Physiological responses of selected Philippine upland rice genotypes evaluated using drought and salinity stress

  • Screening for drought and salinity tolerance was undertaken for selected Philippine upland rice landraces during germinative and seedling stages to identify varieties which can potentially be grown in marginally dry and saline soils. While increasing PEG and NaCl concentrations caused obvious signs of injury to all rice genotypes, considerable varietal differences were noted in the nature of responses providing evidence that these genotypes possess broad intraspecific genetic variations for drought and salt tolerance. Inconsistent responses of these varieties during both growth stages highlight complexities involved in stress responses and underscore the futility of utilizing a single stage in the rice plant's life cycle for physiological screening. Notwithstanding these perplexing responses, G_Katiil and Ml-Pilit Tapul were observed to thrive relatively well despite increased salt and drought stress during early growth stages and may therefore possess genes needed in crop improvement efforts for drought and salinity tolerance. While these results do not reflect the entire spectrum of adaptive expression to drought and salinity stress during the life cycle of the upland rice plant, they nonetheless provide an easy, reliable and reproducible method for preliminary identification of drought and salt tolerant rice varieties.

A Study on the Decondensation and Pronucleus Formation of Sperm Nucleus in the Mouse Oocyte

  • To investigate the ability to decondense sperm head penetrated into cytoplasm of the oocytes and the relationship between this ability and the level of glutatione (GSH) in mouse oocyte at various maturing stages. The fertilizability of oocytes at various stages of maturation the decondensation of sperm nucleus and the formation of male pronucleus, were observed and the levels of GSH were measured in oocyte at same stages. Besides, the relation between fertilizability and level of GSH in oocyte cytoplasm treated with L-buthionine-S, R-sulfoxmine (L-BSO), the inbitor of biosynthesis of GSH, was determined. The decondensation of sperm head was not found in GV stage and L-BSO treated oocytes. In maturing oocytes (GVBD, MI), the decondensation was found, but the formation of male pronucleus was not. The levels of GSH in oocyte cytoplasm were measured; 2.2 pmol per oocyte in the ovulated and the matured in vitro each, 1.0 pmol in GV intact oocyte, 1.3 pmol in GVBD, and 1.5 pmol in MI phase oocyte. In L-BSO treated oocytes the levels of CSH were measured 0.08~o.09 pmol per oocyte, slightly lower than GV stage oocyte. In conclusion, GSH in oocyte is supposed to be synthesized and storaged in cytoplasm during maturation. The failure of decondensation in the cytoplasm of GV stage and L-BSO treated is suggested that GSH is an essential factor in decondensing the sperm head and that the a certain level of GSH, more than in GV oocyte cytoplasm, is required in decondensation.

The Control Mechanism of Gonadotropin-Releasing Hormone and Dopamine on Gonadotropin Release from Cultured Pituitary Cells of Rainbow Trout Oncorhynchus mykiss at Different Reproductive Stages

  • The mechanism by which gonadotropin-releasing hormone (GnRH) and dopamine (DA) control gonadotropin (GTH) release was studied in male and female rainbow trout using cultured pituitary cells obtained at different reproductive stages. The mechanisms of follicle-stimulating hormone (FSH) release by GnRH and DA could not be determined yet. However, basal and salmon-type GnRH (sGnRH)- or chicken-II-type GnRH (cGnRH-II)- induced luteinizing hormone (LH) release increased with gonadal maturation in both sexes. LH release activity was higher after sGnRH stimulation than cGnRH-II stimulation at maturing stages in both sexes. The GnRH antagonist ([Ac-3, 4-dehydro-$Pro^1$, D-p-F-$Phe^2$, D-$Trp^{3,6}$] GnRH) suppressed LH release by sGnRH stimulation in a dose-dependent manner, although the effect was weak in maturing fish. The role of DA as a GTH-release inhibitory factor differs during the reproductive cycle: the inhibition of sGnRH-stimulated LH release by DA was stronger in immature fish than in maturing, ovulating, or spermiated fish. DA did not completely inhibit sGnRH-stimulated LH release, and DA alone did not alter basal LH release. Relatively high doses ($10^{-6}$ or $10^{-5}M$) of domperidone (DOM, a DA D2 antagonist) increased LH release, which did not change with reproductive stage in either sex. The potency of DOM to enhance sGnRH-stimulated LH release was higher in maturing and ovulated fish than in immature fish. These data suggest that LH release from the pituitary gland is controlled by dual neuroendocrine mechanisms by GnRH and DA in rainbow trout, as has been reported in other teleosts. The mechanism of control of FSH release, however, remains unknown.

Liposuction in the Treatment of Lipedema: A Longitudinal Study

  • Background Lipedema is a condition consisting of painful bilateral increases in subcutaneous fat and interstitial fluid in the limbs with secondary lymphedema and fibrosis during later stages. Combined decongestive therapy (CDT) is the standard of care in most countries. Since the introduction of tumescent technique, liposuction has been used as a surgical treatment option. The aim of this study was to determine the outcome of liposuction used as treatment for lipedema. Methods Twenty-five patients who received 72 liposuction procedures for the treatment of lipedema completed a standardized questionnaire. Lipedema-associated complaints and the need for CDT were assessed for the preoperative period and during 2 separate postoperative follow-ups using a visual analog scale and a composite CDT score. The mean follow-up times for the first postoperative follow-up and the second postoperative follow-up were 16 months and 37 months, respectively. Results Patients showed significant reductions in spontaneous pain, sensitivity to pressure, feeling of tension, bruising, cosmetic impairment, and general impairment to quality of life from the preoperative period to the first postoperative follow-up, and these results remained consistent until the second postoperative follow-up. A comparison of the preoperative period to the last postoperative follow-up, after 4 patients without full preoperative CDT were excluded from the analysis, indicated that the need for CDT was reduced significantly. An analysis of the different stages of the disease also indicated that better and more sustainable results could be achieved if patients were treated in earlier stages. Conclusions Liposuction is effective in the treatment of lipedema and leads to an improvement in quality of life and a decrease in the need for conservative therapy.

Effects of Continuous Application of CO2 on Fruit Quality Attributes and Shelf Life during Cold Storage in Cherry Tomato

  • 'Unicon' cherry tomato (Solanum lycopersicum) is one of the most highly perishable horticultural crops due to its high water content and respiration rate. This study was carried out to assess the effect of continuous application of $CO_2$ (control [air], 3%, and 5%) on the quality and shelf life of cherry tomato fruits stored at $10^{\circ}C$ and $85{\pm}5%$ relative humidity (RH) at two maturity stages (pink and red). Continuous application of $CO_2$ did not affect the soluble solids content (SSC) or titratable acidity (TA) of the fruit at either maturity stage during storage. However, there was a significant difference among treatments in terms of flesh firmness, cell wall thickness, pectin content, vitamin C content, skin color, lycopene content, weight loss, ethylene production rate, respiration rate, and acetaldehyde and ethanol production. Fruits treated with 5% $CO_2$ maintained their high quality with regards to vitamin C, skin color ($a^*$), lycopene content, weight loss, physiological parameters (ethylene production rate, respiration rate, and volatile compounds), flesh firmness, cell wall thickness, and pectin content at both maturity stages compared with 3% $CO_2$ treatment and the control. Continuous application of $CO_2$ (5%) reduced the ethylene production rate and the production of volatile compounds during storage. Therefore, cherry tomato 'Unicon' fruit can be stored for two weeks without losing fruit quality at both maturity stages under continuous application of 5% $CO_2$ as a postharvest treatment.

NELL2 Function in Axon Development of Hippocampal Neurons

  • Neurons have multiple dendrites and single axon. This neuronal polarity is gradually established during early processes of neuronal differentiation: generation of multiple neurites (stages 1-2); differentiation (stage 3) and maturation (stages 4-5) of an axon and dendrites. In this study, we demonstrated that the neuron-specific n-glycosylated protein NELL2 is important for neuronal polarization and axon growth using cultured rat embryonic hippocampal neurons. Endogenous NELL2 expression was gradually increased in parallel with the progression of developmental stages of hippocampal neurons, and overexpression of NELL2 stimulated neuronal polarization and axon growth. In line with these results, knockdown of NELL2 expression resulted in deterioration of neuronal development, including inhibition of neuronal development progression, decreased axon growth and increased axon branching. Inhibitor against extracellular signal-regulated kinase (ERK) dramatically inhibited NELL2-induced progression of neuronal development and axon growth. These results suggest that NELL2 is an important regulator for the morphological development for neuronal polarization and axon growth.

Response of Grain Yield and Milled-Rice Protein Content to Nitrogen Rates Applied at Different Growth Stages of Rice

  • Response of grain yield and milled-rice protein content to nitrogen (N) rates at various growth stages is critical for quantifying real-time and real-amount of applied N requirement for target grain yield and protein content. An experiment including 10 N rate treatments at transplanting, tillering and panicle initiation stages with four rice cultivars in 2003, 6 N treatments with two rice cultivars in 2004 and 2005 was conducted. Increase of N rates at PIS significantly increased both grain yield and milled-rice protein content but increase of N rates at tillering stage significantly increased grain yield but not milledrice protein content. Therefore, high grain yield and low milled-rice protein content would be difficult to obtain only by adjusting N rates at PIS. Internal N use efficiency (INUE) was 60.5 kg grain/kg N accumulation on an average over N treatments, cultivars, and experimental years, showing considerable reduction especially at high shoot N accumulation in the experimental year of low sunshine duration. Milled-rice protein content tended to increase almost linearly with increasing shoot N accumulation, but it revealed big variation even at the same shoot N accumulation at harvest. Milled-rice protein content decreased with increasing INUE. N accumulation in the milled rice increased at an almost constant proportion of 45.5 percent of the shoot N accumulated at harvest, showing slight decresing proportion with the increasing shoot N accumulation.

Otolith Microstructural Organization in the South Georgia Icefish Pseudochaenichthys georgianus (Channichthyide) and Cautious Considerations on How Otoliths Can Provide Clues on a Species' Distribution and Migration in Antarctic Waters

  • How in the Antarctic icefish, generally, and Pseudochaenichthys georgianus, in particular, otoliths increase in size and build new material as the fish ages and passes through different life phases is largely unexplored. Morphometric details of 3418 otoliths of Ps. georgianus from S. Georgia and 318 from S. Shetland, were processed and proportions of the amounts of collagen and aragonite removed by EDTA were determined for different age groups. Microstructural investigations showed that characteristics of the 3-dimensinal collagen net are the reason for the radial direction and orientation of the aragonite needles of approximately 1.0 ㎛ in length in larval and 2.3 ㎛ in length in adult specimens. Earlier generated increment layers from the primordial centre (PC) in the dorsal direction restrict those of the secondary centre (SC), causing new growth layer accretion in different directions. In the otoliths of larval Ps. georgianus, aragonite layers are 0.89 ㎛ wide while in juveniles and adults they measure 1.45-2.86 ㎛. Otoliths change from a sphere shape in the larvae to a longish object of irregular outline in the older stages. It is tentaively suggested that the observed otolith shape differences at distinct growth stages are due to physical effects related to swimming speeds at particular water depths and locations. To confirm that otoliths, apart from being useful for age analyses, could also serve to establish correlations between developmental stage and the oceanic environment the fish spend time in, further analyses using additional species and state-of-the-art methods like µCT imaging to evaluate otolith volumes and shapes are required.

잎담배 재배농가의 생산비 분석 (Production Cost Analysis of Leaf tobacco farm Households)

  • This study had carried out an analysis of leaf tobacco production cost by cost items, growing stages, and farm sizes per 10a to provide the basic data for determination of the purchasing price of leaf tobacco by KT&G. Considering the survey results of 12 tobacco farm households, the composition rates of production cost by items revealed as 7-10% for land service, 5-22% for depreciation, 13-25% for material costs, 50-65% for labour cost respectively. The production cost of leaf tobacco by growing stages were shown as 15.3% in nursery bed period, 32.3% in main growing period in field, 30.8% in harvesting period and 21.6% in packing period. The magnitude of wage expenditure was appeared as harvesting stage, packing stage, growing stage on main field and nursery bed stage in order. The amount of material costs were revealed as the growing stage in main field, harvesting stage, nursery bed stage and packing stage respectively. The production costs of leaf tobacco per 10a by farm sizes were shown as 1,615,879won for small farm, 1,446,896won for medium farm and 1,454,408won for large farm respectively. The production cost of leaf tobacco had shown decreasing tendency according to increasing farm sizes. To promote the international market competitiveness of leaf tobacco producing farms, labour saving production technologies and cost effective farm size to decrease tobacco production cost should be developed.

제주땃쥐(Crocidura dsinezumi)의 세정관 상피주기 (Seminiferous Epithelium Cycle of Crocidura dsinezumi)

  • 제주땃쥐(Crocidura dsinezumi)의 세정관 상피 주기와 정자세포의 형태적인 특징을 알아보기 위하여 조사하였다. 제주땃쥐의 세정관 상피 주기는 12 stages로 구분되어졌다. Ad형 정원세포는 모든 단계에서 관찰되어졌으며, In형 정원세포는 IV단계에서, B형 정원세포는 V단계와 VI단계에서 관찰되었다. 정자세포의 첨체 발달과 핵의 형태적 변화는 14 steps로 구분되어졌다. 골지, 두모, 첨체, 성숙 및 이탈단계는 각각 $1{\sim}2,\;3{\sim}6,\;7{\sim}10,\;11{\sim}14$ 및 14 step으로 구분되어 나타났다. 이러한 우리의 결과들은 제주땃쥐의 정자 변태 과정에 대한 금후의 연구에 대한 기초를 제공해준다.
