The goal of this study was to identify new bioactive peptides in extracts derived from Tenebrio molitor (T. molitor) larvae for the development of functional foods. After extraction from freeze-dried T. molitor larvae with various solvents on time course, the extracts showed the highest 2,2,1-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity at 5 and 10 hr per total protein and solid contents, respectively. When the water extract was fractionated, a high methanol concentration led to a reduced level of high-molecular-weight proteins in the centrifugal supernatant, whereas increased DPPH activity in the supernatants suggests low-molecular-weight peptides may mediate antioxidant activity in the supernatant. Most of the organic solvent partitions, excluding butanol, showed similar activities in the water phases, and the organic solvent partition fraction exhibited a 28~44% decrease in activity following heat treatment, implying that some components in the fraction become unstable in the presence of heat. The addition of proteinase K to the water extract increased DPPH activity by 10~20%, suggesting that peptides, when released from total proteins, partially increase antioxidant activity. Therefore, we suggest that the antioxidants in T. molitor larval extracts make them a potential source of functional animal food.
This study was carried out to investigate the developmental characteristics and life cycle of sciarid fly (Lycoriella sp.), an important mushroom pest. The sciarid fly was reared ina growth chamber at 21$\pm$$1^{\circ}C$ and 55$\pm$5% of relative humidity with a 16h high, 8h dark cycle and Flammulina velutipes was used at diet. The egg was oval with 0.17 mm in diameter and 0.27 mm in length. Average egg period was 4 days. Body length of each instar from the 1st to 3rd was 0.7, 1.5, and 4.5, mm. respectively. Average weight of each instar was 1.9, 15.4 and 93.6 $\mu\textrm{g}$, respectively. In the 4th instar larvae, however, developmental characteristics of female and male became different by body length, weight and segment size. Body length and weight of the 4th instar female larvae were 5 mm and 162$\mu\textrm{g}$, and those of the male larvae were 3.5 mm and 90 $\mu\textrm{g}$, respectively. The larval periods of female and male were respectively 13.5 and13 days. Pupal length of female and male was 3.5 and 2.7 mm, and average weight was 136 $\mu\textrm{g}$ and 6.5$\mu\textrm{g}$, respectively. Adults were emerged from female and male pupae on 5.5 and on 5 days after pupation, respectively. Average longevity of male and female were 7 and 5 days, respectively. The weight of female was two-fold heavier than the male. After copulation, female laid approximately 130 to 150 eggs on/in the sporophore of mushroom. In conclusion, total lifespan of male and female were 29 and 28 days, respectively.
1. On the basis of the samples collected on the eastern coast of Koje-Do from May to September, 1969, studies have been made on the growth and the relationships between the carapace length and the body length, and between the carapace length and the body weight of Penaeus japonicus Bate. 2. The mean carapace length of P. japonicus was 51mm in May, 57mm in June, 47mm in July and 50mm in September respectively. 3. As a result of the present studies two populations of P. japonicus exist in waters around Koje-Do, namely the spring and fall spawning populations. 4. The relationship between the carapace length ($\iota$) and the body length(L) and between the carapace length and the body weight (W) are indicated by the following equations: May $$L=2.6544{\iota}+3.1258$$$$W=1.892{\iota}^{1.9844}$$ June $$L=2.8659{\iota}+2.1796$$$$W=1.082{\iota}^{2.4323}$$ July $$L=2.5840{\iota}+3.3090$$$$W=1.290{\iota}^{2.3094}$$ September $$L=2.4234{\iota}+4.5775$$$$W=1.599{\iota}^{2.1857}$$ 5. With regard to the relationships between the carapace length and the body length and between the carapace length and the body weight there is no significant difference between the populations spawning in June and September. 6. The relationships between the carapace length ($\iota$) and the body length (L) and between the carapace length and the body weight (W) for the samples cultured at three different localities are indicated by the following equations: Koje-do $$L=3.7738{\iota}+0.0805\;(r=0.934)$$$$W=0.4690{\iota}^{3.0713}$$ Oma-do $$L=2.993{\iota}+1.6455\;(r=0.990)$$$$W=0.6328{\iota}^{2.6579}$$ Kumdang-do $$L=3.2749{\iota}+0.9055\;(r=0.983)$$$$W=0.5768{\iota}^{2.8076}$$ 7. During the larval stages the relationship between the body length (L) and the rearing day (D) is indicated by the following equations: Zoeal stages (1-3) L=0.1279D+0.2686 (r=0.979) Mysis (1) - Post larva (6) L=0.1697D+0.5634 (r=0.994) Post-larvs (7) - Post larvs (21) L=0.1344D+1.9501 (r=0.978)
Enterobacteria were isolated in the gut of the predacious multicolored Asian ladybird beetle, Harmonia axyridis, and their effects to the development of H. axyridis were examined. Populations of H. axyridis in this experiment were collected from Kimjae at Cheonbuk province (JK population), Geumsan at Chungnam province (CK population) and laboratory population at Laboratory of Insect Physiology in Chungnam National University, Daejeon. Thirty-four enterobacteria isolates were purified and isolated from the digestive tract of H. axyridis, and a total of 4 strains were classified into group by analysis of 16S rRNA gene sequences. About 70% of total isolates were phylogenetic groups of Bacillus genus and Staphylococcus genus, and they were commonly separated from the digestive tract of H. axyridis. After investigating their susceptibility against antibiotics with 18 representative enterobacteria isolates, ofloxacin and penicillin were selected for examination in this study of their ability to inhibit the growth of all of isolates. In order to remove the enterobacteria from the aphids, ofloxacin and penicillin were given to the green peach aphid, Myzus persicae, and the turnip aphid, Lipaphis erysimi. These aphids were provided to H. axyridis as prey. The weight of pupa, developmental periods of each larval instar, the number of eggs and their hatching ratio of H. axyridis with treatment aphids were lower compared with non-treatment aphids. Staphylococcus saprophyticus is a representative enterobacteria and commonly isolated from the digestive tract of H. axyridis. In the absence of S. saprophyticus, the developmental periods of each larval instar increased; however, the weights of pupa, the number of eggs, and their hatching ratio decreased.
Since 1966, practical use of active heavy analogs of the hormones has been also worked out as an insecticide and brought the features of it to the light as cocoon producer. On the other hand, it is expected to afford the increase of silk productivity resulted from control of the fifth larval period by delaying normal development. With these regards, some of analogs have been tried to apply practically to the silkworm for an increase of cocoon productivity. One of the synthesized juvenile hormone available is "Manina". And it is presently used for the increase of silk productivity in Korea. For the practical use, it is very essential the varietal differences in the increase of silk productivity by topical application was tested and the obtained results are summarized as follows. 1. It was evident that the fifty larval period was extended by topical application after 48 hrs. of the last ecdysis, ranging from 8 hrs. to one day, as compared to the control. 2. In pupal rates, there is no significance between control and treatments. It proved that there was no toxicity to silkworm by topical application in general, except jam 120. With regards to an increase of cocoon yield in Japanese, it was resulted from 17∼24% of cocoon yield from 10,000 larvae, as compared to that of control. In case of Chinese, the incrasing rates were varied from 15∼26% of cocoon yield, 17.8kg of it with 26% increase for Jam 122 and 16.7kg of it with 25% increase for Jam 118. In case of all hybrids, an increase of the cocoon yield took places from 20% to 31% and the weight of cocoon layer for the Japanese increased by 6 to 14%, those for the Chinese by 4 to 7% and those for the hybrids ranged from 21 to 29% increase. 3. It was recognized that the hybrid vigor rate took places with the hybrids between high responsing parents to juvenile hormones.
During the periods from lily to October, 2000 in Hongseong and lucy to October, 2001 in Taean in the west coast of Korea, the following environmental conditions prevailed : water temperature : 22.0~26.817, salinity 27.23 ~30.80%, dissolved oxygen 4.12 ~6.26 ml/l, pH 7.89 ~8.09, phosphate 0.39 ~0.65 $\mu m$ , inorganic nitrogen 5.05~9.26 $\mu m$, suspended solid 5.4~20.8 mg/l and chemical oxygen demand 1.12~1.87 mg/l. The B-shaped veliger larvae of the Ark shell occurred in maximum number at $25^{\circ}C$ prevailing from mid-August at Hongseong and Taean. Full grown larvae reached maximum abundance from late August. To identify the effectiveness of the substratum for spat collection, raschel net were tested to Larval settlement. The most effective depth to collect the larvae in natural environment was the collectors suspended at 7~8 m depth. At these depths, about 49 to 94 spats were found on the collector (40$\times$50 cm), The growth of shell height (Y) to shell length (X), and total weight (W) to shell length (L) could be formulated as follows respectively: Hongseong: SH = 0.7168 SL -0.6466 ( $r^2$ = 0.9839), TW = $0.0001SL^{3.1705}$ ($r^2$ = 0.9882) Taean: SH = 0.736 SL -0.8824 ($r^2$ : 0.9899), TW : 0.00005 $SL^{3.3731}$ ($r^2$ : 0.9899)
Hatching rate, larval deformation and growth rate of selected olive flounder (Paralichthys olivaceus) for rapid growth were compared to those of the unselected fish. Fish were spawned on the same day and cultured under the similar conditions. The selected fish had a significantly higher eggs hatching rate, and lower larval deformation. The selected fish grew significantly faster, and at the end of the experiment (8 weeks after hatching) averaged 50.49${\pm}$2.67 mm in total length, 16.30${\pm}$0.08 mm in body height, and 1.036${\pm}$0.118 g in weight, compared to 40.55${\pm}$3.13 mm, 13.50${\pm}$0.96 mm, and 0.557${\pm}$0.073 g for unselected fish, respectively. The selected fish had a significantly higher body shape index, however lower condition factor than the unselected fish. The results of the present study demonstrate that the selected fish of the olive flounder for rapid growth had superior growth and improved body shape.
Investigation of atmospheric data and rearing results was conducted to analyze the effects of atmospheric factors such as temperature and precipitation on silkworm in 8 rearing places in which local adaptation test was being conducted with different mulberry growth condition, soil and atmosphere during spring and autumn rearing season of 2006. The atmospheric characteristics of spring rearing time are as follows. The average temperatures of young silkworm, old silkworm, mounting were $17.7^{\circ}C$, $19.8^{\circ}C$, $21.5^{\circ}C$ respectively, and $1^{\circ}C$ higher than normal year. The precipitation of young silkworm, old silkworm, mounting were 15.1 mm, 6.9 mm, 7.0 mm, respectively, and 22.9 mm lower than normal year in old silkworm and mounting. The daylight hours in larval stage was 1.7 hour shorter than normal year, but no difference in mounting. Thus precipitation was lower and temperature was higher than normal year in 2006. The rearing results of 2006 were 1 kg lower than normal year in cocoon yields per 10,000 3 rd molted larvae, single cocoon weight and cocoon shell percentage were a little higher. The atmospheric characteristics of spring rearing season are as follows. The average temperatures of young silkworm, old silkworm, mounting were $25.1^{\circ}C$, $20.5^{\circ}C$ and $19.9^{\circ}C$ respectively, temperature in young silkworm was $1^{\circ}C$ higher than normal year, and temperature in old silkworm was $1.3^{\circ}C$ lower than normal year. The precipitation of young silkworm, old silkworm, mounting were 110.2 mm, 4.6 mm, 3.7 mm, respectively and there were little differences compared to normal year. The atmospheric condition of 2006 which was similar to normal year did not affect the autumn rearing results of 2006. Namely, the single cocoon weight and cocoon shell weight were not different from normal year, and the same was cocoon shell percentage.
Silk protein is synthesized in the silkgland of silkworm, Bombyx mori L. It is evident that silk productivity is one of the high heritable characters from the genetical aspects. It is also changed with the environmental circumstances. With this regard, this paper dealt with the varietal patterns of silkgland development and the factors concerning to the silk productivity of silkgland of silkworm by the synthesis of nucleic acids, profiles of amino acids and histological basis, using the eight parent silkworm varieties and their F$_1$ hybrids. 1. The weight of silkgland per larva increased proportionally in the F$_1$ hybrids which were crossed between longer silk yielding varieties. The silk content to the weight of the silkgland was higher in the longer silk yielding varieties than that in the shorter silk yielding varieties. 2. It was observed that the morphological changes of nuclei took place in the posterior silkgland cells with the larval development of the 5th instar. In varietal aspect, Jam 107 and Jam 108, longer silk yielding varieties, showed more branches in nuclei than the $N_2$ and $C_{60}$ which were shorter yielding ones. 3. It was observed that there was a high correlation between RNA content per unit weight of silkgland on the 6th day stage of 5th instar and silk productivity both in the parents and their F$_1$ hybrids. 4. RNA and DNA synthesis brought about thirty percent increase in the posterior silkgland of the longer silk yielding varieties during the 2nd day to the 4th day stages of the 5th instar, when compared with those in the posterior silkgland of the shorter silk yielding varieties. 5. RNA/DNA ratio in the posterior silkgland on the 2nd day and 4th day stages of the 5th instar was more increased in the longer silk yielding varieties than the shorter silk yielding varieties. 6. It was shown that DNA content for the longer silk yielding varieties came to be 374$\mu\textrm{g}$ per larva in the posterior division of silkgland on the 4th day stage of 5th instar, whereas it was 199$\mu\textrm{g}$ per larva for the shorter silk yielding varieties. 7 There was 34.8% Alanine, 22.8% Glycine, 9.1% Serine and 7.3% Tyrosine in the posterior division of silkgland as major amino acids. It is noticed that there was a little differences between the amino acids composition of posterior silkgland and silk fibroin. 8. There was some differences in the amino acids composition of posterior silkgland between pure lines and their hybrids. Glycine, Serine and acidic amino acids, essential to silk formation, seemed to be increased in the F$_1$ hybrids, whereas other amino acids such as Valine, Iso-leucine, Leucine, Lysine. Phenylalanine, Histidine and Arginine were reduced. 9. The content of Glycine, Alanine and Serine in the posterior division of silkgland was elevated in the longer silk yielding varieties than the others. Consequently. these three amino acids in the posterior silkgland seemed to be related to the silk yielding ability in the longer silk yielding varieties.s.
Biological characteristics of the almond moth, Cadra cautella, of which eggs will be substitute diets for Orius spp. and Trichogramma spp., were investigated and a mass-rearing system for the moth was developed. At 25$^{\circ}C$, egg, larval, and pupal period was 4.2, 29.8 and 8.3 days, respectively, and adult longevity was 5.8 days for female and 4.8 days for male. Total number of eggs at 20, 25 and 30$^{\circ}C$ was 128.9,207.9 and 139.9, respectively. The moth could be successfully reared with all food substrates tested, of which rice bran (50%)+chick feed (50%) assumed to be proper for massrearing in view of cost. Eggs could be stocked at 9 C for 7 days, representing 82% hatchability. In the rearing cage (16${\times}$24${\times}$9cm) used, 1,000 eggs was better for initial level of inoculation, showing relatively high emergence rate and adult weight. Mass-rearing procedures were explained in detail.
