Trigeminovascular system plays an important role for the cerebral memodynamics. The aim of this study was to investigate the alterations in cerebrovascular reactivity by trigeminovascular system injury in rats. Trigeminovascular system of male Sprague-Dawley rats was injured by either denervation of nasocilliary nerve or neonatal capsaicin treatment. Trigeminovascular system was stimulated by controlled hemorrhagic hypotension or somatosensory (whisker) stimulation. Changes in regional cerebral blood flow (rCBF) and pial arterial diameter were continuously measured by laser-Doppler flowmetry and videomicroscopy, respectively. Nitric oxide synthase (NOS) activity in cerebral cortex was determined by measuring the conversion of $L-^3H-arginine\;to\;L-^3H-citrulline$. Cyclic GMP levels in cerebral cortex and pial artery were determined using the cyclic GMP $^{125}I$ scintillation proximity assay system. rCBF autoregulation was impaired or almost abolished by trigeminovascular system injury. rCBF response to whisker stimulation was significantly attenuated by trigeminovascular system injury. NOS activity as well as cyclic GMP level in cerebral cortex and pial artery were significantly reduced in the group of trigeminovascular system injury. These results suggest that trigeminovascular system injury causes prominent alterations in cerebrovascular reactivity, and that NO, which is generated by neuronal NOS in the trigeminovascular system, is implicated in the regulation of rCBF.
The rodent model has been used frequently to understand stroke pathophysiology, due to its low cost and the large spectrum of genetic strains available. Here, we present a diffuse speckle contrast analysis system (DSCA) with a $1{\times}2$ optical switch that was used to non-invasively assess cerebral blood flow (CBF) changes in the rat during intraluminal suturing for middle cerebral artery occlusion (MCAO) surgery. The blood flow index (BFI) in the left hemisphere was lower than that in the right hemisphere because the left middle cerebral artery was occluded. Furthermore, the performance of the DSCA system was compared with that of commercial laser Doppler flowmetry. The changes in the BFI measured by the two systems were correlated strongly. The DSCA system was less sensitive to motion artifacts and able to measure relatively deep tissue flow in the rat's brain. In conclusion, the DSCA system secured CBF monitoring during surgery in a rodent model without craniotomy.

Scutellaria Radix, originated from Scutellaria baicalensis Georgi, is one of the most important medicine in traditional Oriental medicine, and possesses anti-bacterial activity and sedative effects, can be applied in the treatment of a range of conditions including diarrhea and hepatitis. It is reported that chronic global ischemia induces neuronal damage in selective, vulnerable regions of the brain, especially the hippocampus and cerebral cortex. In the present study, to investigate the effect of Scutellaria Radix extract on cerebral disease, the changes of regional cerebral blood flow and pial arterial diameter on ischemia/reperfusion state was determinated by Laser-Doppler Flowmetry and some parameters concerned with oxidative stress also measured. When SRe were administered for five days with the concentration of 100 mg/kg, GSH activity significantly increased. But SRe administeration showed no significant change in lipid peroxidation. When the activities of CAT, Cu, Zn-SOD and GSH were measured, CAT and GSH were activated by SRe administration. When 1 and 3 ㎍/㎖ SRe was applied to the neuronal cell cultures, the quantities of LDH was significantly reduced when compared with cultures treated only with NMDA. Through this study, it can be concluded that the ischemia/reperfusion induced brain stress may have contributed to cerebral damage in rats, and the present study provides clear evidence for the beneficial effect of SRe on ischemia induced brain injury.
1. Purpose : For many years, Soumin Seonghyangjeongkisan(少陰人 星香正氣散, SJ) has been used in Sasang(四象) constitutional medicine as a therapeutic agent for cerebral disease. The effects of SJ on the vascular system is not well-know. The purpose of this study is to make it clear the effects of SJ and composing drugs on blood pressure(BP) and regional cerebral blood flow(rCBF). 2. Method : We measured BP with Pressure Transducer, rCBF with Laser-Doppler flowmeter. Propranolo and methylene blue were used to determine the mechanisms of SJ effects. 3. Results and Conclusion : BP was not affected by SJ in rats, and rCBF was significantly increased by SJ in rats. rCBF was increased by Folium Perillae(蘇葉), Rhizoma Atractylodis(蒼朮), Pericarpium Arecae(大腹皮) and Rhizoma Arisaematic(南星), but decreased by Radix Saussurea(木香) in rats. The increase of rCBF is mediated by adrenergic ${\beta}-receptor$ and guanylyl cyclase which is enzyme producing cyclic GMP.
1. Purpose For many years, Taeumin Chungsimyoinjatang(太陰人 淸心蓮子湯, CS) has been used in Sasang(四象) constitutional medicine as a therapeutic agent for cerebral disease. The effects of CS on the vascular system is not well-known. The purpose of this study is to make it clear the effects of SJ and composing drugs on blood pressure(BP) and regional cerebral blood flow(rCBF). 2. Method We measured BP with Pressure Transducer, rCBF with Laser-Doppler flowmeter. Propranolol and methylene blue were used to determine the mechanisms of CS effects. 3. Results and Conclusion BP was not affected by CS in rats, and rCBF was significantly increased by CS in rats. rCBF was increased by Nelumbinis Semen(蓮于肉), Ophiopogonis Radix(麥門冬), Asparagi Radix(天門冬), Polygalae Radix(遠志), Zizyphi Spinosae Semen(酸棗仁), Longnae Arillus(龍眼肉), Raphani Semen, Chrysanthemi Flos(甘菊) in rats. The increase of rCBF is mediated by adrenergic ${\beta}$- receptor and guanylyl cyclase which is enzyme producing cyclic GMP.
We evaluated the effect of nicotinamide on cellular $O_{2}$ consumption and metabolic status i.e., adenylate phosphates and $NAD^{+}$in-vitro, and changes in blood flow in-vivo, to determine whether changes in cellular metabolism or increased oxygen availability, was responsible for increased tumor oxygenation. Thirty min, pre-incubation of cells with$\∼$4mM (= 500mg/kg) nicotinamide resulted in no change in cellular $O_{2}$ consumption. Similarly neither the adenylate Phosphates nor the cellular $NAD^{+}$levels were altered in the presence of $\∼$4mM nicontinamide. In-vivo, nicotinamide (500mg/kg) increased $O_{2}$ availability as estimated by changes in relative tumor blood flow (RBC flux). The changes in RBC flux measured by the laser Doppler method, were tumor volume dependent and increased from$\∼$35$ \% $ in$\∼$ 150$mm_{3}$tumors to$\∼$~75$ \% $ in$\∼$500$mm^{3}$ tumors. In conclusion, these observations indicate a reduction in local tissue $O_{2}$ consumption is not a mechanism of improved tumor oxygenation by nicotinamide in FSa II murine tumor model. The primary mechanism of increased $pO_{2}$ appears to be an increased local tumor blood flow.

To study the effect of Korea red ginseng (KCG) on the skin and cerebral blood flow, we evaluated the change of skin perfusion rate and cerebral perfusion rate after the intravenous, intraperitoneal, and oral administration of crude saponin (CS) and saponin-free fraction (SFF) of KRG in the rats. The change of skin perfusion rate and cerebral perfusion rate was measured laser doppler flowmetry. The intravenous injections of CS or SFF of KRG and intraperitoneal injection of SFF of KRG did not change the relative skin and cerebral blood flow in the rats. When the rats were treated by the intraperitoneal injection of CS of KRG, relative cerebral blood flow was significantly increased with a time-dependent manner, however, relative skin blood flow was not influenced by the them. Oral administration of CS of KRG slightly increased skin blood flow in the rats. Also, the change of cerebral blood flow by transient bilateral carotid arterial clamp in the CS-treated rats was significantly decreased, compared with control groups. From the above results, it was suggested that Korea red ginseng have a increasing property of cerebral blood flow in the rats.
The aims of the study were to evaluate the effect of epinephrine-containing local anesthetics on pulpal blood flow (PBF) and to investigate its effect on cavity preparation-induced PBF change. PBF was recorded using a laser Doppler flowmeter (Perimed Co., Sweden) from canines of nine cats under general anesthesia before and after injection of local anesthetics and after cavity preparation. 2% lidocaine hydrochloride with 1 : 100,000 epinephrine was administered by local infiltration given apical to the mandibular canine at the vestibular area and the same volume of isotonic saline was injected on the contralateral tooth as a control. A round carbide bur was operated at slow speed with isotonic saline flushing to grind spherical cavities with increasing depth through the enamel and into the dentin on both teeth. The obtained data was analyzed with paired t-test. Cavity preparation caused significant increase of PBF (n = 9, p < 0.05). Local infiltration of lidocaine with epinephrine resulted in decreases of PBF (n = 9, p < 0.05), whereas there was no significant change of PBF with the physiologic saline as a control. Cavity preparation on tooth anesthetized with lidocaine with epinephrine caused significantly less increase of PBF than in control tooth (p < 0.05). Therefore, the result of the present study demonstrates that local infiltration of 2% lidocaine with 1 : 100,000 epinephrine effectively reduces PBF increase caused by cavity preparation.

Citri Reticulatae Viride Pericarpium extract(CRVP) have been used in oriental medicine for many centuries as a therapeutic agent for Soothing the liver and regulating the circulation of qi(疏肝理氣), and promoting digestion and removing stagnated food(消積化滯). The effects of CRVP on the vascular system is not known. The purpose of this Study was to investigate the effects of CRVP on the pial arterial diameter and regional cerebral blood flow(rCBF) in normal rats and ischemic cerebrovascular pathologic model rats. The changes in rCBF was determinated by Laser-Doppler Flowmetry(LDF), and the changes in pial arterial diameter were determinated by video microscopy methods and video analyzer. The results were as follows ; 1. Pial arterial diameter was significantly increased by CRVP in a dose-dependent manner. 2. Pretreatment with L-NNA significantly inhibited CRVP induced increased rCBF and pial arterial diameter. 3. Both the methylene chloride fraction and the hexane fraction of CRVP dose-dependently improved the altered cerebral hemodynamics of cerebral ischemic animal by increasing rCBF. 4. Pretreatment with L-NNA and indomethacin significantly inhibited CRVP(MC) induced increased rCBF. 5. Pretreatment with L-NNA and indomethacin significantly inhibited CRVP(hexane) induced increased rCBF. 6. Pretreatment with CRVP maredly stabilized the changes rCBF and pial arterial diameter during the period of cerebral reperusion. In conclusion, CRVP causes a diverse response of rCBF and pial arterial diameter, and CRVP dose-dependently improved the altered cerebral hemodynamics of cerebral ischemic animal by increasing rCBF and pial arterial diameter. These results suggest that the improvement of cerebral hemodynamics is also mediated by nitric oxide synthase and cyclooxygenase.

Jaeumgenby-tang(JGT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo caused by deficiency of qi(氣) and blood(血). Effect of Aurantii Fructus(AF) take off the phlegm by promoting the circulation of qi, Gastrodae Rhizoma(GR) has effects treating for headache, vertigo by calming the liver and suppressing hyperactivity of the liver-yang(陽). And, I designed to investigate whether injection of JGT adding AFㆍGR extract(JGTAG) affects cytotoxicity in vitro, cerebral hemodynamics [regional cerebral blood flow(rCBF), pial arterial diameter(PAD), mean arterial blood pressure(MABP)] in normal and cerebral ischemia rats by MCA occlusion method. The changes of rCBF and MABP were determinated by laser-doppler flowmetry(LDF), and the change of PAD was determinated by video microscope and width analyzer. The results were as follows in normal rats; JGTAG was not cytotoxicity in brain cells. And JGTAG was significantly increased rCBF, PAD and MABP. This results suggest that JGTAG increased significantly rCBF by dilating PAD. And the results were as follows in cerebral ischemic rats; The changes of rCBF and PAD were increased stably by treatment with JGTAG(10mg/kg, i.v.) during the period of cerebral reperfusion, and pretreatment with propranolol and indomethacin were increased JGT AG induced increase of rCBF and PAD during the period of cerebral reperfusion. We suggest that JGTAG has an anti-ischemic effect through the improvement of cerebral hemodynamics.
