Effect of Fused Superphosphate Levels in Various Root Media Containing Micronutrient Mixes on Growth of Marigold Plug Seedlings (미량원소복합제가 혼합된 각종 상토에서 용과린의 시비수준이 매리골드 플러그 묘의 생육에 미치는 영향)

  • Objective of this research was to determine the effect of application level of fused superphosphate (FSPP) in root media containing granular micronutrient fertilizers (MF) on growth of marigold 'Orange boy' in plug culture. To achieve this, three granular micronutrient mixes such as MF 1, MF 2, and MF 3 were produced and incorporated into three root media, peatmoss+composted pine bark (1:1, v/v, PB), peatmoss+composted rice-hull (1:1, v/v, PR), and peatmoss+composted saw-dust (1:1, v/v, PS), at a rate of $0.3g{\cdot}L^{-1}$. Elevated application level of FSPP in PS medium containing each of MF resulted in increase of plant growth such as plant height, plant width, stem diameter, fresh weight and dry weight at 35days after sowing. The dry weight in the treatment of MF 1, MF 2, MF 3, and control fertilized with $9.0g{\cdot}L^{-1}$ of FSPP were 0.066g, 0.103g, 0.077g, and 0.095g per plant, respectively. These results indicated that each of MF affected marigold plug seedlings differently. The application level of FSPP resulted in the heaviest fresh and dry weight in PR medium were $9.0g{\cdot}L^{-1}$ in MF 1, $3.0g{\cdot}L^{-1}$ in MF 2, $9.0g{\cdot}L^{-1}$ in MF 3, and $6.0g{\cdot}L^{-1}$ in control. Elevated application level of FSPP in PB medium containing each of the micronutrient mix increased fresh and dry weight. The general trends in response of plant growth to elevated FSPP were linear and quadratic. The dry weight in the treatment of MF 1, MF 2, MF 3, and control fertilized with $9.0g{\cdot}L^{-1}$ of FSPP in PB medium were 0.131g, 0.104g, 0.137g, and 0.111g per plant at 35days after sowing. These results indicated that MF 1 and MF 3 performed better than MF 2 and control treatments in growth of marigold plug seedlings.

Effect of irradiation on cell cycle, cell death and expression of its related proteins in normal human oral keratinocytes (방사선조사가 사람 정상 구강각화 세포의 세포주기, 세포사 및 수종 단백질의 발현에 미치는 영향)

  • Purpose: To investigate the radiosensitivity of the normal human oral keratinocytes (NHOK), and the effect of irradiation on cell cycle and protein expression. Materials and Methods: To evaluate the radiosensitivity of NHOK, the number of colonies and cells were counted after irradiation and the SF2 (survival fraction at 2Gy) value, and the cell survival curve fitted on a linear-quadratic model were obtained. LDH analysis was carried out to evaluate the necrosis of NHOK at 1, 2, 3, and 4 days after 2, 10, and 20 Gy irradiation. Cell cycle arrest and the induction of apoptosis were analyzed using flow cytometry at 1, 2, 3, and 4 days after 2, 10, and 200y irradiation. Finally, proteins related cell cycle arrest and apoptosis were analysed by Western blot. Results: The number of survived cell was significantly decreased in a dose-dependent manner. The cell survival curve showed SF2, α, and β values to be 0.568, 0.209, and 0.020 respectively. At 200y irradiated cells showed higher optical density than the control group. After irradiation, apoptosis was not observed but G2 arrest was observed in the NHOK cells. 1 day after 10 Gy irradiation, the expression of p53 remained unchanged, the p2l/sup WAF1/Cipl/ increased and the mdm2 decreased. The expression of bax, bcl-2, cyclin B1, and cyclin D remained unchanged. Conclusion: These results indicate that NHOK responds to irradiation by G2 arrest, which is possibly mediated by the expression of p21/sup WAFl/Cipl/, and that cell necrosis occurs by high dose irradiation.

Effects of L-glutamine supplementation on degradation rate and rumen fermentation characteristics in vitro

  • Objective: Two follow-up studies (exp. 1 and 2) were conducted to determine the effects of L-glutamine (L-Gln) supplementation on degradation and rumen fermentation characteristics in vitro. Methods: First, rumen liquor from three cannulated cows was used to test L-Gln (50 mM) degradation rate and ammonia-N production at 6, 12, 24, 36, and 48 h after incubation (exp. 1). Second, rumen liquor from two cannulated steers was used to assess the effects of five levels of L-Gln including 0% (control), 0.5%, 1%, 2%, and 3% at 0, 3, 6, 12, 24, 36, and 48 h after incubation on fermentation characteristics, gas production, and degradability of nutrients (exp. 2). Results: In exp. 1, L-Gln degradation rate and ammonia-N concentrations increased over time (p<0.001). In exp. 2, pH was reduced significantly as incubation time elapsed (p<0.001). Total gas production tended to increase in all groups as incubation time increased. Acetate and propionate tended to increase by increasing glutamine (Gln) levels, whereas levels of total volatile fatty acids (VFAs) were the highest in 0.5% and 3% Gln groups (p<0.001). The branched-chain VFA showed both linear and quadratic effects showing the lowest values in the 1% Gln group particularly after 6 h incubation (p<0.001). L-Gln increased crude protein degradability (p<0.001), showing the highest degradability in the 0.5% Gln group regardless of incubation time (p<0.05). Degradability of acid detergent fiber and neutral detergent fiber showed a similar pattern showing the highest values in 0.5% Gln group (p<0.10). Conclusion: Although L-Gln showed no toxicity when it was supplemented at high dosages (2% to 3% of DM), 0.5% L-Gln demonstrated the positive effects on main factors including VFAs production in-vitro. The results of this study need to be verified in further in-vivo study.

The apoptotic fragment assay in rat peripheral lymphocytes and crypt cells with whole body irradiation with 60Co ϒ-rays and 50 MeV cyclotron fast neutrons (코발트-60 감마선과 50 MeV 싸이크로트론 고속 중성자선에 전신조사된 랫드의 말초 임파구와 음와 세포의 아포토시스 유도를 이용한 생물학적 선량 측정 모델 개발 연구)

  • Here, we compared the effectiveness of 50 MeV($p{\to}RBe^+$) cyclotron fast neutrons versus $^{60}Co$ ${\gamma}$-rays by the apoptotic fragment frequency in both rat peripheral lymphocytes and crypt cells to check a radiobiological endpoint. The incidence of apoptotic cell death was increased in all irradiated groups, and radiation at all doses trigger rapid changes in both crypt cells and peripheral lymphocytes. These data suggest that apoptosis may play an important role in homeostasis of damaged radiosensitive target organ by removing damaged cells. The curve of dose-effect relationship for these data of apoptotic fragments frequencies was $y=0.3+(6.512{\pm}0.279)D(r^2=0.975)$ after neutrons, while $y=0.3+(4.435{\pm}0.473)D+(-1.300{\pm}0.551)D^2(r^2=0.988)$ after ${\gamma}$-rays. In addition, $y=3.5+(118.410{\pm}10.325)D+(-33.548{\pm}12.023)D^2(r^2=0.992)$ after ${\gamma}$-rays in rat lymphocytes. A significant dose-response relationship was found between the frequency of apoptotic cell and dose. These data show a trend towards increase of the numbers of apoptotic cells with increasing dose. Dose-response curves for high and low linear energy transfer (LET) radiation modalities in these studies were different. The relative biological effectiveness (RBE) value for crypt cells was 1.919. In addition, there were significant peaks on apoptosis induction at 4 and 6h after irradiation, and the morphological findings of the irradiated groups were typical apoptotic fragments in crypt cells that were hardly observed in the control group. Thus, apoptosis induction in both crypt cells and peripheral lymphocytes could be a useful endpoint of rat model for studying screening test and microdosimetic indicator to evaluate the biological effects of radiation-induced cell damage.

Determining Nitrogen Topdressing Rate at Panicle Initiation Stage of Rice based on Vegetation Index and SPAD Reading (유수분화기 식생지수와 SPAD값에 의한 벼 질소 수비 시용량 결정)

  • The core questions for determining nitrogen topdress rate (Npi) at panicle initiation stage (PIS) are 'how much nitrogen accumulation during the reproductive stage (PNup) is required for the target rice yield or protein content depending on the growth and nitrogen nutrition status at PIS?' and 'how can we diagnose the growth and nitrogen nutrition status easily at real time basis?'. To address these questions, two years experiments from 2001 to 2002 were done under various rates of basal, tillering, and panicle nitrogen fertilizer by employing a rice cultivar, Hwaseongbyeo. The response of grain yield and milled-rice protein content was quantified in relation to RVIgreen (green ratio vegetation index) and SPAD reading measured around PIS as indirect estimators for growth and nitrogen nutrition status, the regression models were formulated to predict PNup based on the growth and nitrogen nutrition status and Npi at PIS. Grain yield showed quadratic response to PNup, RVIgreen around PIS, and SPAD reading around PIS. The regression models to predict grain yield had a high determination coefficient of above 0.95. PNup for the maximum grain yield was estimated to be 9 to 13.5 kgN/10a within the range of RVIgreen around PIS of this experiment. decreasing with increasing RVIgreen and also to be 10 to 11 kgN/10a regardless of SPAD readings around PIS. At these PNup's the protein content of milled rice was estimated to rise above 9% that might degrade eating quality seriously Milled-rice protein content showed curve-linear increase with the increase of PNup, RVIgreen around PIS, and SPAD reading around PIS. The regression models to predict protein content had a high determination coefficient of above 0.91. PNup to control the milled-rice protein content below 7% was estimated as 6 to 8 kgN/10a within the range of RVIgreen and SPAD reading of this experiment, showing much lower values than those for the maximum grain yield. The recovery of the Npi applied at PIS ranged from 53 to 83%, increasing with the increased growth amount while decreasing with the increasing Npi. The natural nitrogen supply from PIS to harvest ranged from 2.5 to 4 kg/10a, showing quadratic relationship with the shoot dry weight or shoot nitrogen content at PIS. The regression models to estimate PNup was formulated using Npi and anyone of RVIgreen, shoot dry weight, and shoot nitrogen content at PIS as predictor variables. These models showed good fitness with determination coefficients of 0.86 to 0.95 The prescription method based on the above models predicting grain yield, protein content and PNup and its constraints were discussed.

Effects of Enzyme Complex Supplementation on Growth Performance, Nutrient Digestibility, Blood Profiles and Economic Feed Cost in Growing Pigs (복합효소제의 첨가가 육성돈의 생산성, 영양소 소화율, 혈액성상 및 사료비용에 미치는 영향)

  • This study was conducted to evaluate effects of enzyme complex on growth performance, nutrient digestibility, blood profiles and feed cost in growing pigs. Ninety-six pigs [(Landrace ${\times}$ Yorkshire) ${\times}$ Duroc, $22.96{\pm}0.79$ kg average initial body weight] were used in 42d growth assay. Dietary treatments included:1) HC (high energy and nutrient density diet), 2) CON (control, basal diet), 3) CE1 (CON + 0.05% enzyme complex) and 4) CE2 (CON + 0.1% enzyme complex). Four pigs were allotted per pen with six replicate pens per treatment by completely randomized design. The ADG was higher in CE1 and CE2 treatments than CON treatment (P<0.05). The ADFI was linearly increased by CE treatments compared to HC treatment. The CE1 treatment had highest DM, N and GE digestibility (P<0.05). Digestibility of DM, N and GE were quadratic enhanced by enzyme complex level. No differences were found among treatments for creatinine and BUN. The enzyme complex treatments (CE1 and CE2) showed lower feed cost/body weight gain than HC treatment. In conclusion, enzyme complex can improve ADG and reduce feed cost/body weight gain when low energy diet was used. Furthermore adding 0.05% enzyme complex had highest nutrient digestibility.

Development of a Planting Density-Growth-Harvest Chart for Common Ice Plant Hydroponically Grown in Closed-type Plant Production System (식물 생산 시스템에서 수경재배한 Common Ice Plant의 재식밀도-생육-수확 도표 개발)

  • In this study, a planting density-growth-harvest (PGH) chart was developed to easily read the growth and harvest factors such as crop growth rate, relative growth rate, shoot fresh weight, shoot dry weight, harvesting time, marketable rate, and marketable yield of common ice plant (Mesembryanthemum crystallinum L.). The plants were grown in a nutrient film technique (NFT) system in a closed-type plant factory using fluorescent lamps with three-band radiation under a light intensity of $140{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ and a photoperiod of 12 h. Growth and yield were analyzed under four planting densities ($15{\times}10cm$, $15{\times}15cm$, $15{\times}20cm$, and $15{\times}25cm$). Shoot fresh and dry weights per plant increased at a higher planting density until reached an upper limit and yield per area was also same tendency. Crop growth rate, relative growth rate and lost time were described using quadratic equation. A linear relationship between shoot dry weight and fresh weights was observed. PGH chart was constructed based on the growth data and making equations. For instance, with within row spacing (= 20 cm) and fresh weight per plant at harvest (= 100 g), we can estimate all the growth and harvest factors of common ice plant. The planting density, crop growth rate, relative growth rate, lost time, shoot dry weight per plant, harvesting time, and yield were $33plants/m^2$, $20g{\cdot}m^{-2}{\cdot}d^{-1}$, $0.27g{\cdot}g^{-1}{\cdot}d^{-1}$, 22 days, 2.5 g/plant, 26 days after transplanting, and $3.2kg{\cdot}m^{-2}$, respectively. With this chart, we could easily obtain the growth factors such as planting density, crop growth rate, relative growth rate, lost time and the harvest factors such as shoot fresh and dry weights, harvesting time, marketable rate, and marketable yield with at least two parameters, for instance, planting distance and one of harvest factors of plant. PGH charts will be useful tools to estimate the growth and yield of crops and to practical design of a closed-type plant production system.

In Vitro Intrinsic Radiosensitivity Of Human Squamous Cell Carcinoma in Primary Culture (인체 상피 세포암의 일차 배양을 이용한 방사선 민감도 측정)

  • There are a number of reports suggesting that there may be a correlation between the clinical response to radiotherapy in various tumors and the clonogenic survival of cell lines derived from these tumors following exposure to 2 Gy(SF2). Authors conducted this study to determine SF2 for cells in primary culture from surgical specimens. The tumor tissues with squamous cell carcinoma of uterine cervix and head and neck were obtained. The tumor tissues were disaggregated to single cells by incubating with collagenase type w for 2 hours with constant stirring. Single cell suspensions were inoculated in four 24-well plates precoated with cell adhesive matrix. After 24 hours of incubation at 37$ ^{\circ}C $, rows of four wells were then irradiated, consisting of control set and five other sets each receiving doses of 1,2,3,4, and 6 Gy. After incubation for a total of 13 days, the cultures were stained with crystal violet and survival at each dose was determined by quantitative image analysis system, To determine whether cell growth was of epithelial origin, immunocytochemical staining with a mixture of cytokeratin and epithelial monoclonal antibodies were performed on cell cultures. During the period of this study, we received 5 squamous cell carcinoma specimens of head and neck and 20 of uterine cervical carcinoma. Of these, 15 yielded enough cells for radiosensitivity testing. This resulted an overall success rate of 60$ \% $. The mean SF2 value for 15 tumours was 0.55$\pm$0.17 ranging from 0.20 to 0.79. These results indicate that there is a broad range of sensitivities to radiation in same histologic type. So with a large patient population, we plan to determine whether a different SF2 value is associated with tumours that are controlled with radiotherapy than those that are not.

Environmental Controls on Net Ecosystem CO2 Exchange during a Rice Growing Season at a Rice-Barley Double Cropping Paddy Field in Gimje, Korea (김제 벼-보리 이모작 논에서 벼 재배기간 동안의 순생태계 CO2 교환량에 대한 환경요인 분석)

  • Using the Eddy Covariance technique, we analyzed seasonal variation in net ecosystem $CO_2$ exchange (NEE) and investigated the effects of environmental factors and aboveground biomass of rice on the $CO_2$ fluxes in a rice-barley double cropping paddy field of Gimje, Korea. Quality control and gap-filling were conducted before this investigation of the effects. The results have been showed that NEE, gross primary production (GPP), and ecosystem respiration (Re) during the rice growing period were -215.6, 763.9, and $548.3g\;C\;m^{-2}$, respectively. Relation between NEE and net radiation (Rn) could be described by a quadratic equation, and about 65 % of variation in NEE was explained by changes in Rn. On the other hand, an exponential function relating Re to soil temperature accounted for approximately 43 % of variation in Re under the flooded condition of paddy field. Aboveground biomass showed significant linear relationships with NEE ($r^2=0.93$), GPP ($r^2=0.96$), and Re ($r^2=0.95$), respectively.
