Antioxidant and Antidiabetic Activities of Jerusalem Artichoke Composites Containing Gynura procumbens, Momordica charantia, and Curcuma longa via AMPK Activation (명월초, 여주 및 울금을 포함한 돼지감자 복합물의 항산화 및 AMPK 활성화를 통한 항당뇨 활성)

  • This study was performed to identify the antioxidant and ${\alpha}$-glucosidase inhibitory activities of water and 70% ethanol extracts of the three following herbs: G. procumbens, M. charantia, and C. longa. In addition, the antioxidant and antidiabetic activities of five types of Jerusalem artichoke composites (JA1 - 5), which were prepared by adding ethanol extracts of several herbs to Jerusalem artichoke concentrate, were studied and compared. The results showed that the total phenol and flavonoid contents of the ethanol extracts were higher than those of the water extracts. The DPPH and ABTS radical scavenging activities and reducing power depended on the total phenol and flavonoid contents. The antioxidant activities of ethanol extracts from G. procumbens and C. longa were comparable. Moreover, the ${\alpha}$-glucosidase inhibitory activity of the ethanol extracts ($2,000{\mu}g/ml$) from each herb was found to be over 50%. In contrast, the five types of JA composites showed higher total phenol and flavonoid contents than those of JA concentrate. In addition, increased antioxidant and ${\alpha}$-glucosidase inhibitory activities were observed, with that of JA1 being the highest. However, all concentrations ($1{\sim}100{\mu}g/ml$) of JA tested did not affect the cell viability of Chang cells. In addition, JA induced the activation of AMP-activated protein kinase (AMPK) in Chang cells and significantly increased the glucose uptake in C2C12 cells. Therefore, it could be concluded that the JA composites (JA1 - 5) mixed with G. procumbens, M. charantia, and C. longa extracts were effective in increasing the extracts' antioxidant and antidiabetic activities.

  • This study reports a court dress culture in 1719 shown in the Scene of a Royal Banquet given by the King at Gyeong-hyeon-dang(景賢堂) of Gisagyecheop(耆社契帖). It can be summarized as follows: First, King Suk-jong(肅宗) attending a Royal Banquet for members of the Hall of Elder Statement wore the Ikseonkwan decorated with flowers, red Gollyongpo, Okdae, and Heukpiwha. The Crown Prince(王世子) also wore the Ikseonkwan decorated with flowers, black Goliyongpo, Okdae, and Heukpiwha. Second, senior statesmen(耆老臣) over the age 70 wore Sarno, Heukdallyeong with a crane rank tablet, belt, and black boots. Royal family members and officers also wore Samo, Heukdallyeong with a crane or silver pheasant rank tablet, belt, and black boots. Third, Sakum(司禁) of King's body guards wore Yungbok and Muyaebyulgam(武藝別監) wore Ja-geon and the green Jikryeong. Ui-jang-gun(儀仗軍) wore Pirip and Hongui, Haengjeon, and Unhae. Saboksikwan(司僕侍官) wore Samo, Heukdallyeong, belt, black boots, and a sword. Fourth, boy dancers(舞童) wore Buyongkwan, Kwanbok, skirt, belt, and black boots. Cheoyong-dancers(處容舞童) wore outfits in fine colors, wide pants, skirt, mask with an earring, gold colored belt, and white colored sandal. Hyeopryulrang(協律郞) wore Sarno and Heukdallyeong. Jipbak(執拍) wore Morabokdu and Noksaui, and Ak-kong(樂工) wore Whawhabokdu, Hongju-ui, Ojungdae, and Heukpiwha. Singer(歌童) wore Jajeok-dugeon, nokju-ui, and jajeokdae.

Effect of Deposition Conditions on the Morphology of MPECVD Diamond Thin Films (MPECVD 다이아몬드 박막의 표면 형상에 미치는 증착조건의 영향)

  • 다이아몬드 형성에 미치는 MPECVD 증착조건에 관하여 연구하였다. 증착 실험 기판은 Si p-type (100)wafer를 사용하였으며 다이아몬드 박막은 다음과 같은 조건하에서 증착되었다. 메탄 농도:0.75%(3scm)-3%(12scm),산소 농도:0%-0.5%(2scm), 반응 압력:20torr-80torr, 반응 온도:$600^{\circ}C$-90$0^{\circ}C$. 낮은 증착온도($600^{\circ}C$)에서는 (100)의 우선성장면을 보였고 온도가 증가함에 따라 (100)과 (111)이 혼재된 cubo-octahedron이 형성되었고 90$0^{\circ}C$에서는 (111)의 우선성장면을 가징 octahedron이 형성되었다. 산소가 첨가됨에 따라 높은 메탄농도에서도 양질의 다이아몬드가 형성되었다. 낮은 압력하(200torr)에서는 비정질탄소가, 높은 압력하(80torr)에서는 양질의 다이아몬드가 형성되었다.

자궁암 치료 시 고선량률 근접 치료기와 사용될 초음파 온열치료기 개발

  • 자궁암 치료시 고선량률 강내 근접치료기와 병행하여 사용할 초음파 온열 치료기가 개발되었다. 온열치료기는 텐덤형, 오보이드 형, 원통형으로 디자인 되었다. 각각의 치료기들에 사용될 초음파원 으로 PZT물질이 사용되었다. 텐덤형의 경우 외부직경이 2mm 이고 길이가 24.5mm 인 PZT5A가 사용되었고 오보이드형은 직경 20mm, 길이 20mm 인 원통형의 PZT 8 물질을 반으로 나누어 사용하였으며 원통형 치료기는 외경 25.4mm, 길이가 15.2 mm 인 원통형 PZT 8물질이 이용되었다. 각각의 트랜스듀서의 초음파 발생효율을 측정하였다. 텐덤형의 경우 3.2MHz 로 작동하였고 평균효율성은 16.7% 이다. 오보이드형의 작동 주파수는 1.5MHz 이며 효율성을 32${\sim}$46% 이다. 마지막으로 원통형의 작동주파수는 1.7MHz 이며 측정?? 효율성의 범위는 55${\sim}$65%이다. 특성분석 결과 온열치료에 사용될 초음파원으로 사용가능함이 입증되었다. 따라서 3개의 텐덤형 드랜스듀서를 연결하여 자궁내막의 길이가 73.5mm까지 치료가능한 텐덤형 온열치료기가 제작되었고 2개의 반원통형 트랜스듀서를 연결하여 치료길이가 40mm 인 오보이드형 치료기가 제작되었다. 마지막으로 4개의 원통형 트랜스듀서를 연결하여 질암등에 사용이 가능하고 61mm 까지 치료가 가능한 원통형 온열 치료기가 제작되었다.

  • To temperature-compensate the Bragg wavelength of fiber grating filters two materials with different thermal expansion coefficients were depolyed for packaging. After temperature-compensation packaging the maximum difference of the Bragg wavelength in the temperature range of $-10^{\circ}C$ to $70^{\circ}C$ was 0.03nm, which is only about one thirtiety of the Bragg wavelength shift of the temperature-uncompensated fiber grating filter.

  • The variations of crystalline structures and physical proerties of serpentine and talc mineral which was treated in a planetary ball mill, wre investigated by means of X-ray diffraction, FT-IR, DSC-TG, particle size analysis and SEM observation. The crystalline peaks of serpentine and talc in the XRD patterns were gradually reduced with the increase of grinding time and after 120 minutes turned into the amorphous phase, which is attributed to the disordering of the local structure around magnesium. The endothermic peaks for the ground serpentine and talc sample are observed at a considerably lower temperatures than that for the starting sample. A morphological change from the originally flat and irregular shape to spherically agglomerated particles was clearly observed.

  • Leuconostoc mesenteroides B-512FMCM, 742CB3, 1299C의 dextransucrase들의 glycosyl기 전이 특성을 수용체 반응과 transglycosylation반응을 통해 확인하였다. 수용체 반응의 경우 10% sucrose에 수용체로 4% maltose를 첨가하여 반응시켰고 transglycosylation반응은 다른 크기, 다른 농도 그리고 다른 종류의 가지 결합의 dextran 을 합성하는 효소들을 이용하여 수행하였다. 각각의 효소들은 maltose를 이용한 수용체 반응에서 유사한 종류의 수용체 산물들을 합성한 것에 비해 세 dextransucrase들 (512FMCM, 742CB3, 1299C) 을 일정 비율로 혼합하여 maltose를 이용한 수용체 반응 결과 512FMCM 효소의 활성 비율을 줄이고 742CB3, 1299C 효소의 활성 비율을 증가시켰을 경우에는 ${\alpha}-1{\rightarrow}$3 의 가지결합이 많은 dextran 을 합성하였다. 또한, 세 가지 다른 구조의 dextran(T40, 742CB, B1299)에 100mM maltose을 수용체로 첨가해 각각의 dextransucrase(512FMCM, 742CB3, 1299C)와 transgly cosylation을 수행한 결과 1299C 효소가 세 종류의 dextran(T40, 742CB, B1299) 에 모두 가지 결합이 많은 dextran을 합성함을 확인하였다. 또한 ${\alpha}-1{\rightarrow}$6 결합으로 주로 이루어진 2%, 5% dextran(T10, T40, T7O, T500, T2000)에 dextransucrase(512FMCM, 742CB3, 1299C)를 반응시켜 기존의 dextran 보다 가지 결합이 더 많이 형성된 transglycosylation 산물을 합성하였다. 이때 maltose를 첨가했을 경우 이 수용 체에 의해 많은 ${\alpha}-1{\rightarrow}$6 가지 결합의 dextran 을 합성함을 확인하였다.

Cloning, Expression, and Characterization of a Glycoside Hydrolase Family 118 ${\beta}$-Agarase from Agarivorans sp. JA-1

  • We report a glycoside hydrolase (GH)-118 ${\beta}$-agarase from a strain of Agarivorans, in which we previously reported recombinant expression and characterization of the GH-50 ${\beta}$-agarase. The GH comprised an open reading frame of 1,437 base pairs, which encoded a protein of 52,580 daltons consisting of 478 amino acid residues. Assessment of the entire sequence showed that the enzyme had 97% nucleotide and 99% amino acid sequence similarities to those of GH-118 ${\beta}$-agarase from Pseudoalteromonas sp. CY24, which belongs to a different order within the same class. The gene corresponding to a mature protein of 440 amino acids was inserted, recombinantly expressed in Escherichia coli, and purified to homogeneity with affinity chromatography. It had maximal activity at $35^{\circ}C$ and pH 7.0 and had 208.1 units/mg in the presence of 300 mM NaCl and 1 mM $CaCl_2$. More than 80% activity was maintained after 2 h exposure to $35^{\circ}C$; however, < 40% activity remained at $45^{\circ}C$. The enzyme hydrolyzed agarose to yield neoagarooctaose as the main product. This enzyme could be useful for industrial production of functional neoagarooligosaccharides.

MLL5, a histone modifying enzyme, regulates androgen receptor activity in prostate cancer cells by recruiting co-regulators, HCF1 and SET1

  • In prostate cancer, the androgen receptor (AR) transcription factor is a major regulator of cell proliferation and metastasis. To identify new AR regulators, we focused on Mixed lineage leukemia 5 (MLL5), a histone-regulating enzyme, because significantly higher MLL5 expression was detected in prostate cancer tissues than in matching normal tissues. When we expressed shRNAs targeting MLL5 gene in prostate cancer cell line, the growth rate and AR activity were reduced compared to those in control cells, and migration ability of the knockdown cells was reduced significantly. To determine the molecular mechanisms of MLL5 on AR activity, we proved that AR physically interacted with MLL5 and other co-factors, including SET-1 and HCF-1, using an immunoprecipitation method. The chromatin immunoprecipitation analysis showed reduced binding of MLL5, co-factors, and AR enzymes to AR target gene promoters in MLL5 shRNA-expressing cells. Histone H3K4 methylation on the AR target gene promoters was reduced, and H3K9 methylation at the same site was increased in MLL5 knockdown cells. Finally, xenograft tumor formation revealed that reduction of MLL5 in prostate cancer cells retarded tumor growth. Our results thus demonstrate the important role of MLL5 as a new epigenetic regulator of AR in prostate cancer.
