몇 가지 뽕품종에 따른 오디의 형태 및 화학적 성분의 특성 (Morphological and Chemical Characteristics of Mulberry(Morus) Fruit with Varietes)

  • The morphological and chemical characteristics of leaves and fruits were observed in the three mulberry varieties, including Daeryugppong(Morus Lhou(Ser.) koidz), Kugsang 20(Morus Lhou(Ser.) koidz) and Cataneo(Morus alba L.). The leaf development in spring was earliest in Cataneo and latest in Kugsang. Content of total nitrogen and Mg in leaf was the highest in Cataneo while that of P2O5, K and Ca in Daeryukppong. Flowers started to bloom from May 8 and in full bloom around May 15. Mature fruits began to set from June 10 to 15 and lasted by July 10 in Cataneo. Average fruit weight was heaviest in Kugsang 20(3.52 g/fruit), while lowest in Daeryukppong(1.61 g/fruit). In fruits, glucose and fructose were the major sugars. Citric acid was the most abundant organic acid in three varieties with its average content from 0.8 to 0.14%. The major pigment in fruit was anthocyanin and its content varied among varieties. The stability of anthocyanin was evaluated under various pH, temperature, and sugar concentrations. Rutin was the major flavonol glycoside present in fruits and its content varied from 0.92 to 3.36 mg/gDW. Other flavonol glycosides such as isoquercitrin and quercitrin were also detected in fruit.

목련잎의 염색성에 관한 연구 (Dyeing Properties of Magnolia liliflora Leaf Extract on Fabric)

  • This study investigated the dyeability on silk, wool and rotten fabrics dyed with Magnolia liliflora leafs. In addition, the fastness of washing, perspiration, rubbing, drycleaning and the effects of its pigment on bacterial reduction and uv-B protection were also investigated. The results were as follows : It was found that uv-visible absorption spectrum showed two strong absorption peaks in the range of $250{\sim}340nm$. The optimum dyeing condition of the pigments extracted from the Magnolia liliflora leafs was dyeing with 0.5% mordants and three repeated dyeing at $95^{\circ}C$ for 1hr. When the wool fabric was dyed with Magnolia liliflora leaf, dyeing properties was the best among the three fabrics. Washing fastness of dyed fabrics was very low, drycleaning fastness was good and the other fastness were good. Light fastness of three fabrics dyed by Magnolia liliflora leafs increased by mordant treatment, especially copper sulfate treatment. The bacterial reduction and uv-B protection of dyed wool fabric with Magnolia liliflora leafs also increased.

  • 인삼(Panax ginseng C.A. Meyer) 엽소병에서 chorophyll의 bleaching 현상에 대한 생리적 반응기작을 조사, 분석한 결과 chorophyll의 bleaching 현상은 singlet oxygen ($^1O_2$)에 의한 photooxidation 과 catalase와 peroxidase의 inactivation에 의한 $H_2O_2$의 autooxidation이었다. Chorophyll의 bleaching 현상에 원인으로 추리된 $H_2O_2$를 농도별로 인삼잎 절편에 처리하였던 바, chorophyll의 함량과 chorophyll의 흡수스펙트럼은 자연광 처리시 유발된 bleaching 현상과 일치하였으며, saponin 첨가로 인하여 bleaching 현상은 현저히 촉진되었다.

  • 국립중앙박물관이 소장하고 있는 부석사 괘불(浮石寺 掛佛, 1684년, K969)은 사찰의 야외 행사에 사용하기 위해 제작한 대형 불화이다. 하단에 석가모니불을 중심으로 비로자나불, 약사불, 아미타불의 부처가 그려져 있으며 현존하는 괘불 중에서 가장 복잡한 구성을 보여준다. 본 연구는 부석사 괘불에 대해 바탕직물의 특성과 괘불 제작방법을 조사하고 채색에 사용된 재료를 판별하기 위한 비파괴 조사 결과를 종합하여 채색기법의 특징을 연구한 것이다. 부석사 괘불은 좌·우로 11폭, 상·하단으로 각 1폭씩 연결되어 총 13폭이며 중심 부분의 9폭은 생초(生綃), 사면을 두르는 4폭은 주(紬)를 사용하여 그림의 바탕을 만들었다. 그림을 그리는 데 사용된 채색 재료는 엑스선형광분석 방법으로 조사하였고 적색, 황색, 녹색, 청색, 흰색의 무기안료이며 황색과 청색의 일부에서는 연백을 먼저 칠하고 그 위에 유기안료로 색상을 표현했음을 확인하였다. 또한 검은 선은 먹, 가사의 문양에는 금박이 사용되었다. 엑스선조사는 안료의 주성분과 안료층의 두께에 따라 밝고 어둡게 보이는 차이를 판독하는 것으로 안료의 색상에 따른 채색 위치와 기법을 확인할 수 있었다.

  • 찹쌀풀과 생참기름 또는 밀가루풀과 콩기름을 사용하여 제조한 연근, 김, 깻잎 부각의 텍스쳐, 지방질 산화, 색소를 평가하고 튀김유의 변화를 평가하였다. 찹쌀풀과 생참기름으로 제조한 연근, 김, 깻잎 부각이 밀가루풀과 콩기름으로 제조한 부각에 비하여 유의하게 높은 경도를 나타냈으며, 기름 흡수량은 다소 높았으나 공액이중산값과 아니시딘값으로 평가한 지방질 산화는 적었다. 부각 지방질의 산화 정도는 튀김유의 산화에 의해 결정되었으며 부각의 지방산 조성은 튀김유의 지방산 조성과 유사하였다. 김 부각과 깻잎 부각은 클로로필과 카로티노이드를 다량 함유하였으며 찹쌀풀과 생참기름 또는 밀가루풀과 콩기름 등 제조 방법은 부각의 색소 함량에 큰 영향을 주지 않았으며 찹쌀풀과 밀가루풀 등의 튀김옷은 김과 깻잎 색소가 기름으로 용출되는 것으로부터 보호해 주었다.

인삼 틸라코이드 막의 지질과 산화 (Lipid Peroxidation of Ginseng Thylakoid Membrane)

  • In order to elucidate the mechanism of the leaf-burning disease of ginseng (Panax ginseng C.A. Meyer), the relationships between thylakoid membrane peroxidation and chlorophyll bleaching were investigated in comparison with the ones of soybean (Glycine max L). When I measured the rate of lipid peroxidation in the thylakoids of ginseng and soybean by irradiation of light(60 w.m-2), it was identified that, the remarkably lower rate of lipid peroxidation was found in the ginseng thylakoid than the case of soybean. When lipid peroxidation of ginseng thylakoid was induced in the dark, chlorophyll contents of thylakoid was not changed. The results suggest that lipid peroxidation does not affect the chlorophyll bleaching in ginseng thylakoid. Thylakoid membrane peroxidation as well as chlorophyll bleaching was closely related with photosynthetic electron transport. But, according to the quenching experiment active oxygen species induced lipid peroxidation may be different species in the case of chlorophyll bleaching.

자색고구마 습관성캘러스로부터 안토시아닌 고생합성 세포주 선발 (Selection of High Anthocyanin-Producing Cells from Habituated Callus Derived from Purple Sweetpotato)

  • 자색고구마 (Ipomoea batatas cv. Jami) 잎 조직배양으로 유도된 캘러스는 계대배양시 생장량이 감소하였다. 빠르고 간편하게 생장하는 습관성캘러스를 $1{\mu}M\;2,4-D$ 첨가된 MSBM에서 계대배양 없이 6개월간 지속배양하여 선발하였다. 습관성캘러스는 생장조절제가 첨가되지 않는 MSBM의 고체, 액체 배지에서 계대배양 횟수의 제한 없이 빠르게 지속생장이 가능하였다. 암조건의 액체배양 6 개월 후에는 자색생합성 부위가 생겼으며, 이를 2년 여간 분리하여 자색습관성세포괴를 선발하였다. 안토시아닌 함량은 자미의 괴근에서 추출되는 1.5 mg/ml에 근접한 1.0 mg/ml으로 나타났다. 자색습관성 세포괴는 안토시아닌 기내대량에 효과적 세포주로 사용 될 수 있을 것으로 기대한다.

백더덕, 홍더덕, 청더덕의 외부형태적 특성 및 유전적 차이 분석 (Morphological and genetic differences among white-, red- and blue colored root lines in Codonopsis lanceolata)

  • In general, the root color of Codonopsis lanceolata is white, but red or blue-colored root is found at a low frequency in nature. Red or blue-colored roots have scarcity value, thus farmers wish to produce colored roots. The factors for determining the color of roots are unclear whether the color is controlled by genetically or simply by environmentally such as soil environment. Using in vitro culture system which is advantageous for setting of the same culture condition, we analyzed the physiological and morphological characteristics and genetic differences among red-, blue- and white lines of C. lanceolata. In the red colored roots, stems of in vitro cultured plantlet were colored in dark red pigment. Histological analysis revealed that the red pigment was accumulated in the outer cortex layer of the stem and determined as anthocyanin. Chlorophyll contents in red root lines were higher than those in white- and blue root lines. Plantlets from red roots were smaller in both shoot length and total leaf area than those from white- and blue roots. Genetic differences among the three different colored C. lanceolata were determined by RAPD (Randomly Amplified Polymorphic DNA) analysis. Each line of colored roots had clear DNA polymorphism. These results indicate that the occurrence of red- and blue colored roots in nature was determined by genetic factors rather than soil enviromental conditions.

평안감사향연도(平安監司饗宴圖)의 채색 재료 특성 연구 -연광정연회도(練光亭宴會圖)를 중심으로- (A Study of the Characteristics of Painting Materials Used in Welcome Feast for the Pyeongan Governor: Focusing on Banquet at Yeongwangjeong Pavilion)

  • 본 연구에서는 조선 후기 기록화인 《평안감사향연도》 총 3점 중 <연광정연회도>를 분석하여 채색 재료를 규명했다. 포인트 위주의 분석으로 인해 발생 가능한 문제를 최소화하고자 우선 파장별 이미지를 촬영했다. 엑스선을 통해 먹, 금, 유·무기 안료를 분류했고, 적외선으로 먹과 구리계 안료를 파악했다. 초분광으로는 흑색, 청색, 녹색 재료 중에서 안료와 유기 염료를 구별해냈다. 색상별 선정 지점을 분석하여 백색(연백), 흑색(먹/쪽), 적색(연단+진사), 분홍색(염료), 자주색(염료), 갈색(산화철), 황색(석황/염료), 녹색(석록/석록+염료/쪽)과 청색(석청/연백+쪽/쪽), 자색(쪽+연지), 금색(금박) 등의 재료를 규명했다. 향후 파장별 라이브러리가 확보되면 더 효율적인 분석이 가능할 것으로 기대된다.

Secondary Metabolites and Morphological Diversity in the Leaves of Perilla Landrace from Korea

  • Screening and identification of genetic resources based on their phytoconstituents and morphological characters potentially provide baseline data for researchers, breeders, and nutraceutical companies who wish to formulate a nutrient-dense diet and health beneficial supplement. Thus, we evaluated the amount of total phenolic content and major phenolic compounds; examined if phenolic compounds could be used as distinguishing factors for perilla genetic resources; and investigated the relation between some quantitative and qualitative morphological characters with the contents of phenolic compounds in 360 accessions obtained from National Agrobiodiversity Center gene bank, Jeonju, Korea. Total phenolic content (TPC) was estimated using Folin-Ciocalteu colorimetric assay. Individual phenolic compounds were determined using an Ultra-High Performance Liquid Chromatography system equipped with Photodiode Array detector. Considerable variations were observed in TPC (7.99 to 117.47 mg GAE/g DE), rosmarinic acid (RA) (ND to 19.19 mg/g DE), caffeic acid (CA) (ND to 0.72 mg/g DE), apigenin-7-O-diglucuronide (ADG) (ND to 1.24 mg luteolin equivalent (LUE)/g DE), scutellarein-7-O-glucuronide (SG) (ND to 4.32 mg LUE/g DE), and apigenin-7-O-glucuronide (AG) (ND to 1.60 mg LUE/g DE). RA was the most dominant phenolic compound in most accessions (95.3%) followed by SG. The adaxial leaf color was light green, green and dark green in 13.8%, 65.0%, and 21.1 % of the accessions, respectively. 78.8% of the accessions had light green color at the abaxial side with the remaining being described as green. Most of the accessions (96.9%) were cordate shape, the remaining being eclipse. Intensities of green pigment at abaxial and adaxial leaf surfaces were correlated with contents of individual phenolic compounds and TPC whereas leaf length and width had no correlation with TPC, CA and RA, and negatively correlated with ADG, AG, and SG. Leaf shape was not related with content of phenolic compounds, color of leaves, or the length or width of leaves. Accessions IT57426, IT157434, IT267710, and IT267712 which contained relatively high contents of TPC and major phenolic compounds (RA and SG) could be used for further research in breeding and bioassay test. Our study result showed the contents of total phenolics and individual phenolic compounds along with the morphological characters could be useful distinguishing factors for perilla genetic resources.
