Physiological Effects of TOPE, a Photo - independent Diphenylether, on Higher Plants (비광요구형 디페닐에테르계 TOPE 의 생리적 작용에 관한 연구)

  • Several physiological responses were investigated in plants treated with TOPE as a preliminary step to know its action site. Unlike photo-dependent diphenylethers, herbicidal activity of TOPE appeared slowly and its typical symptoms were both burning of leaf blades and abnormal division of meristem in grasses, Similarly, both leakage of cell electrolytes and the curling of cotyledon margin were also shown in cucumber(Cucumis sativus L.). Biosynthesis of chlorophyll in etiolated cucumber cotyledon was not inhibited directly by treatment of TOPE at low light intensity(5.5${\mu}$ mol $m^{-2}s^{-1}$ PAR) and protoporphyrin IX was not also accumulated. The contents of phytoene, phytofluene and ${\beta}$-carotene were abnormaly increased. Photosynthesis was inhibited only at high concentration. Mitochondrial respiration was inhibited at high concentration but rather increased significantly at 10${\mu}$M of TOPE. However, respiration inhibitors did not alleviate the two symptoms of TOPE in cucumber cotyledon. In the same experiments, using inhibitors of protein or nucleic acid biosynthesis, only one of the two symptoms was alleviated by chloramphenicol and cycloheximide. In contrast, both symptoms were alleviated by actinomycin-D and hydroxyurea, suggesting that nucleic acid metabolism might be preferentially related to the mode of action of TOPE. DNA, RNA and protein contents were accumulated in both cucumber cotyledon and rice (Oryza sativa L.) routs treated with TOPE, and the DNA of them was increased at first. Thus, it is conjectured that TOPE increase nucleic acid metabolism directly or indirectly, and then disturb various metabolic pathways causing abnormal physiological and morphological effects followed by final death.

Effects of NaCl on the Growth and Physiological Characteristics of Crepidiastrum sonchifolium (Maxim.) Pak & Kawano (NaCl 처리가 고들빼기의 생장과 생리적 특성에 미치는 영향)

  • Background: This study was conducted to investigate the effects of NaCl concentration on the photosynthetic parameters, chlorophyll fluorescence and growth characteristics of Crepidiastrum sonchifolium. Methods and Results: As treatments, we subjected C. sonchifolium plants to four different concentrations of NaCl (0, 50, 100 and 200 mM). We found that the photosynthetic parameters maximum photosynthesis rate (PN max), net apparent quantum yield (Φ), maximum carboxylation rate (Vcmax), and maximum electron transport rate (Jmax) were significantly reduced at an NaCl concentration greater than 100 mM. In contrast, there was an increase in water-use efficiency with increasing NaCl concentration, although in terms of growth performances, leaf dry weight, root dry weight, stem length, and total dry weight all decreased with increasing NaCl concentration. Furthermore, leakage of electrolytes, as a consequence of cell membrane damage, clearly increased in response to an increase in NaCl concentration. Analysis of the polyphasic elevation of chlorophyll a fluorescence transients (OKJIP) revealed marked decrease in flux ratios (ΦPO, ΨO and ΦEO) and the PIabs, performance index in response to treatment with 200 mM NaCl, thereby reflectings the relatively reduced state of photosystem II. This increase in fluorescence could be due to a reduction in electron transport beyond Q-A. We thus found that the photosynthetic parameters, chlorophyll fluorescence and growth characteristics of C. sonchifolium significantly increased in response to treatment with 200 mM NaCl. Conclusions: Collectively, the findings of this study indicate that C. sonchifolium shows relatively low sensitivity to NaCl stress, although photosynthetic activity was markedly reduced in plants exposed to 200 mM NaCl.

Plant regeneration from protoplasts-derived from embryogenic callus of Citrus (감귤 embryogenic callus 원형질체 배양에 의한 식물체 재분화)

  • This study describes conditions for plant regeneration from protoplasts-derived from embryogenic callus of satsuma mandarin. Plants were generated via somatic embryogenesis. Protoplasts isolated directly from nucellar callus induced from immature ovule of satsuma mandarin cv. Okitsu (Citrus unshiu Marc.) were cultured in 0.6M $BH_3$ medium. Cell division and plating efficiency were affected by protoplast culture method. The liquid over solid method was the most effective for formation of microcalli. Most of microcalli grew rapidly and transferred onto embryoid formation medium. Optimum embryoid formation medium was MT medium containing 1.5 g/L malt extract, 0.146 M sucrose and the medium for plantlet regeneration was MS medium containing 0.09M sucrose, 1.0 mg/L $GA_3$. No differences were noticed in growth habits and leaf characters such as shape, thickness, and colour between protoplast-derived plants and nucellar seedlings. This plant regeneration system from protoplasts-derived from embryogenic callus provides an alternative way for producing new scion and rootstock cultivar from citrus species which can not be crossed.

Mutagenesis of Nodulation and Other Growth Characters with Ethyl Methanesulfonate in Soybean (EMS 처리에 관한 콩 근류착생 및 기타 형질의 돌연변이 출현 양상)

  • The aims of the work were to examme the variability induced by EMS (ethyl methanesulfonate) mutagenesis of soybean plants, and to isolate mutants altered in nodulation and other growth characters. Seeds of two soybean cultivars, ‘Hwanggeumkong’ and ‘Baegunkong’ were treated with 30 and 50mM EMS(pH 7.0) for 6 hours and were planted directly in the field. Field emergency of$M_1 seed was averaged to be 61.0%, and frequency of plants with chlorophyll-deficient sectors of the first trifoliolate is about 0.7%. Regardless of varieties and does of EMS, $M_1 plant injury at harvest was present in plant height, pod and seed number per plant when compared to those of original-type soybean plants. The $M_2 variability of nodulation process induced by EMS treatment was found to be narrower than that of shoot dry weight. On the basis of the occurrence of chlorophyll-deficient plants, mutated cell frequency within $M_1 seed ranged from 5.3% to 84.2%, suggesting that mutation frequency on the $M_1 seed induced by EMS occurred partly and randomly regardless of varieties and doses of EMS. The putative mutant, which had more nodulation than original-type plant, was short in plant height. Sparse-nodulating soybean mutant was lower in leaf chlorophyll content and showed reduced growth.

Effects of Diniconazole Application on Anatomical and Biochemical Characteristics Related to Stress Tolerance in Lilum davuricum (날개하늘나리에 있어서 디니코나졸 처리가 스트레스 내성과 관련된 해부학적 및 생화학적 특성에 미치는 영향)

  • This study was carried out to examine the effects of foliar-sprayed diniconazole on the morphological characteristics and the contents of polyamines (PA) and proline, and to investigate their relationship with stress tolerance in Lilium davuricum native to Korea. Plants with 5 cm mean height were sprayed with $50mg{\cdot}L^{-1}$ diniconazole or distilled water (control) and grown in a greenhouse maintained at 23/$18^{\circ}C$ (day/night) for 30 days. Diniconazole decreased plant height, leaf length, diameter and length of the pith and cortical cell of the stem, while it increased the thickness and epicuticular wax of leaves measured at 30 days after treatment. In polyamines contents, diniconazole increased spermidine content at 5-10 days after treatment and spermine content after the 10th day of treatment as compared with the control. Diniconazole decreased proline content after the 10th day of treatment as compared with the control. From these results, it was concluded that foliar-sprayed diniconazole might morphologically and biochemically improve the stress tolerance of this plant species.

Pyracantha Extract Acts as an Antioxidant Agent to Support Porcine Parthenogenetic Embryo Development In Vitro (돼지 단위 발생 난자의 체외 발달에 있어서 피라칸타 추출액의 처리 효과)

  • Pyracantha is a genus of thorny evergreen large shrubs in the family of Rosaceae, with common names Firethorn or Pyracantha. It's extract has also been used in cosmetics as a skin-whitening agent and functioning through tyrosinase inhibition. Recent studies have shown that pyracantha extract possesses antioxidant activities and may significantly improve lipoprotein metabolism in rats. Although the mode of action of Pyracantha extract is not fully understood, a strong relationship was observed between antioxidant and apoptosis in some types of cells. Thus, the aim of this study was to evaluated the effect of pyracantha extract on blastocysts formation and their quality of the porcine parthenogenetic embryos. After parthenogenetic activation by chemicals, presumptive porcine parthenogenetic embryos were cultured in PZM-3 medium supplemented with extracts of pyracantha leaf, stalk and root for 6 day (1, 5 and $10{\mu}g/ml$, respectively). In our results, the frequency of blastocyst formation in pyracantha root extract ($5{\mu}g/ml$) treated group had increased that of other groups. Furthermore, blastocysts derived from pyracantha root extract ($5{\mu}g/ml$) treated group had increased the total cell numbers and reduced apoptotic index. Blastocyst development was significantly improved in the pyracantha root extract ($5{\mu}g/ml$) treated group when compared with the $H_2O_2$ treated group (p<0.05). Subsequent evaluation of the intracellular levels of ROS in pyracantha root extract ($5{\mu}g/ml$) treated groups under $H_2O_2$ induced oxidative stress were decreased (p<0.05). In conclusion, our results indicate that treatment of pyracantha root extract may improve in vitro development of porcine parthenogenetic embryos through its antioxidative and antiapoptotic effects.

First Report of Stem Rot in Statice Caused by Rhizoctonia solani in Korea (Rhizoctonia solani에 의한 스타티스 줄기썩음병)

  • Stem rot of perennial statice (Limonium sinuatum) was observed in Un bong, Jeonbuk from 2006 to 2007. Affected plants were randomly distributed in the greenhouses and infection rate was more than 10%. Stem and leaf of statice at soil line were dried and turned brown, initially. As the disease became severe, other stem parts and crowns were turned dark brown and then sunken. The fungal isolates were showed initially white aerial mycelium and turned brown with age. They produced few sclerotia which small, irregularly shaped with pinpoint sized. Mycelia were branched at $90^{\circ}$ angles and multinucleate in one cell. The pathogenicity of causal organism was proved according to Koch's postulates. The causal fungus of stem rot was identified as Rhizoctonia solani based on the cultural and morphological characteristics. This is the first report on stem rot of statice by R. solani in Korea.

Seasonal Changes of Water Relations Parameters of the Korean Mistletoe (Viscum album var. coloratum) Leaves (겨우살이 엽의 계절별 수분특성)

  • This study was conducted to study the seasonal changes of relative water relations parameters by pressure-volume curves of Korean mistletoe (Viscum album var. coloratum) leaves for understanding ecological characteristics. In growing season, the osmotic potentials at full turgor (${\Psi}_o^{sat}$) and at incipient plasmolysis (${\Psi}_o^{tlp}$) decreased, while increased the maximum bulk elastic modulus of the cell wall ($E_{max}$) and relavive water content ($RWC^{tlp}$). Korean mistletoe in Quercus variabilis and Korean mistletoe in Quercus mongolica in November showed best maximum perssure potential (${\Psi}_{P,\;max}$). Pressure potential (${\Psi}_P$) and water potential (${\Psi}_L$) in Korean mistletoe in Quercus variabilis were rapidly decreased with decreasing of relative water content. The values of $RWC^{tlp}$ in November were all above 84% showing that the function of osmoregulation is somewhat better, and symplastic water content (Vo/DW) and maximum water content (Vt/DW) were variable seasonally. Thus, responses to water relations of Korean mistletoe in Quercus variabilis and Korean mistletoe in Quercus mongolica such as ${\Psi}_o^{sat},\;{\Psi}_o^{tlp},\;E_{max},\;{\Psi}_{P,\;max},\;RWC^{tlp}$ showed increaing drought tolerance with increasing of leaf aging.

Growth Suppression of Tomato Plug Seedlings as Affected by Material Type for Brushing Stimulation (브러싱 소재에 따른 토마토 공정묘의 생장억제)

  • This study was conducted to find proper material of brushing stimulation for height suppression of tomato plug seedlings. The tomato seeds were sown in 40-cell plug tray filled with commercial seedling medium and brushing stimulation was started at 18 days after sowing. Acrylic, polypropylene film, and weaving film were used by materials of brushing stimulation and, non-treatment and diniconazole treatment were used as the control. In acrylic treatment, the plant height was the shortest and the stem diameter was the thickest. Leaf growth was the lowest in diniconazole treatment. However, the SPAD value was the greatest in diniconazole treatment. The dwarf rate was the greatest in acrylic treatment. In acrylic treatment, the T/R ratio was the lowest and compactness was the greatest. In conclusion, use of the acrylic as material for brushing stimulation has higher dwarf rate than diniconazole treatment, and has advantages height suppression and seedling quality.

Effect of Cold Acclimation and Deacclimation on the Freezing Tolerance, Total RNA, Soluble Protein and Soluble Sugar in Chinese Cabbage (저온순화 및 탈순화가 배추의 내동성 및 total RNA, soluble protein, soluble sugar 함량에 미치는 영향)

  • This study was conducted to determine the change in freezing tolerance of Chinese cabbage (Brassica campestris L. ssp. penkinensis). Four-week old seedlings grown in a greenhouse at $25^{\circ}C$$\pm$0.5 were transferred to 5, 10 or 15$^{\circ}C$ condition for acclimation for one day, and then transferred to a lethal temperature of -3$^{\circ}C$ condition. Optimal acclimation temperatures were 5$^{\circ}C$ and 1$0^{\circ}C$. Freezing tolerance of leaf tissues was investigated during a week-long cold stress. Development of freezing tolerance was shown by survival ratio, and amounts of total RNA, soluble protein and soluble sugar. LT$_{50}$ value increased from -3 to -4$^{\circ}C$ after 5 days acclimation at 5 or 1$0^{\circ}C$, and this was considered to be the first indication of enhanced freezing tolerance. Plants, cold acclimated for five days, reached to a survival ratio of 60%. This increase in freezing tolerance was found to be associated with the increased levels of total RNA, soluble sugar and soluble protein. These metabolic changes imply the association of adjustment of growth and cell metabolism with low temperatures at the beginning of cold acclimation in chinese cabbage.e.
