A Tomato spotted wilt virus (TSWV-KP) was isolated from Paprika (Capsicum annuum var. grossum) showing necrosis spot on the leaves and malformation of the fruit in Yesan, Korea. The virus infected Chenopodium amaranticolor, C. quinoa, Petunia hybrida, Nicotiana glutunosa, Gomphrena globosa, and Physalis floridana. Ten plants including tomato were observed to have systemic TWSV-KP infection. The virus produced necrosis or necrotic ring spots on the inoculated leaves and mosaic, vein necrosis or death on the upper leaves of Datura stramonium, N. clevarandii, N. rustica, and N.tabacum cvs. Thin sections of the infected leaf tissue contained spherical to oval particles, a characteristic of a Tospovirus. The virion contained three molecules of genomic RNAs, which were approximately 9.0, 4.9 and 3.0 kb. The nucleocapsid (N) protein of the purified virion migrated as a single band with molecular weight of about 29 kDa in SDS-PAGE. The N gene of TSWV-KP showed 96.5-97.2% and 97.7-98.5% identities to the three different TSWV isolates of Genbank Database at the nucleotide and amino acid, respectively.
In this study, we report a unrecorded plant on the Korean peninsula, Potamogeton × anguillanus Koidz. from Yeondangcheon (stream) in Yeondang-ri, Nam-myeon, Yeongwol-gun, Gangwon-do province. Potamogeton × anguillanus is a natural hybrid between P. wrightii Morong. and P. perfoliatus L. It is distinguished by its characteristic that the petiole is shorter than P. wrightii and that the leaf blade is narrower and longer than P. perfoliatus also, it didn't fully wrap the stem. Considering that P. × anguillanus is a natural hybrid, the name is given to it as 'Eol-chi-gi-dae-ga-rae'. We provide descriptions of the morphological characteristics, photographs, and key to allied taxa in Korea of P. × anguillanus.
The essential oils were isolated by hydrodistillation and their composition determined capillary GC method with standards. The essential oil content showed significant differences between the two populations on the vegetative organs. The essential oil level of the leaves and roots was considerably higher in the Korean population at full flowering and waxy ripening stage but essential oil content of the roots was significantly higher in the Hungarian taxon at leaf rosette stage. We observed the essential oil accumulation tendency was mianly dependent on plant organs and intra-specific taxon during the vegeation period. Butylidene-phthalide was proved to be the main component of the oil in both population roots (50.9-73.3%), while dimethyl-acetate was showed as a major compound on the over-ground parts (56.7-62.0%). The qualitative composition of the essential oil in the reproductive organs concerning the identified compounds was the same as the vegetative parts with the main component ${\alpha}-phellandrene$ (4.8-28.1%) and butylidene-phtalide (9.7-16.1%), The quantitative composition showed some changes during the ontogenesis phases. Most characteristic ones are the decreasing proportion of dimethyl-acetate (from 7.3% to 1.1%) and the appearance of ${\alpha}-pinene$ (from 0.5% to 1.5%) only after fruit setting in both population.

After long exposure to low-density $SO_2$ the five tree species showed different changes in their visible damage: Ailanthus altissima showed no visible damage; Acanthopanax sessiliflorus, Populus alba ${\times}$ Populus glandulosa, and Platanus orientalis showed typical damage of yellow spots on their leaves; and Liriodendron tulipifera showed serious damage on the end of leaf tip. The photosynthesis rate of Liriodendron tulipifera and Acanthopanax sessiliflorus was usually lower than the control group: Ailanthus altissima, Populus alba ${\times}$ Populus glandulosa, and Platanus orientalis showed no difference from the control group, while Acanthopanax sessiliflorus and Liriodendron tulipifera showed large changes. In regard to the light compensation points, Ailanthus altissima showed similar values, Populus alba ${\times}$ Populus glandulosa and Platanus orientalis exposed to $SO_2$, showed 3 to $5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ higher values than the control group, and Acanthopanax sessiliflorus and Liriodendron tulipifera exposed to $SO_2$ showed approximately twice higher light compensation points.
Recently the word "digital" is widely used in almost every field and is dominating this generation. Digital has become the most significant characteristic representing the 21st century, and is leading change across wide range of our life-styles in our culture and thoughts. New art is in harmony with digital in the 21st century. Digital photography is simpler, faster and newer than the analog system of the past. From ancient times, the nature has been the subject of art, and many designers have studied the ways to create beauty from nature. In this study, I chose the lotus as the subject material of textile design development. The lotus invokes a sense of stillness, and nestles many fluid elements, including the curved fluid lotus, rhythmic lotus petal, sinuous lotus leaf, radial vein, lotus pip and oval seed. Therefore, I tried to use these elements of lotus for development of textile design. For this purpose, I photographed the lotus with a digital camera as equipment of design development. Then, on computer, I have developed six textile designs through the process of modification and editing by using the adobe illustrator program.
Lily symptomless virus (LSV), Lily mottle virus (LMoV), and Cucumber mosaic virus (CMV) are the most prevalent viruses infecting lilies in Korea. Leaf and bulb samples showing characteristic symptoms of virus infection were collected in 2012, and 80 field samples were analyzed by reverse transcription polymerase chain reaction (RT-PCR). The infection frequencies were 79% for LMoV, 5% for LSV, and 3% for CMV. The LMoV coat protein gene was amplified and cloned into the pET21d(+) expression vector to develop serological diagnostic tools to detect LMoV. The resulting carboxy-terminal His-tagged coat proteins were expressed in Escherichia coli strain BL21 (DE3) by induction with IPTG. The recombinant proteins were purified using Ni-NTA agarose beads and used as an antigen to produce polyclonal antibodies in laying hens. The resulting egg yolk immunoglobulin (IgY) specifically recognized LMoV from infected plant tissues in immunoblotting assays and had comparable sensitivity to that of a mammalian antibody. In addition, method of immunocapture RT-PCR using this IgY was developed for sensitive, efficient, and rapid detection of LMoV. Based on these results, large-scale bulb tests and detection of LMoV in epidemiological studies can be performed routinely using this IgY. This is the first report of production of a polyclonal IgY against a plant virus and its use for diagnosis.
Excessive soil water at vegetative growth stages during the rainy season induces yield losses in soybeans. Our objectives were to obtain basic information about the cultivar differences and to understand the stress-tolerance process for due to excessive soil water. Previous experiments revealed soybean genotypic differences in tolerance to excessive soil water. A field experiment was conducted at the Research Farm of Korea University near Seoul on 21 May 1998. Soybean[Glycine max (L.) Merrill] cultivars, 'Hannamkong' (sensitive) and 'Taekwan-gkong'(tolerant) were planted in vinyl-lined plots(1.2 x 4.2 x 0.3 m deep) and control plots. Drip irrigation began at VI growth stage to submerge the soil surface. Three weeks of excessive soil water treatment reduced all growth parameters measured to soybean plants. Excessive soil water stress resulted in decreases of N, P, K, Ca, Mg and Cu, and increases of Fe and Mn contents in soybean leaves. The stress index of tolerant cultivars under excessive soil water showed no large difference in soybean growth characteristics measured at three growth stages. However, K, Ca, Mg, Fe and Mn contents in soybean leaves appeared to differ between sensitive and tolerant cultivars. From the above results, stress and tolerance indices are proposed for a method to test cultivar differences in plant responses within a species under adverse growth environments.
The surface ultrastructure of Acanthotrema felis (Trematoda: Heterophyidae) adults, recovered from a kitten experimentally infected with the metacercariae, was observed using a scanning electron microscope. The worm was leaf-like, ventrally concave and covered with scale-like multi-pointed tegumental spines. The spines on the anterior surface were short but broad, and had 10-12 pointed tips. The cytoplasmic processes protruded around the spines, like pockets for the spines. The ventrogenital opening was crescent, or kidney-shaped, and had protuberances with minute spines on its surrounding tegument. The spines on the posterior surface were long, but narrow, with 6-8 pointed tips. The cytoplasmic processes on this tegument were ridge-like, and elevated along the row of the spines. The surface ultrastructure of A. felis is generally similar to that of other heterophyid flukes, but some features are characteristic, and may be of taxonomic and bio-ecological significance.
This study was conducted to develop new lines expressing the characteristic of early flowering by introducing MdMADS2 gene in chrysanthemum (Dendranthema grandiflorum (Ramat.) Kitamura) ‘Zinba'. Transformation of chrysanthemum was conducted by Agrobacterium tumefaciens LBA4404 harboring the binary vector containing MdMADS2 controlled by double CaMV 35S promoters. Ninety three shoots were regenerated from 1,463 leaf segment explants cultured on the first selection medium (MS basal salts + 1.0 mg/L BA + 0.5 mg/L IAA + 10 mg/L kanamycin + 400 mg/L cefotaxime, pH 5.8) after co-cultivation, and 20 out of the 93 shoots rooted on the second selection medium containing 20 mg/L kanamycin and 400 mg/L cefotaxime. Many escapes (98.6%) were removed on the selection stage for rooting. Nineteen lines were confirmed as transgenic plant with transgene by PCR analysis. Six transgenic plants flowered 2-11 days earlier than non-transgenic plant without big change of phenotype, and especially, 3 (Mo-7, Mo-11, Mo-17) out of 6 transgenic lines showed a significant reduction in days to flowering compared to non-transgenic plant. Introduction and expression of MdMADS2 gene in them were confirmed by Southern and real-time PCR analyses, respectively.

Multiple viruses such as Lily symptomless virus (LSV), Lily mottle virus (LMoV), and cucumber mosaic virus (CMV) are the most prevalent viruses infecting lilies in Korea. Leaf samples and bulbs showing characteristic symptoms of virus infection were collected from Gangwon, Chungnam, and Jeju provinces of Korea in 2008-2011. Coat protein (CP) genes of LSV and LMoV were amplified from collected samples by reverse transcription-polymerase chain reaction (RT-PCR) and cloned into a pET21d(+) expression vector to generate recombinant CPs. The resulting carboxy-terminal His-tagged CPs were expressed in Escherichia coli strain BL21(DE3) by isopropyl-1-thio-${\beta}$-D-galactoside induction. The recombinant proteins were purified using Ni-NTA agarose beads, and the purified proteins were used as an immunogen to produce polyclonal antibodies in rabbits. The resulting polyclonal antisera recognized specifically LSV and LMoV from infected plant tissues in Western blotting assays. Indirect enzymelinked immunosorbent assay and immunocapture RT-PCR using these polyclonal antisera were developed for the sensitive, efficient, economic, and rapid detection of Lily viruses. These results suggest that large-scale bulb tests and economic detection of Lily viruses in epidemiological studies can be performed routinely using these polyclonal antisera.
