SDS-PAGE OF THE LINGUAL EPITHELIUM WITH SPECIAL REFERENCE TO TASTE BUDS (미뢰함유설상피와 비함유설상피 단백질에 관한 SDS-PAGE적 비교 연구)

  • As a study to elucidate whether taste buds contain specific proteins, rat dorsal lingual epithelium was analysed by electrophoresis. The epithelium of the vallate papilla (with numerous taste buds), the area of the fungiform papilla 9with a few taste buds), and the area between vallate papilla and large filiform papilla (not containing taste buds) were strippled off by treatment with 0.7% EDTA. The epithelial protein was extracted by 1% SDS and 1% Mercaptoethanol in 0.01M phosphate buffer (pH7.2). Extracts were analysed by disc SDS-PAGE. Because the patterns of protein composition from each site were similar with each other as a whole, it is concluded that taste buds do not contain specific protein detected by SDS-PAGE in adult rat. But a protein on M.W. 49000 which lies in the area of molecular weight of keratin molecules was found only in the epithelium containing taste buds. This results suggested that the epithelium containing taste buds differentiate dissimilarly to the epithelium not containing taste buds.

Morphological study on the tongue of Korean native goat (한국재래산양 혀에 관한 형태학적 연구)

  • This studies were carried out to identify the characteristics of the tongue of Korean native goat(Capra hircus) by macroscopy, microscopy and scanning microscopy. Korean native goat had torus linguae, median lingual sulcus, lingual fossa and ventral median fissure but did not have glossoepiglottic fold and terminal sulcus in the tongue. The whole length of tongue was $11.51{\pm}0.76cm$. The length of tongue apex, tongue body, tongue root and the torus linguae were $2.62{\pm}0.28$, $7.39{\pm}0.27$, $1.56{\pm}0.26$ and $6.37{\pm}0.29cm$, respectively. The width of tongue apex, torus linguae and tongue root were $3.41{\pm}0.24$, $3.74{\pm}0.29$ and $3.68{\pm}0.11$, respectively. The thickness of tongue apex was $1.60{\pm}0.10$, and the height of torus linguae was $1.52{\pm}0.15cm$. Filiform papillae were present at the tongue apex and the tongue body rostral to torus linguae. Fungiform papillae were scattered from tongue apex to rostral portion of torus linguae, being in abundance at the tongue apex. Vallate papillae were showed at the lateral portion of torus linguae, while lentiform papillae were present at its central portion. Conical papillae were located between vallate and lentiform papillae. The numbers of filiform, fungiform, conical, vallate and lentiform papillae were $46,980{\pm}1070.98$, $446.8{\pm}36.97$, $818.4{\pm}43.99$, $34.8{\pm}2.77$, and $255.6{\pm}39.30$, respectively. The average numbers of taste bud were $8.3{\pm}2.04$ in a fungiform papilla and $247.3{\pm}37.44$ in a vallate papilla. The filiform papilla had secondary and tertiary papillae. The height of filiform papilla was about $150{\mu}m$ and the diameter was $100{\mu}m$. The diameters of fungiform papillae were 350 to $550{\mu}m$. The long and short diameters of maximum-sized lentiform papilla were 4000 and $3000{\mu}m$, respectively, while those of minimum-sized papilla were 700 and $600{\mu}m$, respectively. The height of conical papillae was 450 to $600{\mu}m$ and diameter was 250 to $450{\mu}m$. The vallate papilla was round or oval in shape and its diameter was 500 to $850{\mu}m$. It had well-developed papillary groove around itself. The modified conical papillae were not observed in the tongue of Korean native goat.

CHANGE OF TASTE PREFERENCE AND TASTE BUD AFTER UNILATERAL LINGUAL NERVE TRANSECTION IN RAT (백서 편측 설신경 손상 후 미각 및 설유두의 변화에 대한 연구)

  • Purpose of study: Lingual nerve damage can be caused by surgery or trauma such as physical irriatation, radiation, chemotherapy, infection and viral infection. Once nerve damage occurred, patients sometimes complain taste change and loss of taste along with serious disturbance of tongue. The purpose of this study was to evaluate the effects of unilateral lingual nerve transection on taste as well as on the maintenance of taste buds. Materials & Methods: Male Sprague-Dawley rats weighing 220-250g received unilateral transection of lingual nerve, subjected to the preference test for various taste solutions (0.1M NaCl, 0.1M sucrose, 0.01M QHCl, or 0.01M HCl) with two bottle test paradigm at 2, 4, 6, or 8 weeks after the operation. Tongue was fixed with 8% paraformaldehyde. After fixation, they were observed with scanning electron microscope(JSM-$840A^{(R)}$, JEOL, JAPAN) and counted the number of the dorsal surface of the fungiform papilla for changes of fungiform papilla. And, Fungiform papilla were obtained from coronal sections of the anterior tongue(cryosection). After cryosection, immunostaining with $G{\alpha}gust$(I-20)(Santa Cruz Biotechnology, USA), $PLC{\beta}2$(Q-15)(Santa Cruz Biotechnology, USA), and $T_1R_1$(Alpha Diagnostic International, USA) were done. Immunofluorescence of labeled taste bud cells was examined by confocal microscopy(F92-$300^{(R)}$, Olympus, JAPAN). Results: The preference score for salty and sweet tended to be higher in the operated rats with statistical significance, compared to the sham rats. Fungiform papilla counting were decreased after lingual nerve transaction. In 2 weeks, maximum differences occurred. Gustducin and $T_1R_1$ expressions of taste receptor in 2 and 4 weeks were decreased. $PLC{\beta}2$ were not expressed in both experimental and control group. Conclusion: This study demonstrated that the taste recognition for sweet and salty taste changed by week 2 and 4 after unilateral lingual nerve transection. However, regeneration related taste was occurred in the presence of preserving mesoneurial tissue and the time was 6 weeks. Our results demonstrated that unilateral lingual nerve damage caused morphological and numerical change of fungiform papilla. It should be noted in our study that lingual nerve transection resulted in not only morphological and numerical change but also functional change of fungiform papillae.

Analysis of the embrasure dimensions between maxillary central incisors in relation to the topography of the interdental papilla

  • Purpose: To analyze the dimensions of the embrasure space between the maxillary central incisors as potential factors influencing interdental papilla fill and height. Methods: The embrasure dimensions between the maxillary central incisors of 100 subjects (40 females/60 males) were assessed with clinical, study model, and radiographic examinations. Variables of the complete and deficient papilla fill groups were compared. Multiple regression analyses were performed to investigate potential influence of the distance between the contact point and bone crest (CP_BC), horizontal interdental distance (HID), and facio-lingual thickness (FLT) at the papilla base on complete/deficient papilla fill and papilla height (PH). Results: CP_BC was the only variable that showed a significant difference between the complete and deficient papilla groups (P<0.05). When the CP_BC was less than 5 mm, the embrasure spaces between the maxillary central incisors were completely filled with interdental papilla. Multiple regression analyses revealed that a significant predictor for complete/deficient papilla fill was CP_BC, and significant predictors for PH were CP_BC and HID (P<0.05). Conclusions: The chances of complete papilla fill increased as CP_BC decreased, while PH increased as CP_BC and HID increased. However, the FLT of the papilla base did not appear to affect papilla fill or PH. From an esthetic perspective, CP_BC as well as HID should be considered as factors influencing the topography of interdental papilla.

Effect of immediate provisional restoration on the preservation of gingival contour (발치 창에 삽입한 임시 수복물의 치은형태 보존 효과)

  • One of the central components of periodontal therapy is the improvement of esthetics. The presence and appearance of interdental papillae plays an important role of periodontal esthetics. The aim of the present study was to investigate how immediate provisional restoration preserve the shape of interdental papilla around the extraction socket and the width of bucco-lingual of gingiva. Another aim was to investigate the change in the interdental papilla and the amount of vertical bone fill of a extraction socket in relation to the interdental alveolar bone levels adjacent the alveolar socket. A total of 19 patients (11 male, 8 female, mean age of 50.57${\pm}$8.16), who visited the Department of Periodontology, Pusan National University and had more than one anterior tooth scheduled to be extracted due to an advanced periodontal disease were included in the present study. After initial periodontal therapy, the extracted teeth were reshaped of the root and placed into the socket followed by splinting with adjacent teeth with self-curing resin. The width of hucco-lingual of gingiva and interdental papilla height were measured at baseline, 1, 3, 6, 9 and 12 month and the periapical radiographic examination were taken at baseline, 6 and 12month following the extraction. The amount of vertical bone fill in the extraction socket were calculated. At 12 months following the extraction, the changes in mesial and distal interdental papilla and the width of bucco-lingual showed -1.06${\pm}$0.48mm, -0.844${\pm}$0.50mm, -1.50${\pm}$0.96m, relatively. The positional change in the interproximal papillae was significantly associated with the interdental bone level adjacent to the extraction socket(p=0.028). The higher the interproximal bone level adjacent the extraction socket, the greater the amount of bone fill in the extraction socket(p<0.001). In conclusion, it was thought that immediate provisional restoration could minimize the loss of the width of bucco-lingual and interproximal papillae around the extraction socket. In addition, the higher the interproximal bone level adjacent the extraction socket, the greater the amount of bone fill and the smaller the reduction of papillary height around the extraction socket.

The Relationship of Two-Point Discrimination Threshold and the Number of Fungiform Papilla According to Anatomical Location of Tongue (해부학적 위치에 따른 혀의 촉각식별능의 차이와 심상유두 개수의 관계)

  • The lingual branch of the trigeminal nerve transmitts general sensation from anterior two thirds of the tongue, also bearing within sheath fibers of chordal tympani branch of the facial nerve. Chorda tympani nerve carries special taste sensations from the anterior two thirds of the tongue and sub-serves the existing trigeminal pathway. Chorda tympani nerve and the lingual nerve innervate to fungiform papilla and distribution of fungiform papilla on tongue dorsum is variable according to anatomical location. The purpose of this report is to assess that the relationship of the number of fungiform papilla and the ability of two-point discrimination of tongu dorsum. Twenty-six healthy students(male:female=13:13) whose mean age was $30{\pm}3$ participated in our study. Two-point discrimination thresholds were measured to evaluate the spatial acuity of touch sensation. The measurement was carried out at the tip and posterolateral region of dorsal tongue. After two-point discrimination test, we took the pictures of their dorsal tongue dyed with methylene blue with digital camera. There were no significant differences between the number of fungiform papilla and the two-point discrimination threshold. But, we found that there were the intraregional and intersubject variations of spatial acuity of the tongue. During the test on the posterolateral region of the dorsal tongue, students appealed the difficulty of discrimination of one point and two point.

THE EFFECTS OF IRRADIATION AND HYPERVITAMINOSIS $D_2$ ON THE ODONTOGENESIS IN THE RAT INCISOR (Vitamin $D_2$의 과량투여와 방사선조사가 치아 발육에 미치는 영향에 관한 실험적 연구)

  • 150 rats weighting about 150gm were devided into control group of 80 and experimental group of 70. Control group was subdivided into the irradiated vitamin D injection group and X-ray irradiated group. Experimental group was given 2.0mg ergocalciferol by four intramuscular injection prior to X-ray irradiation with single 800 rads and 1,500 rads respectively. Experimental animals from each group was sacrificed after 1, 3, 7, 14, and 28 days and their incisors were investigated by histopathological examination. The results were as follows; 1. In the irradiated groups, it showed dentin hypoplasia and formation of dentinoid substance caused by degeneration of odontoblast at the early stage. Especially, 1,500 rads group which was severely effected showed formation of osteoid dentin at the apical portion and severe injuries of dental papilla at the first week. 2. In the vitamin D2 administration group, it showed thinned dentin layer at the early stage but, taking time, predentin and dentin layer was thickened. At the fourth week, dentin was chiefly composed of interglobular dentin, especially in the lingual portion. 3. Using in combination of overdose vitamin D2 administration and X-ray irradiation, it effected severely odontoblast, undifferentiated mesenchymal cells around tooth germ and pulp tissue. At the early stage, dentin layer was thinned but, taking time, it was thickened and composed of interglobular dentin caused by calcification of predentin layer. 4. In 800 rads irradiation after the overdose vitamin D2 administration, it showed formation of osteoid dentin in the lingual portion at the first week. In the 1,500 rads irradiation after the overdose vitamin D2 administration, it showed formation of osteoid dentin and degeneration of ameloblast in both buccal and lingual portion at the first week, and enamel hypoplasia caused by edema and loss of polarity of ameloblasts at the second week. 5. By the entire experiment, the overdose vitamin D2 administration and X-ray irradiation effected severely odontoblasts, undifferentiated mesenchymal cells of dental papilla, and primitive cells of tooth germ among the dental tissue. Especially using combination of overdose vitamin D2 administration and X-ray irradiation also effected ameloblasts, resulting in enamel hypoplasia.

Anatomic study of the incisive canal in relation to midpalatal placement of mini-implant (정중구개부의 미니 임플랜트 식립과 관련된 절치관의 해부학적 연구)

  • Objective: The purpose of this study was to reveal the position of the incisive foramen in relation to the incisive papilla and cusp tips. Methods: Plaster models and CT images of 25 adult orthodontic patients were used to measure the width of the incisive canal and positions of the anterior and posterior borders of the incisive foramen in relation to the incisive papilla. Results: The palatal surface distance from the interdental papilla between the maxillary central incisors to the posterior border of the incisive foramen along the palatal surface was 1.7 fold of the distance from the interdental papilla between the central incisors to the posterior border of the incisive papilla. The distance between the posterior border of the incisive papilla and posterior border of the incisive foramen along the palatal surface was 6.15 ${\pm}$ 1.75 mm. The anteroposterior position of the posterior border of the incisive foramen was slightly anterior to the lingual cusp tips of the maxillary 1st premolars. The width of the incisive foramen was 4.03 ${\pm}$ 0.64 mm, therefore it is recommended to position the mini-implant more than 3 mm laterally when placing a mini-implant lateral to the incisive foramen, from the center. Conclusions: These results can be used as a reference in presuming the position of the incisive foramen when placing mini-implant in the anterior palate area.

A CASE REPORT OF THE MANDIBULAR INFECTED BUCCAL CYST

  • The author observed a cystic lesion which had new radiographic entities in the 6 years old male patient and obtained the following features. 1. The main clinical feature is the swelling. 2. The lesion is situated on the buccal surface of a erupting mandibular first molar. 3. The affected molar is tilted, so that the apices are adjacent to the lingual plate of the mandible. 4. The affected molar is of normal morphology, caries-free and vital. 5. The radiographic feature is the cystic radiolucency which is extended over the buccal surface of the roots of the affected molar and slightly inferior to the root apices. 6. The dental papilla of the affected molar shows the lamina dura of relatively normal density and definitition. 7. The buccal surface shows the laminated new bone formation. 8. The inferior concave border of the lesion is delineated by a thick and calcified layer of cortical bone. 9. The lesion causes displacement of the developing second molar of which anterior cortex is partially destroyed.

Comparative Morphology of the Tongue of Miniopterus schreibersi fuliginosus and Pipistrellus savii (긴가락박쥐(Miniopterus schreibersi fuliginosus)와 검은집박쥐(Pipistrellus savii) 혀의 비교형태)

  • A SEM study on morphology of lingual papillae of Korean long-fingered bat (Miniopterus schreibersi fuliginosus) and Savi's Pipistrelle (Pipistrellus savii) was conducted. Three kinds of lingual papillae were observed: filiform, fungiform, circumvallate papillae. Filiform papillae were divided into two types; the type 1 had a group of needle-like projections, and was distributed throughout the front half of the tongue; the type 2 had a smooth and thick body, and was found in rear half of the tongue. 35 to 45 fungiform papillae were found on the dorsal surface of the tongue in both species. They were observed along the lateral margins and were also found on front and rear end part of the tongue. There were two to three noticeably large fungiform papillae arranged in a straight line on the region between lingual prominence and circumvallate papillae. There were two circumvallate papillae close to the rear end of the tongue. They were large and round, each having two layers of pads. The overall morphology of lingual papillae of M. schreibersi fuliginosus and P. savii was found to be similar with other Chiroptera. However, few but noticeable differences were found among the filiform papillae and fungiform papillae. Type 2 filiform papillae differed in that bifid and trifid configuration were found in M. schreibersi fulginosus unlike in P. savii. In addition, numbers of large fungiform papillae located in the center of posterior region of the tongue were different with M. schreibersi have three while P. savii having only two.
