplatelet and anticoagulant properties have not been studied. In the present study, we evaluated the effects of an ethanol extract of Artemisia princeps Pampanini (EAP) and its major flavonoids, eupatilin and jaceosidin, on platelet aggregation and coagulation. To determine the antiplatelet activity, arachidonic acid (AA)-, collagen- and ADP (adenosine diphosphate)-induced platelet aggregation were examined along with serotonin and thromboxane A2 ($TXA_2$) generation in vitro. The anticoagulant activity was determined by monitoring the activated partial thromboplastin time (aPTT) and prothrombin time (PT) in vitro. The data showed that EAP and its major flavonoids, eupatilin and jaceosidin, significantly reduced AA-induced platelet aggregation and the generation of serotonin and $TXA_2$, although no significant change in platelet aggregation induced by collagen and ADP was observed. Moreover, EAP significantly prolonged the PT and aPTT. The PT and/or aPTT were significantly increased in the presence of eupatilin and jaceosidin. Thus, these results suggest that EAP may have the potential to prevent or improve thrombosis by inhibiting platelet activation and blood coagulation.
