Lilies (Lilium spp.) are one of the most important ornamental flower crops grown in Korea. Most viral diseases in lilies are transmitted by infected bulbs, which cause serious economic losses due to reduced yields. Various diagnostic techniques and high-throughput sequencing methods have been used to detect lily viruses. According to Oxford Nanopore Technologies (ONT), MinION is a compact and portable sequencing device. In this study, three plant viruses, lily mottle, lily symptomless, and plantago asiatica mosaic virus, were detected in lily samples using the ONT platform. As a result of genome assembly of reads obtained through ONT, 100% coverage and 90.3-93.4% identity were obtained. Thus, we show that the ONT platform is a promising tool for the diagnosis and characterization of viruses that infect crops.
This study was carried out to examine tne incidences of virus diseases in lily plants cultivated in highland areas, and to develop an effective detection method. Viral symptoms on lilies in the highland areas were differentiated into mosaic, crinkle, mottle, stripe and line pattern. The distribution of symptoms on infected plants was 43.8% of mosaic, 29.2% of crinkle, and 10.9% of mottle symptoms. Six viruses such as Lily symptomless vires(LSV), Cucumber mosaic virus (CMV), Lily mottle virus (LMoV), Lily virus X (LVX, Potexvirus), Tabacco mosaic virus (TMV,Tobamovirus), and Tabacco rattle virus (TRV,Tobravirus) were detected from the infected lilies. Infection rate of Lilium oriental (cvs. Casablanca and Marcopolo) was 2~4 times higher than that of L. asiatic (cvs. Solemio and Prato). Virus detection on lilies by one-step RT-PCR (by using reverse transcription and polymerase chain reaction simultaneously) was more rapid rapid and reliable than by the conventional RT-PCR method.
This work was undertaken to study the effect of stigmatic and cut-style pollination on self seed set in Lilium longiflorum and L. ${\times}$ formolongi, and their crosses as the female parent with other cultivars/genotypes. Stigmatic pollination of L. longiflorum cv. Gelria and Lorina resulted in cent per cent fruit set with mean number of seeds/fruit of 189 and 70, respectively. However, cut-style pollination resulted in 87% and 40% fruit set in Gelria and Lorina, respectively. The corresponding mean number of seeds/fruit was 53 and 20. In L. ${\times}$ formolongi, stigmatic pollination set 80% fruits with 736 seeds/fruit. On the other hand, cut-style pollination resulted in 47% fruit set with 81 seeds/fruit. The intraspecific stigmatic pollination of L. longiflorum cv. Gelria and Lorina with other cultivars formed 60% fruits with a mean number of 18 seeds/fruit. However, only 20% fruit set with mean number of seeds/fruit of 7 was recorded when cut-style pollination of L. longiflorum cultivar were pollinated with other cultivars/genotypes. In the intraspecific pollination of L. ${\times}$ formolongi cv. Raizan with Novia, fruit set resulting from stigmatic and cut-style pollination was 75% and 50%, respectively with the corresponding mean number of seeds/fruit of 579 and 98. It was concluded that self as well as intraspecific seed set in the two species of Lilium gets considerably reduced as a result of cut-style pollination.
Lily mottle virus (LMoV) causes flower quality reduction in Lilium spp. The coat protein gene was RT-PCR-amplified from total RNA extracted from infected lily leaves and the amplified fragment was cloned into the pRSET expression vector tagged with a His-MBP. The plasmid of recombinant coat protein was used to transform an Escherichia coli strain pLysS and was expressed. The coat protein was purified by affinity chromatography using a Ni-NTA resin. The identity of the purified protein was confirmed by SDS-PAGE. The in vitro-expressed protein was used for immunization of mice. The polyclonal and monoclonal antibodies reacted specifically for the detection of LMoV in lily extracts in Western blot. Moreover the monoclonal antibodies reacted with lily extracts in DAS-ELISA with no unspecific or heterologous reactions against other non-serologically related viruses, but the polyclonal antibodies revealed a weak reaction against both infected lily and healthy control.

We investigated the effects of shading on the plant growth, flowering and the bulbil formation of Lilium species. As the shading treatment, we used full sunlight and various light intensity, 30%, 50%, 75%, 90% compared to full sunlight with shading net. The results obtained were as follows. The flowering rate and flower number of 'Roma' and 'Georgia' were not effected by various shading treatment. Days to flower in 'Roma' and Tiger lily showed similiar tendency like this. One of the shading effect was stem elongation of 'Connecticut king' markedly reduced compared with other cultivars in flowering rate and flower number. Leaf number of all cultivars was not consistent in tendency. But leaf number of Tiger lily and 'Connecticut king' reduced. The tendency of other experimented Lilium species weren't consistent. Bract leaf number reduced gradually as shading rate increased, plant height and internode length increased. Especially the height of 'Georgia' showed eminant increase compared with others. Total number and weight of bulbil of 'Enchantment' and Tiger lily reduced as shading rate increased.. But bulbil weight from bract part of 'Enchantment' in creased. The bulbils of 'Connecticut King' and 'Roma' appeared only on upper stem parts when treated serious shaded conditions, 75% and 90%. Only 'Connecticut King' and 'Enchantment' showed leaf emergence from bulbil among the L. elegans species.
The study aimed to identify effect of shading rate and duration on the growth and flowering of Lilium species in faring culture. Plant height of 'Gelria' and 'Casablanca' increased by higher shading at early growth stage, whereas that of 'Jolanda' and 'Dream Land' decreased. Also, plant height of 3 cultivars except 'Gelria' were longer by higher shading at whole growth stage, that of 'Gelria' was shorter as compared to in untreated plants. In cultivar 'Gelria', blasting increased and days to flowering shorted in higher shading only except 100% shading treatment at early growth stage. In 100% shading treatment, Blasting of 'Jolanda' was occurred 15.7% and the number of flower buds of 'Dream Land' and 'Casablanca' were reduced. Flower buds of 'Gelria' were not induceded in continued shading condition regardless of shading degree, the number of flower buds and flower size of 'Jolanda' and 'Dream Land' was reduced by 75% and 50% shading, respectively and in 'Casablanca', days to flowering was longer by shading at whole growth stage. Growth of daughter bulbs in 'Gelria', 'Jolanda' and 'Dream Land' was not influenced by shading at early growth stage but those of 'Casablanca' was inactive. Whereas that of 4 cultivars were inactive in continused shading condition.

A FA intersectional hybrid lily cultivar 'Cherry Pink' was developed in 2006 at National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA) Korea. The cross and immature embryo rescue was conducted between female parents Lilium FA hybrid 'Supia' and male parents L. Asiatic hybrid 'A94-9' by cut style pollination method (CSM) at Suwon in 1999. The first selection was done and was tentatively named as 'FA01-17' in 2001. After in vitro multiplication, bulbing production of 'FA01-17' line, growth and flowering characteristic tests were conducted from 2002 to 2005. The evaluation of characteristics and consumer preferences were surveyed at a lily flower show of NIHHS at Taean in 2006. 'Cherry Pink' flowered in the middle of June and grew more than 120 cm stem in length. Flowers bloomed facing upward, a little spotted in petals and deep pink (RHS, RP27D). The pollen of 'Cherry Pink' was sterile. Year-round flowering can be done by storing the bulb under $-1.5^{\circ}C$ conditions. It was needed to control Botrytis disease in wet season.

After in vitro multiplication, bulb-producing ability, line, growth and flowering characteristic of 'FA04-24' were evaluated from 2005 to 2007. The evaluation of characteristics and consumer preferences were surveyed at a lily flower show of NIHHS in 2008. 'Golden Center' flowers in the middle of June and grows up to 144 cm high in length. Flower blooms facing upward, with light yellow petals (RHS, GW157C). The pollen of 'Golden Center' is sterile. Year-round flowering can be done by storing the bulb under $-1.5^{\circ}C$ conditions. It is needed to control Botrytis disease in wet season.
The Lily mottle virus (LMoV) impedes the growth and quality of lily crops in Lanzhou, China. Recently Arabis mosaic virus (ArMV) has been detected in LMoV-infected plants in this region, causing plant stunting as well as severe foliar symptoms, and likely posing a threat to lily production. Consequently, there is a need to develop simple, sensitive, and reliable detection methods for these two viruses to prevent them from spreading. Reverse transcription (RT) loop-mediated isothermal amplification (LAMP) assays have been developed to detect LMoV and ArMV using two primer pairs that match six conserved sequences of LMoV and ArMV coat proteins, respectively. RT-LAMP assay results were visually assessed in reaction tubes using green fluorescence and gel electrophoresis. Our assays successfully detected both LMoV and ArMV in lily plants without the occurrence of viral cross-reactivity from other lily viruses. Optimal conditions for LAMP reactions were 65℃ and 60℃ for 60 min for LMoV and ArMV, respectively. Detection sensitivity for both RT-LAMP assays was a hundredfold greater than that of our comparative RT-polymerase chain reaction assays. We have also found this relatively rapid, target specific and sensitive method can also be used for samples collected in the field and may be especially useful in regions with limited or no laboratory facilities.
We studies seed set in the interspecific F1 /backcross hybrids of Lilium species. In the interspecfic hybrid of L. longiflorum cv. Gelria with L.${\times}$ fomolongi cv. Raizan,93% fruit set was obtained by stigmatic pollination in comparison to 53% from cut-style pollination. Accordingly, the number of seed set resulting from stigmatic and cut-style pollination was 147 and 53, respectively. Pollination o( both stigmatic and cut-style pollination resulted in 47% fruit set in the hybrid of L. longiflorum cv. Lorina with L.${\times}$ fomolongi cv. Raizan. However, stigmatic pollination formed 413 seeds, whereas only 24 seeds were obtained by cut-style pollination in this cross. The hybrid of L.${\times}$ fomolongi cv. Raizan with L. longiflorum cv. Come set 40% fruit with a total of 43 seeds by stigmatic pollination. However, no fruit set was observed in cut-style pollination in this hybrid. Backcrossing the F1 hybrid by cut-style pollination of L. longiflorum cv. Lorina ${\times}$ Asiatic hybrid cv. Chicago with the latter parent led to 53% fruit set, and 109 embryos were obtained. Likewise, backcrossing following cut-style pollination of L. longiflorum cv. Lorina ${\times}$ Asiatic hybrid cv. Corsia with the latter parent formed 67% fruits and 107 embryos. However, in the remaining interspecific hybrids, cut-style pollination set no fruit.
