This study was designed to investigate the possible utilization of Ligularia stenocephala as a source of functional ingredients in white bread. In terms of color values, as more Ligularia stenocephala leaf powder was added, 'lightness,' 'redness,' and 'yellowness' decreased in the crust, while in the bread crumb 'lightness' and 'yellowness' decreased, but 'redness' increased. When 5.0%, 7.5%, or 10.0% Ligularia stenocephala leaf powder was substituted for an equivalent amount of strong flour, the ratio of the volume and specific volume of white bread decreased while its weight increased. In the texture measurements for the white breads, the hardness decreased slightly with the addition of 2.5% Ligularia stenocephala leaf powder, but increased with the addition of 5.0%, 7.5%, or 10.0% Ligularia stenocephala leaf powder. The addition of the Ligularia stenocephala leaf powder also increased the springiness and gumminess of the white bread. The highest sensory scores for color, flavor, taste, texture, and overall acceptance for both the student group and the baker group were obtained from white bread with 5.0% Ligularia stenocephala leaf powder. The highest sensory scores for the housewife group for taste, texture, and overall acceptance were obtained from control bread. Through the results of these experiments, we conclude that the highest quantity of Ligularia stenocephala leaf powder content should be no more than 5.0% when making bread with added Ligularia stenocephala leaf powder.
The purpose of this study was to investigate the volatile flavor compounds of Ligularia stenocephala and Ligularia fischeri, edible wild plants. The volatile flavor compounds were isolated by the simultaneous distillation extraction method and analyzed by GC-MSD (gas chromatography-mass selective detector). Forty-eight volatile flavor compounds were identified in the extracts from L. stenocephala and the major compounds were sabinene, cis-ocimene, trans-caryophyllene, and β-elemene. Fifty-one volatile flavor compounds were identified in the extracts from L. fischeri and the major compounds were α-pinene, germacrene-D, transcaryophyllene, endo-1-bourbonanol and 1-limonene. The common volatile flavor compounds between two plants were transcaryophyllene, sabinene, β-elemene and β-cubebene etc. However, α-phellendrene and myrcene were identified in L. stenocephala, but were not identified in L. fischeri. However α-pinene, germacrene-D and limonene were identified in L. fischeri but were not identified in L. stenocephala.
In the previous study, we reported the antioxidative activity and inhibition of melanogenesis of Ligularia stenocephala extracts. In this study, inhibitory effect on elastase and production type I procollagen were investigated. The compounds which was investigated were 4-hydroxyacetophenone, vanillin, chlorogenic acid, caffeic acid isolated from Ligularia stenocephala. They were slightly mild on elastase inhibition activity but 4-hydroxyacetophenone, vanillin exhibited good inhibition activity on collagen production. These results suggest that Ligularia stenocephala may have potential as an anti-aging ingredient in cosmetics.
본 연구는 산마늘, 곰취, 곤달비를 대상으로 피음 수준을 전광 처리구(상대 투광율; 100%), 약피음 처리구 (상대 투광율; 64~73%), 보통피음 처리구(상대 투광율; 35~42%), 강피음 처리구(상대 투광율; 9~16%)의 4단계로 달리하여 이들의 생장 특성 및 ascorbic acid 함량을 조사 분석하였다. 산마늘의 엽면적과 총 물질생산량은 보통피음 처리구에서 가장 높았지만 곰취와 곤달비는 엽면적과 총 물질생산량이 약피음 처리구에서 가장 높았다. 또한 세 식물의 엽두께는 피음 수준이 증가할수록 감소하였으며 피음 수준이 증가할수록 지하부의 생장보다 지상부의 생장이 양호하여 T/R율은 증가하였다. ascorbic acid 함량은 산마늘이 곰취와 곤달비보다 많았으며, 산마늘은 보통피음 처리구에서, 곰취와 곤달비는 약피음 처리구에서 가장 많은 함량을 보였다.
본 연구는 곤달비(Ligularia stenocephala)의 생리 효능을 체계적으로 밝히기 위한 기초 자료를 얻고자 80% 에탄올 추출물과 순차적 분획물에 대하여 항산화 및 항균 효과를 검증하였다. 곤달비 에탄올 추출물의 추출수율은 21.36%이었으며, 이에 대하여 용매별로 분획한 수율은 water 분획물이 60.35%로 가장 높았으며, n-butanol, n-hexane, ethyl acetate 순이었다. 곤달비 추출물 및 분획물의 총 폴리페놀 함량은 ethyl acetate 분획물에서 254.00 mg/g으로 가장 높게 나타났다. DPPH radical 소거활성을 측정한 결과, 곤달비 ethyl acetate 분획물에서 $IC_{50}$ 값이 0.28 mg/mL로 높은 활성을 나타내었다. 0.5 mg/mL 농도에서의 ABTS 라디칼 소거활성은 ethyl acetate 분획물에서 96.67%의 활성을 보였다. FRAP 활성 역시 1 mg/mL 농도에서 ethyl acetate 분획물이 2.70 mM의 가장 높은 활성을 나타내었다. 항균활성은 B. cereus 균주에 대해 ethyl acetate 분획물 5 mg/disc에서 10.2 mm의 저해환을 나타내었다. 이상의 결과에서 곤달비의 추출물 및 분획물 중 ethyl acetate 분획물의 경우 높은 항산화 활성과 약간의 항균활성을 나타내었으며 천연물소재로서 식품첨가물 및 화장품 원료로의 가능성이 있는 것으로 사료된다.
Ligularia stenocephala has been widely used as a traditional medicine for the treatment of lots of diseases such as asthma, arthritis, and jaundice. In this study, we evaluated the anti-nocicepitve activities of methanolic extract of L. stenocephala (MLS) using various pain models including thermal nociception as well as chemical nociception methods. MLS showed significant increase in latency time in the tail immersion test and hot-plate test. In addition, the number of acetic acid-induced abdominal constrictions was decreased by MLS. MLS also attenuated paw licking time in the formalin test. The combination test using naloxone revealed that the anti-nociceptive properties of MLS was not associated with opioid receptor activation. The present results demonstrate that MLS may possibly used as valuable analgesic agent for the treatment of nociceptive pain.
Ligularia stenocephala has been used as a traditional medicine for the treatment of asthma, arthritis, jaundice, and hyperpiesia. In this study, we investigated the anti-metastatic and hypnotic effects of the methanolic extract of L. stenocephala (MLS). Gelatin zymographic analysis revealed that MLS suppresses matrix metalloproteinase-2 (MMP-2) and MMP-9 activities in B16F10 cells. The gene expressions of MMPs were also down-regulated by MLS treatment in a dose-dependent manner. In addition, cancer cell invasion and migration were attenuated by MLS via suppression of NF-${\kappa}B$ activation. The in vivo lung metastasis of B16F10 melanoma cells was also inhibited by the treatment of MLS. These findings show that MLS has anti-metastatic properties, and, therefore, it might be applicable as a valuable anti-metastatic agent.
Screening was conducted using 200 kinds of plant extracts to explore herbicide-activated components of plant origin. We separated and purified active substances and elucidated chemical structures using Ligularia stenocephala M., which has strong activity and has not yet been studied. When the solvent fractions of the leaves of Ligularia stenocephala M. were tested for their herbicidal activity, ethyl acetate and chloroform layer showed an inhibition rate of 95.2% and 94.1%, respectively. In particular, the chloroform layer exerted more than 50% herbicidal activity at 10 ppm. From the chloroform layer with the highest herbicidal activity, we isolated three herbicidal active compounds using stepwise chromatography, specifically silica gel or octadecyl silica (ODS) column chromatography, Sep-pak cartridges, and high performance liquid chromatography (HPLC). Based on the analysis of the active compounds using electron ionization mass spectroscopy (EI-MS), 1H-NMR, and 13C-NMR, we identified the active compounds as euparin, 5,6-dimethoxy-2-isopropenylbenzofuran, and liguhodgsonal. When the herbicidal activity of the identified compounds was tested, euparin showed selective herbicidal activity for lettuce at 10-3 M, and both liguhogsonal and 5,6-dimethoxy-2-isoprophenylbenzofuran exerted selective activity for rice and Echinochloa crus-galli.
The purpose of this study was to evaluate the antioxidant and anti-adipogenic activities of Ligularia stenocephala (L. stenocephala) extract. The contents of the total polyphenol of the extract was 55.950 mg GAE/g residue. Antioxidant activities of L. stenocephala were evaluated by free radical scavenging ability and a reducing power test. 2,2'azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and ${\alpha}$-${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) free radical scavenging activities of the extract were approximately 90% and 70%, respectively. Reducing power of the extract was 258.833 mg TE/g residue. The anti-adipogenic activity of L. stenocephala extract was examined in 3T3-L1 cells. During adipocyte differentiation, the 3T3-L1 cells were treated both with and without the extract. L. stenocephala extract suppressed the lipid accumulation in a concentration-dependent manner in the 3T3-L1 cells. The L. stenocephala extract inhibited the expression of peroxisome proliferator activated receptor ${\gamma}$ ($PPAR{\gamma}$) and adipocyte protein 2 (aP2) proteins, compared with control adipocytes. These results indicate that L. stenocephala could be regarded as a potential source natural antioxidant and an anti-obesity agent.
In this study, we evaluated anti-oxidation and whitening effects of Ligularia stenocephala extract for use as the cosmeceuticals. L. stenocephala was extracted by three different solvents which was n-Hexane, ethyl acetate, $H_{2}O$. The free radical (1,l-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity of extract of L. stenocephala was in the order: ethyl acetate fraction of leaf ($IC_{50}$ value of 10.512ug/mL) > ethyl acetate fraction of stem ($IC_{50}$ value of 31.877ug/mL) > $H_{2}O$ fraction of leaf ($IC_{50}$ value of 129.194ug/mL). Reactive oxygen species (ROS) scavenging activity of extract of L. stenocephala was in the order: ethyl acetate fraction of leaf ($IC_{50}$ value of 0.230mg/mL) > ethyl acetate fraction of stem ($IC_{50}$ value of 0.528mg/mL) > $H_{2}O$ fraction of leaf ($IC_{50}$ value of 0.799mg/mL). Tyrosinase inhibition activity of L. stenocephala extracts was reduced 29.477% on ethyl acetate fraction of leaf, 13.583% on ethyl acetate fraction of stems. Therefore, L. stenocephala extracts may be useful as a new antioxidant and whitening agent to inhibit melanogenesis.
