  • A high performance liquid chromatographic method was developed for the determination of glycyrrhetinic acid contained in licorice powder. Glycyrrhetinic acid which is hydrolysate of glycyrrhizin extracted from licorice powder, was determined with good result by HPLC using 2-bromoacetyltriphenylene labeling reagent. The glycyrrhetinic acids were labeled with 2-bromoacetyltriphenylene in acetonitrile using 18-crown-6-ether and KOH as a catalyst. Derivatized glycyrrhetinic acids were separated from the extracted licorice powder on a reversed-phase column (chemopak $C_{18}$) using 100% acetonitrile as a mobile phase and monitored by an UV-detector at 268nm. Linearity of calibration curve was obtained between 5 ng and 20 ng, and the lower limit of detection was 2 ng. The recovery of glycyrrhetinic acid to licorice powder was about 99.3%. This method was sensitive, reliable and useful for, determination of glycyrrhetinic acid.

감초 추출물의 산지별 화학성분 비교 및 전자코 장치를 이용한 산지 판별 분석 (The Comparison of Chemical Components of Licorice Extracts and Discrimination Analysis of Licorice Cultivation Areas by Electronic Nose)

  • This study was carried out to compare the quality of licorices from various cultivating areas. licorice samples used in this study were collected from North-east and Sinkiang area in China, Mongolia, Uzvekistan and Kazahstan. The chemical components of licorice samples were analyzed and the signal patterns of the electracts were detected by the electronic nose. Contents of glycyrrhizin and glicyrrhizic acid, the key components of licorice were distributed in the region of 16.7~25.2% and 5.8~10.2%, respectively and were various according to the samples of the collected areas. In glycyrrhizin contents, root of Sinkiang showed the lowest value of 16.7%, and that of North-east the highest of 25.2%. In glycyrrhizic acid contents, root of Sinkiang showed the lowest of 5.8 %, and Kazahstan showed the highest of 10.2 %. Composition ratio of glycyrrhizin to glycyrrhizic acid was not always limear. As other components is other components affecting quality, contents of ash, starch and gums were 2.4~3.7%, 0.2~3.9%, respectively. When the headspace volatiles of licorices were analyzed using Electronic Nose System and the obtained data were interpreted using statistical method of MANOVA, characteristic patterns of licorices were different from each other according to collected area and its p value showed 0.0001. These results suggest that licorices may be discriminated from the collected areas by using Electronic Nose System.

다양한 젤화제와 감초추출액을 이용한 감초젤리의 특성 (Characteristics of Licorice Jellies using a Water Extract of Licorice Root and Various Gelling Agents)

  • This study was conducted to investigate the usefulness of a hot water extract of licorice root as a source for production of healthy food. The electron donating capacity of the hot water extract of licorice root was very strong. This activity decreased by $6.9\%$ after keeping it in 5 days of cold storage, but it was not significantly different. Ten types of licorice jellies were prepared, using agar, agar-pectin, agar-cellulose, 2 different proportions of agar-pectin-cellulose as a gelling agent, and 2 levels of sugar. Among the 5 jellies containing $10\%$ sugar, the elasticity and overall acceptance of the agar jelly obtained the highest sensory score, but the color and flavor were not affected by the type of gelling agents. As the sugar concentration increased to $15\%$, the elasticity of the agar jelly was also evaluated as being the best one among the 5 jellies, but the overall acceptance was not significantly different from the others.

생약중의 감초에 관한 연구(I) HPLC에 의한 Glycyrrhizin 및 Glycyrrhetinic acid의 정량 (Studies on Licorice in Drug Preparations(I) Determination of Glycyrrhizin and Glycyrrhetinic acid by HPLC.)

  • Glycyrrhizin (GA) content in licorice was determined by a couple of methods using HPLC, respectively. In Method(I), GA content itself was determined from the licorice aqueous extract, while in Method (II) glycyrrhetinic acid (GHeA ; the aglicone of GA) content corresponding to the quantity of GA was measured from the chloroform extract of the hydrolyzed product of licorice aqueous extract. A reverse phase column Hibar Lichrosorb RP-18 (E. Merck) was used as the stationary phase. As the mobile phase MeOH: $H_{2}O$(0.05M-$NaH_{2}PO_{4}$)=58 : 42 solution in Method (I), and MeOH: $H_{2}O $: AcOH=78; 19: 3 solution in Method (II) were suitable, respectively. The value obtained by Method (II) appeared slightly higher than that by Method (I). The effect of some other herbal drugs on the assay of GA quantity in mixed sample was also observed in both above two methods. By Method (I) Cassiae Cortex, Rehmaniae Rhizoma, Paeoniae Radix, and Angelicae Radix gave the subtractive effect on the amount of GA compared with the value from licorice alone. In the case of Method (II) Cassiae Cortex and Rehmaniae Rhizoma appeared to have subtractive effect but Paeoniae Radix and Angelicae Radix scarcely showed any influence. Pachymae Fungus did not affect the GA content at all. It seems that glycyrrhizin in licorice interacts with certain components of other herbal drugs.

감초추출물을 이용한 다진 마늘의 갈변 저해 효과 (Antibrowning Effect of Licorice (Glycyrrhiza glabra) Extracts on Chopped Galic)

  • 다진 마늘의 제조 중 발생하는 효소적 갈변을 막기 위해 산업적으로 적용되는 방법은 주로 화학적인 갈변저해제를 이용하는 경우가 다수이나, 최근 소비자의 천연물에 대한 수요 증대를 감안하여 천연물 유래의 갈변저해제를 탐색할 필요가 있다. 따라서 본 연구에서는 천연물 유래의 갈변저해제 중 효과적으로 나타난 감초 추출물의 갈변저해 효과를 조사하였다. 또 기존 다진 마늘의 갈변저해에 효과적인 것으로 알려져 있는 구연산과의 병용 효과를 동시에 조사하여 효율적인 응용이 가능하도록 하였다. 다진 마늘 제조 시 구연산과 감초추출물을 다양한 조성비로 적용하고 냉장저장 중 다진 마늘의 색도를 측정하여 L값의 변화를 중심으로 갈변 저해 효과를 조사하였다. 그 결과 저장 초기에는 감초추출물 단독으로도 갈변저해 효과가 있었으나 저장 후기로 갈수록 감초추출물과 구연산을 혼합한 조성물이 매우 효과적으로 갈변을 저해함을 알 수 있었다. 천연물인 감초추출물은 구연산과 상승작용을 일으켜 갈변을 효과적으로 저해하는 것으로 추측된다. 또한 감초추출물과 구연산의 혼합비를 조정하여 실험한 결과에 따르면, 감초추출물:구연산의 혼합비가 2:8 내지 5:5일 때 효과적으로 갈변을 저해하며, 특히 감초추출물:구연산의 혼합비가 2:8인 경우 가장 뛰어난 갈변 저해 효과를 나타냄을 알 수 있었다. 이 경우 구연산을 단독으로 처리한 경우와 동등한 수준으로 뛰어난 갈변 저해 효과를 나타낼 뿐만 아니라, 구연산 단독 처리시의 문제점인 신맛 등 관능적 품질 저하 문제를 해결할 수 있을 것으로 판단된다.

Comparison of the effect of three licorice varieties on cognitive improvement via an amelioration of neuroinflammation in lipopolysaccharide-induced mice

  • BACKGROUD/OBJECTIVES: Neuroinflammation plays critical role in neurodegenerative disorders, such as Alzheimer's disease (AD). We investigated the effect of three licorice varieties, Glycyrhiza uralensis, G. glabra, and Shinwongam (SW) on a mouse model of inflammation-induced memory and cognitive deficit. MATERIALS/METHODS: C57BL/6 mice were injected with lipopolysaccharide (LPS; 2.5 mg/kg, intraperitoneally) and orally administrated G. uralensis, G. glabra, and SW extract (150 mg/kg/day). SW, a new species of licorice in Korea, was combined with G. uralensis and G. glabra. Behavioral tests, including the T-maze, novel object recognition and Morris water maze, were carried out to assess learning and memory. In addition, the expressions of inflammation-related proteins in brain tissue were measured by western blotting. RESULTS: There was a significant decrease in spatial and objective recognition memory in LPS-induced cognitive impairment group, as measured by the T-maze and novel object recognition test; however, the administration of licorice ameliorated these deficits. In addition, licorice-treated groups exhibited improved learning and memory ability in the Morris water maze. Furthermore, LPS-injected mice had up-regulated pro-inflammatory proteins, such as inducible nitric oxide synthase (iNOS), cyclooxygenase-2, interleukin-6, via activation of toll like receptor 4 (TLR4) and nuclear factor-kappa B ($NF{\kappa}B$) pathways in the brain. However, these were attenuated by following administration of the three licorice varieties. Interestingly, the SW-administered group showed greater inhibition of iNOS and TLR4 when compared with the other licorice varieties. Furthermore, there was a significant increase in the expression of brain-derived neurotrophic factor (BDNF) in the brain of LPS-induced cognitively impaired mice that were administered licorice, with the greatest effect following SW treatment. CONCLUSIONS: The three licorice varieties ameliorated the inflammation-induced cognitive dysfunction by down-regulating inflammatory proteins and up-regulating BDNF. These results suggest that licorice, in particular SW, could be potential therapeutic agents against cognitive impairment.

Effects of Modifiers on the Supercritical $CO_{2}$ Extraction of Glycyrrhizin from Licorice and the Morphology of Licorice Tissue after Extraction

  • Optimal conditions for the supercritical carbon dioxide $(scCO_{2})$ extraction of glycyr­rhizin from licorice (Glycyrrhiza glabra) were investigated, with an emphasis on the types and levels of modifiers. The morphology of the licorice tissue remaining after the $scCO_{2} $ extraction of glycyrrhizin was examined by scanning electron microscopy, coupled with measurements of ab­solute density. Conventional organic solvent extraction was also carried out for purpose of quantitative comparison. At 50 MPa and $60^{circ}C$ glycyrrhizin could not be extracted with pure $scCO_{2}$, while a considerable amount of glycyrrhizin was extracted when water was added to $scCO_{2}$ as a modifier. The highest recovery was found to be about $97\%$ when $70\%$ aqueous methanol was added to $scCO_{2}$ at a concentration of $15\%$. The optimal pressure and temperature for the supercritical fluid extraction of glycyrrhizin were observed to be 30 M Pa and $60^{circ}C$, respectively. Under these conditions, the percentage recovery of glycyrrhizin attained a maximum value of 102.67\pm$ $1.13\%$ within 60 min. Furthermore, in the case of $scCO_{2}$ modified with $70\%$ aqueous methanol, the licorice tissue obtained after extraction was found to be severely de­graded by excessive swelling, and the absolute density of the licorice residues was observed to be the highest.

감초가 포함된 한약 복용 시 혈중 포타슘 수치의 저하에 영향을 미치는 요인 (Effect of Herbal Medicines Including Licorice on Serum Potassium Levels)

  • Licorice is a very useful herbal medicine frequently prescribed, but glycyrrhizin, one of its components, can cause pseudoaldosteronism presenting hypokalemia, metabolic alkalosis and hypertension as a result of prolonging a biological half-life of systemic steroid by strongly inhibiting the enzyme $11{\beta}$-hydroxysteroid dehydrogenase type 2. The risk factors for this side effect are still unclear. Here, 479 hospitalized patients taking herbal medicines including licorice for more than five days were recruited to analyze their serum potassium levels, under following parameters ; age and gender of patient, dose and period of licorice, disease state such as hypertension and with or without diuretics. They were divided into the non-occurrence group (over 3.5 mEq/L) and the occurrence group (under 3.5 mEq/L) of hypokalemia based on serum potassium levels during hospitalization. The average ages of the non-occurrence group and the occurrence group were $52.6{\pm}17.8$ and $68{\pm}10.5$, respectively (p < 0.001). But there were no noticeable differences in the daily dose of licorice, days of taking, and total dose between groups. Regression analysis showed that odd ratio of age (>60) and taking diuretics inducing hypokalemia was 3.5 (95%CI 1.5-8.1), 4.6 (95%CI 2.0-10.9) that indicates significant correlation with hypokalemia. Based upon this analysis, it is imperative to monitor regularly the risk of pseudoaldosteronism or hypokalemia not only when administered licorice of high dose for long period but also when combined it with diuretics causing hypokalemia to the elderly patients over 60 year old.

Inhibitory Effects of Licochalcone A and Isoliquiritigenin on Monocyte Adhesion to TNF-$\alpha$-activated Endothelium

  • Numerous natural herbal compounds have been reported to inhibit adhesion and migration of leukocytes to the site of inflammation Licorice extracts, which have been widely used in traditional Chinese medicinal preparation, possess various pharmacological effects. Isoliquiritigenin, a biogenetic precursor of flavonoids with various pharmacological effects, is a natural pigment present in licorice. We attempted to explore whether licorice extracts and isoliquiritigenin mitigate monocyte adhesion to tumor necrosis factor-$\alpha$ (TNF-$\alpha$)-activated human umbilical vein endothelial cells (HUVEC). In addition, it was tested whether the inhibition of monocyte adhesion to the activated HUVEC accompanied a reduction in vascular cell adhesion molecule-l expression(VCAM-l). Dry-roasted licorice extracts in methylene chloride but not in ethanol markedly interfered with THP-l monocyte adhesion to INF-$\alpha$-activated endothelial cells. licochalcone A compound isolated from licorice extract in methylene chloride appeared to modestly inhibit the interaction of THP-l monocytes and activated endothelium. In addition, isoliquiritigenin abolished the monocyte adhesion with attenuating VCAM-l protein expression on HUVEC induced by INF-$\alpha$. These results demonstrated that non-polar components from dry-roasted licorice extracts containing licochalcone A as well as isoliquiritigenin were active in blocking monocyte adhesion to cytokine-activated endothelimn, which appeared to be mediated most likely through the inhibition of VCAM-l expression on HUVEC. Therefore, licorice may hamper initial inflammatory events on the vascular endothelium involving induction of endothelial cell adhesion molecules.

감초(甘草) Alkaloidal Fraction 의 평활근(平滑筋)에 대(對)한 Acetylcholine 길항작용(拮抗作用) -감초 알카로이드에 관한 연구 (제 3 보)- (Antimuscarine-like Action of Licorice Alkaloidal Fraction on Intestinal Smooth Muscle -Studies of Alkaloid of Glycyrrhiza glabra L. III-)

  • Antimuscarinic agent like antispasmodic actions of licorice alkaloidal fraction, obtained from the Glycyrrhiza glabra L., was compared with that of atropine quantitatively. For this purpose, the author calculated the kinetic constants and $ED_{50}$ for acetylcholine antagonism by these drugs on rat ileum and guinea-fig ileum longitudinal muscle according to Paton's theoretical equations describing the combination of an antagonist drug with its receptors. The results are as follows. 1. On rat ileum. a) Licorice alkaloidal fraction $K_1$ (association rate constant)=$4.078{\times}10^2\;(s^{-1}\;gm^{-1}\;ml)$ $K_2$ (dissociation rate constant)=$6.986{\times}10^{-4}\;(s^{-1})$ $ED_{50}(K_2/K_1)=1.772{\times}10^{-6}(gm/ml)$ b) Atropine $K_1=5.136{\times}10^6$, $K_2=7.714{\times}10^{-4}$, $ED_{50}=1.408{\times}10^{-10}$ 2. On guinea-pig ileum longitudinal muscle a) Licorice alkaloidal fraction $K_1=1.30{\times}10^2$, $K_2=1.25{\times}10^{-3}$ $ED_{50}=9.58{\times}10^{-6}$ b) Atropine $K_1=5.75{\times}10^6$, $K_2=1.54{\times}10^{-3}$ $ED_{50}=2.68{\times}10^{-10}$ Above results present that 1 r of licorice alkaloidal fraction has equal fotency of acetylcholine antagonism with $8.5{\times}10^{-5}r$ of atropine on rat ileum, $2.8{\times}10^{-5}r$ on guinea-pig ileum longitudinal muscle. This facts suggest that the site and numbers of licorice alkaloid receptors of guinea-pig ileum are different from that of rat ileum. Besides, it also gives a suggestion that licorice alkaloidal fraction may be a partial antagonist on guinea-pig ileum in this experimental conditions.
