Action Spectra of Apoptosis Induction and Reproductive Cell Death in L5178Y cells in UV-B Region

  • It is important to determine the action spectrum of UV-B radiation contained in the sunlight to estimate the risk of skin cancer. We have investigated action spectra for induction of apoptosis and reproductive cell death in L5178Y cells using the Okazaki Large Spectrograph at NIBB. L5178Y cells were exposed to light at different wavelengths in UV-B or UV-A region. Frequencies of apoptosis induction and reproductive cell death were determined by counting cells with chromatin condensation, and by the colony formation assay, respectively. The measured sensitivity spectra for the two end-points were in very good agreement. Sensitivity decreased steeply with increase of wavelength in UV-B region and remains nearly constant in UV-A region. The action spectra were also slightly steeper than that for the minimum erythematic dose (MED), but very similar to the light absorption spectrum of DNA in UV-B region. On the other hand, the spectra for both endpoints were similar to MED spectrum but not DNA spectrum in the UV-A region. Also different time-course and morphological difference of apoptosis were found between UV-B (long time, fragmentation) and UV-A (short time, shrinkage) region. These results suggest that DNA damage induced by UV-B light triggers apoptosis and reproductive cell death, but other damaged targets (membrane, protein and so on) trigger these effects in UV-A region.

Callus Induction and Plant Regeneration from Stolon in Zoysiagrass (한국잔디류에서 포복경 배양을 통한 캘러스 유기와 재분화에 관한 연구)

  • This study was carried out to induce and maintain callus from 59 zoysiagrass lines, to know the effective disinfestation method for zoysiagrass stolon as explant and the difference in the response of callus induction among 59 lines, and to investigate the effect of medium, growth regulators, light, temperature, stolon part and internode position on callus induction and emhryogenic callus(E.C.) formation. The treatment of 0.lmg/L $HgCl_2$for 15 min resulted in no contamination and the highest callus induction(46.6%). Callus was induced from the 59 zoysiagrass lines. The callus growth of Z. japonica and Z. sinica was generally better than Z. matrella Ten cell lines whose callus and stolon grow fast in culture and in field, respectively were selected to he used for breeding. Callus induction was the most effective at 2.0mg /L of both 2, 4-D and picloram in MS medium. MS medium was the best for callus induction and growth while LS medium was the best for embryogenic callus and shoot formation. Callus induction and growth was better at 28, 31$^{\circ}C$. than 25$^{\circ}C$. and dark condition was better than light condition in MS me-dium containing 2mg/L 2,4-D. While callus induction was better with node part as explant than with internode part, callus growth and embryogenic callus formation was better with internode part. In 'Japonica 1', the first internode was the most effective in callus induction, but third internode was the best in '$M_2$ X $S_2$'.

Induction Motor with Adjustable Windings for High Efficiency Drive in Light Load Operation

  • Heavy load start but light load operation is a common case in practical drive applications. When an induction motor is employed for such applications, its rated power is usually chosen according to the heavy load start. Then, during light load operation, its efficiency and power factor are low. To solve this problem, it is proposed to adjust the motor windings from the startup to the normal operation conditions. In this paper, arrangement of the adjustable windings is introduced, air gap field with different windings is investigated, and steady state operation performance under various loads is examined. It can be seen that by using proper winding arrangement both startup and operation performances are satisfactory.

Photoperiodic Floral Induction in Pharbitis Cotyledons Affected by Polyamines and Ethylene

  • Exogenous putrescine of 0.5 mM or higher concentratons applied during a 16 h inductive dark period could elevate putrescine content in cotyledons of Pharbitis nil Choisy cv. Violet, a short-day plant, resulting in complete blocking of photoperiodic floral induction. Titers of putrescine, spermidine and spermine in the cotyledons were traced throughout a 16 h dark period. While non-induced cotyledons under continous light slightly increased levels of polyamines, induced tissue maintaiend its putrescine, spermidine and spermine levels as low as 66.4%, 60.9% and 84.9% of non-induced levels respecitvely. Endogenous polyamines kept at lower levels in the inductive dark period were found to upsurge by a night break treatment of 10 min light in the middle of the dark and consequently the inductive dark effect was canceled. Elevation of polyamine titers could also be induced by 100 $\mu$L/L ethylene treatment which completely suppressed floral induction. Compared to untreated cotyledons, ehtylene-treated tissues increased putrescine content by as much as 136.5% in 12 h and spermidine level by up to 130.1% in 8 h. Ethylene-treated cotyledons not only increased endogenous polyamine content but also liberate ethylene in the second half of the inductive dark period accumulating up to three to fourfold level supporting a hypothesis that ethylene-treated tissues are stimulated to produce ethylene which in turn accelerates polyamine biosynthesis in the tissues. It is postulated that substantially low polyamine titers in the inductive dark period would be one of the necessary factors controlling photoperiodic induction of flowering in Pharbitis nil and the inhibitory effects of night break and exogenous ethylene treatment may be atributed to their action to stimulate endogenous polyamine production.

A New Approach to Improve Induction Motor Performance in Light-Load Conditions

  • Induction motors often reach their maximum efficiency at the nominal load. In most applications, the machine load is not equal to the nominal load, thus reduces the motor efficiency and turns a greater percent of power into loss. In this paper, the induction motor control problem has been investigated to reduce the system losses. The Field Oriented Control method (FOC) has been employed in this paper. In this research, the mathematical equations related to system losses are calculated in relation to torque and speed, and then the q- and d-axis are summarized according to the current components. After that, the proposed method is applied along with d- and q-axis. In the recent three decades, many techniques have been suggested to improve the induction motor performance using smart and non-smart methods. In this paper, a new PSO-Fuzzy method have used in real time. The fuzzy logic method serves as speed controller in q-axis and PSO algorithm controls the optimum flux in d-axis. It will be proved that the use of this combined method will lead to a significant improvement in motor efficiency.

In vitro Shoot Propagation Derived from Stem and Shoot Tip in Hovenia dulcis var. koreana Nakai by Plnat Growth Regulators and Light Resources (식물생장조절제 및 광원처리에 따른 헛개나무 줄기와 경정유래 신초의 기내증식)

  • This study was conducted to examine effects of plant growth regulators and light resources on the formation of multiple shoot and plant regeneration of Hovenia dulcis var. koreana Nakai. Stem and shoot tip were cultured on MS medium or WPM supplemented with various plant growth regulators. At the single treatment, the highest shoot formation was obtained when stem explants were cultured on WPM supplemented with kinetin $1.0mg{\cdot}L^{-1}$. MS medium containing NAA 0.1 and TDZ $0.1mg{\cdot}L^{-1}$ gave the best results for shoot induction rate and shoot growth in combination treatments. Of the BAP and kinetin tested, BAP $0.5mg{\cdot}L^{-1}$ on WPM was found to be more effective for shoot growth from shoot tip. Under white fluorescent light treatment, shoot growth was much higher than blue, red LED treatments. Root induction from in vitro growth of plantlet was the best on WPM supplemented with $1.0mg{\cdot}L^{-1}$ IBA. The results suggest that selection of plant growth regulators and light resources could be important factor to achieve an efficient in vitro growth.

Light-Dependent Chilling Injury on the Photosynthetic Activities of Cucumber Cotyledons (저온처리한 오이의 자엽에서 광합성 활성의 광의존성 저해)

  • The photosynthetic activities in relation to oxygen evolution rates, quantum yield, CO2 uptake rates and room temperature chlorophyll fluorescence were investigated in cotyledons of cucumber seedlings exposed to low temperature (at 4$^{\circ}C$) for 24 h. Light-chilling caused more inhibition on light-saturated maximum oxygen evolution rates, quantum yield, and CO2 uptake rates than dark-chilling did in the cucumber plant. Light-chilling induced more marked increase in Fo and decrease in (Fv)m/Fm than dark-chilling did in the room temperature chlorophyll induction kinetics. The above results affected by chilling in the light are considered to be associated with the partial damage of the reaction center of PS II and the decreased photosynthetic activities. There occurred a large decrease in qQ with little change in qNP in the light-chilling plant. When light- and dark-chilled plants were recovered at room temperature for 24 h and their chlorophyll fluorescences were induced with light doubling technique, light-chilled plants showed more smaller magnitude and rate of fluorescence relaxation than dark-chilled plants. These suggest that light-chilling might cause some alterations in transthylakoid pH formation, and that photosynthetic apparatus of cucumber cotyledons is more susceptible to light-chilling. In the fast fluorescence induction kinetics, FR was decreased by 60% in the light-chilled plants with reference to $25^{\circ}C$ light-grown plants, while the dark-chilled plants showed a decreased rate of only 20% with reference to $25^{\circ}C$ dark-treated plants for 24 h, indicating that cucumber seedling is very sensitive to chilling stress. So, it is certain that chilling injury to the photosynthetic apparatus is strongly dependent on the presence of light in cucumber seedlings.

  • This paper is concerned with the simulation and determination of the input voltage and frequency for the optimal efficiency operation of induction motors. In general, induction motors have a specific character that operation efficiency is dropped sharply at the light roads condition. Consequently, if the induction motor is controlled by high efficiency using the VVVF(variable voltage variable frequency) control methods at optimal values, the entire system can obtain the substantial energy savings from the efficiency improvement in induction motors. In this paper, optimal slip is derived from the modeling of an induction motor and the optimal hybrid-control method is suggested by the simulation of the proposed algorithm for a 3-phase induction motor.

A Study on the Flow Field Characteristics of Air Induction System for Reducing the Signal-to-Noise in the MAFS Output

  • This study presents the flow visualization results, velocity and turbulence intensity measurements made within an air filter cover and entry region of a mass air flow sensor (MAFS) which is used in an induction system of 3.8L engine. Flow structure in two air filter cover assemblies were examined. The first was a clear plastic replica of the production cover while the second was a modified clear plastic cover with a geometry configured to reduce fluctuations. High speed flow visualization and laser doppler velocimetry (LDV) systems were used to reveal and analyze the flow field characteristics encountered in the sensor design process under steady flow conditions. A 40-watt copper vapor laser was used as a light source. Its beam is focused down to a sheet of light approximately 1.5mm thick. The light scattered off the particles was recorded by a 16mm high speed rotating prism camera at 5000 frames per second. A comparison of the flow patterns and LDV measurements in the original and modified air filter covers is presented to illustrate the controlling effect of the cover design on the turbulence structure formation near the bypass and on the sensor output signal. In both axial and radial planes of the main passage it was found that the turbulence flow pattern is remarkably influenced by the air filter cover and main passage configuration.

  • Light Rail Transit (LRT) System is an urban transit system which has approximately an intermediate transportation capacity between conventional subway and bus ($5,000{\sim}25,000$ persons per hour and per direction). It is a high-tech system which operational capacity, punctuality and mobility are remarkably improved. There are some types of LRT systems such as monorail, tramway, AGT(Automated Guideway Transit), and so on. The LRT systems have been applied and being operated in about a hundred lines around the world and many projects that apply the LRT systems in Korea are being proceeded and scheduled. For the efficient management, economical construction, and safe operation of various LRT systems, the establishment of national standard is necessary such as vehicle standardized specification, vehicle performance test standard, vehicle safety standard, construction guide, operation regulation, etc. of LRT systems. This paper presents the standard specification of electrical equipment of linear induction motor type light rail vehicle, that is LIM AGT(AGT system propelled with linear induction motor) vehicle. The LIM AGT system has been applied in Japan subway and ART(Advanced Rapid Transit) system of Canada and Yongin LRT is currently under construction.
