Microarray analysis of hypoxia-induced changes in gene expression in BV-2 microglial cells (BV-2 microglia 세포주에서 저산소증의 유전자 발현에 대한 마이크로어레이 분석)

  • 목적 : 허혈시 발생되는 저산소중 상태에서는 세포독성을 유발한다고 알려져 있으나 정확한 기전은 아직 규명되지 않았다. 본 연구에서는 뇌허혈로 인한 세포독성의 기전을 유전자 발현을 통하여 살펴보고자 하였다. 방법 : 본 실험에서는 BV-2 microglia 세포주에 12시간 동안의 저산소 상태에서의 유전자 발현을 분석하기 위하여 마이크로에레이를 시행하였다. 결과 : 저산소 상태에서는 정상에 비하여 cathepsin F, growth factor independent 1, calcitonin/calcitonin-related poly, leucine-rich repeat LGI family membrane, dublecortin, cyclohydrolase 1, Ia-associated invariant chain, carbohydrate kinase-like과 erythrocyte protein band 4.1-like 3 등의 유전자 발현이 3배 이상 증가하였다. 한편 neuronal guanine nucleotide exchange factor, Bcl-2-related ovarian killer protein, chemokine (C-X-C motif) ligand 5, RNA binding motif protein 3, interleukin 2 receptor, alpha chain, crystallin zeta, cytochrome P450 subfamily IV B, asparagine synthetase과 moesin 등의 유전자 발현은 0.2배 이하로 감소하였다. 결론 : 이상의 결과는 저산소중에 관여하는 유전자 및 저산소중과 관련된 뇌경색 등의 질환의 기전을 밝히는데 기초적 자료로 이용될 수 있을 것이다.

Optical Resolution of DABS-Amino Acids with Mobile Chiral Chelate Addition (키랄킬레이트 이동상첨가법에 의한 답실아미노산의 광학이성질체 분리)

  • Optical isomers of DABS-amino acids have been separated in a reversed phae high performance liquid chromatography by adding Cu (Ⅱ)-L-Proline chelate to the mobile phase. The retention behaviors for the DABS-amino acids are discussed in terms of pH of the mobile phase and the concentrations of acetonitrile, Cu (Ⅱ) complex, and buffer. The selectivity of the optical isomers of DABS-amino acids increases with the pH of the mobile, and the concentration of the chelate, but decreases with concentration of the oganic modifier. The concentration of buffer does not affect the optical separation selectivity. A separation mechanism is illustrated by cis and trans formation based on the steric effect of the ligand exchange reaction between DABS-amino acids and the copper chelate.

Separation of Lithium Isotopes by Tetraazamacrocycles Tethered to Merrifield Peptide Resin

  • Tetraazamacrocyclic ion exchangers tethered to Merrifield peptide resin (DTDM, TTTM) were prepared and the ion exchange capacity of these was characterized. The isotope separation of lithium was determined using breakthrough method of column chromatography. The isotope separation coefficient was strongly dependent on the ligand structure by Glueckauf's theory. We found that the isotope separation coefficients were increased as the values of distribution coefficients were increased. In this experiment the lighter isotope, 6Li was enriched in the resin phase, while the heavier isotope, 7Li in the solution phase. The ion radius of lighter isotope, 6Li was shorter than the heavier isotope, 7Li. The hydration number of lithium ion with the same charge became small as mass number was decreased. Because 6Li was more strongly retained in the resin than 7Li, the isotopes of lithium were separated with subsequent enrichment in the resin phase.

  • Cobalt(III) complex containing ethylenediamine-triacetate and trimethylenediamine has been isolated from the reaction of ethylenediamine-tetraacetatocobaltate(III) with trimethylene-diamine in aqueous solution by Dowex 50W-X8, cation exchange resin in $H^+$ form. The ethylenediamine-triacetate($EDTRA^{3-}$) ligand coordinates to the cobalt(III) ion as a quadridentate with a free acetate branch. It has been observed that the complex has trans(O-O) (1) structure via the elemental analysis, UV, IR and NMR data.

  • The concern for dissolved phosphate in water/wastewater has been increasing because of the risk for eutrophication. A variety of conventional and advanced technologies were applied to meet the enforced new regulation of phosphate around the world. However, there still remained a lot of challenge because most introduced/developed method, for example, biological and physic-chemical treatment is not easy to satisfy the new regulation of phosphate in water. In order to meet the new regulation, the application of ion exchanger has been tried which showed that the removal efficiency for phosphate was strongly determined by in the presence of the competing ion, especially sulfate. As results, a new class of ion exchanger governed by ligand exchange was developed and investigated to increase the selectivity for phosphate. The current study using organic/inorganic anion exchanger developed with Lewis acid-base interaction confirms the selectivity for phosphate over sulfate. According to isotherm test and column test, the value of the maximum phosphate uptake (Q) showed 64 mg/g as $po{_4}^{3-}$ and the breakthrough for phosphate occurs after 1000 min and completely finishes at 2500 min, respectively.

  • InP quantum dots capped by myristic acid (InP-MA QDs) were synthesized by a typical hot injection method using MA as stablizing agent. The current density across the InP-MA QDs thin film which was fabricated by spin-coating method is about $10^{-4}A/cm^2$ at the electric field of 0.1 MV/cm from I-V measurement on a metal-insulator-metal (MIM) device. The low conductivity of the InP-MA QDs thin film is interpreted as due to the long interdistances among the dots governed by the MA molecules. Therefore, replacing the MA with thioacetic acid (TAA) by biphasic ligand exchange was conducted in order to obtain TAA capped InP QDs (InP-TAA). InP-TAA QDs were designed due to: 1) the TAA is very short molecule; 2) the thiolate groups on the surface of the InP-TAA QDs are expected to undergo condensation reaction upon thermal annealing which connects the QDs within the QD thin film through a very short linker -S-; and 3) TAA provides better passivation to the QDs both in the solution and thin film states which minimizing the effect of surface trapping states.

  • The reactions of O,O-diphenyl Z-S-phenyl phosphorothiolates with X-pyridines have been studied kinetically in acetonitrile at $35.0{^{\circ}C}$. The Hammett plots for substituent (Z) variations in the leaving group (log $k_2$ vs. $\sigma$ Z) are biphasic concave downwards with breaks at Z = H. The large magnitudes of ${\rho}X(\rho_{nuc})$, ${\beta}X(\rho_{nuc})$, and the cross-interaction constant, $\rho$XZ, suggest frontside nucleophilic attack toward the leaving group. The sign reversal of $\rho$Z from positive in $\sigma$ Z $\leq$ 0 to negative in $\sigma$ Z $\geq$ 0 is interpreted as the change in mechanism from concerted to stepwise with rate-limiting expulsion of the leaving group. The anomalous negative sign of $\rho$ Z for leaving groups with electron-withdrawing substituents is interpreted as the intramolecular ligand exchange process of the leaving group from the equatorial position in the intermediate to the apical position in the TS.

Thioacetic-Acid Capped PbS Quantum Dot Solids Exhibiting Thermally Activated Charge Hopping Transport

  • Size-controlled lead sulfide (PbS) quantum dots were synthesized by the typical hot injection method using oleic acid (OA) as the stabilizing agent. Subsequently, the ligand exchange reaction between OA and thioacetic acid (TAA) was employed to obtain TAA-capped PbS quantum dots (PbS-TAA QDs). The condensation reaction of the TAA ligands on the surfaces of the QDs enhanced the conductivity of the PbS-TAA QDs thin films by about 2-4 orders of magnitude, as compared with that of the PbS-OA QDs thin films. The electron transport mechanism of the PbS-TAA QDs thin films was investigated by current-voltage (I-V) measurements at different temperatures in the range of 293 K-473 K. We found that the charge transport was due to sequential tunneling of charge carriers via the QDs, resulting in the thermally activated hopping process of Arrhenius behavior.

  • The one-dimensional coordination polymers, $[Cu^{II}(L)(NO_3)_2]_n$ (1) and {$[Cu^{II}(L)(NO_3)]{\cdot}2H_2O}_{2n} (2), were synthesized from $Cu(NO_3)_2{\cdot}3H_2O$ and 2-[(pyridin-3-ylmethyl)amino]ethanol (L, PMAE) in methanol by controlling the molar ratio of copper(II) salt. Copper(II) ion in 1 has one pyridine group of PMAE whose an aminoethanol group coordinates adjacent copper(II) ion. As the pyridine group is bonded to neighboring copper(II) ion, 1 becomes a one-dimensional chain. Contrary to 1, the structure of 2 shows that the oxygen atom of ethoxide group is bridged between two copper(II) ions, which forms a dinuclear complex. Additionally, the pyridine group of PMAE included one dinuclear unit is coordinated to the other dimeric one each other, which leads to a one-dimensional polymer. Due to the structural differences, 1 exhibits weak antiferromagnetic interaction, while 2 shows strong antiferromagnetic interaction. Due to direct spin exchange via oxygen of PMAE 2 has a much strong spin coupling than 1.

The Characterization of Borohydride-Stabilized Nanosilvers in Laponite Sol Using 1H NMR: Its Ligand Exchange Reactions with MUA and TOP

  • In borohydride-protected nanosilvers in laponite sol, the silver particles aggregate to form short chains and a dumbbell shape. The $^{1}H$ NMR measurements in this study represent, to our knowledge, the first observation of proton resonances of borohydride-protected nanosilvers in aqueous solution. Borohydride on nanosilver can be exchanged with 11-mercaptoundecanoic acid (MUA) or trioctylphosphine (TOP). Transmission electron microscopy and UV-Vis spectroscopy data show that the number of aggregated silver nanoparticles decreases upon addition of aforementioned ligands due to the formation of silver MPCs (monolayer-protected clusters). Adsorption of MUA or TOP on nanosilver is confirmed through the observation of broad proton resonances of MPCs in $^{1}H$ NMR spectra.
