A Preliminary Study on Growth and Habitat Characteristics of Zostera marina (Zosteraceae) in Gamak Bay, Yeosu

  • This study was performed to obtain basic information on the ecology of Zostcra marina and to promote efficient conservation of this species which has been decline in Gamak Bay, Yeosu, Korea. Whater column characteristics and eelgrass morphology at Anpori, Jangsuri and Wonpori were investigated every month from December 1999 to November 2000. The water temperature, salinity and pH at the three sites were 10.0-27.0${\circ}C$,29.4-34.7% and 8.1-8.5, respectively. The water temperature at Anpori tended to be slightly lower than that at the other locations; the salinity at Wonpori from July to November was a little lower than that of the other locations. The concentrations of $NO_2$-N, $NO_3$_N, $NH_4$-N, $PO_4$-P and $Si(OH)_4$-Si at the three sites were 0.9-1.3, 2.0-6.2, 7.8-9.0, 3.0-3.6 and 22.2-30.2 uM, respectively. The concentration of $NO_3$-N at Wonpori from June to November was somewhat lower than that at the other locations; that of NH4-N at Jansuri was somewhat lower than the others. The mean shoot height and leaf width of the Anpori, Jangsuri and Wonpori populations were 80.6 cm and 0.9 mm, 90.0 cm and 1.0 mm, and 95.3 cm and 1.0 mm, respectively. The mean total shoot weight of the Anpori, Jangsuri and Wonpori ones was 24.5,31.0 and 29.7 & respectively. The mean leaf and branch numbers of the Anpori, Jangsuri and Wonpori populations were 16.5 and 2.6, 16.1 and 2.4 and 15.4 and 2.6 individuals, respectively. The correlation coefficients between shoot height and water temperature, leaf width and total shoot weight, leaf number and branch number, and $Si(OH)_4$-Si and $NO_3$-N were 0.726, 0.692, 0.862, and 0.693, respectively. The coefficients between shoot height and $NO_3$-N, total shoot weight and NO_3$-N, water temperature and $Si(OH)_4$-Si, water temperature and salinity, and water temperature and $NO_3$-N were -0.716, -0.536, -0.775, -0.685 and -0.685, respectively. The first four principal components explain 71.1% of the total sample variance. For axis 1, shoot height and water temperature tended to correlate with the population of Jansuri, followed by the Wonpori population, and $Si(OH)_4$-Si and $NO_3$-N tended to correlated strongly with the Anpori population. For axis 2, total weight, leaf width, leaf number and branch number showed a tendency to correlate with the Anpori and Jangsuri populations. For axis 3, the Anpori population tended to be influenced by $NO_2$-N and $PO_4$-P. For axis 4, the Wonpori and Jangsuri populations tended to be affected by salinity. The tendency, however, differed according to season.

Antioxidative and Anticancer Activities of Various Solvent Fractions from the Leaf of Camellia japonica L. (동백나무 잎 용매분획물의 항산화 및 항암 활성)

  • To obtain basic information on the potential use of Camellia japonica leaf as a raw material in functional food, leaf antioxidant and anticancer activities were investigated. The radical-scavenging activity of various solvent fractions from the leaf, as shown by the DPPH radical test, increased in a dose-dependent manner, with the water fraction showing the highest activity. The reducing power of various solvent fractions from the leaf was also dose-dependent, and, again, the water fraction showed the highest reducing power. The water fraction showed strong antioxidant activity in the linoleic acid test and was also capable of scavenging nitrite in a dose-dependent manner. Proportions of 92.15% and 95.61% of available nitrite were scavenged by the water and butanol fractions, respectively, at levels of $1,000{\mu}g/mL$. Both butanol and water fractions exhibited strong inhibitory effects on the growth of human lung and colon cancer cells. The total phenolic contents of the butanol and water fractions were 216.26 mg/g and 220.68 mg/g, respectively. High-performance liquid chromatography (HPLC) showed that quercetin and epicatechin were the predominant phenolic compounds in the water fraction. The activities of this fraction are attributable to the presence of these phenolic compounds, particularly quercetin and epicatechin.

Study on the Quality Characteristics of Steamed Bread Using Bamboo and Lotus Leaf Powder (죽엽과 연잎 분말을 첨가한 찐빵의 품질 특성에 관한 연구)

  • The purpose of this study was to investigate the quality characteristics of steamed bread using mixed flour containing bamboo leaf and lotus leaf powders. Properties of the steamed breads were tested by colorimeter, water activity, rheometer, SEM, and sensory tests. The L values and a values were higher in steamed bread with bamboo leaf powder. On the other hand, the b values of streamed bread with bamboo leaf powder were lower than lotus leaf powder. During storage, the hardness of steamed bread with bamboo leaf and lotus leaf powders increased control. Furthermore, steamed bread with lotus leaf powder showed hardness than bamboo leaf powder. The springiness of all steamed breads decreased with storage, and steamed breads with bamboo leaf powder showed lower values than lotus leaf powder. The cohesiveness of the control was the highest, and all samples showed significant differences each other. The gumminess of the control was higher than steamed bread with bamboo leaf and lotus leaf powders. Moreover, longer storage time yielded higher gumminess. control relatively smoother surface than that of 1% bamboo leaf powder. The steamed bread with lotus leaf powder had a rougher surface than bamboo leaf powder. sensory characteristics of steamed bread, green color of the samples with bamboo leaf powder was darker than lotus leaf powder. Leaf odor was similar throughout. Freshness of steamed bread containing 1% bamboo leaf powder the highest. The overall acceptability of consumer acceptance was the control, followed by steamed bread containing 1% lotus leaf powder.

Effects of Artificial Acid Mist on Leaf Injury and Surface Wettability of Several Broad-Leaved Species (인공산성연무(人工酸性煙霧)의 처리(處理)가 몇 활엽수종(闊葉樹種)의 엽피해(葉被害)와 엽표면(葉表面)의 친수성(親水性)에 미치는 영향(影響))

  • To seek effective methods for evaluating air pollution and acid rain injury, artificial acid mist(pH 2.5, 3.5 and 4.5) and ground water(pH 6.5) were treated on the potted seedlings of Ligustrum obtusifolium, Cercis chinensis, Hibiscus syriacus and Sophora japonica. Leaf chlorophyll contents, characteristics of leaf-injury, wettability-measurement of diameter of water-droplets on the leaf surface-among treatments were investigated. The results were summarized as follows. 1. Chlorophyll contents of Ligustrum obtusifolium and Hibiscus syriacus measured on June 3 were highest in pH 2.5 plot, but those of Cercis chinensis and Sophora japonica were relatively low level. Chlorophyll contents of Ligustrum obtusifolium measured on August 24 was highest in pH 2.5 plot, but those of Cercis chinensis, Hibiscus syriacus and Sophora japonica were highest in the control. 2. Changes of chlorophyll contents with acid mist treatments were differed among tree species. 3. For all the tested species, leaf injury(injured leaf number and rate, and injured leaf area) increased with decreasing pH levels of acid mist. 4. Leaf tissue injury seemed to be related with the wettability of the leaf surface. Measurement of diameter of water-droplets on the leaf surface might be useful criteria for acid rain or acid mist injury for the glabrous leaved species, such as, Cercis chinensis, Sophora japonica, etc.

Effects of Storage Temperature and Packaging Treatment on the Quality of Leaf Lettuce (상추 저장에 미치는 온도, 포장재료 및 유공 필름 영향)

  • The effects of packaging treatments with either low density polyethylene (LDPE) or polypropylene (PP) films on the quality of leaf lettuce were evaluated. It was also stored within a plastic bag (LDPE and PP films) with 4 pelf orated holes (6mm diameter). Changes in weight loss, color difference, anthocyanin, chlorophyll, water content, and general appearance (shape of lettuce) ware investigated Total weight of leaf lettuce packaged within non-pelf orated films decreased slowly over the storage periods at 5 and $20^{\circ}C$. The weight loss of leaf lettuce showed no significant difference between non-pelf orated and perforated films at $20^{\circ}C$. General appearance in leaf lettuce stored at $20^{\circ}C$ was not significantly affected by packaging treatments with LDPE and PP films. The shelf life of non-packaged leaf lettuce at $5^{\circ}C$ was 8 days, whereas packaging treatments with LDPE and PP films showed freshness in leaf lettuce for 30 days of storage. Total chlorophyll and water contents of leaf lettuce packaged within pelf orated and non-pelf orated films decreased gradually during storage. However, no remarkable changes in color difference, levels of chlorophyll and anthocyanin, and water contents were observed in leaf lettuce packaged within perforated and non-pelf orated films. In this experiment, leaf lettuce packaged within LDPE or PP films without any punching holes at $5^{\circ}C$ was the most desirable for extending the quality. These results suggest that non-pelf orated packaging treatment in combination with low storage temperature could be effective in prolonging the shelf life of leaf lettuce

Effects of Water Stress on Leaf Orientation, Apparent Photosynthetic Rate, Transpiration Rate, Yield and Its Related Traits in Soybean Plants (한발조건이 콩식물체의 엽운동, 광합성능, 증산량, 수량 및 관련 형질에 미치는 영향)

  • To investigate effects of water stress on apparent photosynthetic, transpiration rates, leaf orientation, yield and its related traits, four soybean varieties were planted on the Wagner pots in a plastic house covered with polyethylene film. As the light intensity and leaf temperature in a day increased, the movement of central leaflet in the second leaf of main stem occurred earlier than that of the lateral leaflet. The apparent photosynthetic rate of the central leaflet was higher than that of the lateral leaflet, but light intercept and leaf temperature of lateral leaflet were higher than those of the central leaflet. The apparent photosynthetic rate had highly positive correlation with the photon flux density, stomatal conductance and temperature, respectively. The photon flux density, stomatal conductance, transpiration and photosynthetic rates in the control were significantly higher than those in the water stress plot. The yield and its related traits in the water stress plot became decreased significantly in comparison with the control.

Water Extracts of Anthriscus sylvestris Leaf induces Apoptosis in FaDu Human Hypopharynx Squamous Carcinoma Cells

  • Anthriscus sylvestris (L.) Hoffm. is a perennial herb found widely distributed in various regions of Korea, Europe, and New Zealand. The root of A. sylvestris have been extensively used in the treatment for antitussive, antipyretic, cough remedy in Oriental medicine, but the physiologically active function of the leaf of A. sylvestris is as yet unknown. In this study, we investigated the anti-cancer activity and the mechanism of cell death of water extracts of leaf of Anthriscus sylvestris (WELAS), on human FaDu hypopharyngeal squamous carcinoma cells. Our data showed that WELAS treatment inhibited cell viability in a concentration- and time-dependent manner. In addition, the treatment of WELAS markedly induced apoptosis in FaDu cells, as determined by the viability assay, DAPI stain and FACS analysis. WELAS also increased the proteolytic cleavage of procaspase-3, -9 and PARP (poly(ADP-ribose) polymerase). In addition, exposure to WELAS decreased the expression of Bcl-2 (an anti-apoptotic factor), but increased the expression of Bax (a pro-apoptotic factor), suggesting that mitochondria-dependent apoptotic pathways are mediated in WELAS-induced apoptosis. Taken together, these results indicate that water extracts of leaf of A. sylvestris inhibits cell growth and induces apoptosis via the mitochondrial-dependent apoptotic pathway in FaDu human hypopharyngeal squamous carcinoma cells. Therefore, we propose that the water extracts of leaf of A. sylvestris is a novel chemotherapeutic drug, having growth inhibitory properties and induction of apoptosis in human oral cancer cells.

Quality Characteristics of Pork Patties Prepared with Mugwort, Pine Needle and Fatsia Leaf Extracts (쑥, 솔잎 및 두릅 추출물을 첨가한 돈육 Patty의 품질 특성)

  • In order to investigate the possibility of functional property improvement of meat products, four kinds of pork patties were prepared with water 10% as control, mugwort extract 10%, pine needle extract 10% and fatsia leaf extract 10%, respectively. In case of control moisture content was higher, but crude fat was lower, compared to patties treated with plant extracts. Crude ash content of pine needle extract treatment showed higher level than that of other patties. pH range of patties revealed to 5.92∼5.978. In raw patties Hunter's L-and a-value of control were higher than those of plant extract treatment, and a-value of raw patties showed higher level than that of cooked. Yield, water holding capacity, salt soluble protein extractability and gel strength among patties did not show significant differences. In control patty, fat retention was lower compared to other plant extract treatments, but water soluble protein extractability was higher compared to pine needle. Values such as hardness, springiness, cohesiveness, gumminess and chewiness were not significantly different among the patties. In sensory scores such as aroma, juiciness and palatability, significant differences were not observed among cooked patties, but texture score was higher in the order pine needle, mugwort, fatsia leaf and control.

Physiological Activities of Leaf and Twig Extracts from Lindera obtusiloba Blume (생강나무 잎 및 가지 추출물의 생리활성)

  • We investigated the physiological activities of extracts from Lindera obtusiloba Blume leaf and twig (LLW: water extract from Lindera obtusiloba Blume leaf, LLE: 50% ethanol extract from Lindera obtusiloba Blume leaf, LTW: water extract from Lindera obtusiloba Blume twig, LTE: 50% ethanol extract from Lindera obtusiloba Blume twig). Total polyphenol and total flavonoid contents of LTE were 445.38 mg/g and 302.09 mg/g, respectively. The electron donating ability (95.38%) of LTE was higher than that of the LLE (93.76%), LTW (88.09%), and LLW (82.06%). The oxygen radical absorbance capacity of extracts were improved with 50% ethanol condition, rather than hot water. Superoxide radical scavenging activity and FRAP activity of the extracts were improved with an increase of treatment concentration. All the extracts($1,000{\mu}g/mL$) stimulated a production of nitric oxide (NO) in macrophage RAW264.7 cells. In particular, the NO stimulating activity of LTE was superior to that of LLE, LTW, and LLW. The antitumor activity of LTE ($500{\mu}g/mL$) in A549, HeLa and SNU719 was 55.63%, 83.87% and 68.11%, respectively. The UVB-induced MMP-1 production in HS68 cells was suppressed by the treatment of LTE (88.28%), LLE (83.96%), LTW (80.59%) and LLW (76.08%).

Evaluation of Chemical Composition in Reconstituted Tobacco Leaf using Near Infrared Spectroscopy (근적외선 분광분석법을 이용한 판상엽 화학성분 평가)

  • Near InfraRed Spectroscopy(NIRS) is a quick and accurate analytical method to measure multiple components in tobacco manufacturing process. This study was carried out to develop calibration equation of near infrared spectroscopy for the prediction of the amount of chemical components and hot water solubles(HWS) of reconstituted tobacco leaf. Calibration samples of reconstituted tobacco leaf were collected from every lot produced during one year. The calibration equation was formulated as modified partial least square regression method (MPLS) by analyzing laboratory actual values and mathematically pre-treated spectra. The accuracy of the acquired equation was confirmed with the standard error of prediction(SEP) of chemical components in reconstituted tobacco leaf samples, indicated as coefficient of determination($R^2$) and prediction error of sample unacquainted, followed by the verification of model equation of laboratory actual values and these predicted results. As a result of monitoring, the standard error of prediction(SEP) were 0.25 % for total sugar, 0.03 % for nicotine, 0.03 % for chlorine, 0.16 % for nitrate, and 0.38 % for hot water solubles. The coefficient of determination($R^2$) were 0.98 for total sugar, 0.97 for nicotine, 0.96 for chlorine, 0.98 for nitrate and 0.92 for hot water solubles. Therefore, the NIRS calibration equation can be applicable and reliable for determination of chemical components of reconstituted tobacco leaf, and NIRS analytical method could be used as a rapid and accurate quality control method.
