The Production of Glucose-1-phosphate from Sucrose by Leuconostoc sp. (Leuconostoc sp.에 의한 Sucrose로부터 Glucoes-1-phosphate의 생산)

  • For the production of glucose-1-phosphate from sucrose, bacteria having sucrose phosphorylase were isolated from Kimchi. Among them, JS-05, newly isolated strain having high activity of sucrose phosphorylase was selected and identified as Leuconostoc sp. The specific activity of sucrose phosphorylase of Leuconostoc sp. JS-05 was the highest when the strain was cultured at 25C for 20 hrs in the medium (pH 7.5) containing 10 g sucrose, 5g corn steep liquor, and 2.5g yeast extract per liter.

  • The liquid of ripened Kimchi was spread on phenylethylalcohol sucrose medium and incubated at 20$^{\circ}C$ for 2 days in order to isolate Leuconostoc sp. strains. Twenty isolated colonies were identified as Leuconostoc sp. strains from sugar fermentation test. Dextransucrase activities of the isolated strains were determined and the strain J-2 showed highest activity. The morphological, cultural and physiological studies on these 5 strains showed that gram(+), spores(-), motility(-) and produced gas from glucose, acid in Whittenbury C. Only Y-1 strain produced ammonia from arginine.

Plasmid-associated Bacteriocin Production by Leuconostoc sp. LAB145-3A Isolated from Kimchi

  • Leuconostoc sp. LAB145-3A isolated from kimchi produced a bacteriocin which was active against food pathogens, such as Listeria monocytogenes, Enterococcus faecalis, and E. faecium. Bacteriocin production occurred during the early exponential phase of growth and was stable upto the late stationary phase of growth. Optimum conditions for bacteriocin production were $37^{\circ}C$ with an initial pH of 7.0. The bacteriocin of LAB145-3A was sensitive to proteases, but stable for solvents, pH change and heat treatment. It was stable even at autoclaving temperature for 15 min. The bacteriocin exhibited a bactericidal mode of action against Lactobacillus curvatus LAB170-12. The bacteriocin produced by Leuconostoc sp. LAB145-3A was purified by CM-cellulose cation exchange column chromatography and Sephadex G-50 gel filtration. The purification resulted in an approximate 10,000-fold increase in the specific activity. Approximately 4% of the initial activity was recovered. Purified bacteriocin exhibited a single band on the SDS-PAGE with an apparent molecular weight of 4,400 daltons. This bacteriocin was named leucocin K. Leuconostoc sp. LAB145-3A had two residential plasmids with molecular sizes of 23 kb and 48 kb. A comparison of plasmid profiles between LAB145-3A and its mutants revealed that the 23 kb plasmid (pCA23) was responsible for bacteriocin production and immunity to the bacteriocin in Leuconostoc sp. LAB145-3A.

Changes in Lactic Acid Bacteria of Squid with Low Salt during Fermentation (저염 오징어젓갈 숙성중 젖산균의 변화)

  • Isolation and identification of predominant microorganisms in fermented squid with low salt were carried out during fermentation at $10^{\circ}C$. Dominant strains were lactic acid bacteria(80%) including Lactobacillus plantarum, Lactobacillus brevis, Leuconostoc sp., Pediococcus sp. and Streptococcus sp. Leuconostoc, Pediococcus and Streptococcus were shown in the early stage of fermentation and gradually increased until optimum stage of squid fermentation, and then decreased rapidly. Lactobacillus grew lastingly during fermentation. Yeasts were detected in the middle stage of fermentation and shown rapid increase after the last stage of fermentation, suggesting that yeasts participate in putrefaction of fermented squid with low salt.

  • A new approach to the solubilization of excess activated sludge by the inoculation of lactic acid bacteria was studied to reduce the amount of sludge produced in the activated sludge treatment process. Aerobic microorganism in sludge was lysed in anaerobic condition and the cytoplasmic substance eluted was utilized as a carbon source by lactic acid bacteria. On the basis of sludge solubilization efficiency, Lactobacillus brevis and Leuconostoc mesenteroides subsp mesenteroides were selected the best candidates among five kinds of Lactobacillus sp. and seven kinds of Leuconostoc sp. The sludge solubilization efficiency by heterofermentative lactic acid bacteria was more efficient than that of homofermentative bacteria. Initial value of soluble COD (sCOD) was 1050 mg/L at the initial inoculation time increased to 3070 mg/L (192% solubilization) at 96 h of the incubation time. The inoculation of lactobacillus brevis to the sludge resulted in 2824% increase in sCOD value after 96 h of incubation than the control experiment. Leuconostoc mesenteroides subsp mesenteroides showed 152% increase of solubilization and 30% increase of S-COD/T-COD on 96 h of incubation time. Considering the increase of S-COD by the inoculation of Leuconostoc sp. on 24 h, 10% inoculation of lactic acid bacteria to the sludge was most effective.

  • This study was carried out to characterize the antilisterial substances produced by Leuconostoc sp. W65 and to evaluate the effects of pH, temperature, and time on inhibitory activity using response surface methodology. Leucocin W65, an antilisterial substance produced by Leuconostoc sp. W65, markedly inhibited the growth of Listeria monocytogenes, L. innocua, and L. ivanovii, whereas other pathogens including Gram-negative bacteria were not susceptible. The pH was the most effective factor with regard to bacteriocin activity, while temperature and time of heat treatment had no significant effect. Fifty percent of inhibitory activity remained after 22.8 min at pH 4.2 and $121^{\circ}C$. Leucocin W65 was purified by ammonium sulfate precipitation, hydrophobic interaction chromatography, and tricine-SDS-PAGE. Compositional analysis originally estimated the peptide to be 56 amino acids in length without asparagine, glutamine, and tryptophane. The sequence of partial N-terminal amino acid residues of purified bacteriocin was identified as follows: $NH_{2}-XGXAGVXPXGGQQPXVPLXYP$.

Evaluation of Physiochemical Properties and Fermentation Qualities of Kimchi Supplemented with Yacon (야콘 첨가 김치의 이화학적 특성과 발효 품질 평가)

  • This study aimed to evaluate the physiochemical properties and fermentation qualities of kimchi supplemented with yacon (Smallanthus sonchifolius) during fermentation. Kimchi supplemented with 0%, 0.1%, 0.5%, 1%, and 5% yacon were investigated. As fermentation proceeded, pH decreased from 5.5~5.7 on the first day to 4.6~4.8 on day 21, whilst total titratable acidity (TTA), lactate levels, and number of microbes increased. The kimchi with 5% yacon showed a higher production of lactate concentration at 7 days fermentation than other groups, which is consistent with the observed high numbers of viable Leuconostoc sp. cells at 7 days fermentation. These observations demonstrate that 5% yacon stimulates the growth of kimchi microbes, particularly of Leuconostoc sp..

Effect of High Pressure Carbon Dioxide on Inactivation of Leuconostoc sp. (고압 이산화탄소에 의한 Leuconostoc sp.의 살균 효과)

  • Inactivation of Leuconostoc sp. isolated from kimchi using carbon dioxide under pressure was investigated in terms of operating parameters in order to evaluate its feasibility as a novel nonthermal process. Inactivation rates increased with increasing pressure, temperature and exposure time, but with decreasing working volume. Microbial reduction of 3 log cycles was achieved within 150 min under a $CO_2$ pressure of $60\;kg/cm^2$ at 30^{\circ}C$. It was confirmed that microbial inactivation by the high pressure $CO_2$ was governed essentially by the characteristic of $CO_2$ mass transfer and thus penetration of $CO_2$, into cells was a rate limiting step to determine efficiency of the inactivation process. The experimental results suggested that the high pressure $CO_2$ treatment could be used as one of the effective nonthermal methods for preserving foods.

Fermentation Patterns of Leek Kimchi and Chinese Cabbage Kimchi (부추김치와 배추김치 발효양상)

  • For the comparison of fermentation pattern of leek kimci with chinese cabbage kimchi, the changes of total viable cell number, Leuconostoc sp. bacteria, Lactobacillus sp. bacteria, pH and total sugar content of twotypes kimchies were investigated during fermentation at $20^{\circ}C$ and $10^{\circ}C$. In chinese cabbage kimchi at $20^{\circ}C$ fermentaion, the numbers of total viable cell, Leuconostoc sp. bacteria and Lactobacillus sp. bacteria reachedthe maximum level on 2nd day and reduced slowly. But in leek kimchi, the maximum numbers of total via-ble cells, Leuconostoc sp. bacteria and Lactobacillus sp. bacteria were obtained after 3 days fermentation,and the cell number of Lactobacillus sp. maintained at the maximum level oyer 15 days. At $10^{\circ}C$ fer-mentation, in both kimchies, the viable cell number of lactic acid bacteria more slowly increased anddecreased than at $20^{\circ}C$. The pH of chinese cabbage kimchi was 4.2 on 3rd day (optimal ripening phase) andmere decreased to 3.5 after 5 days, but in leek kimci the pH 4.2 could be reached after 10 days at $20^{\circ}C$. At $10^{\circ}C$, the optimal ripening pH 4.2 of chinese cabbage kimchi was reached after 6 days, but in leek kimchieven though after 24 days, the pH was maintained oyer 4.3. The total sugar contents of chinese cabbage him-chi and leek kimci were decreased continuously during fermentation. From these results, we know that thefermentation of leek kimchi proceed more slowly than chinese cabbage kimchi by the retardation of lacticacid bacteria growing in leek kimchi.

Effect of Lithospermum erythrorhizon, Glycyrrhiza uralensis and Dipping of Chitosan on Shelf-life of Kimchi (김치의 보존성 증진을 위한 자초.감초의 혼합 첨가와 Chitosan 침지 효과)

  • The studies were carried out to investigative effects of Lithospermum erythrorhizon, Glycyrrhiza uralensis 3% (LG) with and without dipping of salted Chinese cabbage in 1% chitosan solution(LGDC) on fermentation of kimchi at $10^{\circ}C$ during 25 days. The pH and titratable acidity of kimchi with LG and LGDC were higher and lower, respectively, than that of control. Viable cells of total bacteria, lactic acid bacteria, Leuconostoc sp. and Lactobacillus plantarum in kimchi added with LG and LGDC were shown inhibitory effect about $1.6{\sim}2.1,\;1.2{\sim}2.9,\;0.8{\sim}2.2,\;0.7{\sim}1.6$ log10 cycle, respectively. Specially Leuconostoc sp. and L. plantarum was very inhibited than in control from 0 day. The sour taste of LG and LGDC added kimchi was changed more slowly than that of control during fermentation of kimchi. But flavor, color and overall acceptability did not show significant difference(P<0.05) between treatments. The shelf-life of LGDC added kimchi was extended over 10 days compared with control.
