납 중독 랫드의 말초신경내 myo-inositol 수송 체계에 관한 연구 (A study on myo-inositol transport system in peripheral nerve isolated from lead-intoxicated rat.)

  • In our previous studies, we reported that lead intoxicated nerve cell by inhibition of the Na$^{+}$-K$^{+}$ ATPase activity and reduction of myo-inositol in nerve cell. As the second series of experiments, in order to understand toxic mechanism of lead for nerve cell, the characteristics of myo-inositol transport system and the effect of lead on its system have been studied in the sciatic nerves of control and lead-treated rats. A lead intoxicated animal model was induced by feeding diet containing lead to Sprague-Dawley rat for two weeks. Four weeks aged Sprague-Dawley rats were divided into three group : normal control group, 10ppm-lead treated group, 100ppm-lead treated group. All rats were sacrified at the end of two weeks. The rate o myo-inositol transport by sciatic nerve isolated from lead-treated rat was significantly decreased compared with that of control rat. This deficit results from that myo-inositol transport system which is carrier mediated and sodium-potassium dependent was inhibited by the lead treatment (both 10ppm and 100ppm) due to increase of the Km value without affecting Vmax value for myo-inositol carrier. These observations suggest that the toxic mechanism of lead on nerve myo-inositol transport system might be a change of affinity without change of maximum transport velocity for carrier.

  • Nramp2, also known as DMT1 and DCT1, is a 12-transmembrane domain protein responsible for dietary iron uptake as well as metal ions such as lead, manganese, zinc, copper, nickel, cadmium, and cobalt. High expression of DMT1 increase lead uptake, and DMT1-dependent lead transport was H -dependent and inhibited by iron ions. The molecular mechanism of lead transport in CNS is as yet unknown. although interactions between iron and lead at the level of absorption have been known for some time. The process of lead uptake into astrocytes was not known yet. Nramp2 may mediate transport of heavy metal into astrocytes. We investigated whether Nramp2 mediate transport of lead into astrocytes. And we do whether Nramp2 was expressed highly by deprivation of iron in Astrocytes, and lead uptake into astrocytes was influenced by expression of Nramp2. Immortalized human fetal astrocyte(SV-FHA) cells were cultured in medium containing Dulbecco's modified Eagle's medium and treated with Deferoxamine. Northern blot analysis was done for determining mRNA level of DMT1 and lead uptake assay was done in incubation condition of pH 5.5 and 7.4.

  • A chloroform membrane system containing a given mixture of benzylaza-12-crown-4 and oleic acid is introduced for the selective and efficient transport of $Pb^{2+}$ ion. The transport was capable of moving metal ions ‘up-hill’. In the presence of ${S_2O_3}^{2-}$ion as a suitable metal ion acceptor in the receiving phase, the amount of lead ion transport across the liquid membrane after 150 minutes is (95.0 ${\pm}$ 1.7)%. The selectivity and efficiency of lead transport from aqueous solution containing $Tl^+,\;Ag^+,\;Mg^{2+},\;Ca^{2+},\;Co^{2+},$ $Ni^{2+},\;Cu^{2+},\;Zn^{2+},\;Hg^{2+}$ and $Cd^{2+}$were investigated. In the presence of thiosulfate as a suitable masking agent in the source phase, the interfering effects of $Ag^+\;and\;Cu^{2+}$ were diminished drastically.

  • We have fabricated and tested a Bi-based high temperature superconducting current lead system. Ag sheathed Bi-2223 mono-filament tapes of $I_c=8.4$ A at 77 K under self-field condition were fabricated using powder-in-tube(P1T) method. Multi-layer current leads can be made by stacking of Ag sheathed Bi-2223 mono-filament wires. The critical current of this 10-layer current lead is about 68 A. The contact resistance across the copper-current lead interface has been studied using current-voltage characteristics. At temperature below critical temperature the resistive contribution of the interface to the total contact resistance dominates. We have measured AC transport losses in a current lead at 77 K, 60 Hz by a transport method.

  • Lead and cadmium are potent environmental toxicants that affect populations living in Europe, Americas, and Asia. Identifying transporters for lead and cadmium could potentially 1 help us better understand possible risk factors. The iron transporter divalent metal transporter 1(DMT1) mediates intestinal transport of cadmium, and lead in yeast and fibroblasts overexpressing DMT1. In human intestinal cells knocking down expression of DMT1 attenuated uptake of cadmium and iron but not lead. A possible explanation is the expression of a second transporter for lead in intestine. In astrocytes, however, DMT1 appears to transport lead in an extracellular buffer at pH value. At neutral pH, transport was not mediated by DMT1 but rather by a transporter that is stimulated by bicarbonate and inhibited by 4,4'-diisothiocyanatodihydrostilbene-2,2'-disulfonic acid. The identity of this lead transporter is under study.

  • As the risk of the various external risk was increased, a study on the integrity assessment of the nuclear transport cask was needed. In this paper, an integrity assessment of the nuclear transport cask under the ballistic impact was studied. The projectile with L/D = 5 was used in simulation. The applied head shapes of the projectile were five types such as flat shape, conical shape, hemispherical shape, truncated conical and sliced flat shape, respectively. The range on the velocity of the projectile was 85 m/s to 680 m/s. The cask body of the nuclear transport cask was not penetrated by the projectile speed up to Vprojectile = 510 m/s. As the cask body was penetrated by the all types projectile with Vprojectile = 680 m/s and the cask lead in the nuclear transport cask was collided with the projectile. As the projectile moved to 31.3 mm in the cask lead, the cask lead was not penetrated by the projectile with Vprojectile = 680 m/s. The integrity assessment on the nuclear transport cask under ballistic impact up to Vprojectile = 680 m/s was obtained.

납(Pb)이 생쥐 신장세포에 미토콘드리아 미세구조 및 전자전달계에 미치는 영향 (Effects of Lead on the Ultrastructure ana the Electron Transport System of Mitochondria of Mouse Kidney)

  • To investigate the effects of lead on the electron transport system and ultrastructure of mouse kidney mitochondria, various lead acetate concentrations were treated in vitro and respiration rate, enzyme activities were measured. Ultrastructural changes at state IV respiration were also observed. To compare with in vivo experiments, mouse were injected intraperitoneally of 100 mg lead acetate per kg body weight and state IV respiration rate and enzyme activities were measured. Ultrastructure of renal proximal tubular cells were also observed. In in vitro treatement, decreased state IV respiration, decreased enzyme activities, ruptured membranes and inhibition of condensed to orthodox transformation were observed. In in vivo treatment, decreased state IV respiration and decreased enzyme activities were observed after 24 hrs of i.p. injection. Cytochrome c oxidase activity showed twice the inhibition compared to NADH-CoQ reductase activity at 24 hrs. Continuous decreased state IV respiration was observed after 48 and 72 hrs of injection, however, the enzyme activities were increased to control level. Lead-protein complex which probably inhibits the toxic effects of lead appeared. To conclude, dominant effect of lead on the electron transport system appeared at cytochrome c oxidase activity, and the increased enzyme activities may be a result of appearance of lead-protein complex.

Development and Assessment of a Dynamic Fate and Transport Model for Lead in Multi-media Environment

  • The main objective was to develop and assess a dynamic fate and transport model for lead in air, soil, sediment, water and vegetation. Daejeon was chosen as the study area for its relatively high contamination and emission levels. The model was assessed by comparing model predictions with measured concentrations in multi-media and atmospheric deposition flux. Given a lead concentration in air, the model could predict the concentrations in water and soil within a factor of five. Sensitivity analysis indicated that effective compartment volumes, rain intensity, scavenging ratio, run off, and foliar uptake were critical to accurate model prediction. Important implications include that restriction of air emission may be necessary in the future to protect the soil quality objective as the contamination level in soil is predicted to steadily increase at the present emission level and that direct discharge of lead into the water body was insignificant as compared to atmospheric deposition fluxes. The results strongly indicated that atmospheric emission governs the quality of the whole environment. Use of the model developed in this study would provide quantitative and integrated understanding of the cross-media characteristics and assessment of the relationships of the contamination levels among the multi-media environment.

Validation of the neutron lead transport for fusion applications

  • Lead is an important material, both for fusion or fission reactors. The cross sections of natural lead should be validated because lead is a main component of lithium-lead modules suggested for fusion power plants and it directly affects the crucial variable, tritium breeding ratio. The presented study discusses a validation of the lead transport libraries by dint of the activation of carefully selected activation samples. The high emission standard 252Cf neutron source was used as a neutron source for the presented validation experiment. In the irradiation setup, the samples were placed behind 5 and 10 cm of the lead material. Samples were measured using a gamma spectrometry to infer the reaction rate and compared with MCNP6 calculations using ENDF/B-VIII.0 lead cross sections. The experiment used validated IRDFF-II dosimetric reactions to validate lead cross sections, namely 197Au(n, 2n)196Au, 58Ni(n,p)58Co, 93Nb(n, 2n)92mNb, 115In(n,n')115mIn, 115In(n,γ)116mIn, 197Au(n,γ)198Au and 63Cu(n,γ)64Cu reactions. The threshold reactions agree reasonably with calculations; however, the experimental data suggests a higher thermal neutron flux behind lead bricks. The paper also suggests 252Cf isotropic source as a valuable tool for validation of some cross-sections important for fusion applications, i.e. reactions on structural materials, e.g. Cu, Pb, etc.
