The Effects of Artificial Light Sources on Lettuce Seedling Vigor and Growth

  • The aim of this study was to investigate the effects of artificial light sources on the germination and seedling vigor of lettuce, aiming to identify the optimal artificial light source for producing high-quality seedlings. Lettuce cultivar of 'Tomalin' and 'Seonpunggold' exhibited the highest seed emergence in the Metal halide(MH) lamp and High-pressure sodium(HPS) lamp treatment group, while the emergence rate parameter, T50, was the fastest in the HPS lamp treatment group. Both cultivars showed good growth characteristics such as number of leaves, root length, and stem diameter under RGB-LED, and their seedling vigor was excellent as well. The plant height was smallest in the Red + Green + Blue LED treatment, but the leaves were round and thick, resulting in higher biomass and dry weight. Single light sources of Red LED and Blue LED led to reduced growth compared with that under the mixed light treatments. Chlorophyll content in lettuce varied with the type of artificial light, with both cultivars exhibiting the highest chlorophyll content in the Red + Green + Blue LED treatment. The most suitable artificial light for lettuce seedling growth was the Red + Green + Blue LED treatment.

Accumuation Pattern of Nitrate-Nitrogen in Sorghum And Maize Plants as Affected by Morphological Characteristics And Environmental Temperature (Sorghum 및 옥수수의 형태적 특성과 재배온도가 Nitrate-Nitrogen 축적에 미치는 영향)

  • Sorghum cv. Pioneer 93 1, sorghum-sudangrass hybrid cv. Sioux and maize plant cv. Blizzard were assayed for toxic concentrations of nitrate-nitrogen ($NO_3$-N) and their relationship to morphological characteristics and environmental temperature in a field and phytotron trial. In the phytotron, sorghum and maize plants ranging from emergence to heading stage, were grown under different day/night temperatures of 30125, 25/20,28/18 and 1818 degree C. Nitrate-nitrogen in sorghum and maize plants was accumulated mainly in stems. Therefore nitrate concentration in the young plants was increased as development of stalks advanced and was highest at the stage of 3-4 leaves, when the plants had a leaf weight ratio 0.78-0.80 g/g plant weight. However, nitrate concentrations of the plant decreased as morphological development progressed, especially from the stage of growing point differentiation. Correlation coefficients showed a positive correlation of nitrate concentration with leaf weight ratio, leaf area ratio and specific leaf area, while plant height, dry matter percentage and absolute growth rate showed a negative association with TEX>$NO_3$-N ($P{\le}0.1$%). Cyanogenic glycosides, total nitrogen and crude protein were close associated with nitrate accumulation, and positively significant ($P{\le}0.1$%). High temperature over 30/25^{\circ}C.$ for 3 weeks increased N-uptake and dry matter accumulation, but reduced nitrate concentration. Under cold temperature below 18/8^{\circ}C.$ concentration of nitrate-N was increased in spite of its limited nitrogen uptake and plant growth.

  • These experiment were conducted to know the seedling growth pattern and to deter­mine the appropriate seeding amount for producing the small and middle sized uniform seedlings of which known to have low bolting in the cultivation of Angelica gigas. Cotyledons of seedling were emerged by 18 days after seeding and the first and the second leaf appeared at 5.0 days and 9.8 days respectively after the anterior leaf emergence. Leaf development from the 3rd to the 6th in order needs about 13 days respectively after the emergence of anterior leaf. Growth of small and middle sized seedling of which required $60{\sim}87days$ were 3.0 to 7.0mm in root head diameter, 6.3 to 10.3cm in root length, 0.4 to 1.3g in fresh root weight per plant with 3.9 to 5.0 leaves. The highest production ratio of small and middle sized seedlings was shown in $15,000seeds/m^2$ of seeding amount as 52.8% respectively.

Optimizing a Method for Measuring Firmness of Chinese Cabbage (Brassica rapa) and Comparing Textural Characteristics among Cultivars (배추 조직감 측정 방법의 최적화와 품종 간 물성특성비교)

  • In order to optimize a method to determine the firmness of Chinese cabbage, hardness of midrib tissues was examined based on their chronological order of emergence. Texture measurement using volodkevich bite jaws gave a consistent and highest regression ($r^2=0.85$) between firmness and the order of leaf emergence, while blade set, cylinder probe, and crisp fracture support rig showed a lower coefficient of determination. Thickness of midrib tissue within an individual head from 16 cultivars of Chinese cabbage was positively correlated with the order of emergence, becoming thinner toward inner leaves. Mean thickness of midrib tissue from the head ranged from 7.74 mm for 'CR-shingshing' and 9.28 mm for 'Norangyeorum'. The covariance of leaf thickness within a head was highly cultivar-dependent, ranging from 23.6% for 'Chihili' and 5.8% for 'Bulam'. Firmness of the midrib tissue, defined as maximum peak height per tissue thickness, became higher from outer to inner leaves, showing $2^{nd}$ order of regression. Mean firmness of the midrib tissue from individual head varied from 1.58 N for 'Rangno' to 3.46 N for 'CR-shingshing'. The $10^{th}$ or $11^{th}$ leaf brought the best correlation coefficient (r = 0.81) between firmness of an individual leaf and the mean firmness of the entire leaves in a head, suggesting a reliable and rapid method to estimate the firmness of a head in lieu of examining all leaves in the head. The relationship between firmness of midrib tissue and dry mass ($r=0.70^{**}$) as well as cell wall content ($r=0.58^*$) of the head were positively correlated. Results obtained from the present study suggested that a new method to determine midrib firmness would enable to clarify the relationship between textural quality of fresh Chinese cabbage and their processed product, 'Kimchi'. It will also be important to apply this method to screen textural quality of various genotypes under breeding programs.

  • Activity of endogenous growth regulator in 4th year Panax ginseng root was investigated by second leaf sheath test of rice seedling and paper chromatogram of a acidic fraction of methanol extract before (March 28) and after (May 9) emergence of root bud, at the late season (Sept.4) and after leaf fall (November 11). GA$_3$ and ABA were used as reference. According to paper and high performance liquid chromatography of samples and authentic growth regulators the presence of insole acetic acid (IAA), gibberellic acid (GA$_3$) and abscisic acid (cis and trans ABA) was confirmed. These three regulators appeared to consist of major system though the existence of other regulators could not be ruled out. IAA activity seemed little changed through out the seasons. GA activity decreased in the later stages while ABA activity increased.

  • A field trial was initiated to examine the range of inter- and intraspecific variations in morphological and phenological traits with five different accessions of smooth and large crabgrass. In addition, a controlled environment study was conducted to determine the phenotypic plasticity among the accessions of both species in response to 4 daily tempera-ture differentials. In the field experiment, significant inter- and intraspecific variations of smooth and large crabgrass were observed in morphological traits such as leaf length and width. However, most phenological traits were not substantially different between the species and among the accessions of each species. The first seedling emerged at the same time, requiring 9~ 10 days, regardless of the accessions and species. In a controlled environment study, all accessions of each species responded similarly to the 4 temperature differentials in seedling emergence, indicating seedling emergence was not a plastic trait. These results suggest that predicting crabgrass seedling emergence could be independent of geographical regions in the US.

  • The study was conducted to know the differential growth responses of soybean cultivars as affected by various application stages of Metribuzin herbicide, at the University experimental farm of Chonnam Nat'l. Univ, Kwangjoo, 1980. Experimented soybean materials were six cultivars, as Kwanggyo, Dongbuktae, Bongui, Yukwoo 3, Suweon 85, and Suweon 86., . and Laid-out application stages were five as pre-sowing, Pre-emergence, Early Post-emergence, and Late Post-emergence treatment comparing with Control. Results obtained from the study were as follows: Among experimented soybean cultivars, no significantly tolerant cultivar was shown. Highly significant differences in phytotoxicities by Metribuzin treatments were detected among various application stages, and crop tolerances were recognized bigger in order as E. Post-emg. < L. Post-emg. $\ll$ Pre-emg. < Pre-sowing $\leq$ Control. The phytotoxic symptoms of all soybean cuitivars were severely recorded in order of Leaf No. /Plant < Plant height $\ll$ Branch No. /Plant. For the cultivation of most soybean cuitivars, the use of Metribuzin in Pre-sowing or Pre-emergence were recommended, and especially it is expected to follow of more detailed study on soil-incorporation method in Pre-sowing treatment, and rather the tank-mixture system of Metribuzin with others than the mono-application system for the safe-use and enlargement of weeding-spectrum.

  • Plant water status during growth is directly and indirectly associated with seed yield. The objective of the present study was to determine the genotypic differences in leaf water characteristics at an early growth stage of soybean [Glycine max (L.) Merrill] plants through the pressure-bomb technique. Measurements of water potential as well as relative water content (RWC) were made at the third leaf from the fully-expanded top leaf of eight different soybean genotypes grown for 31 to 35 days after field emergence. On the basis of the modified exponential model, pressure-volume (PV) curves were fitted well ($R^2$=0.92** to 0.99** for the curvi-linear region and R=0.67** to 0.96** for the linear region), indicating that a segmented model using PROC NLIN of SAS could be used effectively to estimate the leaf water characteristics. The regression analysis for the pressure-volume (PV) curve revealed genotypic variation in the solute potential at saturation (Ψ$_{s,sat}$ :-10.7 to -14.8 bar), solute potential at incipient plasmolysis (Ψ$_{s,ip}$ : -14.3 to -18.3 bar), RWC at incipient plasmolysis (RW $C_{ip}$ : 83.3 to 91.7%), high integrated turgor pressure from saturation to plasmolysis ( $_1$$^{b}$ : 0.39 to 0.81), and maximum volumetric modulus of elasticity ($\varepsilon$$_{max}$ : 150 to 445 bar).).

  • The effects of temperature on transpiration, chlorophyll content, frequency and aperture of stomata, and leaf temperature of Panax ginseng were reviewed. Temperature changes of soil and air under spade roof were also reviewed. Growth responses of responses of ginseng plant at various temperature were assessed in relation to suseptibillity of ginseng plants. Reasonable management of ginseng fields was suggested based on the response of ginseng to various temperatures. Stomata frequency may be increased under high temperature during leaf$.$growing stage. Stomata aperture increased by high temperature but the increase of both frequency and aperture appears not enough for transpiration to overcome high temperature encountered during summer in most fields. Serial high temperature disorder, i.e high leaf temperature, chlorophyll loss, inhibition of photosynthesis, increased respiration and wilting might be alleviated by high humidity and abundant water supply to leaf. High air temperature which limits light transmission rate inside the shade roof, induces high soil temperature(optimum soil temperature 16∼18$^{\circ}C$) and both(especially the latter) are the principal factors to increase alternaria blight, anthracnose, early leaf fall, root rot and high missing rate of plant resulting in poor yield. High temperature disorder was lessen by abundant soil water(optimum 17∼21%) and could be decreased by lowering the content of availability of phosphorus and nitrogen in soil consequently resulting in less activity of microorganisms. Repeated plowing of fields during preparation seems to be effective for sterilization of pathogenic microoganisms by high soil temperature only on surface of soils. Low temperature damage appeared at thowing of soils and emergence stage of ginseng but reports were limited. Most limiting factor of yield appeared as physiological disorder and high pathogen activity due to high temperature during summer(about three months).

Pseudorecombination between Two Distinct Strains of Cucumber mosaic virus Results in Enhancement of Symptom Severity

  • Recently, a Cucumber mosaic virus (CMV) strain, named as CMV-209, was isolated from Glycine soja. In this study, symptom expression of CMV-209 was analyzed in detail in Nicotiana benthamiana by comparing with that of CMV-Fny, which is a representative strain of CMV. Using infectious cDNA clones of CMV strains 209 and Fny, symptom expression of various pseudorecombinants between these two strains were examined in the early and late infection stages. In the early infection stage, the pseudorecombinants containing Fny-RNA2 induced stunting and leaf distortion on the newly emerged leaves whereas the pseudorecombinants containing 209-RNA2 caused no obvious symptoms. In the late infection stage, the pseudorecombinants containing 209-RNA1 and Fny-RNA2 induced severe leaf distortion and stunting, while CMV-209 induced mild symptom and CMV-Fny caused typical mosaic, general stunting, and leaf distortion symptoms, indicating that RNA 2 encodes a symptom determinant(s) of CMV, which is capable of enhancing symptoms. Furthermore, our results support the possibility that natural recombination between compatible viruses can result in emergence of novel viruses causing severe damages in crop fields.
