Effects of Bulblet Formation on Scale Tissue Culture and Growth of Characteretis in Edible Lilium brownii spp. (식용 백합 Lilium brownii의 생육특성과 인편조직배양이 자구 형성에 미치는 영향)

  • This experiment was carried out to establish growth characterestics and number of bulblet, bulb weight, and diameter of bulblet on scale tissue culture in Lilium brownii. Flowering date of Lilium brownii was 25th, May with 50cm hight in field condition. Vimyl house condition was flowered 28th May with 54cm hight. Number of bulblet, bulb weight, diameter of bublet was good position of outer>Middle>inner scale in Lilium brownii. Number of bulblet formation was the best with 3.5 in large scale. Concentration of BA was the best at 1.5mg/L with 3.8 bulblet with bulb weight, weight and diameter of bulblet.

  • Cryopreservation is one of the ideal and suitable methods for long-term storage of plant germplasm. The plant contaminated with diseases and pathogens are decreased the multiplication rate, survival rate and high quality of plants after cryopreservation. The aim of this work was to improve a micropropagation method for lily in Korea, which are cultivars and indigenous plant. In the last process of rinsing scales after surface-sterilization, we tried to control the diseases and pathogens lived within the tissue by rinsing in 0.03% sodium hypochlorite (NaClO) instead of sterile distilled water. Bulb scales of Lilium were cultured in vitro on MS medium supplemented with Plant Preservative Mixture (PPM). The newly small bulb formed from bulb-scales was transferred to MS medium. We checked the non-contamination and survival rate after 2 weeks in culture. Non-contamination was shown to be 70 to 90% in formed small bulbs. This study will help to mitigate microbial contamination in Lilium species micropropagation for cryopreservation.

Chemical Constituents of Bulb of Lilium lancifolium Thunberg and Lilium tsingtauense Gilg (참나리와 하늘말나리 인경의 성분분석)

  • In this study, the chemical constituents of bulb from Lilium lancifolium and Lilium tsingtauense were investigated. The content of moisture, crude protein and fiber of bulb from L. tsingtauense was higher than that of L. lancifolium, whereas the content of crude ash and nitrogen free extract of bulb from L. lancifolium was higher than that of L. tsingtauense. HPLC analysis revealed that total free sugar and amino acid content of bulb from L. tsingtauense was higher than that of L. lancifolium. There was no remarkable difference in proximate composition between the two species. The content of p-coumaric acid and ferulic acid was 1.93 mg% and 0.25 mg% in ethyl acetate fraction of L. lancifolium, on the other hand, 2.94 mg% and 0.46 mg% in ether fraction of L. tsingtauense. These results suggest that the bulb of L. tsingtauense can be considered as promising oriental medicine resource likewise L. lancifolium.

Effect of Inorganic Salts in MS Medium, Sucrose, and Activated Charcoal on Bulblet Formation from in Vitro Bulbscales in Lilium Oriental Hybrid 'Casa Blanca' (MS 배지 무기물, 당 및 활성탄의 농도가 Lilium Oriental Hybrid 'Casa Blanca'의 기내인편으로부터 자구형성에 미치는 영향)

  • The effects of MS salt strength, sucrose, and cultural conditions on bulblet formation and growth were investigated to optimize the conditions for micropropagating bulblets from in vitro bulb scales of Lilium Oriental Hybrid 'Casa Blanca'. There was no difference on bulblet formation in the range of 1/2~2 $\times$ strength of MS salt, but it was inhibited remarkably in 3 $\times$ strength of MS salt. The growth of regenerated bulblets was most stimulated on MS basal medium. Favorable bulblet formation and its growth from bulb scales were achieved when grown on the media with 1 : 2 or 1 : 3 in the ratio of NH$_4$^+ : NO$_3$^- , as well as on MS basal medium (NH$_4$^+ : NO$_3$^- = about l : 2). Therefore, MS basal medium was very suitable for bulblet formation and growth from bulb scales. Bulblet formation was inhibited but its growth was stimulated with increase sucrose concentration in the medium. The growth of regenerated bulblets was very effective on the media with 9~12% sucrose. Addition of activated charcoal (AC) to the medium inhibited bulblet formation from bulb scales, but enhanced the growth of regenerated bulblets. Especially, the medium containing 1 g/L AC was most effective on the growth of bulblets. No difference was found on bulblet formation and growth from bulb scales under light and dark conditions. In vitro micropropagation of L. Oriental Hybrid 'Casa Blanca' was supposed very reasonable to enhance the growth of the bulblets after forming of bulblets from in vitro bulb scales, and then, subculture the bulb scales from the grown bulblets on MS medium with 9% sucrose and 1 g/L AC.

Preconditioning for Cryopreservation of in vitro Grown Bulblets of Lily using Droplet-Vitrification

  • This study was conducted to improve and supplement the system of cryopreservation for adventitious bulbs induced by tissue cultured bulb-scales of lily (Lilium spp.) cvs. 'Milky way'. The explants, bulblets and bulb-scale-bulblets, were treated to low temperature (4℃) for 7 days prior to the pre-culture. The adventitious bulbs were pre-cultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3 and 0.7M). The pre-cultured adventitious bulbs were treated to loading solution (LS1 or LS2, C4 or C6) containing 35% of PVS3 (LS1, C4) or 40% of PVS3 (LS2, C6) for 40 min and exposed to dehydration solution (PVS3, B1) containing 50% glycerol and 50% sucrose for 60 min at 25℃. The adventitious bulbs were moved onto droplets containing 3 µl PVS3 on sterilized aluminum foils, and then soaked into liquid nitrogen (LN) for 60 min. The result of highest regrowth rate as 65.7% was obtained in cold treatment (4℃), osmoprotected with LS1 solution, and cultured in PCM3 medium by using bulb-scale-bulblet for cryopreservation. This result shows that droplet-vitrification could be used as a promising method for long-term storage of lily genetic resource.

Bulb of Lilium longiflorum Thunb Extract Fermented with Lactobacillus acidophilus Reduces Inflammation in a Chronic Obstructive Pulmonary Disease Model

  • Chronic obstructive pulmonary disease (COPD), one of the leading causes of death worldwide, is caused by repeated exposure to harmful matter, such as cigarette smoke. Although Lilium longiflorum Thunb (LLT) has anti-inflammatory effects, there is no report on the fermented LLT bulb extract regulating lung inflammation in COPD. Thus, we investigated the protective effect of LLT bulb extract fermented with Lactobacillus acidophilus 803 in COPD mouse models induced by cigarette smoke extract (CSE) and porcine pancreas elastase (PPE). Oral administration of the fermented product (LS803) suppressed the production of inflammatory mediators and the infiltration of immune cells involving neutrophils and macrophages, resulting in protective effects against lung damage. In addition, LS803 inhibited CSE- and LPS-induced IL-6 and IL-8 production in airway epithelial H292 cells as well as suppressed PMA-induced formation of neutrophil extracellular traps in HL-60 cells. In particular, LS803 significantly repressed the elevated IL-6 and MIP-2 production after CSE and LPS stimulation by suppressing the activity of the nuclear factor kappa-light-chain-enhancer of activated B (NFκB) in mouse peritoneal macrophages. Therefore, our results suggest that the fermented product LS803 is effective in preventing and alleviating lung inflammation.

Effects of Bulb Circumference and Cultivar on the Cut Flower Quality in Lily. (백합의 품종 및 구근크기가 절화품질에 미치는 영향)

  • This study was carried out to investigate cut flower quality bulb circumference and cultivars affecting 7 different cultivars to produce high quality cut flower lilies. To determine the effects of bulb circumference on growth and cut flower quality of 7 different cultivars, Lilium oriental hybrid 'Chilli', 'Ismael', 'Medusa', 'Mother's Choice', 'Siberia', 'Sorbonne', and 'Yelloween' were tested. After cut flower quality experiment, the highest 1st flower bud length and cut flower height were 'Medusa' and 'Mother's Choice' respectively. Most plants had over $30^{\circ}$ stem bending angle except for 'Mother's Choice', and 'Ismael'. The study shows that the bigger bulb circumference has the better 1st peduncle diameter, inflorescence length, stem diameter, number of flowers, and fresh weight. Bulb circumference affects the 1st flower bud length, cut flower height and number of flowers. Stem bending angle, however, is affected by neither bulb circumference nor cultivar. In conclusion, growth, cut flower quality and vase life were affected by the bulb circumference rather than cultivar.

Conservation of An Endangered Species of Lilium cernum Komarvo. through in vitro Mass-Propagation (멸종위기 종 솔나리의 기내 대량생산을 통한 보존 연구)

  • This study was conducted to establish the optimal condition for conservation of genetic resources and micropropagation of Lilium cernum. Induction of bulbet of L. cernum was highly effective (9.2 bulb/explant) on 1/2 SH (Schenk and Hildebrandt) medium supplemented with 1.0 mg/L TDZ (Thidiazuron) and 0.1 mg/L NAA (Naphthaleneacetic acid). The treatment of 0.1 mg/L NAA increased root development (6.4 root/explant) under the in vitro condition. In addition, treatments of AC (Activated Charcoal) and ventilation were enhanced to develop number of shoots and to elongate length of leaf, bulb and root. Futhermore, the process of short-term soil acclimatization was promoted to strengthen the plantlets induced under the in vitro condition.

'Honghwa' an Asiatic Hybrid Lily with Unspotted Orange Petals

  • 'Honghwa' an Asiatic hybrid lily was released in 2001 at National Horticultural Research Institute (NHRI), Rural Development Administration (RDA), and Suwon, Korea. The cross was made in 1994 between Lilium Asiatic 'Avignon', a scarlet red colored, and L. Asiatic 'Connecticut King', bright yellow colored. It was preliminarily selected as 'A96-3' in 1996. Multiplication and bulbing, and characteristic tests were performed from 1997 to 2000. The evaluation of characteristics was made investigated as 'Wongyo C1-31' in 2001 at Suwon. 'Honghwa' flowers at the beginning of June and grows to 111.4 cm stem length. Flowers are upward-facing, unspotted with orange petals. Year-round flowering is possible by storing the bulb at $-1.5^{\circ}C$ conditions. It is necessary to add calcium to the fertilizer or remove side scales to prevent leaf scorch. It is needed to control Botrytis disease during wet season.
