Lilium ledebourii Bioss is a wild species of Lilium, which grows naturally in some provinces of Iran. Previous studies on Lilium tissue culture have been linked to direct regeneration and a few studies have been conducted on indirect regeneration, which has been studied under bright conditions. In this study, for the first time in the world, all the stages of indirect regeneration (callus induction, shoot and root induction) have been studied under dark conditions. Callus formation and the regeneration levels of L. Ledebourii Bioss were examined for three replicates in an MS (Murashige and Skoog) medium with different hormonal compositions and by using a factorial experiment in the framework of a completely random plan. For callus initiation, 2,4-D and kinetin hormones were used in five and four levels, respectively, as auxin and cytokinin. Results showed that the highest percentage of the callus was found in $3{\mu}M$ of 2,4-D and $0.5{\mu}M$ of kinetin. In terms of callus wet weight, the highest amount was found in $3{\mu}M$ of 2,4-D and $0.5{\mu}M$ of kinetin. In addition, in terms of diameter, the highest amount was found in $3{\mu}M$ of 2,4-D, and $0.5{\mu}M$ of kinetin. In summary, the 2,4-D hormone had a major impact on the percentage of regeneration increase so that the best response was related to the composition of $3{\mu}M$ of 2,4-D, and $0.1{\mu}M$ of kinetin. This study contended that auxin and cytokinin can induce long shoots and roots through cell elongation in dark condition.
Transient uidA expression was used to optimize parameters required for biolistic transformation of suspension cells of Easter lily, Lilium longiflourm. Maximum uidA expression occurred following bombardment with gold particles as compared to tungsten. A 3hr pre-treatment of suspension cells with 0.125M osmoticum resulted in a 1.5X increase in uidA expression. A helium pressure of 1550 psi combined with a particle travelling distance of 6cm resulted in maximum uidA expression as compared to either 1100, 1200, or 1800 psi. Transient transformation resulted in up to 493 uidA expressing cells/Petri plate. For stable transformation suspension cells of Lilium longiflorum, were co-bombarded with plasmid DNA containing cucumber mosaic virus (CMV) replicase under the rice actin (Act1) promoter and either the bar or PAT genes under the cauliflower mosaic virus (CaMV 355) promoter. Ten regenerated plants contained the transgene as analyzed by PCR, and two of the ten plants were confirmed to contain the transgene by Southern hybridization. The two transgenic plants were independent transformants, one containing the bar gene and the other both the CMV replicase and bar genes. Plants were sprayed at the rosette stage and found to be resistant to 1000 mg/L of phosphinothricin (Trade name-Ignite) indicating expression of the bar gene throughout the leaves when bar was under control of the CaMV 35S promoter.
To investigate the effects of MS medium strength and nitrogen concentration on bulblet formation from bulblet scale segment culture and bulblet growth from bulblet of Lilium oriental hybrid 'Casa Blanca', asiatic hybrid 'Mona', and longiflorum hybrid 'Hinomoto', 0.5∼2.0 strength of MS salts and 30∼120 mM nitrogen concentrations of MS medium were examined in vitro. The number of bulblets from bulb scale segment was favored in the strength of 0.5∼1.0 strength of MS salts or 30 mM total nitrogen concentration of MS medium in three cultivars. But the growth of bulblets formed in vitro was promoted in the 2 strength of MS medium or hight concentration of total nitrogen of MS medium up to 120 mM in three experimented cultivars.

This study was conducted to find out the proper chemicals and doses for controlling the bulb disease of Lilium Oriental hybrids 'Capablanca' and 'Marco Polo' cultivars during storage, and to ascertain any disadvantage of the treatment on shoot growth and flower development. Bulbs of 'Casablanca' and 'Marco Polo' cultivars were dipped in some fungicide solutions, including azoxystrobin, benomyl+thiram, and prochloraz, and culture broth of antagonistic bacteria for one hour. The rot was apparently inhibited by azoxystrobin and prochloraz solution dippings and it was not by benomyl+thiram in bulbs produced domestically. To improve physiological integrity, calcium chloride was treated. However, this treatment had no positive effects on bulb rot of lily. When bulbs were dipped in one-fifth of diluted broth. But the bulb rot was inhibited effectively as compared with untreated during storage. Benomyl+thiram and that with calcium inhibited the sprouting in 'Casablanca' and 'Marco Polo' cultivars during cultivation. Calcium and benomyl+thiram treatments decreased growth of leaf and flower but with no statistically significant difference. Other fungicidal materials treated to bulbs showed no effect on vegetative growth and flower quality.
Bulbs of Lilium Oriental hybrids cv. 'Casablanca' and 'Marco Polo' were grown in a plastic house in alpine area of Pyongchang Kangwondo. Saw-dust, black film, reflective film, transparent film, and white/black double film were used as mulching materials to ascertain the growth of foliage, the quality of flower, and the size of bulbs. Foliage weights of both lilies were higher when lilies were grown with mulching materials than control. There were no differences of bulb weight among the control and mulching treatments, except delayed bulb growth in black film and white/black double film. Mulching cultivation with saw-dust and reflective film stimulated foliage growth, root growth and bulb production, but black film delayed the growth and development. Transparent and white/black double films showed no effects on foliage growth and bulb development than the control. As mulching materials tended to delay lily growth, including foliage and bulb, an extension of growing term might be better to produce high quality lilies than common cultivation in alpine area.

Plants regenerated from in vitro cultures are associated with chromosomal variations, which have been generally found in long-term culture. Reducing plant culture age is one of the ways to reduce genetic and epigenetic changes. The present study focused on the efficient in vitro propagation of lily cultivars and has intensified to speed up bulb propagation for cryopreservation. The multiplication process applied in this experiment uses starting material, which the newly small bulb formed from bulb-scales in two lily cultivars. The adventitious bulb from bulb-scale tissue cultured on three different media following Murashige and Skoog (MS) basal medium supplemented with 1 g/L Charcoal, MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone with or without Charcoal, respectively. After about seven weeks, there is little change in the number of newly propagated bulbs in small bulbs of the two media. Compared to the both mediums, the number of the propagated bulbs is increased 5 times in MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone without Charcoal. After about seven weeks, the results of the propagation showed that the number of the propagated bulbs is increased 5 times in MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone without Charcoal compared to the both mediums. The number of propagated bulbs ranged from 5 to 6 and 4 to 6 with an average of 5 in Tropicalpink and Greenstar cultivars, respectively. There is little change in the number of newly propagated bulbs in small bulbs of other media. The multiplication process applied in this study may save in vitro culture period and effort.

"Hyehwa" an Asiatic lily cultivar was released in 1998 at National Institute of Horticultural and Herbal Science (RDA), Suwon, Korea. The cross was made in 1991 between Asiatic lily "White Bird", a white colored, and "Avignon", an unspotted scarlet red colored. It was preliminarily selected as A93-20 in 1993. Its multiplication, bulbing growth and flowering characteristic tests were conducted from 1994 to 1998. A new cultivar "Hyehwa" flowers in middle of June and grows 98.7 cm in height. Flowers bloom upward-facing, thick orange (RHS, 28A). Year-round flowering is possible by storage of the bulb under $-1.5^{\circ}C$ conditions. For forcing culture, it is necessary to add calcium to the fertilizer or remove side scales to prevent leaf scorch. Botrytis disease control is needed in the wet season.

'Yeeun' an Asiatic hybrid lily was released in 2005 at National Horticultural Research Institute (NHRI), Rural Development Administration (RDA), Suwon, Korea. The cross was made in 1994 between Liliium Asiatic 'White Bird' white colored, and Liliium Asiatic 'Cote d'Azur', pink colored variety. It was preliminarily selected as 'A96-5' in 1996. Multiplication and bulbing, and characteristic tests were conducted from 1997 to 2004. The evaluation of characteristics was investigated as 'Wongyo C1-21' in 2004 at Suwon. 'Yeeun' flowers in the end of June and grows 41.6 cm stem length. Flowers are upward-facing, unspotted in petals and yellow green (RHS, 18C). Year-round flowering can be by storing the bulb under $-1.5^{\circ}C$ conditions. For forcing culture, it is necessary to add calcium to the fertilizer or remove side scales to prevent leaf scorch. It is needed to control Botrytis disease in wet season.
Effects of harvest date, number of the leaves remained on stump after flower cutting, duration of cold treatment on the growth and flowering of L. elegans 'Connecticut King' bulb were tested. Shoot emerging rate increased over 90% by delayed cold treatment, late harvesting time and more leaves remained. Flowering rate increased over 90% when conducted cold treatment of 90 days to the premature bulb at flowering date, the bulb matured for 50 days with 10 leaves and the bulb matured for over 30 days with 20 leaves, and when conducted cold treatment of 50 days to the bulb matured for over 30 days with all leaves. Delayed cold treatment duration shortened days needed to shoot emergence and flowering. Especially days to flower of the premature bulb at flowering date were the shortest of all treatments, and can be flowered within 100 days after planting. In generally, plant height, number of flowers per plant and stem diameter generally increased by longer bulb maturing period after flower cuting, and the premature bulb harvested at flowering date showed poor growing and flowering responses. But, if used the larger size bulb, it is believed that the prematured bulb can be forced to flower normally in current year without bulb maturing period.

In this study, we reduced the duration of long-term culture and repeat subculture using small bulblets derived from bulb scales in two lily cultivars. The adventitious bulblets derived from bulb-scale tissue were cultured on three different media containing Murashige and Skoog (MS) basal medium supplemented with 1 g/L Charcoal, MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone with or without Charcoal, respectively. About seven weeks later, the number of newly propagated multiple shoots in the two media, A and B media, showed little differentiation. Compared to both media, the number of propagated multiple shoots increased 5-fold in MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone without Charcoal (C medium). The number of propagated multiple shoots ranged from 5 to 6 and 4 to 6 with an average of 5 in TropicalPink and GreenStar cultivars, respectively. The flow cytometric measurements indicated no variation in the ploidy level between control and in vitro propagated plants.
