The Anti-Inflammatory Effect of Gabyeobda Tea in High Fat Diet-Induced Obese Mice (가볍다차(茶)가 고지방식이로 유도된 비만 마우스에서 항염증에 미치는 효과)

  • Objectives: The purpose of this study was to investigate the effects of Gabyeobda tea (GT) on anti-inflammation in ice induced high fat diet (HFD). Methods: The C57BL/6 mice fed HFD were administrated with GT once daily for 8 weeks. The changes of body weight, calorie intake levels were measured in mice. The level of serum total cholesterol, triglyceride, high density lipoprotein cholesterol, glutamic oxaloacetic transaminase (GOT), and glutamic pyruvic transaminase (GPT) were measured in mice by enzyme-based assay. It was also observed the histological changes of liver, and fat tissues with hematoxylin and eosin staining. Further real-time polymerase chain reaction (PCR) and enzyme-linked immunosorbent assay were employed to detect inflammatory cytokine levels such as tumor necrosis factor (TNF)-𝛼, interleukin (IL)-6, and IL-1𝛽. Results: HFD+GT group, which was administered with GT with HFD, showed no body weight gain compared with HFD group. However, levels of GOT, GPT, and inflammatory cytokines such as TNF-𝛼, IL-6, and IL-1𝛽 in the blood of HFD+GT group were significantly reduced compared with HFD group. In addition, the messenger RNA (mRNA) expression level of the IL-12 gene was significantly reduced and the mRNA expression level of the IL-10 was increased in the liver. Conclusions: It suggests that Gabyeobda tea can alleviate inflammatory responses induced by high fat diet by inhibiting inflammatory cytokines production.

Biochemical Components and Physiological Activities of Ice Plant (Mesembryanthemum crystallinum) (아이스플랜트(Mesembryanthemum crystallinum)의 생화학적 성분 및 생리활성 효과)

  • The general components and cyclitol compounds of ice plant (Mesembryanthemum crystallinum) were analyzed to examine the possibility of using extracts as functional food materials, the antioxidant effects and antidiabetic activities of the extracts by solvent fraction were tested. Among the mineral contents, contents of K and Na were the largest ($1,213.33{\pm}2.52$ and $545.53{\pm}12.01mg/100g$, respectively), followed by S, Ca, P, and Mg in order of precedence. Among cyclitol compounds, content of D-pinitol was the largest ($4.04{\pm}0.08mg/g$) while contents of chiro-inositol and myo-inositol were relatively small ($2.82{\pm}0.01$ and $0.25{\pm}0.01mg/g$, respectively). Among total phenol contents by solvent fraction, contents of chloroform and ethyl acetate fractions were large ($35.80{\pm}1.33$ and $23.70{\pm}0.62mg\;GAE/g$, respectively). Among antioxidant activity levels examined by DPPH, ABTS, FRAP, and lipid/MA assays, the chloroform fraction commonly showed the highest level of activity while the ethyl acetate fraction showed relatively high levels of activity. The antioxidant activity levels were proportional to total phenol contents by solvent fraction. As for antidiabetic effects, all solvent fractions showed at least 50% ${\alpha}-glucosidase$ inhibitory activity levels while the ethyl acetate, butanol, and chloroform fractions showed high levels activity of $90.33{\pm}0.40$, $87.98{\pm}0.16$, and $86.38{\pm}0.51%$, respectively. The ${\alpha}-amylase$ inhibitory actively levels were in the range of $25.63{\pm}1.45{\sim}60.34{\pm}2.67%$, which was lower than the ${\alpha}-glucosidase$ inhibitory activity levels, but the inhibitory activity levels by solvent fraction were similar. Given the above study results, ice plant can be utilized as a natural material with antioxidative and antidiabetic functionality.

Microbiological Hazard Analysis in Children Snacks around Schools (학교 주변 어린이기호식품의 미생물학적 오염도 평가)

  • This study was conducted to develop an appropriated management for safety of children snacks sold around school. Total 598 items as targeted food were collected; 66 biscuits, 320 candies, 57 chocolates, 40 ice creams and 115 beverages. Microbiological hazards such as total aerobic bacteria, Coliforms, Escherichia coli, Bacillus ceruse, Yeasts & molds were measured by analytical method in Korean food code. Total aerobic bacteria and Yeasts & molds were detected in cookies at the level of less than 2.69 and 2.65 $log_{10}$ CFU/g and the detection rates were 54.55 and 62.12%, respectively. Bacillus cereus was detected in 1 snack only at the level of 1.39 $log_{10}$ CFU/g but it was less than Korean microbial standards and specifications (3 $log_{10}$ CFU/g). Total aerobic bacteria and Yeasts & molds were detected in candies less than 2.86, 3.36 $log_{10}$ CFU/g and the detection rates were 46,8% respectively. Total aerobic bacteria, Yeast & mold were detected in chocolates at the levels less than 2.52 and 1.87 $log_{10}$ CFU/g and the detection rates were 33 and 22% respectively. Total aerobic bacteria in both ice creams and beverages were detected at the levels less than 3.39 and 1.35 $log_{10}$ CFU/g and the detection rates were 82 and 5% respectively. Coliforms were found in one ice cream (1.39 $log_{10}$ CFU/g) only. The result of this study indicated that all children snacks around school were suitable for microbial standard and specifications in Korean Food Code. However, since most children snacks around school are circulated without proper storage temperature and handing condition, consistent microbial management for children snacks are needed.

  • Two experiments were conducted to study the nature of protein degradation in fish muscle postmortem, first one with English sole (Paraphyrus vetulus) followed by another with rockfish (Sebastodes spp.). In the first one, proteolysis was measured by the increase of amino-N in gutted fish during storage in ice and in the homogenates prepared from fish of different ice storage during $20^{\circ}C-incubation$. In order to test the possible involvement of fish muscle a cathepsin, a portion of each homogenate sample was exposed to 0.5 Mrad of gamma radiation to destroy viable microorganisms prior to the incubation. Proteolysis was not detected until viable count reached a level above $10^7$ cells per gm fish flesh, corresponding to 31 days of ice storage. Even if fish flesh were mechanically disrupted by means of homogenization and subsequently incubated at $20^{\circ}C$, proteloysis attributable to muscle cathepsin was not detected. In the second with rockfish muscle aseptically prepared from freshly killed fish, the samples were inoculated with a proteolytic strain of fish spoilage Pseudomonad or irradiated at 0, 0.5 and 3.0 Mrad. The four samle groups were stored at $0-2^{\circ}C$ to compare the spoilage pattern of sterile and non-sterile muscle. In sterile muscle both total-N (extracted in 0.5M KCl) and amino-N $(soluble\;in\;70\%\;ethanol)$ declined slightly while the inoculated muscle showing increase in parallel with the increase of number of inoculated bacterium. The results indicate that proteolysis is a part of normal fish spoilage and the onset of proteolysis is delayed until viable count reaches its maximum level. Contribution of fish muscle cathepsin to protein degradation in white flesh fish muscle post-mortem is nil.

Occurrence and Estimation Using Monte-Carlo Simulation of Aflatoxin $M_1$in Domestic Cow’s Milk and Milk Products (국내산 우유 및 유제품에서의 Aflatoxin $M_1$오염수준 및 Monte-Carlo Simulation을 이용한 발생 추정)

  • In this study, occurrence of aflatoxin M$_1$(AEM$_1$) in domestic milk and milk products was determined. The level of AFM$_1$ in market milk (0.047 ppb) was lower than that in raw milk (0.083 pub) but this looks like that is due to dilution in collecting process rather than the effect of sterilization. In the case of nonfat dry milk, level of AFM$_1$appeared high by 0.24 ppb but it is thought to be not different from market milk actually because nonfat dry milk is diluted at intake. In the case of ice cream, finished products were contaminated with AFM$_1$of 0.020 ppd and also have the possibility of the contamination of AFB$_1$due to secondary raw material such as nuts and almond. On the basis of the results of this study and previous studies, Monte-Carlo simulation is conducted to estimate the contamination level of AFM$_1$in domestic market milk. To consider uncertainty and variability fitting procedure was passed through. And we used beta distribution to estimate the prevalence and triangular distribution to estimate the concentration level of AFM$_1$in milk. As a result, the 5%, 50% and 95% points of the distribution of the probability of AFM$_1$contamination level in milk is 0.0214, 0.0946 and 0.1888 ppb, respectively. Also we estimate that AFM$_1$in almost milk was low more than 0.5 ppb that is American acceptable level but 80.4% exceeded far 0.05 ppb that is European standard.

Prediction of Galloping Accidents in Power Transmission Line Using Logistic Regression Analysis

  • Galloping is one of the most serious vibration problems in transmission lines. Power lines can be extensively damaged owing to aerodynamic instabilities caused by ice accretion. In this study, the accident probability induced by galloping phenomenon was analyzed using logistic regression analysis. As former studies have generally concluded, main factors considered were local weather factors and physical factors of power delivery systems. Since the number of transmission towers outnumbers the number of weather observatories, interpolation of weather factors, Kriging to be more specific, has been conducted in prior to forming galloping accident estimation model. Physical factors have been provided by Korea Electric Power Corporation, however because of the large number of explanatory variables, variable selection has been conducted, leaving total 11 variables. Before forming estimation model, with 84 provided galloping cases, 840 non-galloped cases were chosen out of 13 billion cases. Prediction model for accidents by galloping has been formed with logistic regression model and validated with 4-fold validation method, corresponding AUC value of ROC curve has been used to assess the discrimination level of estimation models. As the result, logistic regression analysis effectively discriminated the power lines that experienced galloping accidents from those that did not.

Alternative splicing and expression analysis of High expression of osmotically responsive genes1 (HOS1) in Arabidopsis

  • High expression of osmotically responsive genes1 (HOS1), a key regulator of low temperature response and flowering time, encodes an E3 ubiquitin ligase in Arabidopsis. Here, we report characterization of a newly identified splice variant (HOS1-L) of HOS1. Comparative analyses revealed that HOS1-L has a longer 5' nucleotide sequence than that of the previously identified HOS1 (HOS1-S) and that its protein sequence was more conserved than that of HOS1-S in plants. HOS1-L transcripts were spatio-temporally more abundant than those of HOS1-S. The recovery rate of HOS1-S expression was faster than that of HOS1-L after cold treatment. Diurnal oscillation patterns of HOS1-L revealed that HOS1-L expression was affected by photoperiod. An in vitro pull-down assay revealed that the HOS1-L protein interacted with the ICE1 protein. HOS1-L overexpression caused delayed flowering in wild-type plants. Collectively, these results suggest regulation of HOS1 expression at the post-transcriptional level.

Rapid Climate Change During the Deglaciation of Lake Hovsgol, Mongolia

  • A 120-cm core recovered from Lake Hovsgol, the northern Mongolia provides evidence for climate variability since the Marine Isotope Stage 3, representing a sharp lithological change. The lowermost part of the core consists of diatom-barren calcareous silty clay without coarse sands, framboidal pyrite, and biogenic components deposited during the MIS 3. Following the last glacial maximum, in-situ moss is included in the sediments, as lake-level was retreated by cold and dry environment with low precipitation. The AMS radiocarbon ages of the plant fragments match a marked lithologic boundary between 14,060 and 14,325 $^{14}C$ yr BP. The contents of coarse sands abruptly increase, indicating probably wind-derived sandy dust or coarse grains contributed from floating icebergs. And abundant framboidal pyrite grains were deposited in an anoxic environment, as reflected by high accumulation of organic matters at a low lake stand. During the deglaciation, quantities of coarse sands, ostracod, shell fragments, framboidal pyrite, and diatom markedly varies by regional and global scale climate regimes. Some allochthonous coarse sands were probably ice-rafted debris derived from floating icebergs. A rapid increase in diatom productivity probably marked the onset of Bolling-Allerod warming. Subsequent high concentration of framboidal pyrite probably represents a dry and cold condition, such as Younger Drays events. Consistent warm period with high precipitation at Holocene is documented by diatomaceous clayey ooze without framboidal pyrite, coarse sands, and ostracod.

A Study on the Automatic Irrigation Control System in the Vinyl-House Cultivation Utilizing Microcomputer (마이크로컴퓨터를 이용한 시설원예작물 재배의 관개자동화에 관한 연구)

  • The purpose of this study was to develop a computer operated automatic drip irrigation system for application in vinyl-house cultivation. The results can be summarized as follows: 1) The T-type ice compensation wire was used to measure the temperature. The voltage level measured up to 0.02 volt was used as input to an 8-bit A/D converter. 2) A specially devised tensiometer was used to content the watering system. When the needle of the pressure gauge reaches the lower threshold position it turns on the pumping system and turns off when it reaches higher threshold position. 3) In order to use the multiple gypsum blocks for one transducer, reed relays and a D/O board were used to make the sequential switching possible. 4) It was possible to automate the trickle irrigation system for the whole growth period of vinyl-house crops with the help of microcomputer. 5) In terms of furrow irrigation, the irrigation water consumption was the smallest, 2.8 times less than conventional method of surface trickle irrigation, 3.4 times less than subsurface trickle irrigation method. 6) In terms of productivity of cucumber, there was a drop in productivity when compared to furrow irrigation method, 7.2% for surface trickle irrigation, 27.4% for subsurface irrigation method.
